Abstract:The full length transferrin (TF) cDNA from the Vibrio anguillarum challenged red seabream was cloned by homology strategy using a pair of degenerated primers designed according to the conserved regions of other animals' TF genes. A 451 bp homologous fragment was obtained by PCR amplification, which was elongated by RACE PCR with the adaptor primer and several gene specific primers to get the full length sequence. The sequence contained 2444 bp including a short 5' untranslated region (UTR) of 31 bp and 3' UTR of 340 bp and an open reading frame (ORF) of 2073 bp which could code a 691 amino acids peptide with putative MW of 74.3 kDa and putative PI of 5.63. The sequence shared high identity and similarity with other animals' transferrin, especially with piscine TF genes. There was the identity of about 65%-75% with other fish, and about 40%-50% with each of the mammalian TFs and lactoferrins, even some identity with the insects. Phylogenetic analysis showed that the gene should be transferrin not lactoferrin. The configuration of the red seabream TF was similar to mammalian TF which was a single strand protein being composed of two similar lobes. The protein had a signal peptide of 18 amino acids but lacked clearly N-glycosylation site. Each of the lobes had three TF family signatures and a MHC molecular signature. From the secondary structure analysis, it was highly conserved in the region of the Fe binding sites and cysteine positions which should involve in constitution of secondary structure, and this indicated that the red seabream TF had a similar function with other animals'. RT-PCR showed that the red seabream TF was constitutively expressed in liver, but it could be regulated by pathogen stimulating, and the distribution of TF expression increased significantly after pathogen stimulating.
