Abstract:Inter-simple sequence repeat (ISSR) marker technology has become a hotspot in DNA molecular technology in recent years, and plays an important role in genetic research. However, this technology is still at an initial stage in animal research, compared with its applications in plant research. This paper introduced the principles and main methods of ISSR marker technology, and summarized its applications in analyzing the genetic structure of animals, such as genetic diversity, resources identification, and genetic relationship, etc. The application prospects of this technology were also discussed, aimed to provide reference for the further applications of this technology in animal research.

Abstract:Key message Plenty of molecular markers have been developed by contemporary sequencing technologies, whereas few of them are successfully applied in breeding, thus we present a review on how sequencing can facilitate marker-assisted selection in plant breeding. Abstract The growing global population and shrinking arable land area require efficient plant breeding. Novel strategies assisted by certain markers have proven effective for genetic gains. Fortunately, cutting-edge sequencing technologies bring us a deluge of genomes and genetic variations, enlightening the potential of marker development. However, a large gap still exists between the potential of molecular markers and actual plant breeding practices. In this review, we discuss marker-assisted breeding from a historical perspective, describe the road from crop sequencing to breeding, and highlight how sequencing facilitates the application of markers in breeding practice.

Abstract:With some technological breakthroughs such as sequencing and microarray in the recent years, genomics, transcriptomics, proteomics, metabolomics and phenomics have made a great progress in the world. Theories and technologies of crop breeding have also been revolutionarized considerably. Traditional crop breeding is based on phenotypic selection, which is often called "empirical breeding" with poor predictability, long breeding cycle and low breeding efficiency. Modern crop molecular breeding including marker-assisted breeding, transgenic breeding and molecular design breeding has become an important direction in the field of plant breeding. It is based on the combination of genotypic selection and phenotypic selection and thus target genes/alleles can be directly selected and pyramided. Therefore, the efficiency of molecular breeding can be improved and the breeding duration can be shortened. The paper defines the concept of molecular breeding, briefly analyzes current status and issues of crop molecular breeding in China, and discusses strategies in the future.

Abstract:Plant improvement and resolving complex traits of plant need to understand the germplasm diversity and hereditary basis of traits.With the development of genomics and biostatistics software,especially the extensive application of molecular markers in plant genetics and breeding,association analysis based on the linkage disequi-librium (LD) offers a new method for genetic study of plants quantitative characters and a new idea for molecular design breeding.In this paper,the basic statistic principles,strategies,characteristics and current successful appli-cations of association analysis in plants were reviewed.The potential application of association mapping to molec-ular dissection of plant traits and molecular breeding were also discussed.

Abstract:With the development of molecular marker technology in the 1980s, the fate of plant breeding has changed. Different types of molecular markers have been developed and advancement in sequencing technologies has geared crop improvement. To explore the knowledge about molecular markers, several reviews have been published in the last three decades; however, all these reviews were meant for researchers with advanced knowledge of molecular genetics. This review is intended to be a synopsis of recent developments in molecular markers and their applications in plant breeding and is devoted to early researchers with a little or no knowledge of molecular markers. The progress made in molecular plant breeding, genetics, genomic selection and genome editing has contributed to a more comprehensive understanding of molecular markers and provided deeper insights into the diversity available for crops and greatly complemented breeding stratagems. Genotyping-by-sequencing and association mapping based on next-generation sequencing technologies have facilitated the identification of novel genetic markers for complex and unstructured populations. Altogether, the history, the types of markers, their application in plant sciences and breeding, and some recent advancements in genomic selection and genome editing are discussed.

Abstract:PCR-SSCP is a method which detects gene mutations.Since its inception,it has undergone its own development and improvement.Initially,it puts isotope into PCR amplification of objects,and then shows results by autoradiography.But it also takes the difficulties to the technology to extend.With the DNA silver staining method and PCR-SSCP combined,especially the application of the direct ethidium bromide-staining method,it makes PCR-SSCP greatly simplified,primitively operate,lessly limitate,low-requirement experimental conditions for general clinical laboratory.It is simple,rapid,sensitive in character.PCR-SSCP is not only used to the detection of gene mutation and short sequence deletion and insertion,but also used to animal breeding,microbes,parasite research fields.Because of the advantages of SSCP,it has been applied widely in recent years.

Abstract:The major approaches of molecular breeding are transgenic and marker-assisted selection breeding.In the present study,the biological base of molecular breeding of rice were stated from several aspects including yield,adaptability,quality,resistance and heterosis.Some important achievements were enumerated as molecular breeding of rice progressed.As a result of the sequencing of rice genome,map-based cloning of genes controlling the important agronomic traits,the gradually perfected transgenic technology,the rapid development of bioinformatics,and the development of software in simulating breeding,breeding by molecular design has increasingly become an important method of rice breeding.

Abstract:Molecular markers, due to their stability, cost-effectiveness and ease of use provide an immensely popular tool for a variety of applications including genome mapping, gene tagging, genetic diversity diversity, phylogenetic analysis and forensic investigations. In the last three decades, a number of molecular marker techniques have been developed and exploited worldwide in different systems. However, only a handful of these techniques, namely RFLPs, RAPDs, AFLPs, ISSRs, SSRs and SNPs have received global acceptance. A recent revolution in DNA sequencing techniques has taken the discovery and application of molecular markers to high-throughput and ultrahigh-throughput levels. Although, the choice of marker will obviously depend on the targeted use, microsatellites, SNPs and genotyping by sequencing (GBS) largely fulfill most of the user requirements. Further, modern transcriptomic and functional markers will lead the ventures onto high-density genetic map construction, identification of QTLs, breeding and conservation strategies in times to come in combination with other high throughput techniques. This review presents an overview of different marker technologies and their variants with a comparative account of their characteristic features and applications.

Abstract:The authors have reviewed one new kind of molecular marker in higher plants, functional markers (FMs), based on comparisons with random DNA markers (RDMs) and gene targeted markers (GTMs) that are usually applied in genetic analysis. FMs include direct functional marker (DFM) and indirect functional marker (IFM). FMs are derived from polymorphic sites within gene motifs causally involved in phenotypic trait variation, which exhibit dominant genetic characters. FMs are independent of molecular genetic mapping and may enhance the efficiency of marker-assisted selection in crops breeding.

Abstract:Molecular markers are an important tool for plant breeding. Since the 1980s, in response to the technology development, molecular marker approaches have been further diversified. The establishment of new-generation sequencing and high-throughput plant phenotyping has greatly decreased the time to genotype large numbers of individuals. For breeders who are not very familiar with molecular techniques and want to catch up with the advances in the field, this review offers basic knowledge. Each molecular marker technology has specific advantages as well as limitations. Molecular marker types, diversity studies, QTL mapping, associative mapping, marker-assisted backcrossing and genomic selection are explored. Marker application in plant breeding is also described. In the genome, molecular markers can detect the genetic architecture of a trait, but also identify candidate genes with an important role in plant breeding programs.

Abstract:There are many single nucleotide polymorphism (SNP) markers available, due to the improved high-throughput genotyping technology. Currently, in the genome research, haplotype analysis has been applied to the association analysis between the SNP loci and complex traits. Existing haplotyping methods include population-based methods, pooled DNA methods, and methods for family and pedigree data. Family and pedigree datasets are abundant and have unique advantages. Currently there are many haplotyping methods for family and pedigree data that have been developed. We review the haplotyping methods and the corresponding computer programs suitable for family and pedigree data, discuss their applications and limitation, and describe the challenges that remain to be addressed.

Genetics Selection Evolution

Abstract:During the last ten years, the use of molecular markers, revealing polymorphism at the DNA level, has been playing an increasing part in animal genetics studies. Amongst others, the microsatellite DNA marker has been the most widely used, due to its easy use by simple PCR, followed by a denaturing gel electrophoresis for allele size determination, and to the high degree of information provided by its large number of alleles per locus. Despite this, a new marker type, named SNP, for Single Nucleotide Polymorphism, is now on the scene and has gained high popularity, even though it is only a bi-allelic type of marker. In this review, we will discuss the reasons for this apparent step backwards, and the pertinence of the use of SNPs in animal genetics, in comparison with other marker types.

Abstract:Abstract Background DNA markers improved the productivity and accuracy of classical plant breeding by means of marker-assisted selection (MAS). The enormous number of quantitative trait loci (QTLs) mapping read for different plant species have given a plenitude of molecular marker-gene associations. Main body of the abstract In this review, we have discussed the positive aspects of molecular marker-assisted selection and its precise applications in plant breeding programmes. Molecular marker-assisted selection has considerably shortened the time for new crop varieties to be brought to the market. To explore the information about DNA markers, many reviews have been published in the last few decades; all these reviews were intended by plant breeders to obtain information on molecular genetics. In this review, we intended to be a synopsis of recent developments of DNA markers and their application in plant breeding programmes and devoted to early breeders with little or no knowledge about the DNA markers. The progress made in molecular plant breeding, plant genetics, genomics selection, and editing of genome contributed to the comprehensive understanding of DNA markers and provides several proofs on the genetic diversity available in crop plants and greatly complemented plant breeding devices. Short conclusion MAS has revolutionized the process of plant breeding with acceleration and accuracy, which is continuously empowering plant breeders around the world.

Because of its reliability and high reproducibility,this technique is widely applied in plant research,for instance,mapping of genetic loci and identifying genes. In the present paper,the principle procedure and application were described. Now this technique has been used in mapping of genetic loci in tea and has expansive prospect in tea breeding.

Abstract:Molecular markers have emerged as the most useful tools in assessment of genetic diversity and characterizing germplasm, for crop improvement. This review paper comprehensively analyzes the applications of molecular markers in diversity analysis and germplasm characterization. It underscores the significance of genetic diversity as the bedrock for plant breeding programs, enabling the development of improved varieties with desirable attributes such as higher yields, stress tolerance, and enhanced nutritional profiles. The paper provides an overview of various types of molecular markers, including hybridization-based markers (e.g., RFLPs) and PCR-based markers (e.g., RAPDs, AFLPs, SSRs, and SNPs). It discusses marker selection strategies, emphasizing the consideration of factors like polymorphism, informativeness, and the potential for multiplexing and high-throughput genotyping. Diversity analysis techniques, including principal component analysis (PCA), cluster analysis methods (UPGMA and Neighbor-Joining), and population structure analysis (model-based approaches like STRUCTURE), are detailed. These methods enable the assessment of genetic relationships, identification of subpopulations, and selection of diverse parents based on molecular marker data. The review further explores the applications of molecular markers in germplasm management, conservation, and utilization. Abstract:With recent developments in DNA technologies, a large number of genetic polymorphisms at DNA sequence level has been introduced over the last decades as named DNA-based markers. The discovery of new class of DNA profiling markers has facilitated the development of marker-based gene tags, mapbased cloning of livestock important genes, variability studies, phylogenetic analysis, synteny mapping, marker-assisted selection of favourable genotypes, etc. The most commonly used DNA-based markers have advantages over the traditional phenotypic and biochemical markers since they provide data that can be analyzed objectively. In this article the main applications of molecular markers in present-day breeding strategies for livestock improvement - parentage determination, genetic distance estimation, genetic diversity, gene mapping and marker-assisted selection have been reviewed.

Abstract:In recent years, great advances have been made in researches on major genes and relevant molecular markers affected lean meat percentage of domestic animals. This review presents the current status and prospects of four relevant genes, including obese gene, leptin receptor gene, insulin-like growth factor(IGF) gene and myostatin gene(GDF-8), and several other relevant genes, such as myogenin gene, pituitary-specific transcription factor I (PIT-I), hormone sensitive lipase(HSL) are also introduced briefly.
