Abstract:The application of cell hybridization to immunology has made the elucidation of the structure and function of T-cell products potentially much more accessible. It was initiated by the experiments of Kohler and Milstein’ who showed that myeloma cells could be fused with antibody-producing cells and that the hybrids secreted the specific antibody of the latter while growing as permanent lines. These “hybridoma” lines are a valuable source of homogeneous antibodies, which have rapidly found a number of important applications.* A similar principle has been applied to T-lymphocytes, using a neoplastic thymus-derived cell as a fusion partner. Thus, successful hybridization of mouse thymoma cells with mouse Tcells has been described, leading to establishment of lines that carry markers characteristic of the normal T-lymphocyte partner.2-E The possible retention of immunological activity by such T-hybrid lines is of considerable interest, as it would facilitate the study of T-cell function and the chemical analysis of T-cell-derived molecules, the great advantages being a continuous supply of T-cell products (lymphokines, factors) and their availability in quantities sufficient for structural studies. Recently, some lines with T-cell activities have been reported.e-lO In this paper we describe a T-hybrid line termed Al , which has retained a characteristic T-cell function, namely antigen-specific suppression of the antibody response. The line releases a suppressor molecule or “factor” that binds to sheep red blood cells (SRBC) and specifically inhibits the development of the antibody response to this antigen in culture. The A1 suppressor factor is not an immunoglobulin, but like the antigen-specific T-cell factors 11l2 it carries determinants of the H-2 complex. Its target cell of action seems to be a Bcell, which carries an appropriate acceptor site. Both factor and acceptor appear to be products of genes to the right of the I J subregion. The structure of the A1 factor has been studied by internal labeling with SH-leucine, followed by polyacrylamide gel electrophoresis (PAGE) in sodium dodecyl sulfate (SDS) . The molecule, which has a molecular weight of about 200,000 daltons, appears to be composed of two polypeptide chain types of sizes 85,000 and 25,000 respectively. While the larger chain binds to SRBC, it lacks H-2 determinants; the latter are presumed to be present on the small factor chain.
