Abstract:Fresh Mentha haplocalyx is a well known traditional Chinese medicinal material (CMM) used in both China and America. This paper reports analysis of the volatile components of fresh Mentha haplocalyx by comprehensive two-dimensional gas chromatography (GCxGC) and high-resolution time-of-flight mass spectrometry (HR-TOF-MS), a combination that provides almost complete chemical separation with elemental composition determination of analytes. 163 ketones and terpenes, including menthol and menthone, were tentatively identified, including enantiomers. This study suggests that GCxGCxHR-TOF-MS is suitable for routine identification of target compounds and enantiomers in CMM.

Abstract:Traditional Chinese medicines (TCMs) have a long history dating back thousands of years. Recently, there has been increasing interest worldwide in the use of TCMs for the prevention and treatment of various illnesses. In China, a large number of analytical tools, especially chromatographic techniques have been used to analyze the constituents of TCMs in order to control their quality and discover new bioactive compounds. In this paper, recent developments in sample preparation techniques for the extraction, clean-up, and concentration of analytes from TCMs are compared. These techniques include headspace solid-phase microextraction (HS-SPME), headspace liquid-phase microextraction (HS-LPME), microwave-assisted extraction (MAE), supercritical-fluid extraction (SFE), pressurized-liquid extraction (PLE), and microwave distillation (MD).

Abstract:Sample preparation is a major task in a regulated bioanalytical laboratory. The sample preparation procedure significantly impacts assay throughput, data quality, analysis cost, and employee satisfaction. Therefore, selecting and optimizing an appropriate sample preparation method is essential for successful method development. Because of our recent expertise, this article is focused on sample preparation for high‐performance liquid chromatography with mass spectrometric detection. Liquid chromatography with mass spectrometric detection (LC‐MS) is the most common detection technique for small molecules used in regulated bioanalytical laboratories. The sample preparation technologies discussed are pre‐extraction and post‐extraction sample processing, protein precipitation (PPT), liquid–liquid extraction (LLE), offline solid‐phase extraction (SPE), and online solid‐phase extraction. Since all these techniques were in use for more than two decades, numerous applications and variations exist for each technique. We will not attempt to categorize each variation. Rather, the development history, a brief theoretical background, and selected references are presented. The strengths and the limitations of each method are discussed, including the throughput improvement potential. Abstract:Envenomation by poisonous animals is a neglected condition according to the World Health Organization (WHO). Antivenoms are included in the WHO Essential Medicines List. It has been assumed that immunoglobulin G (IgG) antivenoms could activate the complement system through Fc and induce early adverse reactions (EARs). However, data in the literature indicate that F(ab')2 fragments can also activate the complement system. Herein, we show that several batches of IgG and F(ab')2 antivenoms from the Butantan, Vital Brazil, and Clodomiro Picado Institutes activated the complement classical pathway and induced the production of C3a; however, only those antivenoms from Clodomiro Picado generated C5a. Different protein profiles (IgG heavy chain, protein contaminants, and aggregates) were observed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot analyses. Our results show that various antivenoms from different producers are able to activate the classical pathway of the complement system and generate anaphylatoxins, and these findings suggest that factors, such as composition, contaminant proteins, and aggregates, may influence the anticomplementary activity of antivenoms in vitro. Therefore, there is a need to further improve antivenom production methods to reduce their anticomplementary activity and potential to cause EARs.

Abstract:The importance of sample preparation methods as the first stage in an analytical procedure is emphasised and examined. Examples are given of the extraction and concentration of analytes from solid, liquid and gas phase matrices, including solvent phase extractions, such as supercritical fluids and superheated water extraction, solid-phase extraction and solid-phase microextraction, headspace analysis and vapour trapping. The potential role of selective extraction methods, including molecular imprinted phases and affinity columns, are considered. For problem samples alternative approaches, such as derivatisation are discussed, and potential new approaches minimising sample preparation are noted.

Abstract:The complete sequence of the horse pancreatic lipase was elucidated by combining polypeptide chain and cDNA sequencing. Among the structural features of horse lipase, it is worth mentioning that Lys373 is not conserved. This residue, which is present in human, porcine and canine lipases, has been assumed to be involved in p-nitrophenyl acetate hydrolysis by pancreatic lipases. Kinetic investigation of the p-nitrophenyl acetate hydrolysis by the various pancreatic lipases and by the C-terminal domain (336-449) of human lipase reveals that this hydrolysis is the result of the superimposition of independent events; a specific linear hydrolysis occurring at the active site of lipase, a fast acylation depending on the presence of Lys373 and a non-specific hydrolysis most likely occurring in the C-terminal domain of the enzyme. This finding definitely proves that pancreatic lipase bears only one active site and raises the question of a covalent catalysis by pancreatic lipases. Moreover, based on sequence comparison with the above-mentioned pancreatic lipases, three residues located in the C-terminal domain, Lys349, Lys398 and Lys419, are proposed as possible candidates for lipase/colipase binding.

Abstract:Although hundreds of papers related to preparative gas chromatography (pGC) have been published since the late 1950s, the success of the GC technique has largely been associated with analytical instead of preparative purposes. Actually, pGC is an ideal alternative technique for the preparation of pure substances, especially volatile compounds. This paper reviews the papers (written in English) associated with pGC published over the period from the 1950s to the 2010s. For large scale preparation, large sample injection and vaporization, a high loading capacity column, a gas splitter at the end of the column and a special collecting device are fundamentally important for a pGC system. The primary components of pGC system, including injector, column, splitter, detector and collection traps, are briefly introduced. Furthermore, the applications of pGC in the separation and purification of volatile compounds from natural essential oils, in addition to the purification of isotopes, isomers and enantiomers are summarized.

Abstract:Sample preparation procedures in use in many application areas are still tedious and manually intensive protocols. These characteristics mean that sample treatment is considered the most time-consuming and error-prone part of the analytical scheme. The increasing demand for faster, more cost-effective and environmental friendly analytical methods is a major incentive to improve these conventional procedures and has spurred research in this field during the last decades. This review provides an overview of the most relevant developments and successful approaches proposed in recent years concerning sample preparation. The current state-of-the-art is discussed on the basis of examples selected from representative application areas and involving conventional instrumental techniques for the final determination of the target compounds. Emphasis will be on those techniques and approaches that have already demonstrated their practicality by the analysis of real-life samples, and in particular on those dealing with the determination of minor organic components. The potential of the latest developments in this field for sample treatment simplification and complete hyphenation and integration of analytical process is discussed and the most pressing remaining limitations evaluated.

Abstract:Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) has emerged as a robust analytical technique for the rapid and sensitive analysis of diverse biomolecules. MALDI-MS effectiveness relies on meticulous sample preparation, with a particular emphasis on achieving a uniform sample surface. Although the dried droplet (DD) method is widely utilized, it encounters challenges such as uneven sample distribution that impacts the ionization process and significantly varies ion signals across sample areas, resulting in poor data reproducibility. The uneven sample distribution can increase the time to finish mass analysis of samples and make automated analysis complicated. Automated sample deposition systems and alternative MALDI sample preparation methods show promise in overcoming the limitations of the DD method, resulting in improved reproducibility and mass spectral quality. These advancements are critical in improving the precision and reliability of quantitative measurements using MALDI-MS, propelling progress in MALDI imaging, quantitative analysis, and biomarker discovery across a wide range of scientific applications.

Abstract:The use of cosmetics and personal care products is increasing worldwide. Their high matrix complexity, together with the wide range of products currently marketed under different forms imply a challenge for their analysis, most of them requiring a sample pre-treatment step before analysis. Classical sample preparation methodologies involve large amounts of organic solvents as well as multiple steps resulting in large time consumption. Therefore, in recent years, the trends have been moved towards the development of simple, sustainable, and environmentally friendly methodologies in two ways: (i) the miniaturization of conventional procedures allowing a reduction in the consumption of solvents and reagents; and (ii) the development and application of sorbent- and liquid-based microextraction technologies to obtain a high analyte enrichment, avoiding or significantly reducing the use of organic solvents. This review provides an overview of analytical methodology during the last ten years, placing special emphasis on sample preparation to analyse cosmetics and personal care products. The use of liquid–liquid and solid–liquid extraction (LLE, SLE), ultrasound-assisted extraction (UAE), solid-phase extraction (SPE), pressurized liquid extraction (PLE), matrix solid-phase extraction (MSPD), and liquid- and sorbent-based microextraction techniques will be reviewed. The most recent advances and future trends including the development of new materials and green solvents will be also addressed.

Abstract:Sample preparation for headspace analysis of volatile organic compounds (VOCs) by gas chromatography (GC) is well understood. Newer direct‐injection mass spectrometry (DIMS) techniques have found application in headspace analysis, but with some necessary modifications due to (i) the removal of the chromatographic column, and (ii) the soft chemical ionization that is used in these techniques. These factors place important constraints on the total load of VOCs—whether of solvents, analytes, or other VOCs in the matrix. This article summarizes the basic differences between DIMS and GC approaches, and describes the strategies by which headspace-DIMS can be successfully adopted.

Abstract:Sample pretreatment is one of the key steps in sample analysis. The design and development of new materials promote advancements in sample pretreatment technology. Deep eutectic solvents (DESs) are a novel material that have been developed in recent years. They possess characteristics such as low toxicity, good thermal stability, simple preparation methods, and low cost. DESs have the potential to replace traditional organic extraction solvents. DESs are formed from a hydrogen bond donor (HBD) and acceptor (HBA). Changing the type of HBA and HBD or their ratio leads to variations in the structure and properties of the resulting DESs. Gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) are the primary analytical techniques used in laboratories. This paper analyzes the selection relationship between DESs and analytes, as well as the steps of sample pretreatment, based on the characteristics of GC instruments, and utilizing DES extractants and extraction materials for sample pretreatment. This paper summarizes the progress of DES-based microextraction methods for GC. It introduces the different classifications of liquid and solid-phase microextraction and the application of DESs in them. The theoretical mechanism and extraction/separation mechanism of DESs are analyzed, and potential application of DESs in extraction/separation technology is discussed.

Abstract:The choice of sample-preparation method is extremely important in metabolomic studies because it affects both the observed metabolite content and biological interpretation of the data. An ideal sample-preparation method for global metabolomics should (i) be as non-selective as possible to ensure adequate depth of metabolite coverage; (ii) be simple and fast to prevent metabolite loss and/or degradation during the preparation procedure and enable high-throughput; (iii) be reproducible; and (iv) incorporate a metabolism-quenching step to represent true metabolome composition at the time of sampling. Despite its importance, sample preparation is often an overlooked aspect of metabolomics, so the focus of this review is to explore the role, challenges, and trends in sample preparation specifically within the context of global metabolomics by liquid chromatography–mass spectrometry (LC–MS). This review will cover the most common methods including solvent precipitation and extraction, solid-phase extraction and ultrafiltration, and discuss how to improve analytical quality and metabolite coverage in metabolomic studies of biofluids, tissues, and mammalian cells. Recent developments in this field will also be critically examined, including in vivo methods, turbulent-flow chromatography, and dried blood spot sampling.

Abstract:This review summarizes literature data from the past 5 years on new developments and/or applications of sample preparation methods for analysis of volatile organic compounds (VOC), mainly in air and water matrices. Novel trends in the optimization and application of well-established airborne VOC enrichment techniques are discussed, like the implementation of advanced cooling systems in cryogenic trapping and miniaturization in adsorptive enrichment techniques. Next, focus is put on current tendencies in integrated sampling-extraction-sample introduction methods such as solid phase microextraction (SPME) and novel in-needle trapping devices. Particular attention is paid to emerging membrane extraction techniques such as membrane inlet mass spectrometry (MIMS) and membrane extraction with a sorbent interface (MESI). For VOC enrichment out of water, recent evolutions in direct aqueous injection (DAI) and liquid-liquid extraction (LLE) are highlighted, with main focus on miniaturized solvent extraction methods such as single drop microextraction (SDME) and liquid phase microextraction (LPME). Next, solvent-free sorptive enrichment receives major attention, with particular interest for innovative techniques such as stir bar sorptive extraction (SBSE) and solid phase dynamic extraction (SPDE). Finally, recent trends in membrane extraction are reviewed. Applications in both immersion and headspace mode are discussed.

Abstract:Metabolomics aims to analyze global intracellular metabolites of organisms. To study the intracellular metabolites of poly-γ-glutamate-producing Bacillus licheniformis, the quenching and extraction conditions were assessed and optimized. It indicated that perchloric acid was an appropriate quenching solution for B. licheniformis by measuring livability of cells, leakage of ATP, energy charge and intracellular metabolites. 0.85% NaCl was an appropriate washing solvent for a sample because it did not lead to serious leakage and would not affect operation of GC–MS. Among the four different extraction methods (cold pure methanol, PM; hot ethanol/water (75:25v/v), HE; cold methanol/water (50:50v/v), MW; and cold chloroform/methanol/water (50:25:25v/v/v), CMW), HE was superior to others on the basis of the energy charge and intracellular metabolites, which could effectively inhibit enzymatic activities and extract more intracellular metabolites. The method could obtain some metabolites which were involved in the most important synthesis pathways of poly-γ-glutamate, including glycolysis, citric acid cycle and glutamate metabolism. It is the first evaluation of a sample preparation of poly-γ-glutamate-producing bacteria, which might be used as one model for the preparation of polymer-producing samples for metabolomics analysis.

Abstract:Food safety and quality have been gaining increasing attention in recent years. Gas chromatography coupled to tandem mass spectrometry (GC–MS/MS), a highly sensitive technique, is gradually being preferred to GC–MS in food safety laboratories since it provides a greater degree of separation on contaminants. In the analysis of food contaminants, sample preparation steps are crucial. The extraction of multiple target analytes simultaneously has become a new trend. Thus, multi-residue analytical methods, such as QuEChERs and adsorption extraction, are fast, simple, cheap, effective, robust, and safe. The number of microorganic contaminants has been increasing worldwide in recent years and are considered contaminants of emerging concern. High separation in MS/MS might be, in certain cases, favored to sample preparation selectivity. The ideal sample extraction procedure and purification method should take into account the contaminants of interest. Moreover, these methods should cooperate with high-resolution MS, and other sensitive full scan MSs that can produce a more comprehensive detection of contaminants in foods. In this review, we discuss the most recent trends in preparation methods for highly effective detection and analysis of food contaminants, which can be considered tools in the control of food quality and safety.

Abstract:Accurate determination of elemental and isotopic compositions of geological samples must be a pre-requisite for successful mineral exploration, basic geochemical and geochronological studies. During rock analysis, sample digestion which involves steps from simple dilution to partial or total digestions, is a crucial step and is the limiting factor many times for achieving reproducible and accurate analytical results. Sample preparation is a critical step in the geochemical analysis, and complete sample dissolution is a pre-requisite for obtaining accurate and precise data for geological materials in most studies. Sample matrix, analytes of interest, the analytical technique, and intended purpose play a major role in selecting the optimum sample preparation method. This review focuses on the recent advances, developments, and trends in sample digestion methods, such as open-vessel acid digestion, closed digestions, microwave digestion, alkali fusion, use of NH4F for both open and closed digestions, sintering with Na2O2, fire-assay methods, infrared radiation digestions, high-pressure asher digestions, and automated sample preparation systems (based on artificial intelligence) along with examples of some important and exciting applications in geochemical and geochronological studies. Various sample decomposition methods have been compared to bring out advantages and limitations for a particular application.

Abstract:Handling and processing of mu l and sub-mu l sized samples are challenging, and have a series of attendant problems, such as loss of sample on the walls of transfer devices, loss by evaporation, lass of components of the sample during manipulation and processing, and obtaining a representative sample from an nonhomogeneous specimen. A. further difficulty is that the expected concentration of the analyte restricts the scale of miniaturization of the sample. Detection of rare cell types (e. g., cancerous cells, fetal cells in maternal circulation, assessment of minimal residual disease) and microorganisms is problematic, and the expected cell frequency or microbial load will determine a sample size compatible with detection. In such cases a specific selection or a preconcentration step is a more logical approach to ensure the presence of the desired cells or microorganisms. In this review the principles and applications of the current range of microfluidic (e.g., pumps, valves) and microfiltration systems (e.g., silicon microfilters) and electronically controlled microstructured devices relevant to the micro-sample preparation are critically discussed.
