Abstract:Objective To investigate the changes of serum malondialdehyde(MDA),superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px)in rats with acute omethoate intoxication and the ameliorative effects of vitamins C.Methods An experimental model of 72 Wistar rats was made by subcutaneous injection with 0 1 LD_ 50 and 0 5 LD_ 50 omethoate.Rats were equally assigned to the control group,0 1 LD_ 50 omethoate group,0 5 LD_ 50 omethoate group and 0.5 LD_ 50 omethoate +VitC(200mg/kg) group.Serum MDA,SOD and GSH-Px were determined at various intervals.Results Serum MDA level increased significantly in 0 1 LD_ 50 and 0 5 LD_ 50 omethoate group compared to the control group (P0 05),and increased significantly in 0 5 LD_ 50 omethoate group compared to 0 1 LD_ 50 omethoate group (P0 05),but decreased significantly in 0 5 LD_ 50 omethoate +VitC group (P0 05).The activity of SOD and GSH-Px decreased significantly in 0 1 LD_ 50 and 0 5 LD_ 50 omethoate group compared to the control group (P0 05),decreased significantly in 0 5 LD_ 50 omethoate group compared to 0 1 LD_ 50 omethoate group (P0 05),and increased significantly in 0 5 LD_ 50 omethoate +VitC group (P0 05).Conclusions The levels of lipid peroxidation go up and the activities of anti-oxidant enzyme go down in rats with acute omethoate intoxication,and treatment with vitamin C can reduce lipid peroxidation caused by omethoate.

Abstract:Isolated rat ventricular myocytes were incubated in different manganese concentrations and after that the reactive oxygen species (ROS) generation, the level of reduced glutathione (GSH), and the changes of mitochondrial membrane potential (Δψ m) were investigated by a flow cytometer (FACScan) as well as laser scanning confocal microscopy (LSCM). Results showed that, although the total ROS in the cell were increased after manganese treatment, hydrogen peroxide (H2O2) was the main elevated species while superoxide anion (O2·−) was nearly unchanged. The generation of H2O2 became obvious when the myocytes were incubated in the higher concentration of manganese. The content of GSH in myocytes also decreased after manganese exposure. When the myocytes were incubated with both manganese and GSH-ethylester (GSH-EE), which is permeable to the cell membrane, the generation of H2O2 decreased greatly. The loss of the mitochondrial membrane potential (Δψ m) was also induced by manganese incubation. However, with the existence of extracellular GSH-EE the Δψ m were rescued. The results suggested that manganese action may lead to the ROS stress upon myocytes which most probably rise from high generation of intracellular H2O2. GSH-EE could effectively clean the over-production of H2O2, indicating that the low level of intracellular GSH was another main reason to the high accumulation of H2O2. The mitochondrial membrane potential was also affected by manganese and rescued by GSH-EE, suggesting that H2O2 was also involved in the manganese damages to the mitochondrial membrane potential.

Abstract:Objective To explore the effect of MnCl2 on level of MDA and the activities of SOD and GSH-Px in PC12 cells.Methods Cultured PC12 cells were exposed to MnCl_2.The survival of the PC12 cells was determined by MTT assay.The product of lipid peroxidation(MDA) was measured by TBA assay method.The activities of SOD and GSH-Px were measured by Kit.Results MnCl_2 could decrease the survival of PC12 cells and this effect showed dose-dependent manner.The level of MDA in manganese-treated PC12 cells increased compared with the control(P0.05),and the activities of SOD and GSH-Px were decreased compared with the control(P0.05).Conclusions Excessive manganese could increase the level of lipid peroxidation,decrease the activities of antioxidase significantly,which could create high level of oxidative stress in cells.This may be the possible mechanism of manganese toxicity in PC12 cells.

Abstract:Manganese (Mn) is an essential trace element found in many enzymes. As is the case for many essential trace elements, excessive Mn is toxic. Individuals suffering from manganese toxicity exhibit several symptoms, which are similar to those frequently observed in cases of Parkinson's disease. In this investigation, we studied the effect of manganese chloride (7.5, 15.0, and 30.0mg/kg body weight) on mitochondrial function and attempted to ascertain the mechanism of manganese-induced mitochondrial dysfunction. The production of reactive oxygen species in mitochondria of rat liver and brain was assayed using 2′,7′-dichlorofluorescin diacetate, and the activities of respiratory chain enzymes were examined spectrophotometrically. Monoamine oxidase (MAO) activity was assayed by measuring reduction of benzylamine. Manganese and calcium content in mitochondria were determined by atomic absorption spectrophotometry. These results indicate that manganese chloride (MnCl2) can decrease MAO activity and inhibit the respiratory chain. Manganese can accumulate in mitochondria and inhibit efflux of calcium. There is a significant inverse correlation between the amount of superoxide radicals and the specific activities of the mitochondria enzymes. Mitochondrial function was significantly affected in both males and females.

Abstract:Manganese (Mn) is an essential trace element involved in various physiological processes, but excessive exposure may lead to toxicity. The vascular endothelium, a monolayer of endothelial cells within blood vessels, is a primary target of Mn toxicity. This review provides a comprehensive overview of the impact of Mn on vascular endothelium, focusing on both peripheral and brain endothelial cells. In vitro studies have demonstrated that high concentrations of Mn can induce endothelial cell cytotoxicity, increase permeability, and disrupt cell-cell junctions through mechanisms involving oxidative stress, mitochondrial damage, and activation of signaling pathways, such as Smad2/3-Snail. Conversely, low concentrations of Mn may protect endothelial cells from the deleterious effects of high glucose and advanced glycation end-products. In the central nervous system, Mn can cross the blood-brain barrier (BBB) and accumulate in the brain parenchyma, leading to neurotoxicity. Several transport mechanisms, including ZIP8, ZIP14, and SPCA1, have been identified for Mn uptake by brain endothelial cells. Mn exposure can impair BBB integrity by disrupting tight junctions and increasing permeability. In vivo studies have corroborated these findings, highlighting the importance of endothelial barriers in mediating Mn toxicity in the brain and kidneys. Maintaining optimal Mn homeostasis is crucial for preserving endothelial function, and further research is needed to develop targeted therapeutic strategies to prevent or mitigate the adverse effects of Mn overexposure.
Manganese induces oxidative damage in the hippocampus by regulating the expression of oxidative stress‐related genes via modulation of H3K18 acetylation

Abstract:Prolonged exposure to manganese (Mn) contributes to hippocampal Mn accumulation, which leads to neurodegenerative diseases called manganese poisoning. However, the underlying molecular mechanisms remain unclear and there are no ideal biomarkers. Oxidative stress is the essential mechanisms of Mn‐related neurotoxicity. Furthermore, histone acetylation has been identified as being engaged in the onset and development of neurodegenerative diseases. Therefore, the work aims to understand the molecular mechanisms of oxidative damage in the hippocampus due to Mn exposure from the aspect of histone acetylation modification and to assess whether H3K18 acetylation (H3K18ac) modification level in peripheral blood reflect Mn‐induced oxidative damage in the hippocampus. Here, we randomly divided 60 male rats into four groups and injected them intraperitoneally with sterile pure water and MnCl2⋅4H2O (5, 10, and 15 mg/kg) for 16 weeks, 5 days a week, once a day. The data confirmed that Mn exposure down‐regulated superoxide dismutase activity and glutathione level as well as up‐regulated malondialdehyde level in the hippocampus and plasma, and that there was a positive correlation between these indicators in the hippocampus and plasma. Besides, we noted that Mn treatment upregulated H3K18ac modification levels in the hippocampus and peripheral blood and that H3K18ac modification levels correlated with oxidative stress. Further studies demonstrated that Mn treatment decreased the amounts of H3K18ac enrichment in the manganese superoxide dismutase (SOD2) and glutathione transferase omega 1 (GSTO1) gene promoter regions, contributing to oxidative damage in the hippocampus. In short, our results demonstrate that Mn induces oxidative damage in the hippocampus by inhibiting the expression of SOD2 and GSTO1 genes via modulation of H3K18ac. In assessing Mn‐induced hippocampal neurotoxicity, oxidative damage in plasma may reflect hippocampal oxidative damage in Mn‐exposed groups.

Abstract:The toxic effect of various concentrations of Mn2+(0、0.09、0.18、0.27、0.36mmol/L)was studied on antioxidant system,active oxygen generation and submicroscopic structure of Potamogeton crispus.The results showed that the content of chlorophyll a,chlorophyll b and total chlorophyll all first increased and then decreased with increasing the concentration of Mn2+.Superoxide dismutase(SOD)activity generally increased,peroxydase(POD)and catalase(CAT)activities enhanced a several fold,but with the manganese concentration increased continuously,they all decreased,which was possibly the reason of accumulation of O-·2 and H2O2.Glutathione(GSH)and antiscorbic acid(AsA)contents changed at the same trend of POD and CAT.The level of lipid peroxidation was measured as the content of malondialdehyde(MDA),which was always increased and was the same as the change of O-·2 and H2O2.Transmission electron microscopy observation indicates that the higher Mn2+ concentration,the more serious damages to the leaf cells,especially chloroplast,mitochondrion and nucleus.The conclusion could be reached that the growth declining even death of plant was resulted from destruction of structure foundation of physiological function,interruption of ion and enzymes equilibrium and disorder of antioxidant system.But the influences of those aspects are in different extent in Potamogeton crispus.

Abstract:Oxidative stress is related to many diseases, but available clinical treatment methods are currently limited. Exploitation of enzymemimicking nanomaterials (nanozymes) is a promising way for scavenging reactive oxygen species (ROS) and treatment of ROS-related diseases. Herein, the catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GPx) mimicking activities are expressed by MnO2 nanoparticles (MnO2-BSA NPs) coated with BSA. Effective (OH)-O-center dot removal activity is also expressed by MnO2-BSA NPs at neutral pH. Apoptosis inhibition and ROS scavenging capabilities of MnO2-BSA NPs are evident on the H2O2-exposed BEAS-2B cells line. Western blot analysis indicates that MnO2-BSA NPs inhibit H2O2-induced apoptosis by mediating the expression of apoptosis-related proteins.

Abstract:Manganese is an indispensable trace element, however, it may be present at high concentrations in water and sediments of aquatic ecosystems due to natural and anthropogenic activities, and can interfere with physiological and biochemical mechanisms in fish. This study was conducted to determine the toxic effects associated with exposure to Mn 2+ (0, 0.5, 1, 2, and 4 mg/L) for 30 d, regarding liver damage and apoptosis in Yunlong grouper ( Epinephelus moara♀×E. lanceolatus♂ ). Expression of superoxide dismutase ( sod ) and catalase ( cat ) genes in the liver was significantly increased on days 10 and 20 following Mn 2+ exposure (4 mg/L), but was reduced on day 30. Similarly, expression of glutathione peroxidase ( gpx ) and glutathione reductase ( gr ) genes was elevated after 10 d of exposure to 2 and 4 mg/L Mn 2+ , but decreased after 20 and 30 d. After 30 d of exposure to high concentrations (2 and 4 mg/L) of Mn 2+ , liver tissue showed hepatic sinusoidal gap congestion, dilatation, cell vacuolation, and necrosis. In addition, the activities of alanine aminotransferase (ALT) and aspartate transaminase (AST) as well as 8-hydroxydeoxyguanosine (8-OHdG) levels were significantly increased after Mn 2+ exposure. Moreover, Mn 2+ exposure altered the expression pattern of some pivotal genes associated to apoptosis ( p53 , bax , bcl-2 , apaf-1 , caspase-9 , and caspase-3 ), which suggested that Mn 2+ exposure induces apoptosis through the mitochondrial pathway. The above results showed that excessive Mn 2+ induced apoptosis and liver damage in grouper through elicitation of oxidative stress. These insights help elucidate the mechanism by which Mn 2+ induces toxicity in marine fish, and provide a new perspective regarding the detrimental effects of heavy metals in fish.

Abstract:Manganese (Mn) is known to induce mitochondrial dysfunction in excessive dose; however the mechanisms underlying its action are not elucidated clearly. To determine if Mn2+ can act directly on mitochondria or indirectly by producing reactive oxygen species (ROS), isolated mitochondria were exposed to different concentration of Mn2+ (5, 50, 500, 1000 μM). ROS generation, respiratory control ratio (RCR), mitochondrial membrane potential (MMP) and respiratory chain complexes activities were investigated. Dose-dependent inhibition of respiratory chain complexes and induction of ROS were observed; these changes were paralleled by decreasing of respiratory control ratio (RCR) both with succinate or glutamate+malate. Further investigation indicated that the membrane potential determined by Rhodamine123 release decreased after MnCl2 exposure at 1000 μM. In addition, effects of the antioxidants NAC (500 μM), GSH (500 μM) and Vitamin C (500 μM) were studied at 500 μM Mn2+. The results indicate that the effect of Mn2+ exposure on respiratory chain is not site-specific, and antioxidants can protect the mitochondria function by reducing the formation of free radicals.

Abstract:Aims: Excessive manganese (Mn) exposure is toxic, and induces lipid deposition, but the underlying mechanisms remain elusive. Herein, we explored how dietary Mn supplementation affects lipid deposition and metabolism in the intestine of vertebrates using the yellow catfish Pelteobagrus fulvidraco as the model. Results: High-Mn (H-Mn) diet increased intestinal Mn content, promoted lipid accumulation and lipogenesis, and inhibited lipolysis. In addition, it induced oxidative stress, upregulated metal-response element-binding transcription factor-1 (MTF-1), and peroxisome proliferator-activated receptor gamma (PPARγ) protein expression in the nucleus, induced PPARγ acetylation, and the interaction between PPARγ and retinoid X receptor alpha (RXRα), while it downregulated sirtuin 1 (SIRT1) expression and activity. Mechanistically, Mn activated the MTF-1/divalent metal transporter 1 (DMT1) pathway, increased Mn accumulation in the mitochondria, and induced oxidative stress. This in turn promoted lipid deposition via deacetylation of PPARγ at K339 by SIRT1. Subsequently, PPARγ mediated Mn-induced lipid accumulation through transcriptionally activating fatty acid translocase, stearoyl-CoA desaturase 1, and perilipin 2 promoters. Innovation: These studies uncover a previously unknown mechanism by which Mn induces lipid deposition in the intestine via the oxidative stress-SIRT1-PPARγ pathway. Conclusion: High dietary Mn intake activates MTF-1/DMT1 and oxidative stress pathways. Oxidative stress-mediated PPARγ deacetylation at K339 site contributes to increased lipid accumulation. Abstract:The impact of a polyphenol-rich 0.1% aqueous extract from Aronia melanocarpa L. berries (AE) on the body status of manganese (Mn) and the activity of this essential element-dependent mitochondrial superoxide dismutase (MnSOD) during treatment with cadmium (Cd) was investigated in a rat model of low-level and moderate environmental human exposure to this xenobiotic (1 and 5 mg Cd/kg diet, respectively, for 3–24 months). The exposure to Cd, dose- and duration-dependently, affected the body status of Mn (apparent absorption, body retention, serum and tissue concentrations, content in some organs and total Mn body burden, and urinary and faecal excretion) and the activity of MnSOD in the mitochondria of the liver, kidney, and brain. The administration of AE during the exposure to Cd prevented or at least partially protected the animals from the perturbation of the metabolism of Mn, as well as ameliorated changes in the activity of MnSOD and the concentration of Mn and protected from Cd accumulation in the mitochondria. In conclusion, AE may protect from disorders in the body status of Mn and influence the antioxidative capacity of cells under chronic exposure to Cd. The findings confirm the protective impact of aronia berries products against Cd toxicity.

Effect of Occupational Manganese Exposure on Plasma Brain-Derived Neurotrophic Factor Levels in Male Workers

Abstract:[Objective]To determine the plasma brain-derived neurotrophic factor(BDNF) levels in male workers with manganese(Mn) exposure at workplace and explore the effect of occupational Mn exposure on plasma BDNF levels.[Methods] A total of 179 male workers of a manganese smelter in Guangxi were chosen as exposure group,and divided into low(84 workers) and high(95 workers) Mn exposure groups by cumulative exposure index(CEI) of Mn.Another 96 male workers of a sugar refinery in the same district were selected as control group.All participants received questionnaires on general information and determination of plasma BDNF.[Results]After adjusted for age and nation,a negative correlation between the plasma BDNF and the Mn exposure were identified(r=-0.200,P<0.05).The plasma BDNF levels of the two exposure groups were statistically lower than that of the control group(P<0.01),and the high Mn exposure group reported a lower level of BDNF than the low Mn exposure group(P<0.05)[.Conclusion]Occupational Mn exposure may decrease plasma BDNF levels in male Mn smelting workers.

Abstract:Exposure to excessive manganese (Mn) is toxic to humans and animals. However, the toxic effects and mechanisms of excessive Mn influencing the vertebrates have been highly overlooked. In the present study, dietary Mn overload significantly increased hepatic lipid and Mn contents, decreased superoxide dismutase 2 (Sod2) activity, increased the Sod2 acetylation level, and induced mitochondrial dysfunction; Mn induced mitochondrial dysfunction through Mtf1/sirtuin 3 (Sirt3)-mediated acetylation of Sod2 at the sites K55 and K70. Meanwhile, mitochondrial oxidative stress was involved in Mninduced lipotoxicity. Mechanistically, Mn-induced lipotoxicity was via oxidative stress-induced Hsf1 nucleus translocation and its DNA binding capacity to the regions of a peroxisome proliferator-activated receptorg (pparg) promoter, which in turn induced the transcription of lipogenic-related target genes. For the first time, our study demonstrated that Mn-induced hepatic lipotoxicity via a mitochondrial oxidative stress-dependent Hsf1 /Pparg pathway and Mtf1/sirt3-mediated Sod2 acetylation participated in mitochondrial dysfunction. Considering that lipid metabolism and lipotoxicity are widely used as the biomarkers for environmental assessments of pollutants, our study provided innovative and important insights into Mn toxicological and environmental evaluation in aquatic environments.
Manganese status, gut endogenous losses of manganese, and antioxidant enzyme activity in rats fed varying levels of manganese and fat

Abstract:We hypothesized that manganese deficient animals fed high vs moderate levels of polyunsaturated fat would either manifest evidence of increased oxidative stress or would experience compensatory changes in antioxidant enzymes and/or shifts in manganese utilization that result in decreased endogenous gut manganese losses. Rats (females in Study 1, males in Study 2, n = 8/treatment) were fed diets that contained 5 or 20% corn oil by weight and either 0.01 or 1.5 mumol manganese/g diet. In study 2, 54Mn complexed to albumin was injected into the portal vein to assess gut endogenous losses of manganese. The manganese deficient rats: 1. Had 30-50% lower liver, tibia, kidney, spleen, and pancreas manganese concentrations than manganese adequate rats; 2. Conserved manganese through approximately 70-fold reductions in endogenous fecal losses of manganese; 3. Had lower heart manganese superoxide dismutase (MnSOD) activity; and 4. Experienced only two minor compensatory changes in the activity of copper-zinc superoxide dismutase (CuZnSOD) and catalase. Gut endogenous losses of manganese tended to account for a smaller proportion of absorbed manganese in rats fed high-fat diets; otherwise fat intake had few effects on tissue manganese concentrations.
Manganese Chloride Exposure Causes Disorder of Energy Metabolism and Induces Oxidative Stress and Autophagy in Chicken Liver

Abstract:Manganese (Mn) pollution is an important environmental problem because of the potential toxicity to human and animal health. However, the effects of Mn on energy metabolism and autophagy are not clear. Consequently, we examined the effects of excessive and chronic exposure to Mn on liver function, oxidative stress, respiratory chain complex activity, and autophagy in chicken liver. Our results indicated that the accumulation of Mn in the liver and levels of AST and ALT in the serum of the Mn-exposed group were significantly higher (P < 0.05) than those in the control group at 90 days; the activities of GSH-Px, SOD, CAT, Na+-K+-ATPase, Mg2+-ATPase, Ca2+-ATPase, and respiratory chain complexes (I, II, III) in the Mn-exposed group were significantly decreased (P < 0.05) compared to the control group. However, the MDA content, NO content, iNOS activity, mRNA and protein levels of iNOS, and autophagy-related genes in the Mn-exposed group were significantly increased (P < 0.05) compared to the control group. In contrast, the mRNA level and protein expression of mTOR were significantly decreased (P < 0.05) compared to the control group. Furthermore, the characteristic autophagic vacuolar organelles were observed in the Mn-exposed group. These results suggested that excess Mn exposure can cause a disorder of energy metabolism by mitochondrial injury and induce oxidative stress and autophagy, which eventually lead to liver damage.

Abstract:In order to study the adverse effects of manganese onneuro-endocrine and cardiovascular system, radioimmunoassay(RIA) was used to test the blood prolactin concentration and plasma renin activity among manganese-exposed workers The result showed that the blood prolactin concentration and Plasma renin activity in manganese exposed workers were significantly higher than those of control group.It suggested that manganese exposure might affect the function at tubero-infundibular area and renin-angiotensin aldosterone system.
The effect of manganese exposure on GnRH secretion via Nrf2/mGluR5/COX-2/PGE2/signaling pathway

Abstract:There are limited studies focused on the precise mechanism of gonadotropin-releasing hormone (GnRH) secretion dysfunction after overexposure to manganese (Mn). The objective of the present study was to explore the mechanism of Mn disruption of GnRH synthesis via nuclear factor erythroid-2-related factor-2 (Nrf2)/metabotropic glutamate receptor-5 (mGluR5)/cyclooxygenase-2 (COX-2)/prostaglandin E2 (PGE 2 ) signaling pathway in vitro and in vivo. Primary astrocytes were cultured and treated with different doses of Mn, tert-butylhydroquinonet (tBHQ; Nrf2 agonists), 3-[(2-methyl-4-thaizolyl) ethynyl] pyridine (MTEP; mGluR5 inhibitor), and celecoxib (COX-2 inhibitor) to measure the levels of COX-2, mGluR5, Nrf2, and Nrf2 target genes. Mice were randomly divided into 11 groups, of which included the control group, 12.5-, 25-, and 50-mg/kg MnCl 2 group, dimethyl sulfoxide (DMSO) group, tBHQ control group, tBHQ pretreatment group, MTEP control group, MTEP pretreatment group, celecoxib control group, and celecoxib pretreatment group. The injection was administered every day for 2 weeks. Then, levels of GnRH, PGE 2 , COX-2, mGluR5, Nrf2, Nrf2 target genes, and morphological changes in the hypothalamus of mice were measured. Mn reduced protein levels of Nrf2 and mRNA expression of Nrf2 target genes and increased mGluR5, COX-2, PGE 2 , and GnRH levels. Meanwhile, injury-related histomorphology changes in the hypothalamus of mice were significantly present. In conclusion, excessive exposure to Mn disrupts GnRH secretion through Nrf2/mGluR5/COX-2/PGE 2 signaling pathway.

Abstract:Manganese oxide nanoparticles (MnOx NPs) are finding applications in several environmentally important areas such as farming and energy storage. MnOx NPs span a range of metal oxidation states that open up a wide range of applications in catalysis as well. As a result, it is important to understand how such materials can impact human health through incidental exposure. In this study, we examined a range of commercially available Mn2O3 NPs and compared our characterization data to those supplied by manufacturers. Discrepancies were noted and then measured values were used to assess the biological impact of these materials on three mammalian cell lines—A549, HepG2 and J774A.1 cells. Cell toxicity assays showed that all Mn2O3 particles exhibited cytotoxic effects that may be correlated, at least in part, to the production of reactive oxygen species. All eight nanoforms also activated caspase 3 but not caspase 1, although the magnitude of these changes varied greatly between materials.

Hematobiochemical Disturbances and Oxidative Stress After Subacute Manganese Chloride Exposure and Potential Protective Effects of Ebselen in Rats

Abstract:The present study aimed to detect the possible disturbances induced by subacute exposure to manganese chloride (MnCl2) on some biomarkers of hematology, clinical chemistry and oxidative stress, serum iron homeostasis, and ferritin status beside the histopathological alterations in hepatic and renal tissues, and the potential protective effects of ebselen on the Mn toxicity were also evaluated. Forty-eight rats were divided into four groups: Group 1 was used as a control. Groups 2, 3, and 4 were administered of ebselen as a single protective dose (15 mg/kg BW) intraperitoneal, daily manganese chloride (50 mg/kg BW) orally, and ebselen plus manganese chloride, respectively. The administrations were conducted for 30 days. Blood and tissue samples were collected at the end of the treatment for various experimental tests. Results revealed that MnCl2 did not significantly change in erythrogram with leukocytosis and neutrophilia but significantly increased serum aminotransferases and alkaline phosphatase activities, bilirubin (total, direct, and indirect), globulins, triglycerides, total cholesterol, creatinine, urea, manganese, iron and ferritin concentrations and hepatic glutathione, renal malondialdehyde and nitric oxide levels and hepatic superoxide dismutase activity, while serum albumin, hepatic malondialdehyde, and nitric oxide concentrations were significantly decreased besides non-statistical change in serum total proteins concentration. Ebselen has reduced the disturbances in these analytes in combined treatment group. Collectively, subacute exposure to MnCl2 causes disturbance in the leukogram, and hepatic and renal functions with marked renal oxidative stress. It also disturbed serum iron homeostasis and ferritin status. Remarkably, ebselen appears to be highly effective in attenuating the various adverse effects of manganese.
