Abstract:In the past decades, many efforts have been made to improve the sensitivity and specificity of electrochemical DNA biosensors. However, it is still strongly required to develop disposable and reliable DNA biosensors for wide and practical application. In this article, we reported superior electrochemical properties of an integrated plastic-gold electrode (PGE) fabricated in-house by chemical plating on polystyrene substrate. PGEs were found having extremely high capacity of DNA immobilization compared with gold electrodes fabricated by standard sputtering based photolithography. Unique nano-structured surface was observed on PGEs through morphology techniques, which would to some extend give an explanation to higher capacity of DNA immobilization on PGEs. A probable mechanism of carboxylic acid produced on polystyrene substrate after exposure to UV irradiation was proposed and discussed for the first time. This biosensor was applied to detection and manipulate of DNA hybridization. Detection limit of 7.2×10−11M and 1–500nM of linearity range was obtained.

Abstract:The target DNA was immobilized successfully on gold colloid particles associated with a cysteamine monolayer on gold electrode surface. Self-assembly of colloidal Au onto a cysteamine modified gold electrode can enlarge the electrode surface area and enhance greatly the amount of immobilized single stranded DNA (ssDNA). The electron-transfer processes of [Fe(CN)6](4-)/[Fe(CN)6](3-) on the gold surface were blocked due to the procedures of the target DNA immobilization, which was investigated by impedance spectroscopy. Then single stranded target DNA immobilized on the gold electrode hybridized with the silver nanoparticle-oligonucleotide DNA probe, followed by the release of the silver metal atoms anchored on the hybrids by oxidative metal dissolution, and the indirect determination of the released solubilized Ag(I) ions by anodic stripping voltammetry (ASV) at a carbon fiber microelectrode. The results show that this method has good correlation for DNA detection in the range of 10-800 pmol/l and allows the detection level as low as 5 pmol/l of the target oligonucleotides.

Abstract:The direct electrochemistry of catalase (Ct) was accomplished at a gold electrode modified with single‐wall carbon nanotubes (SWNTs). A pair of well‐defined redox peaks was obtained for Ct with the reduction peak potential at −0.414 V and a peak potential separation of 32 mV at pH 5.9. Both reflectance FT‐IR spectra and the dependence of the reduction peak current on the scan rate revealed that Ct adsorbed onto the SWNT surfaces. The redox wave corresponds to the Fe(III)/Fe(II) redox center of the heme group of the Ct adsorbate. Compared to other types of carbonaceous electrode materials (e.g., graphite and carbon soot), the electron transfer rate of Ct redox reaction was greatly enhanced at the SWNT‐modified electrode. The peak current was found to increase linearly with the Ct concentration in the range of 8×10 −6 –8×10 −5 M used for the electrode preparation and the peak potential was shown to be pH dependent. The catalytic activity of Ct adsorbates at the SWNTs appears to be retained, as the addition of H 2 O 2 produced a characteristic catalytic redox wave. This work demonstrates that direct electrochemistry of redox‐active biomacromolecules such as metalloenzymes can be improved through the use of carbon nanotubes.
Electronic Biosensors Based on DNA Self-Assembled Monolayer on Gold Electrodes

Abstract:Information concerning the immobilization and hybridization of DNA on a surface is paramount to the development of DNA-based electronic biosensors. This study looks at recent investigations of DNA immobilized on gold surfaces using standard electrochemical techniques such as cyclic voltammetry (CV), potential step chronocoulometry and electrochemical impedance electrochemical impedance spectroscopy (EIS). The thiol-gold linkage is exploited for the immobilization of single- and double-stranded DNA onto gold electrodes. Two redox markers of opposite charge, ferricyanide and ruthenium hexaammine, respectively, are used to probe the environment in the vicinity of thiol-derivatized DNA electrodes. M-DNA is a form of DNA which allows the specific incorporation of certain metal ions into its helical structure under stringent conditions (i.e. low ionic strength and pH of 8.5). Single-stranded DNA monolayer and double strands DNA monolayer resistances were evaluated using EIS, respectively, and CV response were compared each other. The addition of Zn2+, under M-DNA formation conditions, led to a dramatic enhancement of electrochemical response compared to B-DNA.

Abstract:On the basis of gold nanofilm (GNF) electrode and Y junction structure of DNA, a simple and facile single-step DNA sensing protocol with improved sensitivity and lower detection limit was successfully developed. The GNF was prepared via rapid electrooxidization of the gold surface followed by the chemical reduction of the produced gold oxide layer. The capture probe DNA (c-DNA) was firstly immobilized onto GNF electrodes via Au-S bonding, for the subsequent forming of Y junction structure with target DNA (t-DNA) and reporter probe DNA (r-DNA) labeled with methylene blue (MB). Harnessing the unique properties of GNF would allow the improved contact of MB with the electrode surface and hence boost the interfacial electron communication. Experimental results of differential pulse voltammetry (DPV) showed that the peak current of the prepared biosensor was linear with the target DNA concentration from 1.0 x 10(-12) to 1.0 x 10(-9) mol/L and the proposed strategy could detect the target DNA down to the level of 2.4 x 10(-13) mol/L. Comparing with the traditional electrochemical sensors, the present protocol enabled the generation of electrochemical signal from scratch and possessed an improved selectivity against even a single base mismatch. Besides, this DNA sensor exhibited fairly good reproducibility, stability and reusability.

Abstract:Multi-walled carbon nanotube (MWCNT) is successfully immobilized on the surface of platinum electrode by mixing with DNA. The DNA/MWCNT modified electrodes are characterized by electrochemical impedance spectroscopy and cyclic voltammetry. Further research indicates that cytochrome c can strongly adsorbed on the surface of the modified electrode, and forms an approximate monolayer. The immobilized MWCNT can promote the redox of horse heart cytochrome c which gives reversible redox peaks with a formal potential of 81 mV vs SCE.

Abstract:A key challenge in developing DNA-based electrochemical sensors is the rapid and easy immobilization of DNA on different electrode surfaces. The current methods are often laborious and time-consuming. Here, we present a cost-effective and easy-to-perform method for efficiently functionalizing electrode surfaces with DNA in just a few minutes. Our strategy operates in two simple and rapid steps, regardless of the type of electrode, and avoids complicated and time-consuming procedures. The method uses phosphorothioate (PS) modification as an anchoring agent, which is a more cost-effective alternative to thiol (SH) modification, and eliminates the need for complex pretreatment processes. Additionally, a low-pH DNA loading method was used to quickly bind PS DNA to the gold surface without the need for surface-covering agents. We attributed the effect of low pH to the protonation of adenine (A), which leads to the formation of a parallel poly-A duplex (A motif) and facilitates vertical PS-DNA attachment to the electrode surface. Electrochemical techniques were used to confirm the vertical attachment of DNA to the electrode surface, and evaluation of the adsorption stability and hybridization efficiency confirmed the effectiveness of the method. The simplicity and efficiency of this method make it an ideal solution for researchers who require a fast and reliable method for DNA functionalization of electrode surfaces.

Electrochemical Studies of Electron Transfer in DNA Films with Covalently Tethered Intercalators

Abstract:The base stack within double-helical DNA provides an efficient pathway for charge transport. This DNA-mediated charge transport chemistry has been shown to be exquisitely sensitive to minor perturbations in DNA structure and base stacking both in solution and on surfaces. As a result, electrochemical studies on DNA-modified electrodes may provide a novel approach to the development of sensitive, but inexpensive DNA sensing devices. Using intercalated, covalently bound daunomycin as a redox probe, we have examined ground state charge transport in DNA films on gold electrodes. DNA-mediated electron transfer is found to occur over a distance as long as 100 A in the film. Moreover, while the introduction of one or even two breaks in the sugar-phosphate backbone yields no detectable effect on electron transfer, a CA base-pair mismatch significantly attenuates the electron transfer yield. These results confirm that the base pair stack is the pathway for DNA mediated charge transfer, not the sugar-phosphate backbone. Based on these studies, we have developed a method to electrochemically monitor the trapping of double- stranded DNA with a 6-base overhang on a gold electrode modified with double- stranded DNA probes containing a complementary overhang. The trapping of the double-stranded target can be monitored by the reduction of daunomycin crosslinked to the target. A CA mismatch in the target duplex can also be detected by the diminished reduction signal. It has been shown that the electronic coupling between an intercalator and the pi-stack of DNA is critically important for the reduction of the intercalator through DNA-mediated charge transport. Using covalently tethered anthraquinone derivatives as the redox probe, we have investigated also the influence of the binding mode of the intercalator on its reduction in DNA films. The results of these studies underscore the importance of direct interaction between the redox probe and the pi-stack in order to observe efficient DNA-mediated electrochemistry through DNA films. These studies have also shown that the covalent linkage has a significant effect on the intercalation of the probe to the base stack of DNA. In an effort to develop a redox probe that has effective electronic coupling with the pi-stack while being covalently tethered to a DNA strand with a stable linkage, we have crosslinked Nile blue, a redox active DNA intercalator with high DNA binding affinities, to the 5’-end of oligonucleotides. The covalently tethered Nile blue is shown to be sensitive probe for DNA-mediated electron transfer on the gold surface. An intervening CA mismatch has been detected in both tightly packed and loosely packed films of DNA–Nile blue conjugate. We have also coupled the reduction of Nile blue to an electrocatalytic cycle involving freely diffusing ferricyanide, which significantly enhances the sensitivity to intervening mismatches. Furthermore, using Nile blue as the covalently tethered probe, we have developed a method for DNA mismatch detection that eliminates sample modification and has a potential for high throughput assays. These studies may provide a practical approach to diagnostic devices for mutation detection with high sensitivity and low expense.

Abstract:The assembly of synthetic, controllable molecules is one of the goals in nanotechnology. The primary objective of this contribution is to selectively immobilize DNA on gold via electric potential control. The self-assembly monolayer (SAM) was prepared with 2-aminoethanethiol (AET) on the gold electrode. A new approach based on electric potential was firstly used to control DNA immobilization covalently onto the SAM with the activation of 1-ethyl-3(3-dimethyl-aminopropyl)-carbodiimide (EDC) and N-hydroxysulfosuccinimide (NHS) in low ionic strength solution. The influence of electric potential on DNA immobilization was investigated by means of cyclic voltammogram, A.C. impedance, auger electron spectrometer as well as atomic force microscope (AFM) on template-stripped gold surface. The result proves that controlled potential can affect the course of DNA immobilization. More negative potential can restrain the DNA immobilization, while the more positive potential can accelerate the DNA immobilization. It is of great significance for the control of DNA self-assembly and will find wide application in the fields of DNA-based devices.
Modification of Amorphous Poly(ethylene Terephthalate) Surface by UV Light and Plasma for Fabrication of an Electrophoresis Chip with an Integrated Gold Microelectrode

Abstract:Amorphous poly(ethylene terephthalate) (PET), which possess a low softening temperature (Ts=75°C), was exploited to fabricate the electrophoresis chip with an integrated gold electrode for amperometric detection, with emphases being focused on the PET surface modification via UV light and air plasma. Both UV irradiation and plasma treatment were found to be able to improve the surface wettability, enhance the supported electroosmotic flow (EOF), and increase thermal bonding strength of PET sheets, with the latter being more efficient and less time-consuming than the former in the surface modification. Upon treated with plasma for 2min, the PET sheets could be thermally bonded at 65°C. T-peer test showed that the bonding strength increased from 10g/cm for native PET sheets to 1250g/cm for the plasma treated sheets when chips were bonded at the softening point, Attenuated-total-internal-reflection spectrum showed that, after being exposed to the UV light, carboxylic groups site-selectively formed in the UV-exposed region on PET surface. These UV-induced carboxylic groups were further utilized as the scaffold for preparation of micro-gold electrode via electroless gold plating. By using this established UV-directed electroless plating and the plasma-assisted thermal bonding techniques, the full PET electrophoresis chip with an integrated micro-gold electrode could be fabricated in common chemistry laboratory without the need of clean rooms. The fabricated PET chips were demonstrated for separation and detection of model analytes of dopamine (DA) and catechol (CA). Satisfactory resolution of the two analytes was achieved within 40s, and detection limits of 0.87μM and 1.28μM for DA and CA were obtained, respectively.

Abstract:We describe a procedure for reversible adsorption of DNA onto a gold electrode maintained under potential control. The adsorbate can be imaged by scanning probe microscopy in situ. Quantitative control of a molecular adsorbate for microscopy is now possible.We found a potential window (between 0 and 180 mV versus a silver wire quasi reference) over which a gold (111) surface under phosphate buffer is positively charged, but is not covered with a dense adsorbate. When DNA is present in these conditions, molecules adsorb onto the electrode and remain stable under repeated scanning with a scanning tunneling microscope (STM). They become removed when the surface is brought to a negative charge. When operated at tunnel currents below approximately 0.4 nA, the STM yields a resolution of approximately 1 nm, which is better than can be obtained with atomic force microscopy (AFM) at present.We illustrate this procedure by imaging a series of DNA molecules made by ligating a 21 base-pair oligonucleotide. We observed the expected series of fragment lengths but small fragments are adsorbed preferentially.
Fabrication of gold microelectrodes on polystyrene sheets by UV-directed electroless plating and its application in electrochemical detection

Abstract:A novel method for the fabrication of micro gold electrodes on polystyrene (PS) sheets by UV-directed electroless plating was developed. The microelectrode pattern was transferred onto the surface of PS sheets by exposure of the surface to UV-lights emitted from a low-pressure mercury lamp through a photo-mask. The carboxyl moieties photo-chemically generated on the UV-exposed region were used as the scaffold for further surface modifications. After amination of the carboxyl groups with ethylenediamine, adsorption of Au(III) ions by the tethered free amine groups, and reduction of the adsorbed Au(III) with NaBH4, a layer of gold nano-particles that served as seeds to catalyze the electroless plating was formed on the UV-exposed region (so-called being activated). Gold microelectrodes were then selectively plated onto the activated area by using an electroless gold plating bath. Water contact angle, attenuated total reflection fourier transformation infrared (ATR-FT-IR) spectroscopy, X-ray photoelectron spectroscopy (XPS), and chemical mapping were employed to trace the surface changes during the activation process. The prepared microelectrodes showed strong adhesion to the substrates, good stability in strong acidic and alkaline solutions, and good electrochemical properties similar to standard gold-disk electrodes. The fabricated gold microelectrodes were used as the electrode bases of the three-electrode system for amperometric detection in a full PS micro electrophoresis chip, which was demonstrated for separation and detection of model analytes of dopamine (DA) and catechol (CA).

Abstract:<p>The high-performance detection of nucleic acids is increasingly important in the early and accurate disease diagnosis. Herein, we describe a unique label-free electrochemical DNA biosensor, which is based on gold nanoparticles (GNPs) mediated electron transfer through a DNA monolayer modified gold electrode surface. We observed that the different states of DNA monolayers (single-strand DNA or triple-helix DNA) and the different sizes of GNPs have a significant effect on the electron transfer by the DNA modified gold electrodes. This DNA biosensing strategy avoids the complicated fabrication process of the DNA capture probes and minimizes the steps of electrode pre-treatment. Under optimal conditions, this method can sensitively and accurately detect the target DNA ranging from 10 pM to 100 nM with a detection limit of 1.47 pM.</p>

Abstract:Monolayers of double-stranded DNA on polycrystalline gold electrodes were prepared by chemisorbing SS-5'-GTCACGATGGCCCAGTAGTT-3'(1) hybridized with its complementary strand 5'-AACTACTGGGCCATCGTGAC-3' (2) onto gold through a disulfide linkage, giving a monolayer with hydroxylalkyl chains adjacent to the double-stranded DNA (ds-DNA). The system forms a stable and well-behaved ds-DNA monolayer on gold, which reduces the signal transduction between a solution electrophore and the gold surface. Upon addition of Zn2+ to the solution at pH 8.6, Zn2+ associates with the ds-DNA and form M-DNA. In contrast to ds-DNA, the signal transduction is significantly enhanced. The heterogeneous electron-transfer rate (kET) between the ferricyanide electrophore and the surface was determined for bare gold, ds-DNA, and M-DNA monolayers. kET for electron transfer from the solution electrophore to the gold surface through the M-DNA monolayer is 1.2 x 10(-1)+/-0.2 x 10(-4) cm s(-1), which is smaller than for bare gold under the same conditions (k(ET) = 9.1 x 10(-4)+/-0.2 x 10(-4) cm s(-1)). For simple ds-DNA, kET was too slow to be measured.
Redox Buffering Effects in Potentiometric Detection of DNA Using Thiol-Modified Gold Electrodes

Abstract:Label-free potentiometric detection of DNA molecules using a field-effect transistor (FET) with a gold gate offers an electrical sensing platform for rapid, straightforward, and inexpensive analyses of nucleic acid samples. To induce DNA hybridization on the FET sensor surface to enable potentiometric detection, probe DNA that is complementary to the target DNA has to be immobilized on the FET gate surface. A common method for probe DNA functionalization is based on thiol–gold chemistry, immobilizing thiol-modified probe DNA on a gold gate with thiol–gold bonds. A self-assembled monolayer (SAM), based on the same thiol–gold chemistry, is also needed to passivate the rest of the gold gate surface to prevent non-specific adsorption and to enable favorable steric configuration of the probe DNA. Herein, the applicability of such FET-based potentiometric DNA sensing was carefully investigated, using a silicon nanoribbon FET with a gold-sensing gate modified with thiol–gold chemistry. We discover that the potential of the gold-sensing electrode is determined by the mixed potential of the gold–thiol and gold–oxygen redox interactions. This mixed potential gives rise to a redox buffer effect which buffers the change in the surface charge induced by the DNA hybridization, thus suppressing the potentiometric signal. Analogous redox buffer effects may also be present for other types of potentiometric detections of biomarkers based on thiol–gold chemistry.This article has not yet been cited by other publications.

Self-Assembled Multilayer of Gold Nanoparticles for Amplified Electrochemical Detection of Cytochrome C

Abstract:Although different kinds of film materials and some modification techniques are applied for the development of protein-film electrochemistry, the design of a more ordered adsorption platform with improved sensitivity is still required. Here we employ single-strand DNA (ssDNA)-functionalized gold nanoparticles as scaffolds for the construction of a multilayered uniform self-assembled structure via the hybridization of complementary ssDNA. After adsorbing with native conformation onto the uniformly built electrode, cytochrome c responded very well in voltammetry experiments. The peak currents increase with the addition of the number of gold nanoparticle layers, which indicates that the multilayer gold nanoparticles not only provide a compatible microenvironment for the protein to undergo direct electron transfer reactions but also amplify the electrochemical signals by increasing the binding sites for the protein immobilization. Furthermore, ultra-sensitive detection of cytochrome c by using this multilayer gold nanoparticle-modified electrode is carried out. The linear range is from 2 x 10(-9) to 1 x 10(-7) M with a detection limit of 6.7 x 10(-10) M.

Abstract:In this report, we systematically investigated DNA immobilization at gold surfaces with electrochemical techniques. Comparative cyclic voltammetric and chrono-coulometric studies suggested that DNA monolayers immobilized at gold surfaces were not homogeneous. Nonspecific Au-DNA interactions existed even with the treatment of mercaptobexanol, which was known to competitively remove loosely bound. DNA at gold surfaces. While both thiolated and nonthiolated DNA formed monolayers on gold surfaces, their hybridization abilities were distinctly different. In contrast to thiolated DNA probes, nonthiolated DNA probes immobilized at gold surfaces were essentially nonhybridizable. The experimental results presented here might be useful for the design of high-performance electrochemical DNA sensors.

Abstract:Electrochemically modified glassy carbon electrode (GCE) was used to study the electrochemical oxidation and detection of denatured single-stranded (ss) DNA by means of adsorptive stripping voltammetry. The modification of GCE, by electrochemical oxidation at +1.75 V (vs.SCE) for 10 min and cyclic sweep between +0.3 V and -1.3 V for 20 cycles in pH 5.0 phosphate buffer, results in 100-fold improvement in sensitivity for ssDNA detection. We speculated that the modified GCE has a high affinity to single-stranded DNA through hydrogen bond (specific static adsorption). Single-stranded DNA can accumulate at the GCE surface at open circuit and produce a well-defined oxidation peak corresponding to the guanine residues at about +0.80 V in pH 5.0 phosphate buffer, while the native DNA gives no signal under the same condition. ne peak currents are proportional to the ssDNA concentration in the range of 0-18.0 mug mL(-1). The detection limit of denatured ssDNA is ca. 0.2 mug mL(-1) when the accumulation time is 8 min at open circuit. The accumulation mechanism of ssDNA on the modified GCE was discussed.
Potential Control of DNA Self-Assembly on Gold Electrode

Abstract:The self-assembly monolayer (SAM) was prepared with 2-aminoethanethiol (AET) on the gold electrode. A new approach based on potential was first used to control DNA self-assembly covalently onto the SAM with the activation of l-ethyl-3(3-dimethylaminopropyl)-carbodiimide (EDC) and N-hydroxysulfosuccinimide (NHS). The influence of potential on DNA self-assembly was investigated by means of cyclic voltammetry (CV), AC impedance, Auger electron spectrometry (AES) and atomic force microscopy (AFM). The result proves that controlled potential can affect the course of DNA self-assembly. More negative potential can restrain the DNA self-assembly, while more positive potential can accelerate the DNA self-assembly, which is of great significance for the control of DNA self-assembly and will find wide application in the field of DNA-based devices.

Abstract:A single stranded Cecropin CM4 gene (108 bases) was further immobilized at a cysteine modified gold electrode with the help of water soluble 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC). The ssDNA at the modified electrode could undergo hybridization in a hybridization buffer containing single stranded plasmid pLCM-4SN. A DNA minor groove binder, Hoechst 33258, was employed to discriminate between ssDNA and dsDNA. The anodic waves in differential pulse voltammograms (DPVs), of Hoechst 33258 bound to the DNAs, were used as the indicator. This assay procedure was shown to be rapid, sensitive and precise, thus a kind of prototype DNA biosensor was developed.
