Abstract:Dihydromyricetin, one of flavonoids, was greatly resided in the plant of Ampelopsis grossedentata, with many biological and pharmacological activities. This paper puts the emphasis on the anti-tumor activity and four kinds of anti-tumor mechanisms of dihydromyricetin, including induction of neoplasm cell cycle arrest and cell apoptosis、inhibition of invasion, metastasis and angiogenesis of neoplasm cell. The author holds that dihydromyricetin, possess-ing great potentiality to explore and utilization as a anticancer drug, is worthy of further research.

Abstract:Dihydromyricetin is a flavonoid isolated from Ampelopsis grossedentata , which is traditionally used in China. Dihydromyricetin exhibits health-benefiting activities with minimum adverse effects. Dihydromyricetin has been demonstrated to show antioxidative, anti-inflammatory, anticancer, antimicrobial, cell death-mediating, and lipid and glucose metabolism-regulatory activities. Dihydromyricetin may scavenge ROS to protect against oxidative stress or potentiate ROS generation to counteract cancer cells selectively without any effects on normal cells. However, the low bioavailability of dihydromyricetin limits its potential applications. Recent research has gained positive and promising data. This review will discuss the versatile effects and clinical prospective of dihydromyricetin.

Abstract:Dihydromyricetin (DMY) was efficiently extracted from Ampelopsis grossedentata and modified by esterification to obtain two derivatives, which were named as benzoylated derivative (BZ-DMY) and acetylated derivative (AC-DMY). The physicochemical characterizations of BZ-DMY and AC-DMY were investigated by TLC, SEM, HPLC, FT-IR, NMR and MS analysis. Simultaneously, the anti-hepatocellular carcinoma activity was determined by cell experiment to investigate their structure-activity relationship. The results indicated that esterification of DMY were successfully modified by benzoyl and acetyl. The substitute positions were C-3, C-5, C-7, C-3′, C-4′ and C-5′ respectively inferred from NMR analysis. In the experiment of anti-hepatocellular carcinoma activity, two novel derivatives showed better anti-tumor ability than DMY. Therefore, it can be concluded that esterified DMY could significantly induce HepG2 cells apoptosis and improve the anti-tumor activity of DMY. In summary, this research could not only enhance the development and utilization of Ampelopsis grossedentata, but also develop these two derivatives (BZ-DMY and AC-DMY) as potential anti-hepatocellular carcinoma therapeutic drugs.

Abstract:AIM To assess the antioxidative properties and the mechanism of action of dihydromyricetin (DMY) from Ampelopsis grossedentata. METHODS The antioxidative properties of DMY were measured by scavenging 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) and inhibiting lipid peroxidation induced by FeSO4-edetic acid in linoleic acid. The mechanism of antioxidative properties of DMY was tested by measuring the chelating activities of DMY for Fe2+ with ultraviolet spectrum (UV) method. RESULTS The specific absorption of DPPH radical solution at 517 nm was reduced 73.3%-91.5% when DMY was added into the reaction solution in the concentration range from 0.01% to 0.04%. DMY was shown to greatly inhibit the increase of lipid peroxidation (LPO) values in linolei acid system catalyzed by FeSO4-edetic acid. The reaction rates (A532.min-1) of lipid peroxidation were 0.0021-0.0004 in the concentration range from 0.01% to 0.04% and the inhibition activities of DMY was found to be in a concentration-dependent manner. The mechanism of antioxidative properties of DMY was chelating Fe2+ in the Fe(2+)-dependent lipid peroxidation system. CONCLUSION DMY showed great antioxidative effect and would be a good natural antioxidant.

Abstract:[Objective] To optimize the extraction and purification technology of dihydromyricetin from Ampelopsis grossedentata.[Method] Dihydromyricetin was extracted from Ampelopsis grossedentata with water as extracting agent,then purified and determined,the extraction conditions were optimized through single factor test and orthogonal experiment.[Result] The optimum extraction conditions were: extracting temperature was 90 ℃,solid-liquid ratio 1∶ 10 and extracting time 60 min,under which the yield of dihydromyricetin was the largest,the purity of total flavonoids increased from 68.51% to 91.0% after 5 times re-crystallization.[Conclusion] The extraction and purification technology of dihydromyricetin was simple and affordable with convenient source of materials,therefore it had broad prospect in producing antioxidant.

Abstract:Extraction and antioxidant activity in vitro of dihydromyricetin from the wild Ampelopsis grossedentata in Fanjing were studied. The extraction of dihydromyricetin from the wild Ampelopsis grossedentata by ultrasonic method was studied. the scavenging activity of dihydromyricetin was studied by two different free radical systems such as 1,1- diphenly- 2- pieryl- hydrazyl free radical( DPPH·) and hydroxide free radical( ·OH). The results showed that the scavenging activity of dihydromyricetin for DPPH free radical was higher than that for rutin within the experimental concentration range,when concentration of dihydromyricetin reached 50 mg / L,its clearance rate was higher than the Vc,up to72. 58%. With concentration of dihydromyricetin increased,the scavenging activity of dihydromyricetin for hydroxyl free radical was improved gradually. its clearance rate could be up to 55. 05%. It was showed that the dihydromyricetin from the wild Ampelopsis grossedentata had strong antioxidant activity.

Abstract:Ampelopsis grossedentata is rich in flavonoids especially dihydromyricetin.In this paper the purification and crystallization of dihydromyricetin in Ampelopsis grossedentata were discussed.The results showed that the content of total flavonoids increased from 86% to 96.5% after recrystallizing by water for several times,and the impurity could be removed effectively.The crystal shape of dihydromyricetin was needle in water but was radial club-shaped by water isothermal crystallization.When pH value was 4.5,the solubility of dihydromyricetin was minimum and higher recovery rate could be acquired.

Abstract:Dihydromyricetin (DMY) has recently attracted increased interest due to its considerable health-promoting activities but there are few reports on its antibacterial activity and mechanism. In this paper, the activity and mechanisms of DMY from Ampelopsis grossedentata leaves against food-borne bacteria are investigated. Moreover, the effects of pH, thermal-processing, and metal ions on the antibacterial activity of DMY are also evaluated. The results show that DMY exhibits ideal antibacterial activity on five types of food-borne bacteria (Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Salmonella paratyphi, and Pseudomonas aeruginosa). The activities of DMY against bacteria are extremely sensitive to pH, thermal-processing, and metal ions. The morphology of the tested bacteria is changed and damaged more seriously with the exposure time of DMY. Furthermore, the results of the oxidative respiratory metabolism assay and the integrity of the cell membrane and wall tests revealed that the death of bacteria caused by DMY might be due to lysis of the cell wall, leakage of intracellular ingredients, and inhibition of the tricarboxylic acid cycle (TCA) pathway.

Abstract:Dihydromyricetin (DHM) is a natural flavonoid compound extracted from Ampelopsis grossedentata that has been used for centuries in traditional Chinese medicine. DHM has attracted intensive attention due to its numerous beneficial activities, such as hepatoprotection, cardioprotection, antioxidant, and anti-inflammation. In addition, DHM inhibits the progression of cancers such as lung cancer, hepatocellular cancer, breast cancer, melanoma, and malignant reproductive systems through multiple mechanisms, including antiangiogenesis, antiproliferation, apoptosis, and inhibition of invasion and migration. Notably, DHM also activates autophagy at different levels, exerting a dual-regulatory effect on cancers. Mechanistically, DHM can effectively regulate mammalian target of rapamycin (mTOR), noncoding RNA-mediated signaling, phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) pathway, nuclear factor-κB (NF-κB), p53, and endoplasmic reticulum stress (ER stress)-driven signaling in different types of cancers. DHM has also been shown to have inhibitory effects on various regulators that trigger epithelial-mesenchymal transition (EMT). Furthermore, DHM exhibits a remarkable anticancer reversal ability when used in combination with drugs such as adriamycin, nedaplatin, and other drugs. However, the low bioavailability of DHM limits its potential applications, which are improved through structural modification and the exploration of novel dosage forms. Therefore, DHM may become a promising candidate for treating malignancies alone or combined with conventional anticancer strategies used in clinical practice.

Abstract:Myricetin and its glycosides are important flavonols commonly found in plants, and they are natural organic compounds with diverse pharmacological activities. Numerous studies have demonstrated that myricetin and its glycosides are strong antioxidants that have great potential in preventing, alleviating and assisting the treatment of chronic non-infectious diseases such as cancer, diabetes, and cardiovascular diseases. In addition, myricetin and its glycosides also have antiviral, antibacterial, anti-inflammatory, analgesic, liver protection and other pharmacological activities. Myricetin contains more hydroxyl groups in the parent ring structure than other flavonoids, so myricetin and its glycosides have stronger pharmacological activities than other flavonols or flavonoids such as quercetin and kaempferol. Therefore, myricetin and its glycosides have great development and application prospects. In this paper, the classification and distribution of myricetin and its glycosides, their pharmacological activity and mechanism, as well as comparison with other flavonoids were reviewed. In addition, limitations of the current research and application of myricetin and its glycosides were analyzed, and the further studies on pharmacological activities as well as their dose-activity relationship, structure-activity relationship, chemical modification, biosynthesis and application prospects of myricetin and its glycosides were discussed and proposed.

Abstract:Ampelopsis grossedentata is a valuable medicinal and edible plant, which is often used as a traditional tea by the Tujia people in China. A. grossedentata has numerous biological activities and is now widely used in the pharmaceutical and food industries. In this study, two new flavonoids (1–2) and seventeen known compounds (3–19) were isolated and identified from the dried stems and leaves of A. grossedentata. These isolated compounds were characterized by various spectroscopic data including mass spectrometry and nuclear magnetic resonance spectroscopy. All isolates were assessed for their α-glucosidase inhibitory, antioxidant, and hepatoprotective activities, and their structure–activity relationships were further discussed. The results indicated that compound 1 exhibited effective inhibitory activity against α-glucosidase, with an IC50 value of 0.21 μM. In addition, compounds 1–2 demonstrated not only potent antioxidant activities but also superior hepatoprotective properties. The findings of this study could serve as a reference for the development of A. grossedentata-derived products or drugs aimed at realizing their antidiabetic, antioxidant, and hepatoprotective functions.

Abstract:As the most abundant natural flavonoid in rattan tea, dihydromyricetin (DMY) has shown a wide range of pharmacological effects. In addition to the general characteristics of flavonoids, DMY has the effects of cardioprotection, anti-diabetes, hepatoprotection, neuroprotection, anti-tumor, and dermatoprotection. DMY was also applied for the treatment of bacterial infection, osteoporosis, asthma, kidney injury, nephrotoxicity and so on. These effects to some extent enrich the understanding about the role of DMY in disease prevention and therapy. However, to date, we still have no outlined knowledge about the detailed mechanism of DMY, which might be related to anti-oxidation and anti-inflammation. And the detailed mechanisms may be associated with several different molecules involved in cellular apoptosis, oxidative stress, and inflammation, such as AMP-activated protein kinase (AMPK), mitogen-activated protein kinase (MAPK), protein kinase B (Akt), nuclear factor-κB (NF-κB), nuclear factor E2-related factor 2 (Nrf2), ATP-binding cassette transporter A1 (ABCA1), peroxisome proliferator-activated receptor-γ (PPARγ) and so on. Here, we summarized the current pharmacological developments of DMY as well as possible mechanisms, aiming to push the understanding about the protective role of DMY as well as its preclinical assessment of novel application.

Abstract:Diabetic Mellitus (DM), a chronic metabolic disorder disease characterized by hyperglycemia, is mainly caused by the absolute or relative deficiency of insulin secretion or decreased insulin sensitivity in target tissue cells. Dihydromyricetin (DMY) is a flavonoid compound of dihydroflavonol that widely exists in Ampelopsis grossedentata. This review aims to summarize the research progress of DMY in the treatment of DM. A detailed summary of related signaling induced by DMY are discussed. Increasing evidence implicates that DMY display hypoglycemic effects in DM via improving glucose and lipid metabolism, attenuating inflammatory responses, and reducing oxidative stress, with the signal transduction pathways underlying the regulation of AMPK or mTOR/autophagy, and relevant downstream cascades, including PGC-1α/SIRT3, MEK/ERK, and PI3K/Akt signal pathways. Hence, the mechanisms underlying the therapeutic implications of DMY in DM are still obscure. In this review, following with a brief introduction of the absorption, metabolism, distribution, and excretion characteristics of DMY, we summarized the current pharmacological developments of DMY as well as possible molecular mechanisms in the treatment of DM, aiming to push the understanding about the protective role of DMY as well as its preclinical assessment of novel application.

Abstract:As a new type of natural flavonoids, dihydromyricetin (DMY) has attracted more and more attention. It has a series of pharmacological effects, such as anti-inflammatory, anti-tumor, anti-oxidation, antibacterial and so on, and it is almost no toxicity and with excellent safety. Therefore, even if the bioavailability is poor, it is often added to daily food, beverages and even medicines. In recent years, some researchers have found that DMY can treat some diseases by anti-oxidation, anti-inflammation, promoting cell death and regulate the activity of lipid and glucose metabolism. In addition, the mechanism of DMY on these diseases was also related to the signal pathway of AMPK, PI3K/Akt, PPAR and the participation of microRNAs. This review describes the mechanism of DMY in metabolic related diseases from three aspects: metabolic diseases, liver diseases, and cancers, hoping to provide some new ideas for clinical researches.

Abstract:Three techniques--refluxing extraction (RE), ultrasonic-assisted extraction (UAE), microwave-assisted extraction (MAE)--for extraction of dihydromyricetin from Ampelopsis grossedentata, family Vitaceae, were evaluated. A 70% methanol-water solution was used as extraction solvent, and extracts obtained were analyzed by high-performance liquid chromatography (HPLC). Results showed that the MAE method was the most efficient in the shortest time, followed by RE and UAE. Maximum extraction efficiency of dihydromyricetin was obtained by using the MAE technique, which needs only 10 min, whereas UAE and RE require 40 and 300 min, respectively, to gain higher extraction efficiency of dihydromyricetin. The time needed for MAE was 30 times less than that needed for RE. Extracts obtained by using different methods were shown by HPLC analysis to possess similar chemical constituents, and extraction reproducibility was satisfactory for these techniques. Compared with UAE and RE, the MAE is a better alternative extraction technique for large-scale speedy extraction of dihydromyricetin from this plant.

Abstract:Leaves of Ampelopsis grossedentata have been consumed in southern China as health tea and herbal medicine. In this study, two kinds of flavonoid-rich extracts from leaves of A. grossedentata were prepared by traditional solvent extraction (product A) and recrystallization (product B). By analysis of ultraviolet-visible (UV) spectrometry, infrared (IR) spectrometry, electrospray ionization mass spectrometry (ESI-MS), nuclear magnetic resonance (NMR) spectrometry and high-performance liquid chromatography (HPLC)-ESI/MS, the major flavonoid in two products was identified as (+)-dihydromyricetin (DMY). Also, by HPLC, the contents of DMY in leaves, product A and product B were measured to be 32.96 +/- 1.18%, 77.15 +/- 1.98% and 95.12 +/- 2.86%, respectively. Various antioxidant tests (1,1-diphenyl-2-picrylhydrazyl radical scavenging, reducing power, antioxidant activity in linoleic acid system) showed that the antioxidant activity of the flavonoid-rich extracts could be comparable with that of tertiary butylhydroquinone. The flavonoid-rich extracts from A. grossedentata leaves have a promise to be a new natural antioxidant with potential application in food industry.PRACTICAL APPLICATIONLeaves of Ampelopsis grossedentata are consumed in southern China as health tea (Tengcha) and herbal medicine. In this study, the flavonoid-rich extracts from leaves of A. grossedentata, which were rich in (+)-dihydromyricetin, were prepared by traditional solvent extraction and recrystallization. Also, the antioxidant activity of the extracts can be comparable with that of tertiary butylhydroquinone. The results obtained in this study suggest that leaves of A. grossedentata may have potential as a new source of natural antioxidant and health food with great commercial interest in the food and phytopharmaceutical market.
Recrystallization of Dihydromyricetin from Ampelopsis Grossedentata and Its Anti-Oxidant Activity Evaluation

Abstract:A fast and efficient method for purification of dihydromyricetin (3,5,7,3′,4′,5′-six hydroxy-2,3-dihydro flavonol; DMY) from Ampelopsis grossedentata was created by crystallization eight times at 25°C, and a purity of 98% was finally achieved. The purified DMY exhibited high oxygen radical absorbance capacity (ORAC) (30.21 μmol Trolox equiv/mg) and strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (half-maximal inhibitory concentration [IC50]=0.235 μg/mL). The addition of DMY could also effectively attenuate 2,2′-azobis (2-amidinopropane) dihydrochloride (AAPH)-induced human erythrocyte hemolysis and cupric chloride (CuCl2)-induced human plasma lipid peroxidation via inhibition of intracellular reactive oxygen species (ROS) generation. It was also found that DMY (>12 μg/mL) treatment significantly inhibited intracellular malondialdehyde (MDA) formation. Meanwhile, DMY treatment significantly inhibited the obvious increase of anti-oxidant enzymes levels (superoxide dismutase [SOD]; glutathione peroxidase [GPX], and catalase [CAT]) induced by AAPH radicals, suggesting that stress defense mechanisms are associated with protection of DMY against intracellular oxidation.

Abstract:Aim To study chemical constituents from Ampelopsis grossedentata. Methods Separation and purification were performed by using silica gel, polyamide, reverse-phase silica gel, Sephadex LH-20 column chromatographic techniques and silica gel PTLC. Structures were determined by means of physicochemical properties and spectral analysis. Results Four flavonoids were separated and identified from Ampelopsis grossedentata including dihydromyricetin (1), myricetin (2), myricitrin (3), and myricetin-3-O-β-D-galactopyranoside (4). Conclusion Compound 4 was isolated from this genus for the first time, and 2D NMR techniques permitted the correct assignment of 1H and 13C NMR signals of 1.
Identification of Hydroxylation Enzymes and the Metabolic Analysis of Dihydromyricetin Synthesis in Ampelopsis grossedentata

Abstract:Ampelopsis grossedentata leaves are highly rich in dihydromyricetin. They have been used to make tea in China for centuries. Dihydromyricetin has many potential applications in foods and medicine. This are because it has five phenolic hydroxyl groups. However, the hydroxylases involving the biosynthesis of dihydromyricetin have not been identified and characterized. In this study, a series of hydroxylases genes, including flavanone 3-hydroxylase (F3H), flavonoid 3'-hydroxylase (F3'H), flavonoid 3',5'-hydroxylase (F3'5'H), and cytochrome P450 reductase (CPR), were identified after RNA sequencing. The full-length CDSs of AgF3H, AgF3'H, AgF3'5'H, and AgCPR genes were amplified from the cDNA library of leaves. The aforementioned enzymes were expressed and verified in Saccharomyces cerevisiae. Through the substrate specificity assay, the functional AgF3'H, AgF3'5'H, and AgCPR in A. grossedentata were identified. The dihydromyricetin hydroxylation process in A. grossedentata was successfully identified. We found that substantial carbon flux occurred through the Naringenin (NAR)-Eriodictyol (ERI)-Dihydroquercetin (DHQ)-Dihydromyricetin (DHM) and NAR-Dihydrokaempferol (DHK)-DHQ-DHM pathways. This study provides some reference for the development and utilization of the germplasm resources and molecular breeding of A. grossedentata.
