Abstract:A deep understanding upon the relation between anthocyanin extraction performance and physicochemical properties of natural deep eutectic solvents (NADESs) was obtained, and a high-efficiency NADES (Choline chloride-Oxalic acid (ChOa)) was selected and coupled with pulse-ultrasonication for anthocyanin extraction from blueberry pomace. Response surface methodology was adopted to analyzed the significant factors' influence, interactive effects on extraction yield and optimize the extraction conditions. The theoretical optimal conditions with highest yield of total anthocyanins content (TAC) were 3.2 min of ultrasonic time, 325 W of ultrasonic power, 349.15 K of temperature, 60 mL/g of solvent to solid ratio, under which the experimental TAC value (24.27 +/- 0.05 mg C3GE/g DW) was highly correlated with theoretical result (24.28 mg C3GE/g DW). Multi-stability protective effect of ChOa on anthocyanin over acidified ethanol was revealed by stability assays in high temperature, varying pH and light exposure conditions. FT-IR and DSC analysis further characterized the influence of ChOa over acidic ethanol on typical anthocyanins functional groups and thermal behavior of anthocyanin extracts. UPLC-Triple-TOF/MS analysis identified 10 anthocyanins that mainly consisted of delphinidin, petunidin, and malvidin compounds. Overall, this study provided a green, high-efficiency and multistability protective strategy for the recovery of anthocyanins from plant source by-products.
Acidified glycerol as a one-step efficient green extraction and preservation strategy for anthocyanin from blueberry pomace: New insights into extraction and stability protection mechanism with molecular dynamic simulation

Abstract:In present work, green and efficient glycerol solvent system was coupled with pulse-probe ultrasonication for one-step extraction and preservation of anthocyanin from blueberry pomace. Under optimal conditions (40 min, 174 W, 18.6 mL/g, 20% of glycerol fraction), extraction yield was 23.07 ± 0.09 mg C3GE/g DW. The extracted anthocyanins were characterized by UPLC-Triple-TOF/MS and 10 anthocyanins compounds were tentatively identified. Stability of anthocyanins influenced by solvents were evaluated in varying temperature, pH and light exposure conditions, demonstrating higher stability of anthocyanins in glycerol solvent system than methanol one. Furthermore, mechanism of high efficiency extraction and stability of anthocyanin using glycerol were investigated by quantum chemical calculation with molecular dynamic simulation. Larger solvent accessible surface area (127.16 nm2), hydrogen bonds number (228.16) and hydrogen bonds lifetime (4.35 ps), and lower intermolecular interaction energy (-1080.48 kJ/mol) between anthocyanin and glycerol were responsible for better extraction and preservation of anthocyanins using glycerol system.

Abstract:Guizhou Majiang organic blueberries from China are a prevalent fruit in the health industry and have high anthocyanin contents. This study used the ethanol extraction method, ultrasonic-assisted extraction, AB 8 macroporous resin adsorption purification, and the addition of 3% citric acid to extract anthocyanins, and through single-factor tests and orthogonal tests, the optimum extraction conditions of anthocyanins from blueberries was determined. The optimal extraction conditions were as follows: the extraction temperature was 40 °C, the ethanol volume fraction was 80%, the solid-liquid ratio was 1:10, the ultrasonic extraction time was 60 min, and the extraction amount was 2.56 mg/g. Anthocyanins were identified by HPLC, and the effects of storage conditions on their contents were investigated. K+, Cu2+, and Fe2+ had little influence on anthocyanin contents, while hydrogen peroxide had a highly destructive effect. With increasing illumination time, the retention rate of anthocyanins decreased gradually; the higher the temperature was, the stronger the damaging effect on anthocyanin contents, and the appropriate storage temperature was determined to be -4 °C or below. The absorbance was the highest when pH =2. Taken together, the results of our study revealed the optimal extraction process and appropriate preservation conditions of anthocyanins from organic Guizhou blueberries.

Optimization of extraction conditions for main active components in blueberry fruits by response surface methodology

Abstract:This study was designed to optimize the extracting effects of various active components in bluebenry fruits on the conditions of diverse parameters.Analyzing and optimizing the extraction conditions of ethanol concentration,solidto-liquid ratio and extraction temperature by Box-Behnken experiment design based on the results of single factor experiments.The optimal results were determined as follows:1 ∶ 23 (g/mL) solid-to-liquid ratio,71％ ethanol concentration and 60 ℃C,which were in accordance with measured data.The yield of total contents of phenolics,flavonoids and anthocyanins was 89.94％ under these conditions.Besides,the valid model was built to make prediction about the three active components in blueberry fruits.

Abstract:Purpose This study aims to develop a green, economical and efficient ultrasonic-/microwave assisted extraction (UMAE) process for the extraction of anthocyanins. Design/methodology/approach After optimizing the extraction conditions by response surface methodology, three assays including DPPH, ABTS·+, FRAP were applied to analyze the antioxidant activity of the extracted anthocyanins. The stability under different temperatures, reductant concentrations and pHs was also discussed. The components of anthocyanins in blueberry were analyzed by HPLC-QTOF-MS2. Findings The optimal extraction parameters were ultrasonic power of 300 W, microwave power of 365.28 W and solid–liquid ratio of 30 (g/mL). The possible structures can be speculated as Delphinidin-3-O-galactoside, Delphinidin, Petunidin, Delphinidin-3-O-glucoside, Petunidin-3-O-glucoside, Cyanidin-3-O-glucoside. The results demonstrated that the UMAE can improve the yield of anthocyanins in shorter extraction time with higher activity. Originality/value The present study may provide a promising and feasible route for extracting anthocyanins from blueberries and studying their physicochemical properties, ultimately promoting the utilization of blueberry anthocyanins.

Abstract:Abstract Currently, the extraction technology of blueberry anthocyanin includes solvent extraction, enzyme extraction, and ultrasonic extraction. Different methods may damage the internal structure of anthocyanin in the extraction process, and hence the extracted anthocyanin cannot have the maximum nutritional and medicinal value. Therefore, this article analyzes the effects of different extraction methods on the antioxidant properties of blueberry anthocyanin and uses solvent extraction, enzymatic hydrolysis, and ultrasonic extraction methods to extract blueberry anthocyanin. The antioxidative properties of anthocyanins from blueberry by different extraction methods were compared and analyzed. The solvent extraction method, the enzymatic hydrolysis method, and the ultrasonic extraction method were used as experimental comparative extraction methods. The antioxidant properties of blueberry anthocyanins were measured from various angles such as resistance to oil oxidation, reducing power, and ability to scavenge hydroxyl radicals ( ̇OH) performance. From the perspective of antioxidation of fats and oils, the average inhibition rate of the solvent extraction method can reach 90%, and the corresponding inhibition rate of the anthocyanins obtained by the other two extraction methods is about 80%. The measurement results are also consistent with the measurement results of oxidation resistance of oils and fats. Conclusion: Among three different extraction methods of blueberry anthocyanins, the solvent extraction method can preserve the antioxidant properties of blueberry anthocyanins to the greatest extent.

Abstract:In order to ascertain the optimal conditions of extracting anthocyan from purple potato,solvent,alcohol concentration,extraction temperature,extraction time,pH,and liquid to material ratio were studied.Then the optimal conditions were determined by an orthogonal experiment based on single factor test.Results: The obtained optimum extraction conditions are concentration of ethanol 70%,temperature 50 ℃,extraction time 90 min,pH value 1.00,and liquid to material ratio 10∶1(mL/g).In such conditions,the extraction rate is 3.4%.The most significantly affecting factor is the liquid to material ratio,secondly is the concentration of ethanol;extraction time and temperature also has certain influence,but is not remarkable.

Abstract:The objectives of this study focused on the optimum extraction conditions to extract anthocyanin from black-wheat-bran by acidified alcohol.The results showed that the anthocyanin had the higher output of 15.6%when the optimum extraction conditions at the maximal absorption wave-length of 540 nm were as follows:the concentration of acidified alcohol is 40%,pH1.0,material-liquid ratio 1:20,extraction tempersture 70℃,extraction time 80 min,extraction series 2.There were a great deal of anthocyanin in the black wheat bran to supply a new potential pigment material to food natural colorants.

Abstract:Abstract In order to improve the utilization value of the by-product of mulberry, the remaining residue of mulberry wine was used as raw materials to extract anthocyanins from the residue of mulberry wine by enzymatic method, and the response surface method was used to optimize the extraction process. Firstly, the single factor experiment was carried out by controlling four factors: liquid-solid ratio, extraction time, temperature and pH value. Then, Box-Behnken was used to design a three-factor, three-level experiment. Finally, the response surface method was used to optimize the extraction process. The results showed that the optimal extraction process of pectinase was the liquid-solid ratio of 1:20, the extraction time was 58 min, the pH was 5.90 and the extraction temperature was 45 °C, and the extraction effect was the best. The highest anthocyanin content in the extract was 6.040 mg/g.

Abstract:Blueberry wine residues produced during the wine-brewing process contain abundant anthocyanins and other bioactive compounds. To extract anthocyanins from blueberry wine residues more efficiently, a novel procedure of ultrasound-assisted deep eutectic solvent extraction (UADESE) was proposed in this work. The extraction process was optimized by response surface methodology coupled with genetic algorithm. The optimum extraction parameters to achieve the highest yield of anthocyanins (9.32 ± 0.08 mg/g) from blueberry wine residues by UADESE were obtained at water content of 29%, ultrasonic power of 380 W, extraction temperature of 55 °C, and extraction time of 40 min. The AB-8 macroporous resin combined with Sephadex LH-20 techniques was used to purify the crude extract (CE) obtained under optimum extraction conditions and analyze the anthocyanins composition by HPLC-ESI-MS/MS. The cyanidin-3-rutinoside with purity of 92.81% was obtained. The HepG2 antitumor activity of CE was better than that of the purified anthocyanins component. Moreover, CE could increase the intracellular reactive oxygen species levels and the apoptosis, and arrest HepG2 cells in the S phases. These findings provided an effective and feasible method for anthocyanins extraction, and reduced the environmental burden of this waste.

Abstract:Multi-stage countercurrent extraction(MCCE) technique has been developed for the extraction of proanthocaynidins from grape seeds.The effects of extraction temperature,extraction stage numbers,time and solvent to grape seed ratio on the extraction yield of proanthocaynidins were investigated in detail.The process conditions of MCCE were optimized using orthogonal experimental design in three major influence factors: extraction temperature,extraction stage numbers and the ration of grape seed to solvent.The optimum process conditions were as follows: four stages,40 min extracting time for every stage,100 ℃ temperature and 3 mL/g solvent to grape seed ratio.Under those conditions,the extraction efficiency of MCCE has been compared with single pot extraction,and the results showed that the MCCE process offers the higher proanthocaynidins extraction yield and considerable savings in time,energy and solvent consumption.

Abstract:Cranberry contains abundant anthocyanins. In this study, the extraction process for the ultrasound-assisted extraction (UAE) of anthocyanins from cranberry was optimized using response surface methodology coupled with genetic algorithm. The optimum extraction parameters to achieve the highest yield of anthocyanins (7.25 +/- 0.02) mg/g from cranberry via UAE were obtained at an extraction temperature of 53 degrees C, ultrasound power of 310 W, ultrasound time of 33 min, and ethanol concentration of 52%. High-speed countercurrent chromatography was used to separate and enrich main anthocyanins from cranberry. The purified anthocyanin components were analyzed by HPLC-MS2. Seven major kinds of anthocyanins were detected and identified for the first time peonidin-3-(6-malon)-glucoside with a purity of 82.13% and pelargonidin-3-(6-malon)-glucoside with a purity of 85.37% in cranberry. Hence, ultrasound-assisted extraction of anthocyanins was an effective and rapid method. This method has a broad application prospect in the extraction of bioactive substances from natural plant resources. Practical applications Cranberries (Oxycoccos), as a good source of bioactive compounds, include anthocyanins, flavanols, various vitamins, superoxide dismutase, and phenolic acids, and the extraction process for the ultrasound-assisted extraction of anthocyanins from cranberry was optimized using response surface methodology coupled with genetic algorithm in this study. High-speed countercurrent chromatography was used to separate and enrich main anthocyanins from cranberry. This finding provides a new method for the extraction of anthocyanins from cranberry.

Abstract:Anthocyanins are naturally occurring compounds that impart color to fruits, vegetables, and plants. This study aims to optimize the microwave-assisted extraction (MAE) conditions of anthocyanins from mulberry (M. atropurpurea Roxb.) using response surface methodology (RSM). A Box-Behnken experiment was employed in this regard. Methanol concentration, microwave power, and extraction time were chosen as independent variables. The optimized conditions of MAE were as follows: 59.6% acidified methanol, 425 W power, 25 (v/w) liquid-to-solid ratio, and 132 s time. Under these conditions, 54.72 mg anthocyanins were obtained from 1.0 g mulberry powder. Furthermore, 8 anthocyanins were identified by high-performance liquid chromatography-electrospray ionization-mass spectrometry (HPLC-ESI-MS) in mulberry extract. The results showed that cyanidin-3-glucoside and cyanidin-3-rutinoside are the major anthocyanins in mulberry. In addition, in comparison with conventional extraction, MAE is more rapid and efficient for extracting anthocyanins from mulberry.

Abstract:Ultrasonic cell grinder extraction (UCGE), using water as the solvent, was firstly applied to extract anthocyanins from blueberry. Extraction yield was related with four variables, including ratio of solution to solid, extraction power, buffer time, and extraction time. On the basis of response surface methodology (RSM), the optimal conditions were determined to be the ratio of solution to solid as 25:1(mL/g), the extraction power as 1500W, the buffer time as 3.0s, and the extraction time as 40min. The experimental yield of anthocyanins using UCGE was 2.89mg/g higher than that of conventional ultrasound-assisted extraction (CUAE). This study indicated that UCGE was an innovative, efficient, and environment friendly method in ultrasonic extraction fields, and had a potential to effectively extract other bioactive constituents.
Isolation, purification, identification, and stability of anthocyanins from Lycium ruthenicum Murr

Abstract:Response surface methodology was used to investigate the ultrasonic-assisted extraction of anthocyanins from Lycium ruthenicum Murr. A Box-Behnken design of three-level-three-factor was used to optimize extractive factors, including ethanol concentration (A), ultrasonic extraction time (B), and liquid-solid ratio (C) and to achieve high extraction yield of the anthocyanins. The three parameters were optimized at 70% (v/v), 28min, 33: 1 (mL/g), respectively, the best extraction yield of anthocyanins was 7.12 ± 0.093%. The AB-8 resin was further to be chosen for purification the anthocyanins. Anthocyanin composition was analyzed by HPLC-MS2, the possible structures can be speculated as petunidin-3-O-(glucosyl-p-coumaroyl)-rutinoside-5-O-glucoside, pentuidin-3-O-(caffeoyl)-rutinoside-5-O-glucoside, delphinidin-3-O-(p-coumaroyl)-rutinoside-5-O-glucoside, petunidin-3-O-(p-coumaroyl)-rutinoside-5-O-glucoside, malvidin-3-O-(p-coumaroyl)-rutinoside-5-O-glucoside, cyanidin-3-O-rutinoside, and petunidin-3-O-(p-coumaroyl)-rutinoside. The thermal stability indicated the high purity of anthocyanins, and purified anthocyanins combined with whey protein isolate (WPI) can enhance the thermal stability of anthocyanins. The thermal stability of the purified anthocyanins was higher than that of the unpurified anthocyanins; whereas the thermal stability of the purified anthocyanin-WPI synthesis was greater than that of the purified anthocyanins. The thermal stable anthocyanins and anthocyanin-protein synthesis has the potential application in the functional food industry.
Ultrasound Treatment on Stability of Total and Individual Anthocyanin Extraction from Blueberry Pomace: Optimization and Comparison

Abstract:Blueberry pomace is abundant in anthocyanins. This work characterized the anthocyanins in blueberry pomace, discussed the stability of anthocyanins under ultrasound treatment, and compared the extraction conditions for different anthocyanin compositions. Thirteen anthocyanins were identified, and malvidin-3-galactoside (18.56%), which represented the most abundant anthocyanin, was selected as the individual analyte. The general linear model univariate analysis revealed that ultrasound-assisted extraction (UAE) resulted in higher recoveries of both total anthocyanins (TA) and individual anthocyanins (IA) when compared with conventional solvent extraction. The optimized extraction conditions for TA and IA were UAE in pure methanol (12.49 mg/g dry weight) at 25 °C for 30 min and UAE in 70% ethanol (3.57 mg/g dry weight) at 40 °C for 40 min, respectively. Moreover, IA was more vulnerable to degradation compared with TA. Therefore, a specific extraction process of IA is significant for monomer preparation, and harsh conditions should be avoided in UAE.
Extraction and Identification of Anthocyanins from Wild Red Raspberry Fruit

Abstract:In this study,anthocyanins were extracted from wild red raspberries with 0.5% TFA in ethanol solution and were subjected to quantitative analysis.The total anthocyanin content of red raspberry crude extract,which was determined by pH differential method,was(7.548±0.03) mg/g of freeze-dried powder.Furthermore,the extracts were purified on a AB-8 macroporous resin column,the optimum elution reagent was 70% ethanol with TFA(pH 1),elution of ethanol was 6 times column volume at flow rate either on adsorption or desorption of 2 mL/min.Nine major anthocyanins of red raspberry were isolated.All of the peaks are identified by low-resolution electro spray mass spectrometry.HPLC-ESI/MS analysis results showed that the component of anthocyanins of the wild red raspberry fruit are cyanidin-3,5-diglucoside,cyanidin-3-digalactoside,cyanidin-3-glucoside,cyanidin-3-rutinoside,cyanidin,cyanidin-3-arabinoside,cyaniding-3-(6-O-acetyl)-glucoside pelargonidin-3-glucoside and peonidin-3-glucoside.

Abstract:Grape skins produced during the grape juice production and processing contain abundant anthocyanins and other active compounds. Consequently, this study optimized the extraction conditions for ultrasound-assisted enzymatic extraction (UAEE) of anthocyanins from grape skins via response surface methodology coupled with genetic algorithm. The optimum extraction parameters to achieve the highest anthocyanins yield (3.01 +/- 0.04) mg/g from grape skins by UAEE were obtained under an extraction temperature of 50 degrees C, ultrasonic power of 400 W, pectinase dosage of 0.16%, and extraction time of 28 min. The AB-8 macroporous resin combined Sephadex LH-20 techniques were further employed to purify the anthocyanins extracts obtained under optimum extraction conditions (AEOEC), and the main anthocyanins were identified using high-performance liquid chromatography tandem mass spectrometry. The purified anthocyanins contained two anthocyanins in terms of delphinidin-3,5-O-diglucoside and cyanidin-3-O-rutinoside with purity of 91.35% and 92.64%, respectively. Ultimately, we further evaluated the antitumor activity of AEOEC and two purified anthocyanins on breast cancer. The results indicated that the antitumor effect of AEOEC on breast cancer MCF-7 cells was better than that of two purified anthocyanins. In addition, AEOEC could memorably increase intracellular reactive oxygen species levels and apoptosis of MCF-7 cells, and arrest MCF-7 cells in the G2/M phases. The findings provide an effective and feasible method for anthocyanins extraction and reduce the environmental burden of this waste.
Ultrasonic-assisted enzymatic extraction and identification of anthocyanin components from mulberry wine residues

Abstract:Mulberry wine residues produced during the wine-brewing process contain several anthocyanins and other bioactive compounds. Therefore this study optimized the conditions for ultrasound-assisted enzymatic extraction of anthocyanins from mulberry wine residues. A three-level, four-factor Box–Behnken design was used to optimize the extraction conditions. Moreover, anthocyanins were determined using an ultra-performance liquid chromatograph coupled to a mass spectrometer (UPLC-MS). The mathematical model suggested a high coefficient of determination (R2 = 0.9475) for the optimum conditions, namely 52°C, 315W, 0.22% enzyme and 94 min incubation. The yield (5.98 mg/g) was close to the predicted value (5.87 mg/g). The two anthocyanins (cyanidin-3-O-glucoside and cyanidin-3-O-rutinoside) identified are consistent with those present in mulberry. The optimized conditions increased anthocyanin yield, through improved utilization of mulberry wine residues. The findings will potentially lead to a reduction in the environmental burden of this waste and improve the efficiency and productivity of the mulberry fruit processing industry.

Abstract:In order to develop the resource of anthocyanins in purple potato,anthocyanin extraction technology was studied by conducting an orthogonal experiment on the basis of single factor tests.The anthocyanin yield was determined by the spectrophotometric pH differential method.The alternate action of ultrasonic power,extraction temperature,time and the ratio of sample weight to solvent volume were investigated.Citric acid solution was taken as extractant.Results: The sample/solvent ratio,extraction time and temperature have notable effects on anthocyanin yield while ultrasonic power has little effect.The optimum extraction conditions are sample to solvent ratio(m/V) 1∶ 40,extraction time 5 min,extraction temperature 35 ℃,and ultrasonic power 350 W.Additionally,taking citric acid solution as extractant reduces anthocyanins hydrolysis and production cost greatly.
