Agrobacterium tumefaciens-mediated Transformation of Rice with Insecticidal Bacillus thuringiensis Gene and Bioassay of the Transgenic Plants

Abstract:Immature embryos of rice varieties Xiushui 11 and Chunjiang 11 precultured for 4 d were infected and transformed by Agrobacterium tumefaciens strain LBA4404/pGBI4A2B (containing B.t . gene). The resistant calli were transferred onto the differentiation medium and regenerated transformed plants. The stable transformation efficiency reached 44%~70% measured as numbers of Geneticin (G418) resistant calli produced and 27%~64% measured as numbers of transgenic plants regenerated (Table 2). PCR and Southern blotting analysis of the total DNA extracted from transgenic plants indicated that the T DNA had been integrated into the rice genome (Figs.1,2). Bioassays using T 0 and T 1 generation transgenic plants indicated that the mortality of the leaffolder ( Cnaphalocrasis medinalis ) after 7 d of leaf feeding reached 31%~49% and the corrected mortality of the striped stem borer ( Chilo suppressalis ) after 7 d of leaf feeding reached 15%~71%(Tables 3,4). The insect bioassay (Plate Ⅰ 6~10) showed that the transgenic plants expressing the B.t . toxin protein were more resistant to these pests than wild type controls.
