Abstract:Thiostrepton, a natural peptide macrocycle, is of great interest due to its structural complexity and numerous biological activities, including anti-bacterial, anti-tumor, and anti-plasmodial activities. The quinaldic acid (QA) moiety-containing side ring (loop 2) was proven to play an important role in carrying out these functions. Previously, we proposed biosynthetic logic for thiostrepton loop 2 and demonstrated the formation mechanism of QA. Herein, we report the discovery and efficient synthesis of a biologically active alkaloid, that is, a key intermediate involved in the thiostrepton biosynthetic pathway. A chemoenzymatic method was performed to synthesize the molecule, and a series of analogs were prepared for bioassays, which included the examination of anti-bacterial and anti-tumor activities. Abstract:Hydroxamic acid derivatives constitute an interesting novel class of antitumor agents. Three of them, including vorinostat, are approved drugs for the treatment of malignancies, while several others are currently under clinical trials. In this work, we present new vorinostat analogs containing the benzoxazole ring as the cap group and various linkers. The benzoxazole-based analogs were synthesized starting either from 2-aminobenzoxazole, through conventional coupling, or from benzoxazole, through a metal-free oxidative amination. All the synthesized compounds were evaluated for their antiproliferative activity on three diverse human cancer cell lines (A549, Caco-2 and SF268), in comparison to vorinostat. Compound 12 (GK601), carrying a benzoxazole ring replacement for the phenyl ring of vorinostat, was the most potent inhibitor of the growth of three cell lines (IC50 1.2–2.1 μΜ), similar in potency to vorinostat. Compound 12 also inhibited human HDAC1, HDAC2 and HDAC6 like vorinostat. This new analog also showed antiproliferative activity against two colon cancer cell lines genetically resembling pseudomyxoma peritonei (PMP), namely HCT116 GNAS R201C/+ and LS174T (IC50 0.6 and 1.4 μΜ, respectively) with potency comparable to vorinostat (IC50 1.1 and 2.1 μΜ, respectively).

Abstract:Vorinostat (suberoylanilide hydroxamic acid) was the first approved histone deacetylase (HDAC) inhibitor in a group of validated cancer therapeutic agents targeting epigenetics. Riluzole is a drug used to treat amyotrophic lateral sclerosis, the antitumor potency of which has been recently revealed. Herein, a novel hybrid of vorinostat and riluzole (compound 1) was rationally designed, synthesized, and evaluated. Compared with vorinostat, compound 1 exhibited superior total HDAC inhibitory activity and similar HDAC isoform selective profiles. The intracellular HDAC inhibition of compound 1 was confirmed by Western blot analysis. Moreover, compound 1 possessed more potent in vitro antiproliferative activity against all tested solid and hematological tumor cell lines than vorinostat. In vitro metabolic stability evaluation of compound 1 revealed better human plasma stability and comparable human liver microsomal stability than vorinostat. Additionally, compound 1 demonstrated more significant in vivo antitumor activity in a MDA-MB-231 xenograft model than vorinostat, which could be attributed to its superior in vitro antiproliferative activity and metabolic stability. Taken together, the results presented here support further research and development of compound 1 as a promising antitumor agent.

Abstract:Simvastatin is a new antihyperlipidaemic agent. It can be synthesized with the material of lovastatin, hydroxy protecting agent of trimethylchlorosilane, and t-butyl lithium instead of pyrrole lithium. The total yield is about 78%.The structure is identified by UV,IR,MS,NMR spectrums.This method is very easy and the cost is very low, but the yield is high. Most of organic solvents can be recovered in order to be used next time. So this method will have perfect industrialization foreground.

Abstract:Aim To synthesize the antitumor agent sunitinib malate.Methods Starting from tert-butyl acetoacetate,2-tert-butyl-4-ethyl-3,5-dimethyl-1H-pyrrole-2,4-dicarboxylate was obtained via classic Knorr pyrrole reaction.After facile decarboxylation,Vilsmeier formylation,and hydrolysis of the ethyl ester,5-formyl-2,4-dimethyl-1H-pyrrole-3-carboxylic acid was formed.Sunitinib was obtained by one-pot reaction of this intermediate with N,N-diethylethylenediamine and 5-fluoro-1,3-dihydro-indol-2-one,then salt formation.Results and conclusion The target compound was synthesized and its structure was identified by 1H-NMR,MS,elemental analysis,and the overall yield was 28%.

Abstract:Natural products remain the largest resources of lead compounds that can be used to develop novel anticancer drug candidates. Based on deacetylisovaltratum, a natural product with promising anticancer activity, herein we designed and synthesized of a series of valepotriate derivatives with a novel skeleton from commercially available genipin. In addition, a structure-activity relationship study demonstrated the importance of an epoxy group on the C1-position and the preferable size of the sidechain ((5methylhexanoyl) oxy) on the C-7 position of valepotriates for their cytotoxic activities. The most potent compound 1e showed moderate to good IC50 values against various cancer cells, ranging from 10.7 to 50.2 mu M, which are comparable to that of deacetylisovaltratum. Additionally, we demonstrate that mitochondrion-mediated apoptosis would be its mechanism of action, thus enlightening the further development of novel valepotriate derivatives.

Abstract:Two series (14a–d and 21a–h) of novel spin-labeled combretastatin derivatives were synthesized and evaluated for cytotoxicity against four tumor cell lines (K562, SGC-7901, Hela and HepG-2). Simultaneously, a representative compound 21a was selected to investigate the antitumor mechanisms of these synthetic compounds. The results indicated that some of the compounds showed significant cytotoxicity against four tumor cell lines in vitro and were more active than etoposide, a clinically available anticancer drug. Among the newly synthesized compounds, 21a, 21b and 21c displayed the greatest cytotoxicity against three tested tumor cell lines (HEPG-2, BGC-832 and Hela), with IC50 values ranging from 0.15 to 1.05μM, compared with values of 0.014–0.403μM for 3-amino-deoxycombretastatin A-4 (3). In addition, the mechanistic analysis revealed that compound 21a effectively interfered with tubulin dynamics to prevent mitosis in cancer cells, leading to cell cycle arrest and, eventually, dose dependent apoptosis.

Abstract:The catalytic asymmetric synthesis of voriconazole, a synthetically challenging antifungal agent was achieved in nine steps with a 8 % overall yield. This novel strategy relied on the precise synthesis of tetrasubstituted (Z)‐allylic alcohol and the subsequent highly efficient epoxidation to establish the critical contiguous carbon stereocenters in a single step. A concise asymmetric synthesis of triazole antifungal agent voriconazole is described. Salient features of this synthesis include a LiOH‐controlled highly stereospecific formation of (Z)‐enol triflate from 2‐difluorobenzene acetoacetate to access the crucial tetrasubstituted (Z)‐allylic alcohol, and a highly enantioselective VO(OiPr)3/BHA ligand‐catalyzed epoxidation to construct the critical contiguous carbon stereocenters in a single step.

Abstract:Vorinostat (VST) is a chemotherapeutic agent administrated for various types of cancers. However, it suffers from side effects and chemoresistance that reduce its application. Different nanoniosomes comprised Span 20, 60, 65 and 80 were prepared by the thin film hydration method and loaded with VST. The nanoniosomes were physicochemically characterized using particle size analysis and field emission scanning electron microscopy. The best formulation that was prepared using Span 65 (VST-NN-S65) included vesicle size of 127 nm with a narrow size distribution. VST-NN-S65 had an entrapment efficiency and loading capacity of 81.3±5.1 and 32.0±3.9%, respectively. Drug release rate measurements showed that 90% of VST was liberated within 1 h. Cytotoxicity assessments of VST-NN-S65 in HeLa and MCF7 cells indicated significant improvement in the effectiveness of VST, compared to the VST suspension. For VST-NN-S65, IC50 values of 26.3 and 6.6 μg mL -1 were obtained for HeLa and MCF7 cell lines, respectively. In situ apoptosis detection by the TUNEL assay revealed that apoptosis mainly occurred in the cell lines.

Abstract:Using the techniques of computer-aided drug design, the docking of survivin and known active small molecules was simulated and then the key amino acid residue fragments of the target protein were analyzed. It led to the discovery of active groups capable of binding to the critical sites. Through the use of the natural product, oleanolic acid, as a lead compound, the introduction of the active groups onto the A-ring, and the modification of the carboxyl group at the C-28 position using esterification or amidation, 20 new oleanolic acid derivatives had been designed and synthesized. HepG2 and SGC-7901 cells were used to screen the antitumor activity through the standard MTT method. The compounds, II3, III5 and IV4, exhibited more potent cytotoxicity than positive drugs. Western blot experiment demonstrated that compound II3 can effectively inhibit the proliferation of HepG2 cells.

Abstract:A convenient and practical synthetic method for antitumor compound 2-methoxy-N-(3-{4-[3-methyl-4-(6-methyl-pyridine-3-yloxy)phenylamino]quinazolin-6-yl}-E-allyl)acetamide(CP724,714) is accomplished with high overall yields(28%) from 6-methylpyridine-3-ol and high regioselectivity of key step(96:4) based on the Heck coupling reaction which is catalyzed by Pd(OAc)2.Furthermore,the purity of this oncology candidate is 99.8% based on HPLC value.

Abstract:Abstract Background Vorinostat is a histone deacetylase inhibitor suberoylanilide hydroxamic acid (SAHA) with anticancer properties. However, it is plagued by low water solubility, low permeability (BCS class IV drug), and suboptimal pharmacokinetics. The purpose of the present study was to develop a self-microemulsifying drug delivery system (SMEDDS) to enhance the oral bioavailability of vorinostat. Capryol 90, labrasol, and polyethylene glycol (PEG 400) were selected as oil phase, surfactant, and co-surfactant, respectively. The vorinostat self-microemulsifying drug delivery systems were tested for self-microemulsifying time, phase separation, effect of pH, droplet size, zeta potential, dilution study, Fourier-transform infrared (FT-IR) spectroscopy analysis, and field emission scanning electron microscopy (FESEM). A rat model in vivo pharmacokinetic study was conducted for the optimized formulation against vorinostat pure drug powder. Results The results from the characterization studies showed that the optimized formulation (F7) self-microemulsification time was 1.4 ± 0.05 min and no precipitation or phase separation was observed. The mean droplet size, polydispersity index (PDI), and zeta potential of the optimized formulation (F7) were found to be 272.9 ± 82.7 nm, 0.415, and − 57.2 mV, respectively. The pharmacokinetic parameters of the optimized formulation (F7) showed a 1.6-fold increase in maximum concentration ( C max ) and a 3.6-fold increase in area under the curve (AUC (0−∞) ), in comparison with pure drug in suspension. Conclusions The findings suggest that SMEDDS formulation could be an effective method for increasing the oral bioavailability of vorinostat, which is poorly water soluble.

Abstract:Racemic diastereoisomeric spiro-phosphonate derivatives 5 similar to 6 were obtained by tandem double Michael addition and intramolecular nucleophilic substitution of 3-bromo-5-methoxy-5H-furan-2-one and alpha-hydroxylbenzylphosphonate under a warm condition. The target compounds 5 similar to 6 were identified by the elemental analysis, IR, H-1 NMR, C-13 NMR and MS techniques. The stereochemistry and configuration of 5c and 6c were confirmed further by X-ray crystallography. This result could lead to a new strategy for the synthesis of some complicated compounds containing a phosphorus functional group.

Abstract:N-(5-Aryl-1,3,4-thiadiazol-2-yl)-N'-((E)-styryl)ureas 4a similar to 4i were designed and synthesized as antitumor agents, and their structures were characterized by IR, H-1 NMR, C-13 NMR, MS and elemental analysis. The single crystal of compound 4a, C17H13FN4OS, was also determined by X-ray crystallography. Crystal data: monoclinic, space group P21/n, a = 4.9401(8), b = 8.7100(15), c = 36.604(6) angstrom, beta = 93.320(3)degrees, V = 1572.4(5) angstrom(3), Z = 4, F(000) = 704, D-c = 1.438 g/cm(3), mu = 0.228 mm(-1), R = 0.0600 and wR = 0.1448 for 2743 independent reflections (R-int = 0.0418) and 2174 observed ones (I > 2 sigma(I)). The in vitro antitumor activities of compounds 4a similar to 4i were preliminarily evaluated by the standard MTT assay.

Abstract:A series of hydroxamic acids with conjugated structure was designed and synthesized to explore the possible HDAC subtype selectivity by testing these compounds against recombinant human HDAC1 and HDAC4. The most selective compound resulted 5a, with a SI of 11.9. The enzymatic inhibitory activity of these conjugated compounds was relatively weak; however, some of these compounds showed significant effect in inducing apoptosis. Moreover, the anti-proliferative activity in cancer cells resulted quite promising, especially in the HCT119 cell line.

The invention also relates to a synthesis method of the compound, wherein the method adopts a chemical total synthesis route, and the convergent synthesis route can be applied to chemical synthesis of compounds with similar structures and related derivatives, and develops a wide development space for novel antitumor drugs.
