Abstract:Bioenergy is of increasing interest as a renewable, environmentally friendly alternative to energy derived from fossil fuels. Through a variety of processes, biomass can be converted to solid, liquid, or gaseous biofuels. However, various challenges persist to the maintenance and development of biomass-to-energy utilization. This special issue focuses on the most recent advances in the frontier research of bioenergy and biomass utilization and features six selected papers with high quality. The paper titled “Nanofibrillated Cellulose and Copper Nanoparticles Embedded in Polyvinyl Alcohol Films for Antimicrobial Applications” by T. Zhong et al. develops a method to produce hybrids of TEMPO nanofibrillated cellulose (TNFC) and copper nanoparticles. The hybrid material is embedded in polyvinyl alcohol thermoplastic resin and the films are produced using a solvent casting method. The authors evaluate the films in terms of its morphology and thermal and antimicrobial properties and prove that TNFC-copper nanoparticles as antimicrobial nanofillers are valuable for PVA applications. The paper titled “Response of Arbuscular Mycorrhizal Fungi to Hydrologic Gradients in the Rhizosphere of Phragmites australis (Cav.) Trin ex. Steudel Growing in the Sun Island Wetland” by L. Wang et al. assesses the variations of hydrologic gradients in the relationships among AM fungi, reed, and rhizospheric microorganisms. The authors discover water content in soil and reed growth parameters are both positively associated with AM fungi colonization, but only the positive correlations between reed biomass parameters and the colonization could be expected, or both the host plant biomass and the AM fungi could be beneficial. This study could shed light on the mechanisms inside AM fungi-reed symbioses and would be referred to for optimizing the combined phytorhizoremediation. The paper titled “Production by Tobacco Transplastomic Plants of Recombinant Fungal and Bacterial Cell-Wall Degrading Enzymes to Be Used for Cellulosic Biomass Saccharification” by L. Paolo et al. presents work on saccharification of plant biomass. The authors express in tobacco chloroplasts microbial genes encoding five cellulases and a polygalacturonase. Leaf extracts containing the recombinant enzymes show the ability to degrade various cell-wall components under different conditions. In addition, a thermostable xylanase is also tested in combination with a cellulase and a polygalacturonase to study the cumulative effect on the depolymerization of a complex plant substrate. Results demonstrate the feasibility of using transplastomic tobacco leaf extracts to convert cell-wall polysaccharides into reducing sugars, fulfilling a major prerequisite of large scale availability of a variety of cell-wall degrading enzymes for biofuel industry. The paper titled “Enzymatic Saccharification of Lignocellulosic Residues by Cellulases Obtained from Solid State Fermentation Using Trichoderma viride” by T. Sartori et al. presents an approach that uses cellulolytic complex produced by Trichoderma viride in solid state fermentation to hydrolyze lignocellulosic residues. This produced enzyme shows viability in comparison with the commercial cellulase enzyme. The synthesis of cellulases by microorganisms from lignocellulosic residues is a process of great interest, representing the search for renewable sources to replace the fossil energetic matrix. The paper titled “Improving Biomethane Production and Mass Bioconversion of Corn Stover Anaerobic Digestion by Adding NaOH Pretreatment and Trace Elements” by C. Liu et al. describes an approach to improve biomethane production from corn stover. The authors use NaOH pretreatment and trace elements supplementation as a strategy to enhance the biodegradability of corn stover for biomethane production. The approach is compared with only NaOH-pretreated and untreated corn stover, and the paper highlights the supplementation of trace elements. The paper titled “Biochemical Modulation of Lipid Pathway in Microalgae Dunaliella sp. for Biodiesel Production,” by A. F. Talebi et al., explores the use of myoinositol to modulate LP and biodiesel quality. Inclusion of myoinositol in the media indeed improves the total lipid accumulation and biodiesel quality parameters, and this work highlights the fact that the biochemical modulation strategies should be progressively considered in the hope of finding more efficient and economically feasible strategies leading to more viable production systems.

Abstract:Astaxanthin is a highly valued carotenoid with growing market in aquaculture, food, cosmetic and pharmaceutical industries. The market demand for bio-derived astaxanthin and the insufficient efficiency of astaxanthin biosynthesis drive the advances in metabolic engineering of native astaxanthin producers and heterologous hosts. The recent research progress in engineering of astaxanthin biosynthesis in the red-pigmented heterobasidiomycetous yeast Xanthophyllomyces dendrorhous, the food-grade noncarotenogenic yeast Candida utilis, the oleaginous yeast Yarrowia lipolytica and the chassis organism Saccharomyces cerevisiae is reviewed, the advances in the development of pathway engineering tools are discussed, and future strategies for further improving biotechnological production of astaxanthin in yeasts are proposed.
Enhancing Sustainable Production of Biodiesel from Xanthoceras sorbifolia Bunge Oil Using Bio-Based Heterogeneous Catalyst

Abstract:The swift exhaustion of natural oil reserves and worsening environmental issues have prompted the quest for an economical method to produce biofuels. The superiority of heterogeneous catalysis promotes the development of bio-based catalysts. Carbon materials prepared from agricultural and forestry biomass waste have good application prospects in catalysis. In the present study, Xanthoceras sorbifolia shell waste was used as the raw material, Xanthoceras sorbifolia Bunge Carbon (XC) was used as the catalyst carrier, and K2CO3 was used as the activator to prepare a heterogeneous catalyst (KXC). The heterogeneous catalyst was characterized by scanning electron microscopy-energy dispersion X-ray spectroscopy (SEM-EDS), Fourier-transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), X-ray diffraction (XRD), and Brunauer–Emmett–Teller (BET) and X-ray photoelectron spectroscopy (XPS) analysis techniques to evaluate its chemical composition, structure, and physical morphology. EDS and XPS revealed the presence of K metal, which provided an alkaline site for the transesterification reaction to produce biodiesel. The biodiesel yield was observed by gas chromatography−mass spectrometry (GCMS). Under the reaction conditions of a methanol-to-oil molar ratio of 12:1, a reaction time of 90 min, a temperature of 65 °C, and a catalyst loading of 4 wt.% using 25KXC-600-4, the yield of biodiesel can reach 95.13 ± 0.82%. After being repeated five times, the yield was still 58.11 ± 3.80%. The catalyst has no waste generation, and has the characteristics of simple preparation and environmental friendliness, which make it a green heterogeneous catalyst for biodiesel production.

Abstract:Astaxanthin is a red xanthophyll with high economic and industrial value in the pharmaceutical, nutraceutical, cosmetic and food industries. In recent years, the biotechnological production of astaxanthin has attracted much attention as a sustainable alternative to the predominating petrochemical-dependent chemical synthesis. In this regard, Xanthophyllomyces dendrorhous is regarded as a promising microorganism for industrial production of astaxanthin. Unfortunately, biotechnological production of the carotenoid is currently expensive. The present study investigated soy molasses (SM) and residual brewers' yeast as cheap fermentation feedstocks for the cultivation of X. dendrorhous and astaxanthin production. Yeast extract was obtained from residual brewers' yeast using various techniques and then combined with SM to formulate a two-component growth medium which was subsequently used to cultivate X. dendrorhous. Generally, the yeast extract produced from residual brewers' yeast supported X. dendrorhous growth and astaxanthin production at levels comparable to those seen with commercial yeast extract. Overall, cultivating X. dendrorhous in an SM-based medium containing 5% SM and 0.2% yeast extract obtained from residual brewers' yeast resulted in significantly higher (> 20% more) biomass accumulation compared to the control media (YPD). A similar slightly higher astaxanthin output (up to 14% more) was recorded in the SM-based medium compared to YPD. The formulated cultivation medium in this study provides an opportunity to reduce the production cost of astaxanthin from X. dendrorhous while simultaneously reducing the environmental impact related to the disposal of the industrial waste used as feedstock. KEY POINTS: • Cheap culture media were formulated from soy molasses and brewers' spent yeast • The formulated medium resulted in at least 20% more biomass than the control • Up to 14% more astaxanthin was produced in molasses-based medium.
Genetic diversity of Xanthoceras sorbifolium bunge germplasm using morphological traits and microsatellite molecular markers

Abstract:Xanthoceras sorbifolium Bunge has great potential for producing biodiesel. In order to select and evaluate appropriate germplasm to produce biodiesel, we analyzed the genetic diversity of Xanthoceras sorbifolium Bunge germplasm based on morphological traits and simple sequence repeats (SSRs) in this study. Fifty-six germplasm samples were evaluated using nine morphological traits and 23 SSR loci. Significant differences among germplasms were observed in eight morphological characters. The SSR markers analysis showed high genetic diversity among the germplasms. All SSRs had polymorphisms, and we detected 77 alleles in total. The number of alleles at each locus ranged from two to six, averaging 3.35 per marker. The polymorphic information content values ranged from 0.36 to 0.61, averaging 0.49. Expected heterozygosity, observed heterozygosity, and Shannon's information index calculations detected large genetic variations among germplasms. The high average number of alleles per locus and the allelic diversity observed in the set of genotypes analyzed indicated that the genetic base of this species is relatively wide. Thus, microsatellite markers can be used to efficiently distinguish Xanthoceras sorbifolium Bunge germplasms and assess their genetic diversity. Hundred-grain weight and lateral diameter were positively correlated with monounsaturated fatty acids and depended on genotype. These results suggest that seeds with higher hundred-grain weight and lateral diameter could be more suitable to produce biodiesel. Our data will lay a foundation for selecting appropriate germplasm to produce biodiesel based on seed phenotype and will contribute to the conservation and management of this important plant genetic resource.
Potential of five plants growing on unproductive agricultural lands as biodiesel resources

Abstract:Fossil fuels are being heavily depleted due to increasing anthropogenic activities worldwide, and burning them contributes to global climate warming and air pollution. Vegetable oils are one of the main feedstocks for biodiesel: they are non-toxic and environmentally friendly. Rising global population, decreasing arable lands and a decline in crop yields from desertification and salinization demands that biodiesel feedstock be grown on unproductive agricultural lands. To estimate whether five plants growing on such land in China could be used as energy plants, we determined their seed oil content (SOC) and relative fatty acid content, and estimated the cetane number (CN) of the biodiesel produced from these plant oils by a fitted regression between different C18 fatty acids and CN. Results showed that four plants can be developed as energy plants, including Datura candida (SOC = 22.9%, CN = 50.8), Xanthium sibiricum (SOC = 41.9%, CN = 46.5), Kosteletzkya pentacarpos (SOC = 18.6%, CN = 45.9) and Hibiscus trionum (SOC = 17.5%, CN = 46.9). The fifth plant, Rhus typhina, was not adapted as an energy plant because of its low SOC, 9.7%. Our data provide a scientific basis for growing energy plants in unproductive agricultural lands as biodiesel resources.

Abstract:Xanthophyll is an oxidated version of carotenoid. It presents significant value to the pharmaceutical, food, and cosmetic industries due to its specific antioxidant activity and variety of colors. Chemical processing and conventional extraction from natural organisms are still the main sources of xanthophyll. However, the current industrial production model can no longer meet the demand for human health care, reducing petrochemical energy consumption and green sustainable development. With the swift development of genetic metabolic engineering, xanthophyll synthesis by the metabolic engineering of model microorganisms shows great application potential. At present, compared to carotenes such as lycopene and β-carotene, xanthophyll has a relatively low production in engineering microorganisms due to its stronger inherent antioxidation, relatively high polarity, and longer metabolic pathway. This review comprehensively summarized the progress in xanthophyll synthesis by the metabolic engineering of model microorganisms, described strategies to improve xanthophyll production in detail, and proposed the current challenges and future efforts needed to build commercialized xanthophyll-producing microorganisms.

Abstract:In the usage phase, diesel engines fuelled with diesel-biodiesel blends produced lower soot, HC and CO emissions. However, the environmental effects should include the greenhouse gas (GHG) emissions. GHG variations of biodiesel production from three generation feed-stocks were analyzed based on the land use change (LUC). Adequate land use change could keep the biodiversity and did not cause the increase of GHG emissions. The choice for feedstocks of biodiesel in China was comprehensively analyzed based on the land use type, the precipitation and the oil contents and the cultivation conditions of plants. Then, the suggestions for biodiesel development in China were given. Results showed that China should thoroughly abandon the first generation feedstocks due to the lack of arable land. Jatropha curcas (J. curcas), Pistacia chinensis Bunge (P. chinensis), Comus wilsoniana (C. wilsoniana) and Xanthoceras sorbifolium Bunge (X. sorbifolium) were considered as the most promising feedstocks for biodiesel production. It is suggested to plant X. sorbifolium in sand lands in the north and northwest of China with less natural rainfall and the others in south and southwest of China. The concern for microalgae should be transmitted to engineering microalgae cultivated in wastewater. The microalgae biodiesel refinery should be developed together with wastewater proposal industry and coal fired power plants. Lastly, only natural gas-based or biomass-based methanol can be used for biodiesel production. (C) 2020 Periodical Offices of Chang'an University. Abstract:Astaxanthin, a lipophilic carotenoid renowned for its strong antioxidant activity, holds significant commercial value across industries such as feed, food, and cosmetics. Although astaxanthin can be synthesized through chemical methods, it may contain toxic by-products in the synthesized astaxanthin, limiting its application in medicine or functional food. Natural astaxanthin can be extracted from algae, however, the cultivation cycle of algae is relatively longer compared to microorganisms. With the advancement of synthetic biology and metabolic engineering, the method of microbial fermentation has emerged as a promising strategy for the large-scale production of astaxanthin. This article provides a comprehensive overview of the research progress in astaxanthin biosynthesis, highlighting the use of the natural host Xanthophyllomyces dendrorhous , and the heterologous hosts Yarrowia lipolytica and Saccharomyces cerevisiae . Additionally, future research prospects are also discussed.

Transformation of corncob-derived xylose into intracellular lipid by engineered strain of Aureobasidium melanogenum P10 for biodiesel production

Abstract:Lignocellulosic materials such as corn cob are an inexpensive, natural and the most abundant renewable resource in the nature. In this study, it was found that an oleaginous yeast-like fungal strain Aureobasidium melanogenum P10 could grow well on corncob-derived xylose and accumulate 38.27% (w/w) of lipids from it within 120 h. Removal of an oxidoreductase pathway and overexpression of an isomerase pathway from A. melanogenum P10 made the XI-15 strain accumulate 53.74% (w/w) of lipids from the same xylose within 108 h. All the properties of the prepared biodiesel obtained from the crude lipids produced by both P10 and XI-15 strains could totally meet the requirements of US biodiesel ASTM D6751 and EU biodiesel EN 14214. Especially, the cetane numbers (56.82 ± 1.2 and 57.32 ± 1.3 min) of the prepared biodiesels were much higher than those of US biodiesel ASTM D6751 and EU biodiesel EN 14214 and their iodine numbers (80.20 ± 2.1 and 74.71 ± 1.9) were much lower than those of US biodiesel ASTM D6751 and EU biodiesel EN 14214. These meant that the properties of the prepared biodiesels in this study were better than those of the standard ones and the prepared biodiesels had high potential applications in renewable fuel industries. At the same time, the prepared biodiesel was found to be burnt well.

Research on Mechanical–Structural and Oil Yield Properties during Xanthoceras sorbifolium Seed Oil Extraction

Abstract:Products from Xanthoceras sorbifolium Bunge seed have gained extensive attention for various applications, especially in the fields of edible oils and industrial applications. In order to study seed kernel mechanical–structural behavior and oil yield mechanisms during extrusion, we set up a self-developed texture analyzer with in situ microscope observation. Test results indicated that seed kernel oil yield and pressing energy showed an approximately parabolic shape under pressing strain, and maximum oil yield reached 25.7%. Only local tissue damage occurred on seed kernels at strain 45–85%, cracks formed from the kernel edge to the inside zone and small cracks obviously increased in number, corresponding with the oil yield and energy–strain curve. The effect of speed on oil yield showed an opposite trend to strain effect; high pressing speed led to lower oil yield due to the short time for oil precipitation and lower pressing energy. Dwell time obviously promoted oil output within 600 s. Drying temperature had a negative effect due to structural change. Oil yield was almost zero at temperatures below 120 °C. The oil yield and pressing energy relation curve was obtained by polynomial fitting; optimal seed kernel oil pressing conditions were strain 95%, 0.1 mm/s, 20 °C, dwell time 600 s. The research provides in-depth theoretical guidance for Xanthoceras sorbifolium Bunge oil production.

Towards efficient bioethanol production from agricultural and forestry residues: Exploration of unique natural microorganisms in combination with advanced strain engineering

Abstract:Production of fuel ethanol from lignocellulosic feedstocks such as agricultural and forestry residues is receiving increasing attention due to the unsustainable supply of fossil fuels. Three key challenges include high cellulase production cost, toxicity of the cellulosic hydrolysate to microbial strains, and poor ability of fermenting microorganisms to utilize certain fermentable sugars in the hydrolysate. In this article, studies on searching of natural microbial strains for production of unique cellulase for biorefinery of agricultural and forestry wastes, as well as development of strains for improved cellulase production were reviewed. In addition, progress in the construction of yeast strains with improved stress tolerance and the capability to fully utilize xylose and glucose in the cellulosic hydrolysate was also summarized. With the superior microbial strains for high titer cellulase production and efficient utilization of all fermentable sugars in the hydrolysate, economic biofuels production from agricultural residues and forestry wastes can be realized.

Abstract:The significance of producing biodiesel from animal and vegetable oil to the sustainable economy was stressed. The properties and standard as well as research advances and application of biodiesel in the world were introduced, and the methods for production of biodiesel by transesterification including homogeneous chemical catalysts, heterogeneous chemical catalysts, biocatalyst reaction and supercritical technology were reviewed. Moreover, the future prospects of biodiesel in China were pointed out.

Abstract:Tremendous amounts of herbal extraction residues (HERs) derived from the traditional Chinese herb medicine industry are routinely generated annually. However, HERs are generally treated by landfill or incineration, resulting in a great waste of resources and serious environmental issues. Here, the residue of the production of Xuanmai Ganjie Capsule (XGCR) was described for lipid production. Part of the sugar polymers (44.0%) contained in the raw XGCR could be directly hydrolyzed using a fed-batch hydrolysis process at 54% solids loading. More interestingly, sugar yield approached the theoretical maximal value when the residue was pretreated by 1% (w/w) dilute sulfuric acid before enzymatic hydrolysis. Cutaneotrichosporon oleaginosum exhibited high oleaginicity with lipid titer, content, and yield of 8.5 g/L, 40.7%, and 80.2 g/kg of residue supplied, respectively. The fatty acid profiles and the prediction of biodiesel parameters indicated the microbial lipid prepared from the XGCR was a suitable feedstock for biodiesel industry. This study strongly supported that the XGCR-to-lipid route can be a promising way for turning HERs into treasure.

Abstract:Xanthoceras sorbifolium Bunge (Sapindaceae) is a native Chinese plant with promising applications as a biofuel feedstock and a source of novel drugs. Historical records and documents from different periods have mentioned the use of X. sorbifolium and its botanical constituents in treating diseases, highlighting its central role in Chinese and Mongolian traditional medicinal therapies. Phytochemical research has focused on the husks, leaves, trunks, and branches of this herb. A total of 278 chemical compounds have been isolated and divided into 8 categories: triterpenoids, flavonoids, phenylpropanoids, steroids, phenols, fatty acids, alkaloids, and quinones. Modern pharmacological studies on X. sorbifolium have demonstrated positive effects on learning and memory, as well as anti-inflammatory, anti-tumor, and anti-oxidative properties. This review provides a comprehensive analysis of the available research on X. sorbifolium , focusing on the relationship between chemical constituents, traditional uses, and pharmacological effects. We also assess the potential for therapeutic and other applications of this plant in support of further research and development of X. sorbifolium .

Abstract:Succinic acid has multiple practical applications (e.g. synthesis of 1,4-butanediol, tetrahydrofuran, gamma-butyrolactone, and as a monomer of some biodegradable polymers). Bio-based succinic acid is a potential substitute for current petrochemical production. Facing a shortage of crude oil supply and sharply rising oil prices, biological production of succinic acid from abundant and available biomass has become a topic of worldwide interest. Although great progress has been made in recent decades, much needs to be developed further in order to achieve economic viability. This paper reviews developments in technology and updates research progress of biosuccinate production, including pathways, micro-organisms, culture conditions, as well as integrated production with other high-value-added products. Abstract:Biodiesel has gained significant attention as an alternative to petroleum diesel. Currently, there is an increasing interest in obtaining microbial oils from oleaginous micro-organisms for biodiesel production. In the accumulation of microbial oils, carbon substrate constitutes a major portion of the cost. Accordingly, significant efforts have been made to explore suitable carbon sources, and to investigate their effects on cell growth as well as lipid synthesis. This paper reviews the current trend of using various carbon sources for microbial oils production, including carbon dioxide, conventional saccharides, energy crops, C2 compounds, lignocellulosic biomass, and glycerol. The strategies for enhancement of microbial oils production are further discussed. Finally, the constraints and perspectives of using microbial oils for biodiesel production are also highlighted. Abstract:A sustainable process has been developed for synthesis of biodiesel by utilizing seed shell ash and non-edible seed oil from Xanthium strumarium seeds. The catalyst prepared by open air burning (UC), calcinations at 550 °C (C550) and 700 °C (C700) was characterized by powder X-ray diffraction (XRD), fourier transform infrared (FT-IR), field emission scaning electron microscope (FESEM), energy dispersive X-ray (EDX), field emission transmission electron microscope (FETEM), X-ray photoelectron spectrometer (XPS), thermogravimetric (TG) and brunauer-emmett-teller (BET) analysis. Elemental analysis by XPS and EDX displayed the presence of potassium (34 wt%) in high concentrations that can contribute effectively to the transesterification process. The derived catalyst provides a high biodiesel yield (98.3%) at optimum operating conditions viz. 6 wt% catalyst concentrations, 9:1 methanol to oil ratio at 60 °C within 70 min. FFA content reduced from 5.63 mg in the oil to 0.28 mg in the biodiesel due to presence of considerable amount of amphoteric SiO 2 and acidic P 2 O 5 in the catalyst. The catalyst prepared at 550 °C showed the highest pH value of 11.78. The Hammett indicator test provides higher basicity for C550 (0.513 mmol/g) than that of C700 and uncalcined catalyst. The reaction follows pseudo-first-order kinetics and the activation energy was found to be 37.413 kJ mol −1 . The reusability study also showed satisfactory biodiesel yield (82.3%) at 3rd round of transesterification reaction. The prepared biodiesel was characterized by 1 H and 13 C NMR, GC-MS analysis and the fuel parameters are evaluated by the standard protocols.

Abstract:Employing aerobic fermentation, Gram-negative bacteria belonging to the genus Xanthomonas produce the high molecular weight natural heteropolysaccharide known as xanthan. It has various amounts of O-acetyl and pyruvyl residues together with D-glucosyl, D-mannosyl, and D-glucuronyl acid residues in a molar ratio of 2:2:1. The unique structure of xanthan allowed its various applications in a wide range of industries such as the food industry, pharmacology, cosmetics and enhanced oil recovery primarily in petroleum. The cultivation medium used in the manufacture of this biopolymer is critical. Many attempts have been undertaken to generate xanthan gum from agro-based and food industry wastes since producing xanthan gum from synthetic media is expensive. Optimal composition and processing parameters must also be considered to achieve an economically viable manufacturing process. There have been several attempts to adjust the nutrient content and feeding method, temperature, pH, agitation and the use of antifoam in xanthan fermentations. Various modifications in technological approaches have been applied to enhance its physicochemical properties which showed significant improvement in the area studied. This review describes the biosynthesis production of xanthan with an emphasis on the importance of the upstream processes involving medium, processing parameters, and other factors that significantly contributed to the final application of this precious polysaccharide.
