Abstract:Fluorescence-enhancement of 1,2,4-trihydroxyanthroquinone (THAQ)-Be2+ complex enhanced by nucleic acid was studied. Experimental results revealed that double-stranded DNA can enhance remarkably the fluorescence intensity of THAQ-Be2+, while RNA cannot. Based on these results, a fluorescence method for the selective determination of DNA in the presence of RNA was developed. Maximum fluorescence intensity was found in the pH range 3.6-4.5 with maximum excitation and emission wavelengths at 510 and 565 nm, respectively. Under optimum conditions, the calibration graph was linear over the range 0.08-18 micrograms ml-1 for double-stranded fish sperm DNA (fsDNA) with the detection limit being 2.75 x 10(-8) g ml-1. The method was applied for the determination of DNA in synthetic samples. The relative S.D. for five replicates was within 4%. In addition, the interaction mechanism of THAQ-Be2+ with DNA was also discussed.

Abstract:The reaction between nucleic acids and the trihydroxyanthroquinone (THAQ)–Al complex was studied. The nucleic acids used were fish sperm DNA (fsDNA), calf thymus DNA (ctDNA) and yeast RNA (yRNA). Experiments indicated that the addition of a very small amount of double-stranded DNA quenched the fluorescence of the THAQ–Al complex. When the mole ratio R of DNA to Al3+ was 0.53, the fluorescence quenching was a maximum. If R>0.53, the fluorescence increased gradually until a constant value was obtained. If DNA was replaced by RNA or denatured DNA, only a fluorescence quenching effect was observed. Under optimum conditions, the calibration graphs were linear over the range 3.8×10−8–1.75×10−6gml−1 fsDNA, 5.0×10−8–1.5×10−6gml−1 ctDNA and 2.0×10−8–1.5×10−4gml−1 yRNA. The detection limits were 6.6×10−9gml−1 fsDNA, 7.3×10−9gml−1 ctDNA and 5.4×10−9gml−1 yRNA. The method was applied to the determination of DNA or RNA in synthetic samples. The mechanism of the interaction between THAQ–Al and nucleic acids was also discussed.

Abstract:A novel fluorescent probe N-(N-(2-(4-morpholinyl)ethyl)-4-acridinecarboxamide)-alpha-alanine (N-(N-(ME)-4-ACA)-alpha-ALA) was synthesized. The structure was characterized by 1H NMR, MS, elemental analysis, fluorescent and ultraviolet spectra. This new compound exhibited high binding affinity to DNA, intense fluorescence and high water solubility. Experiment indicated that the fluorescent intensity was quenched when DNA was added. A method for DNA determination based on the quenching fluorescence (lambda(ex)=258nm, lambda(em)=451nm) of N-(N-(ME)-4-ACA)-alpha-ALA was established. Under optimal conditions (pH 7.2, CN-(N-(ME)-4-ACA)-alpha-ALA)=3 x 10(-6) mol L(-1)), the linear range is 0.1-4.0 microg mL(-1) for both fish semen (fsDNA) and calf thymus DNA (ct-DNA). The corresponding determination limits are 4.6 ng mL(-1) for fsDNA and 5.1 ng mL(-1) for ct-DNA, respectively. The relative standard deviation is 1.0%. Thus this compound can be used as a DNA fluorescent probe. The experiments proved that the interaction mode between N-(N-(ME)-4-ACA)-alpha-ALA and DNA was groove binding. The modified Rosenthal's graphical method gave the binding constant of 1.0 x 10(6) L mol(-1) and a binding size of 0.31 base pairs per bound drug molecule.

Abstract:Fluorescent sensing methods, because of their simple, cheap, fast, high selectivity and sensitivity , are widely used in the detection of cationic , anionic and other chemicals. Anthraquinone derivatives have many pharmacological activities, rigid planar structure and large ring conjugate system. They have moderate fluorescence emission wavelengths, good optical stabilities and high luminous efficiencies. In this work, anthraquinone derivatives were employed as fluorophore to prepare new fluorescent probes. Based on the mechanism of intramolecular charge transfer (ICT), a series of symmetric bis-1 ,2 ,3-triazole hydrazine derivatives K1 and K2 , which were confirmed by H-1 NMR and C-13 NMR, were designed and synthesized. The spectral properties were also investigated. The structure and theoretical spectrum of K1 were calculated using Gauss View 9. 0 quantization program and time-dependent density functional theory. The results showed that exhibited good selectivity to Ag+ in a mixed solution of DMSO/HEPES (1:7, V/V) . Binding analysis using the method of continuous variations (Job's plot) established that the stoichiometry of K1-Ag+ complex was estimated to be 1:1. The detection limit of K2 for Ag+ was 21. 2 mu mol/L (S/N= 3) . A complexation mechanism of K1-Ag+ was proposed, which was consistent with the calculated results. The fluorescence of K1 was increased dramatically in the presence of Ag+, and it remained nearly constant in the range of pH 6. 0-8.0. Other metal ions , such as Al3+, Ba2+, Cd2+, Co2+, Cr3+, Cu2+, Hg2+, Fe3+, K+, Mn2+, Na+, NH4+, Ni2+, Pb2+ and Zn2+ induced negligible fluorescence increasing for under the same conditions. The synthesized fluorescent probe K1 could be applied to image intracelluar Ag+ in living HepG2 cells , indicating that it was a useful tool in bioanalytical applications.

Abstract:Whereas most conventional DNA probes are flat disklike aromatic molecules, we explored the possibility of developing quadruplex sensors with nonplanar conformations, in particular, the propeller-shaped tetraphenylethene (TPE) salts with aggregation-induced emission (AIE) characteristics. 1,1,2,2-Tetrakis[4-(2-triethylammonioethoxy)phenyl]ethene tetrabromide (TPE-1) was found to show a specific affinity to a particular quadruplex structure formed by a human telomeric DNA strand in the presence of K(+) ions, as indicated by the enhanced and bathochromically shifted emission of the AIE fluorogen. Steady-state and time-resolved spectral analyses revealed that the specific binding stems from a structural matching between the AIE fluorogen and the DNA strand in the folding process. Computational modeling suggests that the AIE molecule docks on the grooves of the quadruplex surface with the aid of electrostatic attraction. The binding preference of TPE-1 enables it to serve as a bioprobe for direct monitoring of cation-driven conformational transitions between the quadruplexes of various conformations, a job unachievable by the traditional G-quadruplex biosensors. Methyl thiazolyl tetrazolium (MTT) assays reveal that TPE-1 is cytocompatible, posing no toxicity to living cells.

Abstract:A new kind of aggregation-induced emission compound was synthesized and used as the probe of nucleic acid. The characterization of this compound was studied. Both the RNA and DNA were detected by using this probe. And the detection scope of DNA and RNA was different. We researched the selectivity of our probe in double and single strand DNA sequences. The visualization of gel electrophoresis and the cell nucleus imaging were researched as well. Compared with the traditional nucleus dye Hoechst 33258, our probe also has the potential to be nucleus dye. And the cell toxicity was well performed by MTT assays.

Abstract:This paper describes a new fluorescence probe technique for the determination of DNA, which is based on a self-quenching course of high concentration of o-hydroxybenzoic acid or o-aminobenzoic acid. Studies involving calf thymus (CT) DNA, salmon (SM) DNA and herring sperm (HS) DNA revealed that the differential value of fluorescence intensity in the presence and absence of nucleic acid was proportional to the concentration of nucleic over the range of about 15 ng/mL similar to 6.0 mu g/mL.

Abstract:Based on the enhancement of the fluorescence intensity of the complex of 5-(3-fluoro-4-chlorophenylazo)-8-sulfonamidoquinoline (FCPBSQ) with Co2+ by nucleic acid in the presence of Tween 80 and in the weakly basic medium, a fluorescence method for the determination of nucleic acid was proposed. The calibration graphs for the determination of denatured calf thymus DNA (ct DNA), fish sperm DNA (fs DNA), and yeast RNA (yt RNA) were obtained in concentration ranges 0.050–4.0, 0.10–3.0, and 0.050–3.0 μg/ml with limits of detection of 0.010, 0.030, and 0.020 μg/ml, respectively. The method has been satisfactorily used for the determination of DNA in wheat cell extraction and ct DNA, fs DNA, and yt RNA in synthetic samples. Investigations on the binding mode by the Scatchard plots method suggested that both intercalation and electrostatic binding modes existed in this system.

Abstract:On the basis of the fluorescence enhancement effect of nucleic acids on the aluminum (III) / s-hydroxyquinoline (8-HQ) complex, a spectrofluorimetric method for nucleic acids is proposed in the present paper. In the pH range 5.8-7.0, the fluorescence of the AI(III)/8-HQ complex, excited at 265 nm or at 365 nm, is enhanced by nucleic acids. The calibration curve was linear in the range 0.25-3.0 mu g ml(-1) for calf thymus DNA, 0.25-3.5 mu g ml(-1) for fish sperm DNA and 0.5-3.5 mu g ml(-l) for yeast RNA, respectively. The limit of determination (3 sigma) was 0.024 mu g ml(-l) for calf thymus DNA, 0.013 mu g ml(-l) for fish sperm DNA and 0.130 mu g ml(-l) for yeast RNA, respectively. Six synthetic samples were determined with satisfaction.

Abstract:A new fluorescent probe for monitoring Zn2+ was synthesized based on the structure of 7-hydroxyquinoline. Compared with 8-substituted quinolines, the new probe exhibited higher selectivity for Zn2+ over Cd2+. Its fluorescence enhancement (14-fold) and nanomolar range sensitivity (Kd = 0.117 nM) were favorable toward biological applications. Experiments also showed that a cell-permeable derivative of the new probe was potentially useful for two-photon imaging in living cells.

Abstract:A novel ratiometric fluorescent probe for oxalic acid was designed and synthesized, based on the zinc-containing [DAQZ@2Zn(2+)] complex. It shows highly selective "on-off" fluorescence changes with a more than 20 nm blue shift in wavelength for oxalic acids in aqueous solution. Moreover, it can fluorescently respond to oxalic acid in living cells.

Abstract:Based on the aggregation-induced emission effect triggered by Al 3+ , a novel probe was constructed for the ratiometric fluorescence visualization detection of fluoroquinolones (FQs) residues. In this probe, Al 3+ cased the glutathione-modified copper nanoclusters (GSH-CuNCs) to aggregate by the interaction with surface carboxyl groups, restricting the intramolecular vibration and rotation of GSH-CuNCs and reducing the non-radiative relaxation of excited states. Significant enhancement of fluorescence and quantum yield were achieved. In the presence of FQs, Al 3+ /GSH-CuNCs coordinated with FQs to form a polymer that inhibited intramolecular proton transfer in the excited state of FQs, thereby enhancing the intrinsic fluorescence of FQs, while the fluorescence intensity of Al 3+ /GSH-CuNCs was not disturbed. The probe solution changed from red to blue (or green) under UV lamp as the concentration of FQs increased, providing the possibility for semi-quantitative visual analysis. It also showed sensitive spectral responses to FQs with detection limits of 7.9 (ciprofloxacin), 6.8 (gatifloxacin), and 9.5 nM (enrofloxacin), respectively. The application potential in the detection of FQs residues in poultry eggs was verified by real sample test. Additionally, the probe has been extended to the development of fluorescent test paper and the smartphone detection platform, contributing to rapid, portable, visual quantitative analysis of FQs.

Methylated Chromenoquinoline As a Novel Nucleus Fluorescent Probe for Nucleic Acid Imaging

Abstract:Two chromenoquinoline-based fluorescent probes 1a-b have been synthesized and investigated. Photofading behaviors of compounds 1a-b showed that at least 89% absorption remained after 6 h irradiating, meanwhile, many of ions and amino acids had negligible impacts on their fluorescence intensity, which meant they had excellent photostability and selectivity. Probes 1a-b exhibited strong absorption and emission in organic solvents with large fluorescence quantum yields, even in water probe 1a still had a relatively large fluorescence quantum yield (20%). Combined with DFT calculation, the influence of alkylation on optical properties of 1b was elucidated. In addition, the fluorescence intensity of probe 1b with red emission enhanced by 5.4-fold and 5.3-fold after DNA and RNA added, and the fluorescence quantum yield increased from 3% to 17% and 14%, respectively, but the neutral molecule 1a had no response to nucleic acid. Furthermore, confocal microscopy imaging of probes 1a-b showed that 1a targeted lipid droplets while the methylated probe 1b to nucleus in living HeLa cells. The results indicated that the subcellular targeting zone could be changed by alkylation of nitrogen atom on chromenoquinoline-based conveniently, which provided a new idea for designing and synthesizing new subcellular labeled probes.

Abstract:We develop a novel single fluorophore-labeled double-stranded oligonucleotide (OND) probe for rapid, nanostructure-free, fluorescence-enhanced nucleic acid detection for the first time. We further demonstrate such probe is able to well discriminate single-base mutation in nucleic acid. The design takes advantage of an inherent quenching ability of guanine bases. The short strand of the probe is designed with an end-labeled fluorophore that is placed adjacent to two guanines as the quencher located on the long opposite strand, resulting in great quenching of dye fluorescence. In the presence of a target complementary to the long strand of the probe, a competitive strand-displacement reaction occurs and the long strand forms a more stable duplex with the target, resulting in the two strands of the probe being separated from each other. As a consequence of this displacement, the fluorophore and the quencher are no longer in close proximity and dye fluorescence increases, signaling the presence of target.

Abstract:1,4-Dithiothreitol (DTT) has wide applications in cell biology and biochemistry. Development of effective methods for monitoring DTT in biological systems is important for the safe handling and study of toxicity to humans. Herein, we describe a two-photon fluorescence probe (Rh-DTT) to detect DTT in living systems for the first time. Rh-DTT showed high selectivity and sensitivity to DTT. Rh-DTT can be successfully used for the two-photon imaging of DTT in living cells, and also can detect DTT in living tissues and mice.
A New Quinone Based Fluorescent Probe for High Sensitive and Selective Detection of Biothiols and Its Application in Living Cell Imaging

Abstract:In view of the vital role of biothiols in many physiological processes, the development of simple and efficient probe for the detection of biothiols is of great medical significance. In this work, we demonstrate the use of 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ), which respond rapidly to biothiols especially to glutathione, as a new fluorescent probe for the selective detection and bioimaging of biothiols. This new fluorescent probe can distinguish glutathione from cysteine and homocysteine easily under physiological concentration and detect glutathione quickly within three minutes. This probe exhibits high selectivity to biothiols and the detection limit was determined to be 3.08 × 10−9 M for glutathione, 8.55 × 10−8 M for cysteine, and 2.17 × 10−9 M for homocysteine, respectively. The sensing mechanism was further explored by density functional theory (DFT) and nuclear magnetic resonance (NMR) experiment; results showed that the interaction forces between the probe and biothiols were electrostatic interaction. In addition, the probe has been successfully applied to the detection of biothiols in Eca9706 cells by fluorescence confocal imaging technology.

Abstract:A novel method was developed for selective determination of deoxyribonucleic acid (DNA) with quinoline sulphate. The fluorescence of quinoline sulphate was greatly quenched by DNA in the pH range of 2similar to4 and the decreased intensity of fluorescence was proportional to the concentration of DNA. The linear range of the calibration curve was 10similar to700 mug/L with the detection limit of 5.9 mug/L. The proposed method was successfully used to determine the plasmid DNA extracted from colibacillus, which was dealt with ribonucleic acidase before determination.

Abstract:In this study, a novel fluorescent probe of acridine derivative N-((N-(2-dimethylamino)ethyl)acridine-4-carboxamide)-α-alanine (N-(ACR-4-CA)-α-ALA) was synthesized. The structure of the new compound was characterized by 1H NMR, MS, elemental analysis, fluorescent and ultraviolet spectra. It was found that DNA had the ability to quench the fluorescence of N-(ACR-4-CA)-α-ALA, and the quenched intensity of fluorescence was proportional to the concentration of DNA. A method for DNA determination based on the quenching fluorescence (λex=260nm, λem=451nm) of N-(ACR-4-CA)-α-ALA was established. Under optimal conditions, the linear range is 0.05–2.0μgmL−1 for both fish semen (fsDNA) and calf thymus DNA (ctDNA). The corresponding determination limits are 9.1ngmL−1 for fsDNA and 8.7ngmL−1 for ctDNA, respectively. The results suggested that the interaction mode between N-(ACR-4-CA)-α-ALA and DNA was intercalative binding. The intrinsic binding constant was determined and the result showed a large binding constant of N-(ACR-4-CA)-α-ALA with DNA.
A Highly Selective and Sensitive Turn-on Probe for Aluminum(iii) Based on Quinoline Schiff's Base and Its Cell Imaging

Abstract:A reversible Schiff's base fluorescence probe for Al3+, (3,5-dichloro-2- hydroxybenzylidene) quinoline-2-carbohydrazide (QC), based on quinoline derivative has been designed, synthesized and evaluated. The QC exhibited a high sensitivity and selectivity toward Al3+ in EtOH-H2O (v/v = 1:9, pH = 6) by forming a 1:1 complex with Al3+ and the detection limit of QC for Al3+ was as low as 0.012 μM. Furthermore, these results displayed that the binding of QCAl3+ was broken by F−, so this system could be used to monitor F− in the future. The enhancement fluorescence of the QC could be attributed to the inhibition of PET and ESIPT and the emergency of CHEF process induced by Al3+. More importantly, QC was not only successfully used for the determination of trace Al3+ in the tap water and the human blood serum, but was valid for fluorescence imaging of Al3+ in the Hela cells.

Abstract:A novel fluorescent probe was designed and synthesized. The probe exhibited much higher selectivity and sensitivity for detecting aluminum(III) compared with other metal ions, anions, and amino acids in C2H5OH/H2O media through a fluorescence enhancement. The mechanism was proposed to form a kind of 2: 1-type complex between probe and aluminum( III) with a detection limit of 0.17 mu M.
