Abstract:It is very significant to research spoilage microorganisms in stored maize.A predominant strain of mold in the stored ice was isolated and identified.According to its morphological observation,phylogenetic analysis of ITS sequence and the reference manual related microbial identification,the strain was preliminary identified as Aspergillus oryzae.The measurement can be used as an accurate,reliable and straightforward method to identify the original species of the stored maize and explore genetic diversity.
Analysis of rice microbial communities under different storage conditions using culture-dependent and -independent techniques

Abstract:Rice is a staple food for over half of the world’s population, and fungal spoilage in stored rice may occur when the moisture content and temperature are conducive. Aspergillus sp. and Penicillium sp. are the most harmful toxigenic species that produce harmful mycotoxins. Molds pose a potential threat to public health and cause a huge economic loss. Therefore, it is of great importance to find out how molds multiply in rice. This study focused on the isolation and identification of fungi presented in rice and their evolution in rice with different moisture contents stored for varying periods of time and under different temperatures. Mold community was detected every month using the culture-dependent and -independent method of polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). Significant differences were detected by the traditional culture method under different storage conditions. For potato dextrose agar (PDA) media, high temperature and moisture were suitable for the dominant strains including Penicillium aurantiogriseum and Penicillium oxalicum. In particular, P. oxalicum competitively inhibited the other fungi. For Rose Bengal medium, no difference was observed under different storage conditions, and only typical strains such as Aspergillus candidus and Alternaria were detected. PCR-DGGE identified some uncultured strains such as Trichoderma sp. and Cladosporium sp., the dominant strains and the flora diversity such as Aspergillus restrictus and Eurotium athecium. These results indicated that storage conditions greatly shape fungal growth. This study provides a foundation for the evolution of fungal flora in rice during storage in China and may help in developing biological control methods to prevent mold contamination in rice.

Abstract:[Objective]To isolate microorganisms in specific ecological environment( rotten straw),and study the degrading abilities of these microorganisms on rice stalk. [Method]By using microcrystalline cellulose powder as carbon source,we isolated and identificated microbes that degrading rice stalks in certain natural ecological environment. [Result] From straw stalks,7 strains were isolated. Morphological observation and biochemical identification showed that 2 isolates belong to penicillium,2 belong to Aspergillus furnigatus,1 belongs to Rhizopus microsporus,1 Gongronella butleri and 1 Mucor racemosus. Furthermore,the ability of these isolates to degrade rice stalks was studied. Among which,the abilities of degrading straw cellulose of 1 Aspergillus furnigatus and 1 Gongronella butleri are strongger,the abilities of degrading straw hemicellulose of another Aspergillus furnigatus and 1 Gongronella butleri are strongger. [Conclusion] The study can provide reference for microbial strains degrading rice stalk.
Comparative transcriptomic analysis and volatile compound characterization of Aspergillus tubingensis and Penicillium oxalicum during their infestation of Japonica rice

Abstract:Volatile organic compounds (VOCs), a byproduct of mold metabolism, have garnered increasing interest because the VOCs can be used to detect food early contamination. So far, the use of VOCs as indicators of rice mildew, specifically caused by Aspergillus tubingensis and Penicillium oxalicum, and the mechanisms of their generation are not well investigated. This study examines the VOCs produced by these molds during paddy storage, utilizing headspace solid-phase micro-extraction gas chromatography-mass spectrometry (HS-SPME-GC-MS). We further elucidate the mechanisms underlying the formation of these VOCs through a comparative transcriptomic analysis. The VOCs characteristic to A. tubingensis and P. oxalicum, identified with a VIP value > 1 in the partial least squares discriminant analysis (PLS-DA) model, are primarily alkenes. Our transcriptome analysis uncovers key metabolic pathways in both molds, including energy metabolism and pathways related to volatile substance formation, and identifies differentially expressed genes associated with alkane and alcohol formation.
Identification and characterization of rhizosphere fungal strain MF-91 antagonistic to rice blast and sheath blight pathogens

Abstract:Aim To examine the inhibition effects of rhizosphere fungal strain MF-91 on the rice blast pathogen Magnaporthe grisea and sheath blight pathogen Rhizoctonia solani. Methods and Results Rhizosphere fungal strain MF-91 and its metabolites suppressed the in vitro mycelial growth of R.solani. The inhibitory effect of the metabolites was affected by incubation temperature, lighting time, initial pH and incubation time of rhizosphere fungal strain MF-91. The in vitro mycelial growth of M.grisea was insignificantly inhibited by rhizosphere fungal strain MF-91 and its metabolites. The metabolites of rhizosphere fungal strain MF-91 significantly inhibited the conidial germination and appressorium formation of M.grisea. Moreover, the metabolites reduced the disease index of rice sheath blight by 35 center dot 02% in a greenhouse and 57 center dot 81% in a field as well as reduced the disease index of rice blast by 66 center dot 07% in a field. Rhizosphere fungal strain MF-91 was identified as Chaetomium aureum based on the morphological observation, the analysis of 18S ribosomal DNA internal transcribed spacer sequence and its physiological characteristics, such as the optimal medium, temperature and initial pH for mycelial growth and sporulation production. Conclusions Rhizosphere fungus C.aureum is effective in the biocontrolling of rice blast pathogen M.grisea and sheath blight pathogen R.solani both in in vitro and in vivo conditions. Significance and Impact of the Study This study is the first to show that rhizosphere fungus C.aureum is a potential fungicide against rice blast and sheath blight pathogens.
Microenvironmental Interplay Predominated by Beneficial Aspergillus Abates Fungal Pathogen Incidence in Paddy Environment

Abstract:Rice fungal pathogens, responsible for severe rice yield loss and biotoxin contamination, cause increasing concerns on environmental safety and public health. In the paddy environment, we observed that the asymptomatic rice phyllosphere microenvironment was dominated by an indigenous fungus, Aspergillus cvjetkovicii, which positively correlated with alleviated incidence of Magnaporthe oryzae, one of the most aggressive plant pathogens. Through the comparative metabolic profiling for the rice phyllosphere microenvironment, two metabolites were assigned as exclusively enriched metabolic markers in the asymptomatic phyllosphere and increased remarkably in a population-dependent manner with A. cvjetkovicii. These two metabolites evidenced to be produced by A. cvjetkovicii in either a phyllosphere microenvironment or artificial media were purified and identified as 2(3H)-benzofuranone and azulene, respectively, by gas chromatography coupled to triple quadrupole mass spectrometry and nuclear magnetic resonance analyses. Combining with bioassay analysis in vivo and in vitro, we found that 2(3H)-benzofuranone and azulene exerted dissimilar actions at the stage of infection-related development of M. oryzae. A. cvjetkovicii produced 2(3H)-benzofuranone at the early stage to suppress MoPer1 gene expression, leading to inhibited mycelial growth, while azulene produced lately was involved in blocking of appressorium formation by downregulation of MgRac1. More profoundly, the microenvironmental interplay dominated by A. cvjetkovicii significantly blocked M. oryzae epidemics in the paddy environment from 54.7 to 68.5% (p < 0.05). Our study first demonstrated implication of the microenvironmental interplay dominated by indigenous and beneficial fungus to ecological balance and safety of the paddy environment.

Abstract:Fusarium species are important seedborne pathogens that cause rice bakanae disease (RBD). In this study, 421 strains were isolated from 25 rice samples collected from Zhejiang, Anhui, and Jiangxi provinces of China. Furthermore, 407 isolates were identified as F. fujikuroi (80.05% isolation frequency), F. proliferatum (8.31%), F. equiseti (5.94%), F. incarnatum (2.61%), F. andiyazi (0.95%), and F. asiaticum (0.48%) based on morphology and translation elongation factor 1-alpha (TEF1-α) gene. Phylogenetic analysis of combined sequences of the RNA polymerase II largest subunit (RPB1), RNA polymerase II second largest subunit (RPB2), TEF1-α gene, and ribosomal DNA (rDNA) internal transcribed spacer (ITS) showed that 17 representative strains were attributed to six species. Pathogenicity tests showed that representative isolates possessed varying ability to cause symptoms of bakanae on rice seedlings. Moreover, the seed germination assay revealed that six isolates had different effects, such as inhibition of seed germination, as well as seed and bud rot. The loop mediated isothermal amplification (LAMP)-based assay were developed for the detection of F. fujikuroi. According to sequences of desaturase-coding gene promoter, a species-specific marker desM231 was developed for the detection of F. fujikuroi. The LAMP assay using seeds collected from field was validated, and diagnostics developed are efficient, rapid, and sensitive.

Abstract:The spoilage molds which caused the corruption of fresh- cut apple were identified,and the rot ability of molds were compared in vivo. 13 Strains of molds were isolated from fresh- cut apples,in which there were 7Penicillium strains,1 Aspergillus strain,3 Cladosporium strains,1 Acremonium strain,and 1 Alternaria strain by morphological character methtods.The different penicillium strains were identified by the cultivation characters and microscopic examination in different medium including CA,CYA,G25 N. The results showed that 5 types of Penicillium were identified, which were P.crustosum, P.expansum, P.miczynskii, P.aurantiogriseum and P.viridicatum.The in vivo experiments were used to determine type of molds spoilage or not. Five spoilage molds were identified,which were P.viridicatum,P.expansum,P.crustosum,A.alternata,and A.niger. And five kinds of molds could not rot apple were indentified as P.aurantiogriseum, A.terricola, C.sphaerospermum,C.cladosporioides,and P.miczynskii. The order of rot ability of 5 spolilage molds was P.crustosum,P.expansum,P.viridicatum,A.alternata,A.niger.
Gram-Positive Bacteria Associated with Rice in China and Their Antagonists Against the Pathogens of Sheath Blight and Bakanae Disease in Rice

Abstract:It is necessary to understand the bacterial populations associated with rice so as to provide more information and natural resources for effective management of major diseases in rice. A survey on screening and identification of gram-positive bacteria was conducted during 1998-2004. Seven hundred and fifty-six rice samples were collected from Zhejiang, Jiangsu, Fujian and Yunnan Provinces, China. Over 1000 bacterial isolates were isolated and tested for colony morphology, pathogenicity, and some characteristics of bacteriology including Gram staining, fluorescent pigment on Kings medium B and microscopic observation for endospore. Together with five standard reference strains, 74 representative gram-positive bacterial isolates were confirmed by Biolog and gas chromatographic analysis of fatty acid methyl esters. Five bacterial species of Bacillus and other three genera were identified and isolates from Bacillus sublitis and Bacillus megaterium, exhibited the most effective inhibition against the pathogens of sheath blight and bakanae disease of rice. A few isolates from Bacillus pumilus and Bacillus megaterium showed weak virulent on rice together with some virulent isolates, risk should be considered when isolates from these species were screened for biocontrol agents.

Abstract:Six molds (Aspergillus niger, Penicillium janthinellum, Penicillium alternatum, Penicillium funiculosum, Aspergillus flavus and Aspergillus parasitica) were isolated from naturally fragrant rice for their characterization. Microbial volatile organic compounds (mVOCs) produced by the molds were determined by gas chromatography – ion mobility spectrometry (GC-IMS). With the help of statistical analysis and digital image processing, specific mVOCs with mildew information were automatically selected, and a discrimination model of moldy fragrant rice was established using chemometrics. The results showed that main mVOCs produced by the mold were alcohols, aldehydes, ketones, and esters. The moldy and normal fragrant rice samples were divided into two clusters in the principal component score diagram. The model developed by quadratic discriminant analysis indicated that the average recognition rate for test sample sets was 95.25%. In conclusion, the composition of mVOCs was used to characterize the mildew of fragrant rice. GC-IMS combined with chemometric tools is effective for the nondestructive discrimination of moldy fragrant rice which is significant for monitoring quality and storage.

Abstract:In this paper,molds of apple were separated,purificated and identificated,as well as different antiseptic to different strains were studied,in order to take further effective measures to extend the shelf life of apple or provide a theoretical basis. Early Identification of Aspergillus Spp. Contamination in Milled Rice by E-nose Combined with Chemometrics.

Abstract:BACKGROUND Rice grains can be contaminated easily by certain fungi during storage and in the market chain, thus generating a risk for humans. Most classical methods for identifying and rectifying this problem are complex and time-consuming for manufacturers and consumers. However, E-nose technology provides analytical information in a non-destructive and environmentally friendly manner. Two-feature fusion data combined with chemometrics were employed for the determination of Aspergillus spp. contamination in milled rice. RESULTS Linear discriminant analysis (LDA) indicated that the efficiency of fusion signals ('80(th) s values' and 'area values') outperformed that of independent E-nose signals. Linear discriminant analysis showed clear discrimination of fungal species in stored milled rice for four groups on day 2, and the discrimination accuracy reached 92.86% by using an extreme learning machine (ELM). Gas chromatography-mass spectrometry (GC-MS) analysis showed that the volatile compounds had close relationships with fungal species in rice. The quantification results of colony counts in milled rice showed that the monitoring models based on ELM and the genetic algorithm optimized support vector machine (GA-SVM) (R-2 = 0.924-0.983) achieved better performances than those based on partial least squares regression (PLSR) (R-2 = 0.877-0.913). The ability of the E-nose to monitor fungal infection at an early stage would help to prevent contaminated rice grains from entering the food chains. CONCLUSIONS The results indicated that an E-nose coupled with ELM or GA-SVM algorithm could be a useful tool for the rapid detection of fungal infection in milled rice, to prevent contaminated rice from entering the food chain. Abstract:Wild filamentous fungi obtained from natural fermented millet catsup were identified as Aspergillus oryzae by morphology after purifying iteratively.The Folin method was used to measure the activity of proteases and find out the most active strain.The result showed that the activity of protease should be concerned,as well as the specific growth rate of spores when selecting and breeding the excellent strains.

Abstract:Rice wine, well known for its unique flavor, rich nutritional value, and health benefits, has potential for extensive market development. Rhizopus and Aspergillus are among several microorganisms used in rice wine brewing and are crucial for determining rice wine quality. The strains were isolated via Rose Bengal and starch as a combined separation medium, followed by oenological property and sensory evaluation screening. The strain exhibiting the best performance can be screened using the traditional rice wine Qu. The strains YM-8, YM-10, and YM-16, which exhibited strong saccharification and fermentation performance along with good flavor and taste, were obtained from traditional rice wine Qu. Based on ITS genetic sequence analysis, the YM-8, YM-10, and YM-16 strains were identified as Rhizopus microsporus, Rhizopus arrhizus, and Aspergillus oryzae. The optimum growth temperature of each of the three strains was 30°C, 32°C, and 30°C, and the optimum initial pH was 6.0, 6.5, and 6.5, respectively. The activities of α-amylase, glucoamylase, and protease of YM-16 were highest at 220.23±1.88, 1,269.04±30.32, and 175.16±1.81 U/g, respectively. The amino acid content of rice wine fermented in a 20-L bioreactor with the three mold strains was higher than that of the control group, except for arginine, which was significantly lower than that of the control group. The total amino acid content and the total content of each type of amino acid were ranked as YM-16 > YM-8 > YM-10 > control group, and the amino acid content varied greatly among the strains. The control group had a higher content, whereas YM-8 and YM-16 had lower contents of volatile aroma components than the control group and had the basic flavor substances needed for rice wine, which is conducive to the formation of rice wine aroma. This selected strain, YM-16, has strong saccharification and fermentation ability, is a rich enzyme system, and improves the flavor of rice wine, thereby demonstrating its suitability as a production strain for brewing.

Abstract:Processed unhusked rice is prone to mildew during storage. In this study, the storage conditions were simulated at temperatures of 20, 30, and 35 °C and a relative humidity of 40%, 60% and 80%, respectively. The water, fatty acid, and total starch content and the peak viscosity, mold colony number, protein secondary structure, and spatial structure of rice were monitored in order to propose the critical point of mildew during storage. In the process of rice from lively to moldy, the water content, fatty acid contents and the peak viscosity were increased. The total starch content decreased and then showed a slow increasing trend, while the microstructure of the powder particles changed from smooth and complete to loosen and hollow. With the increase in storage time, the vibration of the amide I band of the rice samples decreased slightly, indicating that the total contents of β-fold, β-turn, α-helix, and random curl of the rice protein also changed. PCA (Principal Component Analysis) analysis showed that rice mildew index was closely related to temperature and humidity during storage. In our investigation, the best and most suitable temperature and relative humidity for rice storge is 20 °C and 40%, respectively. These results suggested that temperature and environmental humidity are vital factors affecting the physicochemical properties and nutrient changes, which provides a theoretical basis for the early warning of rice mildew during storage.

Identification of the Causal Organism of Bacterial Brown Stripe from Rice Seedlings

Abstract:For elucidating the brown stripe pathogen of rice and differentiating it from watermelon blotch bacterium,four out of six bacterial strains isolated from the diseased rice seedlings were compared with three standard reference strains of Acidovorax avenae subsp.avenae and two standard reference strains of A.avenae subsp.citrulli by the bacteriological tests,colony morphology,pathogenicity,Biolog,FAME analysis,electromicroscope observation and nested-PCR.It has confirmed that A.avenae subsp.avenae is the causal agent of bacterial brown stripe of rice.The results also showed that FAME analysis misidentified some strains of A.avenae subsp.avenae as A.avenae subsp.citrulli whereas Biolog and the nested-PCR were the best way for distinguishing the two bacterial pathogens.

Abstract:Aspergillus flavus is a significant fungus that poses a threat to food safety by producing mycotoxins in various crops. In this study, A. flavus isolates were obtained from storage rice collected from seven provinces in southern China, and their AFB1 production, biosynthesis genes presence, and diversity were detected. Results showed that 56 out of the 81 A. flavus isolates produced detectable levels of AFB1, and 71 isolates (87.6 %) possessed aflR gene in their AF synthesis gene cluster, while only 41 isolates (50.6 %) had the ver-1 gene present. Genetic diversity analysis using inter-simple sequence repeats (ISSR) markers revealed seven main clusters among the isolates and the genetic similarity coefficients of 81 A. flavus isolates ranged from 0.53 to 1.00. Additionally, coculture assays were conducted using two toxigenic and two atoxigenic isolates from the same grain depot to investigate the effect of intraspecific inhibition on AFB1 production and to assess the AFB1 contamination risk of storage rice. The in situ results demonstrated that the atoxigenic isolates effectively inhibited the AFB1 contamination of toxigenic isolates. These findings provide insight into the genetic diversity of A. flavus isolates populations and highlight the potential food safety hazards of them in stored rice grain in China.
Deciphering the Microbiological Mechanisms Underlying the Impact of Different Storage Conditions on Rice Grain Quality

Abstract:Different storage conditions can influence microbial community structure and metabolic functions, affecting rice grains' quality. However, the microbiological mechanisms by which different storage conditions affect the quality of rice grains are not yet well understood. This study monitored the quality (the content of starch, protein, etc.) and microbial community structure of rice grains stored under different storage conditions with nitrogen gas atmosphere (RA: normal temperature, horizontal ventilation, RB: normal temperature, vertical ventilation, RC: quasi-low temperature, horizontal ventilation). The results revealed that the rice grains stored under condition RB exhibited significantly lower quality compared to condition RA and RC. In addition, under this condition, the highest relative abundance of Aspergillus (16.0%) and Penicillium (0.4%) and the highest levels of aflatoxin A (3.77 ± 0.07 μg/kg) and ochratoxin B1 (3.19 ± 0.05 μg/kg) were detected, which suggested a higher risk of fungal toxin contamination. Finally, co-occurrence network analysis was performed, and the results revealed that butyl 1,2-benzenedicarboxylate was negatively correlated (p < 0.05) with Moesziomyces and Alternaria. These findings will contribute to the knowledge base of rice storage management and guide the development of effective control measures against undesirable microbial activities.

Abstract:A survey on isolation and detection of the casual organism of bacterial grain rot of rice was conducted during 1997–2006. In 2006, six pathogenic bacterial strains were isolated from two symptomless seed samples of rice (Oryza sativa L.) originally produced in Hainan Province and then planted in Zhejiang Province, China. They were identified as Burkholderia glumae which is the causal organism of bacterial grain rot of rice by physiological characteristics, colony morphology, pathogenicity test, Biolog, fatty acid methyl ester (FAME) analysis and RAPD-PCR compared with the four standard reference strains. It is confirmed that there is the infection of B. glumae in so-called ‘health looking seeds’.
