Abstract:Hydrophobic charge induction chromatography(HCIC)is a new kind of chromatographic method developed in recent years. The HCIC ligands normally combine the hydrophobic and electrostatic interactions,thus HCIC belongs to a kind of mixed-mode chromatography. HCIC has been widely studied for protein separation,especially for antibody purification,which shows good operation and separation selectivity. This review focuses on the principles,characteristics and applications of HCIC,which would be helpful for the future researches and applications.

Abstract:Due to the shortage of blood donation and safe issues of blood-derived pathogens, recombinant human serum albumin (rHSA) expressed by Pichia pastoris is a potential substitute for plasma-derived HSA (pHSA). In this work, four commercialized mixed-mode resins (Capto adhere, Capto MMC, MX-Trp-650M, and Nuvia cPrime) and two ion-exchange resins (QSepharose FF and SP Sepharose FF) were used for rHSA separation from Pichia pastoris cell culture broth, and loading and elution conditions were optimized via a high-throughput process using microtiter filter plates. The results indicated that resins with cation-exchange groups had better adsorption performance. The loading conditions at pH 4.0 and dilution rate of 1.0 were suitable for SP Sepharose FF, and no dilution worked for mixed-mode resins. The optimal elution pH was determined to be pH 7.0 for SP Sepharose FF, pH 8.0 for MX-Trp-650M and Nuvia cPrime, and pH 9.0 for Capto MMC. Moreover, rHSA separation from Pichia pastoris broth was performed on column under optimal conditions based on the high-throughput screening. The results showed that no feedstock dilution was needed for Capto MMC and MX-Trp-650M, and separation performance of high dynamic binding capacity at 10% breakthrough (similar to 50 mg/mL), elution recovery (>95%), and purity (>96%) as well as excellent removal of pigments (A(350) < 0.080) and HCPs (<581 ppm) could be obtained. The cation-exchange mixed-mode resins (especially MX-Trp-650M) were more suitable for direct capture of rHSA from crude feedstock. The high-throughput method developed can rapidly determine the optimal separation conditions for column chromatographic separation.

Abstract:High-throughput approaches become increasingly important on process development for protein separation. In this work, four mixed-mode resins (Capto adhere, Capto MMC, MX-Trp-650 M and Nuvia cPrime) were investigated for human serum albumin (HSA) separation and compared with two ion-exchangers (Q Sepharose FF and SP Sepharose FF). High-throughput method based on microtiter filter plate was used to determine adsorption properties and optimize separation conditions. For resins with anion-exchange groups alkaline condition would be favorable for HSA adsorption, while acidic condition for resins with cation-exchange groups. Mixed-mode resins showed obviously salt-tolerant properties. Salt addition had little effect on Nuvia cPrime, and could promote HSA adsorption for Capto MMC and MX-Trp-650 M. The elution conditions were also optimized and high recovery could be obtained. In addition, the optimized conditions obtained by high-throughput approaches were verified by lab-scale column chromatography. It was found that Capto MMC and MX-Trp-650 M would be more suitable for HSA separation and DBC10% could reach more than 40 mg/ml under 200 mM NaCl addition. Finally, rHSA separation from P. pastoris culture broth was tested with MX-Trp-650 M, and high purity and recovery could be obtained. This study demonstrated that high-throughput screening method is a powerful tool to optimize the chromatographic separation. Mixed-mode cation-exchange resins showed an efficient practicability for HSA separation with high capacity, suitable salt-tolerance and high elution recovery. (C) 2017 Elsevier B.V. Abstract:High-content screening (HCS) technology combines automated high-speed imaging hardware and single-cell quantitative analysis. It can greatly accelerate data acquisition in cellular fluorescence imaging and is a powerful research technique in traditional Chinese medicine (TCM). An increasing number of laboratories and platforms, including TCM laboratories, have begun utilizing HCS systems. However, this technology is still in its infancy in TCM research and there is a lack of sufficient experience with the associated concepts, instrument configurations, and analysis methods. To improve the understanding of HCS among researchers in the field of TCM, this paper summarizes the concept of HCS, software and hardware configuration, the overall research process, as well as common problems and related solutions of HCS in TCM research based on our team’s previous research experience, providing several research examples and an outlook on future perspectives, aiming to provide a technical guide for HCS in TCM research.

Abstract:Counter-current chromatography (CCC) is a unique support-free liquid-liquid partition chromatography winning wide applications in the separation of various components from natural or synthetic mixtures. It has been one of the prime methods for isolating compounds from Traditional Chinese Medicines (TCM) and other comprehensive natural products. Although early CCC models produced a long-standing false image that CCC is a time-consuming technique, rapid and high-performance CCC devices and methods for high throughput analysis of natural mixtures have been advanced. For instances, multi-channel CCC, dual CCC, elution-extrusion CCC, and solvent simplification protocols can provide high throughput CCC analysis and produce high purity of compounds or large natural product libraries for drug discovery. This review summarizes the recent advancements of CCC in the high throughput analysis of natural product with an emphasis on the developments of instruments and methods.

Abstract:Countercurrent chromatography is a powerful separation technique, which is used as an alternative or complementary technique to other chromatographic methods due to its high efficiency and preparative capacity. This technique does not require a solid stationary phase and relies simply on the partition of a sample between the two phases of an immiscible solvent system. Thus, it eliminates the complications resulting from the solid support matrix, such as irreversible adsorptive sample loss and deactivation, tailing of solute peaks, and contamination. With these advantages, CCC is gaining popularity as an important separation method. This review summarizes the basic principles and gives the most recent CCC applications in various areas, including the isolation and purification of natural products, development of different elution modes and multidimensional methods, progress in pH‐zone‐refining CCC techniques, bioseparation with aqueous polymer two‐phase systems, enantioseparation with various efficient chiral selectors, and on‐line monitoring of the eluate. Abstract:Abstract Polysaccharides, with an extensive distribution in natural products, represent a group of natural bioactive substances having widespread applications in health-care food products and as biomaterials. Devising an efficient system for the separation and purification of polysaccharides from natural sources, hence, is of utmost importance in the widespread applicability and feasibility of research for the development of polysaccharide-based products. High-speed counter-current chromatography (HSCCC) is a continuous liquid–liquid partitioning chromatography with the ability to support a high loading amount and crude material treatment. Due to its flexible two-phase solvent system, HSCCC has been successfully used in the separation of many natural products. Based on HSCCC unique advantages over general column chromatography and its enhanced superiority in this regard when coupled to aqueous two-phase system (ATPS), this review summarizes the separation and purification of various bioactive polysaccharides through HSCCC and its coupling to ATPS as an aid in future research in this direction.

Abstract:<p>In the present study, high-speed counter-current chromatography with a three-phase solvent system composed of <em>n</em>-hexane-acetonitrile-400 mM ammonium sulfate (1:1:1, v/v) was developed to recover four mixed dyes (methyl green, acridine orange, Congo red and oil red O) with diverse polarities. As a result, four dyes were successfully separated in a single HSCCC run using a step-wise gradient elution. Then, elution-extrusion and back-extrusion counter-current chromatography were used to reduce the separation time and solvent consumption. Among them back-extrusion CCC can save more time. Thus, the present study provides an effective method for dye recycling.</p>

Abstract:Alkaloids represent a most widespread group of bioactive natural products. Because of their alkalinity and structural diversity, the fractionation and purification of the alkaloids from herbs can often present a number of practical difficulties using the conventional chromatographic techniques. High-speed counter-current chromatography (HSCCC) is a liquid-liquid partition chromatography with a support-free liquid stationary phase, and is gaining more and more popularity as a viable separation technique for bioactive compounds from natural resources. In the present review, focus is placed on the separation of alkaloids by both conventional HSCCC and pH-zone-refining counter-current chromatography (CCC) techniques from herbs. The review presents the separation of over 120 different alkaloid compounds from more than 30 plant species by the conventional HSCCC and pH-zone-refining CCC. Based on the data from the literature, the proper solvent systems for the separation of alkaloids by the conventional HSCCC and pH-zone-refining CCC are also summarized.
Application of Preparative High-Speed Counter-Current Chromatography for Separation of Chlorogenic Acid from Flos Lonicerae

Abstract:Chlorogenic acid, an ester formed between caffeic acid and quinic acid, is a major phenolic compound in the traditional Chinese medicinal herb Flos Lonicerae. The separation and purification of chlorogenic acid from the crude extract of Flos Lonicerae was achieved by high-speed counter-current chromatography (HSCCC). A high acid, highly polar two-phase solvent system containing n-butanol-acetic acid-water (4:1:5) was run on a preparative scale. The upper phase was used as the mobile phase in the head to tail elution mode. A 300-mg quantity of the crude extract containing 5.97% chlorogenic acid was loaded on a 342-ml HSCCC column. Double separations were performed with the same solvent system yielding 16.9 mg chlorogenic acid at 94.8% purity with approximately 90% recovery.

Abstract:Aiming at exploring the components and the specific molecular structure of natural brown cotton pigments,the high-performance liquid chromatography(HPLC) with polyamide as the column was used to separate pigments extracted from natural brown cotton fiber.The results showed that the pigment extract can be successfully fractionated by polyamide column chromatography,and the pigments consist of two fractions for their different colors and HPLC chromatograms of elution segments.Two colored component and one colorless component were separated from water elution segment by C18-reversed-phase chromatography,but the other segment can not be separated by this chromatography column,and according to the retention action on chromatography columns,this part of pigment may belong to a series of polyphenol compounds that have very similar structures.

Abstract:Countercurrent chromatography (or counter-current chromatography, CCC) is a unique support-free liquid-liquid partition chromatography. Since it was invented by Y. Ito in 1960s, CCC has been widely accepted and applied as popular separation and purification technique for natural and synthetic complex. However, up to date there is little study to address on hydrophobic and hydrophilic interactions in CCC process, although hydrophobic interaction chromatography (HIC) and hydrophilic interaction chromatography (or hydrophilic interaction liquid chromatography, HILIC) as solid-support chromatographic techniques are widely applied at different stages of downstream processing. In fact, hydrophobic and hydrophilic interactions might be more popular in CCC separation than that in the liquid chromatography. For example, adding small solvents or additives in two-phase solvent systems may change significantly hydrophobic or hydrophilic interactions between solvents and solutes. Normally, CCC separation employs a light and hydrophobic organic phase as the stationary phase, and a heavy and hydrophilic aqueous phase as the mobile phase. Hydrophobic interactions between the solvent system and solutes (targets) will increase the partition coefficients (K values) of solutes and lengthen retention time, while hydrophilic interactions will reduce the K values and separation time. In this work, we aim to provide a general insight on the hydrophobic and hydrophilic interactions in CCC separation. We also highlight the current advances in utilizing the hydrophobic and hydrophilic interactions for K-targeted CCC separation and purification.

Abstract:Objective: To introduce a new way, which facilitates the systematic search and evaluation of the solvent systems suitable for high-speed countercurrent chromatography (HSCCC). Methods: The software integrated with the thermodynamics solution functions and theory is directive to calculate the composition, physical parameters, such as volume, viscosity, dielectric constants, and polarity parameters of the upper and lower phases. Results: Separation of Guizhi-decoction Fr. A by HSCCC, one of traditional Chinese medcines, is conducted to prove the applicability of this software in selection and optimization of solvent systems. Conclusion: This software can be used to select and optimize the solvent systems of HSCCC. It is supposed to extend this method.
Separation of six antioxidants from Hypsizygus marmoreus by high-speed countercurrent chromatography utilizing an approach based upon the polarity parameter model

Abstract:For a successful high-speed countercurrent chromatography (HSCCC) separation of multiple components, the suitable solvent system selection is a critical operation. Despite the difference in separation mechanism between HSCCC and HPLC, K values of compounds in the solvent system and the retention factor of compounds in HPLC were connected with polarity, and the polarity mainly depended on the structure and chemical properties of compounds. On the basis of the concept of "like dissolves like", the average polarity of the solvent system is equal to the average polarity of multiple components at the "sweet point" log K = 0. The result of theoretical deduction showed a negative linear correlation between the polarity of each component and the logarithm of the gradient range of the organic phase. Therefore, the average polarity of unknown multiple components could be obtained through the polarity parameter model established in the HPLC analysis. The suitable solvent system composed of n-hexane-ethyl acetate-methanol-water (8:3:8:1, v/v) was selected through the average polarity of multi-components combining K values of the enriched samples. In the context of bioassay, an efficient HSCCC separation procedure was established, and two groups of analogue benzaldehyde derivatives (four main antioxidants, namely, isodihydroauroglaucin, isoaspergin, isotetrahydro-auroglaucin, and flavoglaucin; two minor antioxidants, namely, 6'-oxo-chaetopyranin and chaetopyranin) were obtained from Hypsizygus marmoreus. The predicted polarity values of multi-components were sufficient to meet the HSCCC experimental requirements. The HPLC analysis of reference compounds and multi-components showed a significant consistency with different chromatographic columns. Therefore, the polarity parameter model established in the HPLC analysis was a simple, rapid, and helpful tool for looking an appropriate solvent system, which was a forwarding step for the HSCCC separation of multiple components.
