Abstract:The toxical effects of nonylphenol ethoxylates and nonylphenol were studied with EROD in liver of carp as biomarker under external condition. These two tested substances all had activation on EROD enzyme in liver of carp. Under definite range of concentrations, with the affecting rule of inhibiting at high concentration and inducing at low concentration. The test time and the action temperature had greater influences on inducing EROD enzyme under external condition. More ideal test results would be obtained at 20 min and 20℃ of inducing. Inducing of fish liver EROD enzyme under external condition was certainly a good method to estimate the contaminative effect of chemical substances.
Effects of bisphenol A-related diphenylalkanes on vitellogenin production in male carp (Cyprinus carpio) hepatocytes and aromatase (CYP19) activity in human H295R adrenocortical carcinoma cells

Abstract:The present study investigated the effects of the known xenoestrogen bisphenol A (BPA) relative to eight BPA-related diphenylalkanes on estrogen receptor (ER)-mediated vitellogenin (vtg) production in hepatocytes from male carp (Cyprinus carpio), and on aromatase (CYP19) activity in the human adrenocortical H295R carcinoma cell line. Of the eight diphenylalkanes, only 4,4'-(hexafluoropropylidene)diphenol (BHF) and 2,2'-bis(4-hydroxy-3-methylphenyl)propane (BPRO) induced vtg, i.e., to a maximum of 3% to 4% (at 100 microM) compared with 8% for BPA relative to the maximum induction by 17beta-estradiol (E2, 1 microM). Bisphenol A diglycidyl ether (BADGE) was a potent antagonist of vtg production with an IC50 of 5.5 microM, virtually 100% inhibition of vtg at 20 microM, and an inhibitive (IC50) potency about one-tenth that of the known ER antagonist tamoxifen (IC50, 0.6 microM). 2,2'-Diallyl bisphenol A, 4,4'-(1,4-phenylene-diisopropylidene)bisphenol, BPRO, and BHF were much less inhibitory with IC50 concentrations of 20-70 microM, and relative potencies of 0.03 and 0.009 with tamoxifen. Bisphenol ethoxylate showed no anti-estrogenicity (up to 100 microM), and 4,4'-isopropylidene-diphenol diacetate was only antagonistic at 100 microM. When comparing the (anti)estrogenic potencies of these bisphenol A analogues/diphenylalkanes, anti-estrogenicity occurred at lower concentrations than estrogenicity. 4,4'-Isopropylidenebis(2,6-dimethylphenol) (IC50, 2.0 microM) reduced E2-induced (EC50, 100 nM) vtg production due to concentration-dependent cytotoxicity as indicated by a parallel decrease in MTT activity and vtg, whereas the remaining diphenylalkanes did not cause any cytotoxicity relative to controls. None of the diphenylalkanes (up to 100 microM) induced EROD activity indicating that concentration-dependent, CYP1A enzyme-mediated metabolism of E2, or any Ah-receptor-mediated interaction with the ER, was not a likely explanation for the observed anti-estrogenic effects. At concentrations as great as 100 microM, none of the diphenylalkanes directly inhibited aromatase (CYP19) activity in H295R cells. Environmental exposure of fish to BPA and related diphenylalkanes, depending on the structure, may pose anti-estrogenic, and to a lesser extent estrogenic, risks to development and reproduction.
Enzymatic Biomarker Measurement and Study on Pollution-induced Antioxidant Enzymes Responses in Freshwater Fish Liver, Brocarded Carp

Abstract:The acute toxicity of M-DNB (m-dinitrobenzene) to Brocarded Carp and antioxidant enzyme activities in liver of fish were investigated. The acute toxicity test showed that 24h, 48h, 72h, 96h value of median lethal concentration (LC50) at 20-22 square were 27.19, 20.65, 16.17 and 14.04mg/L, respectively. Fish was exposed to different concentrations (1/6LC50, 1/4LC50, 1/3LC50, 1/2LC50) and killed at 6, 12,18, 24 and 30 day after exposure. The activities of Superoxide dismutase (SOD), Catalase (CAT) and Glutathione peroxidase (GPx) in liver tissue were analyzed. The results indicated the activities of SOD, GPx were induced in all treatment groups at post-exposure, they were higher than those of the control group (p < 0.05). The activity of CAT was significantly induced except for I/6LC50 treatment group. Moreover, the SOD and GPx activities of the control group were stable, except that the CAT activity increased 1-fold. The results demonstrated that the activities of liver antioxidant enzyme in Brocarded Carp might be the promising biomarker of M-DNB exposure. Therefore it was suggested that SOD, GPx and CAT could be used as biomarker of nitroaromatics compounds, when their pollutions affected living beings in aquatic environment. But, the order of the sensibility and stability for 3 kinds antioxidant enzymes was GPx>SOD>CAT.
The Inhibitory Effect Of Typical Brominated Flame Retardants On Cytochrome P450 1a Enzymes In Fish Liver Microsomes

Abstract:Brominated flame retardants (BFRs) are a class of persistent organic pollutants, which are ubiquitous in the environment. Their metabolism in the organism liver is always catalyzed by Cytochrome P450 1A (CYP1A). The in vitro metabolism of 4,4'-dibrominated diphenyl ethers (BDE 15), 2,2', 4,4'-tetrabrominated diphenyl ethers (BDE 47), tetrabromobisphenol A (TBBPA) in crucian carp (Carassius auratus) liver microsomes as well as the inhibition of three compounds on the fish liver microsomal CYP1A were studied. The results showed that BDE 15, BDE 47 and TBBPA were able to be metabolized in fish liver microsmes, the metabolism rate in 6 hours was 14%, 8% and 40%, respectively. BDE 15 and BDE 47 inhibited crucian carp hepatic CYP1A-catalyzed ethoxyresorufin O-demethylase (EROD) activity, while TBBPA didn't have inhibition effect on EROD activity. The IC50 of the BDE 15 and BDE 47 was 10.45 and 57.30 mu M, respectively. The K-i of two compounds were 11.43 and 22.20 mu M, respectively. BDE 15 and BDE 47 were determined to be the competitive type inhibitor of EROD activity. It is indicated that BDE 15 and BDE 47 could combine with CYP1A active site, and be catalyzed by CYP1A. This competitive inhibition also may lead to unexpected toxicological interactions.
Risk Assessment of Fenpropathrin: Cause Hepatotoxicity and Nephrotoxicity in Common Carp (Cyprinus carpio L.)

Abstract:The synthetic pyrethroid pesticide fenpropathrin (FEN) is extensively used worldwide and has frequently been detected in biota and the environment, whilst the negative effects and toxicological mechanisms of FEN on non-target organisms are still unknown. In the present study, healthy immature common carp were treated with FEN (0.45 and 1.35 μg/L) for a duration of 14 days, and the negative impacts and possible mechanisms of FEN on fish were investigated. Biochemical analyses results showed that FEN exposure altered the levels of glucose (GLU), total cholesterol (T-CHO), triglyceride (TG), albumin (ALB), alkaline phosphatase (ALP), alanine transaminase (ALT), and aspartate transaminase (AST) in carp serum, and caused histological injury of the liver and kidney, indicating that FEN may cause hepatotoxicity and nephrotoxicity in carp. In addition, FEN also altered the activities of superoxide dismutase (SOD) and catalase (CAT) in carp serum, upregulated the levels of reactive oxygen species (ROS), and elevated the levels of malondialdehyde (MDA) in the liver and kidney. Meanwhile, tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) levels were also upregulated, indicating that oxidative stress and inflammatory reaction may be involved in the hepatotoxicity and nephrotoxicity caused by FEN. Furthermore, RNA-seq analysis results revealed that FEN treatment induced a diverse array of transcriptional changes in the liver and kidney and downregulated differentially expressed genes (DEGs) were concentrated in multiple pathways, especially cell cycle and DNA replication, suggesting that FEN may induce cell cycle arrest of hepatocytes and renal cells, subsequently inducing hepatotoxicity and nephrotoxicity. Overall, the present study enhances our comprehension of the toxic effects of FEN and provides empirical evidence to support the risk assessment of FEN for non-target organisms.

Abstract:Using brocaded carp (Cyprinus carprio) as test organism exposed to different concentrations of benzidine (6.0,3.0,2.0,1.2,0.6mg/L) for 14d, the influence on the activities of liver glutathione peroxidase (GSH-Px), catalase (CAT), superoxide dismutase (SOD), glutathione s-transferase (GST), and glutamic-pyruvate transaminase (GPT) were studied. Brocaded carp after benzidine exposure even low concentration exposure, the liver GSH-Px activity almost all was inhibited continuously; after the exposure of fourth day, appeared the inducement of CAT, but with continuing of exposure, in seventh and fourteenth days, appeared inhibiliting of enzyme activity; the influence on SOD appeared generally inducing action, but at the fourteen day of high concentration exposure (6.0, 3.0mg/L) appeared inhibiting of enzyme activity; the influence on GST and GPT appeared inducing action under low concentration and inhibiting action under high concentration. GSH-Px was most sensitive to benzidine exposure, possible forming the biomarker of evaluating the benzidine pollution in aquative environment.
Time-Dependent Changes Of Hepatic Enzyme Activities And Thyroid Hormones In Carassius Auratus Exposed To 2,2 ',4,4 '-Tetrabromodiphenyl Ether (Bde 47)

Abstract:As widely used flame retardants, polybrominated diphenyl ethers (PBDEs) are ubiquitous in the environment. The purpose of this study was to evaluate their potential toxicity on freshwater fish crucian carp (Carassius auratus). The fish were given intraperitoneal injection of 2, 2', 4, 4'- tetrabromodiphenyl ether (BDE47) at environmentally relevant concentrations for consecutive 7 days, followed by 7 days depuration period. Liver and serum samples were analyzed for activities ofhepatic ethoxyresorufin-O-deethylase (EROD), ethoxycoumarin-O-deethylase (ECOD), glutathione-S-transferase (GST), and levels of serum total thyroine (T-4), triiodothyronine (T-3). EROD activity was markedly inhibited, and a U-shaped trend with time was observed, while ECOD activity was significant induced, and the most marked induction (1.7 times) was detected in the low-dose group on the 12th day. Reductions of T-4 and T-3 were both noted after BDE47 exposure, and the most significant reduction was 58% and 54.9%, respectively. Correlation analysis revealed that there was an inverse correlation between ECOD activity and T-4 concentration, and a positive correlation between 14 and T-3 concentrations, which indicated that the decrease of thyroid hormone concentration may be attributed to the formation of metabolites through oxygenase catalized reaction involving CYP450. Histological changes on liver have also been detected, and the lesions could not be recovered in shot-time.

Abstract:Fenpropathrin (FEN) is an extensively utilized synthetic pyrethroid insecticide frequently found in aquatic ecosystems. However, the adverse effects and potential mechanisms of FEN on aquatic species are poorly understood. In this work, common carp were treated with FEN at concentrations of 0.45 and 1.35 μg/L FEN for 14 days, after which the tissue structure, physiological alterations, and mRNA transcriptome of the gills were evaluated. Specifically, FEN exposure caused pathological damage to the gills of carp, downregulated the levels of claudin-1, occludin, and zonula occluden-1 (ZO-1), and inhibited Na<sup>+</sup>-K<sup>+</sup>-ATPase activity in the gills. In addition, FEN exposure promoted an increase in reactive oxygen species (ROS) levels and significantly upregulated the levels of malondialdehyde (MDA), 8-hydroxy-2 deoxyguanosine (8-OHdG), and protein carbonyl (PC) in the gills. Moreover, the inflammation-related indices (TNF-α, IL-1β, and IFN-γ) and the apoptosis-related parameter caspase-3 were generally increased, especially in the 1.35 μg/L FEN group, and these indices were significantly greater than those in the control group. These findings suggest that FEN exposure can cause oxidative stress, the inflammatory response, and apoptosis in carp gills. Importantly, the results of RNA-seq analysis showed that 0.45 and 1.35 μg/L FEN could significantly interfere with multiple immune and metabolic pathways, including the phagosome, NOD-like receptor (NLR) signalling pathway, Toll-like receptor (TLR) signalling pathway, necroptosis, and arachidonic acid metabolism pathways, indicating that the effects of FEN on the gills of fish are intricate. In summary, our findings confirm the toxic effects of FEN on common carp gills and provide additional comprehensive information for evaluating the toxicity and underlying molecular mechanisms of FEN in aquatic organisms.

Abstract:Four chlorobenzenes (chlorobenzene, 1,3-dichlorobenzene, 1,4-dichlorobenzene, p-chloro-methylbenzene) were administrated to the crucian carps (Carassius auratus) by peritoneal injections in the laboratory for 30 days. Serum testosterone and 17 beta-estradiol concentrations were detected using radioimmunology assay (RIA), and the activities of two hepatic microsome enzymes, glutathione s-transferase (GST) and UDP-glucuronosyltransferase (UDPGT), were measured using the modified methods as described by Habig and Owens. Results showed that the four chlorobenzenes caused significant increases in serum testosterone concentration in the crucian carps (P < 0.05) compared to the controls, but they caused no significant effect on 17 beta-estradiol level. All test chemicals caused a change in hepatic GST activity in crucian carps, with significant increases in enzyme activity (P < 0.05). Chlorobenzene, 1,3-dichlorobenzene and p-chloro-methylbenzene resulted in a marked inhibition to UDPGT activity in crucian carp (P < 0.05) except 1,4-dichlorobenzene. The changes in hepatic microsome enzyme activities may have resulted in the alterations of serum sex steroids levels in the crucian carps. The results indicated that these four chlorobenzenes may result in the changes of endocrine functions and may affect the reproductive success of this and other species.

Abstract:Fishes have been widely used as a representative to estimate the health of an aquatic ecosystem. In the present study, Labeo rohita was selected for biomarker study against decabromodiphenyl ether (BDE-209), a persistent organic pollutant (POP), as it is a widely used Indian carp. The results suggested significant effects on the optimum metabolism of Labeo rohita . After 48 to 72 h of exposure, most of the biomarkers such as lactate dehydrogenase (LDH), creatine kinase (CK), serum glutamic pyruvic transaminase (SGPT), serum glutamic oxaloacetic transaminase (SGOT), and hepatosomatic index (HSI) increased drastically indicating the higher index of tissue and liver damage. On the contrary, succinic dehydrogenase (SDH), acetylcholinesterase (AChE), and alkaline phosphatase (ALP) showed a reverse trend suggesting the shifting of fish metabolism towards anaerobic respiration mode because of induced stress. Increased catalase (CAT) activity was also observed, which indicated increased abundance of reactive hydroxyl species and therefore a possible oxidative stress in fishes. It is further suggested to understand and examine the biotransformation characteristics and degradation pathways of polybrominated diphenyl ether (PBDE)s, which would be useful to comprehend their environmental fate.
Inhibitory Effect of Nitrobenzene on Oxygen Demand in Lake Sediments

Abstract:Nitrobenzene is an important raw material and product, which presents a heavy threat to the ecosystem. The potential impacts of nitrobenzene on sediment oxygen demand (SOD) were studied in lake sediment simulating reactors receiving relatively low inputs of nitrobenzene. Oxygen microprofiles were measured in these sediment reactors using microelectrodes. After an initial microprofile measurement as a control, nitrobenzene was added to the overlying water resulting in concentrations of 0, 50, 100, and 150 microg/L. Microprofiles were measured on day 1, 2, 4 and 7 following the addition of nitrobenzene. SODs were determined from the microprofiles using a reaction-diffusion model. Results showed that the SODs increased relative to the initial values measured in the pre-treatment period in reactors exposed to all nitrobenzene concentrations on day 1. However, the values decreased gradually on the following days, which eventually resulted in a 50% loss in SODs after 7 days of exposure to nitrobenzene in all reactors. In addition, the inhibition effect of nitrobenzene on SOD exhibited a weak relationship with its concentration. The microscopic observation and count of algae in the sediment showed that the exposure to nitrobenzene did not change the composition of algae greatly, however, it decreased the number of dominant algae species sharply after 7 days of exposure. These results suggested that nitrobenzene could significantly alter SOD in lakes, which could ultimately affect the pollutant recovery in aquatic-sediment systems.
Effects of polybrominated diphenyl ethers on basal and TCDD-induced ethoxyresorufin activity and cytochrome P450-1A1 expression in MCF-7, HepG2, and H4IIE cells

Abstract:Polybrominated diphenylethers (PBDEs) are used as additive flame-retardants in consumer products to reduce the chances of ignition and burning. Levels of certain PBDE congeners have been increasing in fish, wildlife, and human tissues during the last decades. Some PBDEs are lipophilic and persistent, resulting in bioaccumulation in the environment. The structural similarity of PBDEs to other polyhalogenated aromatic hydrocarbons such as PCBs, has raised concerns that PBDEs might act as agonists for the aryl hydrocarbon receptor (AhR). To study the possible AhR-mediated effects of the environmentally relevant PBDEs (BDE47, 77, 99, 100, 153, 154, 183, 209), the induction of cytochrome P450-1A1 (CYP1A1) was studied in human breast carcinoma (MCF-7), human hepatocellular carcinoma (HepG2), and rat hepatoma (H4IIE) cells. 7-Ethoxyresorufin-O-deethylase (EROD) was used as a marker for CYP1A1 activity. Cells were exposed for 72 h to various PBDE concentrations (0.01-10 microM). Positive controls were 2,3,7,8-TCDD (0.001-2.5 nM) and PCB126 (0.01-10 nM). None of these PBDEs was capable of inducing EROD activity; this was confirmed by real time RT-PCR for CYP1A1 mRNA. However, in cells exposed to PBDEs in combination with TCDD, a concentration-dependent decrease in TCDD-induced EROD activity occurred. Co-exposure of BDE153 (10 muM) and a maximally inducing concentration of TCDD (1 nM) reduced EROD activity to 49% of the maximum induction by TCDD alone. All tested PBDEs showed similar effects in each cell line, though quantitative differences were observed. The observed decrease in CYP1A1 activity was not due to PBDE-dependent catalytic inhibition of EROD activity or cytotoxicity, nor were decreased CYP1A1 mRNA levels observed. However, inhibition of luciferase induction in mouse (Hepa) and rat (H4IIE) hepatoma cells containing a stably transfected AhR-responsive luciferase reporter gene, suggests that BDE77 is a weak AhR antagonist or partial agonist.
Resveratrol improves emamectin benzoate-induced pyroptosis and inflammation of Ctenopharyngodon idellus hepatic cells by alleviating oxidative stress/endoplasmic reticulum stress

Abstract:Emamectin benzoate (EMB) is the most widely used pesticide in the world and contributes to water pollution. Owing to the lack of a specific antidote, EMB has a severe negative impact on the health of aquatic organisms. Resveratrol (RES), a substance with antioxidant capacity, is secreted by the fruits of many plants. This study was to explore the protection of RES against EMB-induced pyroptosis and inflammatory response in grass carp (Ctenopharyngodon idellus) hepatic liver (L8824) cells by oxidative stress/endoplasmic reticulum (ER) stress. The results showed that compared to the CON group, EMB induced oxidative stress in L8824 cells with the increase of reactive oxygen species (ROS), methane dicarboxylic aldehyde (MDA), and hydrogen peroxide (H2O2) contents and the decrease of total superoxide dismutase (t-sod) and glutathione peroxidase (gsh-px) activities (P < 0.05). In addition, EMB triggered ERS, increasing the relative mRNA expression of protein kinase R-like endoplasmic reticulum kinase (perk), inositol requiring enzyme 1 alpha (ire1α), glucose-regulated protein 78 (grp78), activating transcription factor 4 (atf4), activating transcription factor 6 (atf6), and CCAAT-enhancer-binding protein homologous protein (chop) and the protein expression of eukaryotic initiation factor 2α (eif2α), chop, atf6, and atf4. Meanwhile, EMB further induced pyroptosis by upregulating the mRNA and protein expression of nlrp3, aptamer protein (asc), caspase-1, gsdmd, interleukin-1β (il-1β), and interleukin-18 (il-18). EMB also induced inflammation in L8824 cells by increasing the mRNA expression of interleukin-2 (il-2), interleukin-6 (il-6), tumor necrosis factor-α (tnf-α), and ifn-γ and decreasing the content of interleukin-10 (il-10). However, compared to the EMB group, the oxidant indices and expression of genes related to ER stress, pyroptosis, and pro-inflammatory factors were significantly down-regulated (P < 0.05), whereas the antioxidant indicators and anti-inflammatory factor were significantly up-regulated in the EMB + RES group (P < 0.05). In conclusion, EMB caused hepatocytes pyroptosis and inflammation in grass carp, and RES could alleviate EMB-induced pyroptosis and inflammation in L8824 cells by ameliorating oxidative stress/ER stress.

Recentstudies have evidenced activation of some PFRs by human CYP enzymes (including CYP2E1) for genotoxic effects. However, the activity of CYPs in fish species toward PFR metabolism remains unclear. This study was aimed on comparing the metabolism of triphenyl phosphate (TPHP) and 4-OH-TPHP in human, rat, and common carp, and the involvement of human CYP2E1 and its orthologs in the metabolism, by using fomepizole (4-MP, CYP2E1 inhibitor) as a modulator, in silico molecular docking and dynamics analyses. The rate of TPHP metabolism was apparently faster with human and rat, microsomes than with fish microsomes, the major metabolites were phosphodiester and hydroxylated phosphate, with 30-80 % of TPHP forming unidentified metabolites in the system of each species. 4-OH-TPHP was readily metabolized by both human and rat microsomes, whereas it was hardly metabolized in carp assays. Meanwhile, with 4-MP the transformation of TPHP to 4-OH-TPHP was enhanced in the human/rat systems while suppressed in the carp system. Moreover, the formation of unidentified metabolites in human and rat systems was mostly inhibited by 4-MP. Through molecular dynamics analysis TPHP and its primary metabolites showed high affinity for human and rat CYP2E1, as well as the carp ortholog (CYP2G1-like enzyme), however, the 4-OH-TPHP bond to the latter was too far from the heme to permit a biochemical reaction. This study suggests that the metabolism/activation of TPHP might be favored in mammals rather than carp, a fish species.
Acute lethal toxicity of environmental pollutants to aquatic organisms

Abstract:The acute lethal toxicity of environment pollutants including chlorophenol, haloalkane, quinone, and substituted nitrobenzene (i.e., nitrophenol, nitrobenzene, nitrotoluene, and aniline) compounds to aquatic organisms was determined. Determination of toxicity of chemicals was performed with chlorella, daphnia, carp, and tilapia. The toxicity of chlorophenols had no relation to the number of chlorine atoms on the benzene ring, but monochlorophenol had lower activity than more chlorine-substituted compounds. The tolerance levels of daphnia and carp to haloalkanes was found to be higher than that of chlorella; toxicity to chlorella was several hundred times higher than to daphnia. The toxicity of naphthoquinone compounds to chlorella and carp was higher than that of anthraquinone. A compound with a monochloride substitution on anthraquinone ring was less toxic to carp than those substituted with amine, hydroxyl, and dichlorine groups. Nitrobenzene compounds with an additional substitution group on the p position were extremely toxic to daphnia and carp.
Synergistic Toxic Effects and Mechanistic Insights of Beta-cypermethrin and Pyraclostrobin Exposure on Hook Snout Carp ( Opsariichthys bidens ): A Biochemical, Transcriptional, and Molecular Approach

Abstract:The extensive utilization of pesticides results in their frequent detection in aquatic environments, often as complex mixtures, posing risks to aquatic organisms. The hook snout carp ( Opsariichthys bidens ) serves as a valuable bioindicator for evaluating the impacts of environmental pollutants in aquatic ecosystems. However, few studies examined the toxic effects of pesticides on O.bidens , let alone the characterization of the combined effects resulting from their mixtures. This study aims to elucidate the toxic effects of beta-cypermethrin and pyraclostrobin on O.bidens , individually and in combination, focusing on biochemical, transcriptional, and molecular responses. By organizing and analyzing the toxicogenomic databases, both pesticides were identified as a contributor to processes such as apoptosis, oxidative stress, and inflammatory responses. The acute toxicity test revealed comparable acute toxicity of beta-cypermethrin and pyraclostrobin on O.bidens , with LC 50 being 0.019 and 0.027 mg/L, respectively, whereas the LC 50 decreased to 0.0057 and 0.0079 mg/L under the combined exposure, indicating potential synergistic effects. The activities of enzymes involved in oxidative stress and detoxification were significantly altered after exposure, with superoxide dismutase (SOD) and catalase (CAT) increasing, while malondialdehyde (MDA) levels decreased. The activity of CYP450s was significantly changed. Likewise, the expression levels of genes ( mn-sod , p53 , esr , il-8 ) associated with oxidative stress, apoptosis, endocrine and immune systems were significantly increased. Combined exposure to the pesticides significantly exacerbated the aforementioned biological processes in O.bidens . Furthermore, both pesticides can modify protein activity by binding to the surface of SOD molecules and altering protein conformation, contributing to the elevated enzyme activity. Through the investigation of the synergistic toxic effects of pesticides and molecular mechanisms in O.bidens , our findings highlight the importance of assessing the combined effects of pesticide mixtures in aquatic environments.

Abstract:Polybrominated diphenyl ethers (PBDEs) and tetrabromobisphenol A (TBBPA) are two major flame retardants that accumulate in fish tissues and are potentially toxic. Their debrominated and oxidated derivatives were also reported in fish tissues although the sources of theses derivatives were unidentified. Our study was to determine whether PBDEs and TBBPA could be metabolized by fish liver subcellular fractions in vitro and to identify what types of metabolites were formed. Liver microsomes and S9 fractions of crucian carp (Carassius auratus) were exposed to 4,4'-dibromodiphenyl ether (BDE 15), 2,2',4,4'-tetrabromodiphenyl ether (BDE 47) or TBBPA solutions for 4h. Exposure of liver subcellular fractions to BDE 15 resulted in the formation of bromophenol and two monohydroxylated dibromodiphenyl ether metabolites. Neither in microsomes nor in S9 studies has revealed the presence of hydroxylated metabolites with BDE 47 exposure which indicated that the oxidation reactions in vitro were hindered by the increased number of bromine substituents on the PBDEs. TBBPA underwent an oxidative cleavage near the central carbon of the molecule, which led to the production of 2,6-dibromo-4-isopropyl-phenol and three unidentified metabolites. Another metabolite of TBBPA characterized as a hexa-brominated compound with three aromatic rings was also found in the liver subcellular fractions. These results suggest that the biotransformation of BDE 15 and TBBPA in fish liver is mediated by cytochrome P450 (CYP450) enzymes, as revealed by the formation of hydroxylated metabolites and oxidative bond cleavage products. Moreover, further studies on the identification of specific CYP450 isozymes involved in the biotransformation revealed that CYP1A was the major enzyme responsible for the biotransformation of BDE 15 and TBBPA in fish liver subcellular fractions and CYP3A4 also played a major role in metabolism of TBBPA.

Abstract:Cypermethrin (CYP, IUPAC name: [cyano-(3-phenoxyphenyl)methyl] 3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carboxylate) is a pyrethroid insecticide that poses a threat to the health of humans and aquatic animals due to its widespread use and environmental contamination. However, the mechanism of CYP on apoptosis, autophagy and inflammation in hepatocytes of carp (Cyprinus carpio) is unknown. We hypothesized that CYP caused damage to hepatocytes through the endoplasmic reticulum stress (ERS) pathway, CCK-8 was used to detect the toxic effects of different doses of CYP on hepatocytes, and finally low (L, 10 μM), medium (M, 40 μM), and high (H, 80 μM) doses of CYP was selected to construct the model. ROS staining, oxidative stress-related indices (MDA, CAT, T-AOC, SOD), AO/EB staining, MDC staining, and the expression levels of related genes were detected using qRT-PCR and western blot. Our results showed that CYP exposure resulted in an increase in ROS production, an increase in MDA content, and a decrease in the activity of CAT, SOD, and T-AOC in hepatocytes; the proportion of apoptotic, necrotic, and autophagic cells increased significantly in a dose-dependent manner. We also found that CYP exposure increased the expression levels of endoplasmic reticulum-related genes (GRP78, PERK, IRE-1, ATF-6 and CHOP), apoptosis (Bcl-2, Bax, Caspase-3, Caspase-9 and Cyt-c) and autophagy-related genes (LC3b, Beclin1 and P62) also showed dose-dependent changes, and the expression levels of inflammation-related genes (NF-κB, TNF-α, IL-1β, IL-6) were also significantly elevated. Thus, we demonstrated that CYP exposure caused apoptosis, autophagy and inflammation in hepatocytes via ERS-ROS-NF-κB axis. This research contributes to our understanding of the molecular mechanisms underlying CYP-induced damage in hepatocytes of carp (Cyprinus carpio).

Abstract:The 7-ethoxyresorufin-O-deethylase (EROD) activity was first time characterized in the neotropical fish Cnesterodon decemmaculatus as a biomarker for assessing environmental health in aquatic ecosystems of the Rio de la Plata Basin impacted by organic pollutants agonist of the aryl-hydrocarbon receptor (AhR). Both laboratory and field studies were conducted. Laboratory experiments were run using β-naphthoflavone (BNF) as an AhR agonist model. A clear concentration-response relationship was found between 1 and 100 μg/L, with a NOEC and LOEC of 1 and 10 μg/L. A fast time-dependent response was observed with a significant induction after 24 h and a plateau from 24 to 48 h up to 264 h of exposure. Differences in basal activity were found between juveniles, females, and males, but induction levels were similar. Both basal activities and induction levels were distinct in the whole body, liver, gill, muscle, brain, and embryos. Fold-change inductions in the respective tissues were: 20, 114, 3, 5, 1, and 14. Maternal transfer and early cyp1a activation were unveiled by embryonic induction. Clear differences in EROD activity were found among juveniles collected in hydrocarbon-polluted streams, beside the La Plata Petrochemical hub, and a reference stream. Similar EROD activities were observed in laboratory and feral fish, usually with values below or above 1,000 pmol/min x mg protein for unexposed or exposed organisms. The study contributes with original information about EROD activity in C. decemmaculatus that encourages the use of both the response as a robust biomarker of exposure and the species as a good sentinel organism to be included in surveillant programs for assessing aquatic pollution by AhR agonist chemicals within the Rio de la Plata Basin within the One Health paradigm.

Abstract:Cypermethrin is a pyrethroid insecticide used in aquaculture farms against ectoparasites. The conventional pesticide formulations are now being found to be greatly ineffective at low dosages, and their indiscriminate usage may cause undesired effects in ecosystems. So, the current study was designed to develop a nanoformulation of cypermethrin that would have many advantages over conventional pesticide formulations, and the toxic effects of this formulation were then tested on the carp Labeo rohita fingerlings. The 96-h LC 50 was estimated at 0.018 mg L −1 after the acute toxicity study. Chronic toxicity studies were carried out, exposing the fish to two sublethal doses via 1/10th (C1) and 1/50th (C2) of 96-h LC 50 for a period of 45 days. The nanoformulation exposure caused a significant reduction in the brain AChE enzyme activity. Catalase and glutathione-s-transferase enzyme activity in the gills and liver increased significantly, as did superoxide dismutase enzyme activity in the kidney. Serum total protein, triglycerides, cholesterol, and some serum enzyme activities decreased. In contrast, white blood cells, red blood cells, haemoglobin, and hematocrit levels decreased only in fish exposed to a higher dose. As baseline information, these findings may aid in understanding the toxic effect of nanoformulated cypermethrin on finfish.
