Abstract:Biocompatible materials and structures with three‐dimensional (3D) architectures establish an ideal platform for the integration of living cells and tissues, serving as desirable interfaces between biotic and abiotic systems. While conventional 3D bioscaffolds provide a mechanical support for biomatters, emerging developments of micro‐, nano‐, and mesoscale electronic and photonic devices offer new paradigms in analyzing and interrogating biosystems. In this review, we summarize recent advances in the development of 3D functional biointerfaces, with a particular focus on electrically and optically active materials, devices, and structures. We first give an overview of representative methods for manufacturing 3D biointegrated structures, such as chemical synthesis, microfabrication, mechanical assembly, and 3D printing. Subsequently, exemplary 3D nano‐, micro‐, and mesostructures based on various materials, including semiconductors, metals, and polymers are presented. Finally, we highlight the latest progress on versatile applications of such active 3D structures in the biomedical field, like cell culturing, biosignal sensing/modulation, and tissue regeneration. We believe future 3D micro‐, nano‐, and mesostructures that incorporate electrical and/or optical functionalities will not only profoundly advance the fundamental studies in biological sciences, but also create enormous opportunities for medical diagnostics and therapies. image

Abstract:The cellular microenvironment is extremely complex, and a plethora of materials and methods have been employed to mimic its properties in vitro. In particular, scientists and engineers have taken an interdisciplinary approach in their creation of synthetic biointerfaces that replicate chemical and physical aspects of the cellular microenvironment. Here the focus is on the use of synthetic materials or a combination of synthetic and biological ligands to recapitulate the defined surface chemistries, microstructure, and function of the cellular microenvironment for a myriad of biomedical applications. Specifically, strategies for altering the surface of these environments using self-assembled monolayers, polymer coatings, and their combination with patterned biological ligands are explored. Furthermore, methods for augmenting an important physical property of the cellular microenvironment, topography, are highlighted, and the advantages and disadvantages of these approaches are discussed. Finally, the progress of materials for prolonged stem cell culture, a key component in the translation of stem cell therapeutics for clinical use, is featured.

Abstract:An appropriate cell microenvironment is key to tissue engineering and regenerative medicine. Revealing the factors that influence the cell microenvironment is a key research topic in the fields of cell biology, biomaterials, tissue engineering, and regenerative medicine. The cell microenvironment consists of not only its surrounding cells and soluble factors, but also its extracellular matrix (ECM) or nearby external biomaterials in tissue engineering and regeneration. This review focuses on six aspects of biomaterial-related cell microenvironments: ① chemical composition of materials, ② material dimensions and architecture, ③ material-controlled cell geometry, ④ effects of material charges on cells, ⑤ matrix stiffness and biomechanical microenvironment, and ⑥ surface modification of materials. The present challenges in tissue engineering are also mentioned, and eight perspectives are predicted.

Exploration of 2D and 2.5D Conformational Designs Applied on Epoxide/Collagen-Based Integrative Biointerfaces with Device/Tissue Heterogeneous Affinity

Abstract:Collagen and multifunctional epoxides, which are respectively the common constituents of natural and polymer interfaces, were combined to fabricate integrative biointerfaces with device/tissue heterogeneous affinity. Further, the traditional 2D and advanced 2.5D conformational designs were achieved on collagen-based biointerfaces. The 2D conformational biointerfaces were formed by the self-entanglement of collagen molecules based on extensive hydrogen bonds among molecules, and the lamellar structures of 2D conformational biointerfaces could act as barriers to protect both biointerfaces and substrates from enzymes and corrosion. The unique stacking structures of 2.5D conformational biointerfaces were formed by cross-linking microaggregates that were established and connected by epoxy cross-linking bonds and provided the extra 0.5D degree of freedom on structure design and functional specialization through artificially manipulating the constituents and density of microaggregates. Besides, the intersecting channels among microaggregates gave 2.5D biointerfaces diffusion behaviors, which further brought good wettability and biodegradability. The integrative biointerfaces behaved well on cell viability and enhanced the cell adhesion strength in vitro, which could be attributed to the collaborations of collagen and epoxy groups. The subcutaneous implant model in rats was utilized to investigate soft tissue response, and the results demonstrated that the tissues around implantation areas healed well and without calcification or infection. The coating of integrative biointerfaces alleviated the fibrosis around implantation areas, and the inflammatory responses and foreign body reactions were improved.

Abstract:As extracellular matrix (ECM) mimics, dynamic biointerfaces with reversible ligand presentation have shown great significance in the field of biology and medicine. However, current systems are trapped in the monotony of bioactivity, which makes it hard to mimic the multipotential of natural ECM. In this work, we reported a dynamic biomaterial interface with optional and diversified bioactivities by the combination of reversible catecholboronate and bioorthogonal click chemistry. Due to the specificity and thoroughness of bioorthogonal reaction, different types of biomolecules including small molecular saccharide, macromolecular peptides and DNA aptamers could be on-demand and reversible binding on biomaterial interfaces through sugar-sensitive catecholboronate interactions. In this design, the obtained dynamic biointerface showed biocompatible sugar-responsiveness and enabled reversible presentation of diversified bioactivities, exhibiting the multipotential to manipulate a variety of cell-biomaterial interactions. Cell capture/release experiments confirmed our dynamic biointerface could reversibly and selectively bind different cancer cells and even the bacterial microorganism. In short, apart from the original significance in ECM mimicking, the optional and diversified bioactivities on our developed dynamic biointerface will also show promising prospects in biomedical science, in particular, the cell isolation area for diagnostics and therapeutics.

Abstract:ABSTRACT Biomimetic materials have long been the (he)art of bioengineering. They usually aim at mimicking in vivo conditions to allow in vitro culture, differentiation and expansion of cells. The past decade has witnessed a considerable amount of progress in soft lithography, bio-inspired micro-fabrication and biochemistry, allowing the design of sophisticated and physiologically relevant micro- and nano-environments. These systems now provide an exquisite toolbox with which we can control a large set of physicochemical environmental parameters that determine cell behavior. Bio-functionalized surfaces have evolved from simple protein-coated solid surfaces or cellular extracts into nano-textured 3D surfaces with controlled rheological and topographical properties. The mechanobiological molecular processes by which cells interact and sense their environment can now be unambiguously understood down to the single-molecule level. This Commentary highlights recent successful examples where bio-functionalized substrates have contributed in raising and answering new questions in the area of extracellular matrix sensing by cells, cell–cell adhesion and cell migration. The use, the availability, the impact and the challenges of such approaches in the field of biology are discussed.

Abstract:The three-dimensional (3D) cultivation of intestinal cells and tissues in dynamic bioreactor systems to represent in vivo intestinal microenvironments is essential for developing regenerative medicine treatments for intestinal diseases. We have previously developed in vitro human intestinal tissue systems using a 3D porous silk scaffold system with intestinal architectures and topographical features for the adhesion, growth, and differentiation of intestinal cells under static culture conditions. In this study, we designed and fabricated a multifunctional bioreactor system that incorporates pre-epithelialized 3D silk scaffolds in a dynamic culture environment for in vitro engineering of human intestine tissues. The bioreactor system allows for control of oxygen levels in perfusion fluids (aerobic simulated intestinal fluid (SIF), microaerobic SIF, and anaerobic SIF), while ensuring control over the mechanical and chemical microenvironments present in native human intestines. The bioreactor system also enables 3D cell culture with spatial separation and cultivation of cocultured epithelial and stromal cells. Preliminary functional analysis of tissues housed in the bioreactor demonstrated that the 3D tissue constructs survived and maintained typical phenotypes of intestinal epithelium, including epithelial tight junction formation, intestinal biomarker expression, microvilli formation, and mucus secretion. The unique combination of a dynamic bioreactor and 3D intestinal constructs offers utility for engineering human intestinal tissues for the study of intestinal diseases and discovery options for new treatments.

Abstract:With the advances in the understanding of cell interactions with their microenvironments, biomaterials mimicking native microenvironments are being developed to display and deliver cell-regulatory signals in a precise and near-physiological fashion, which are used to regulate cell behavior and fate both in vitro and in vivo. Such biomaterials can provide the necessary microenvironmental cues based on chemistry, topography, mechanics, and molecule delivery. This has significantly promoted the applications of biomaterials to tissue engineering and disease treatment. This report has summarized the recent progress made in the field by focusing on native microenvironment-mimetic biomaterials utilized as tissue scaffolding and other implant devices. In particular, material properties that direct cellular behavior through controlled presentation of specific cues in time or in space, such as the material composition and material surface functionality, structure, topology, hydrophilicity/hydrophobicity, charge and energy, as well as the material topology density and mechanics, have been discussed in great detail. It is anticipated that the synergy of cell biology and modern material technologies will have a profound impact on the design and development of new generations of biomaterials.

Abstract:Combining modern methods in microsystem technology with the latest advancements in the life sciences, namely those in tissue engineering and advanced cell culturing, is promoting the development of a promising toolbox for modeling biological systems. The core problem to solve using this toolbox is the design of 3D artificial cellular environments, both in fluidic systems and on solid substrates. The construction of 3D biological fluidic environments involves the use of microfluidic devices where fluid direction and behavior can be tightly regulated in a geometrically constrained environment for advanced cell cultivation. This is used in modern cultivation devices, such as bioreactors and multicompartment systems, including systems with integrated multielectrode arrays in both 2D and 3D. The construction of 3D cell cultures on substrates involves various fabrication techniques that use different polymers and biopolymers processed by micromachining, chemical pattern guided cell cultivation, photopolymerization, and organ printing methods. These methods together have the potential to create an artificial system with the complete hierarchical, geometrical, and functional organization found in an actual biological system. In this review, we describe representative developments in this research area and the fusion of formerly unrelated disciplines that are generating new beneficial applications in life sciences.

Integrated micro/nanoengineered functional biomaterials for cell mechanics and mechanobiology: a materials perspective

Abstract:The rapid development of micro/nanoengineered functional biomaterials in the last two decades has empowered materials scientists and bioengineers to precisely control different aspects of the in vitro cell microenvironment. Following a philosophy of reductionism, many studies using synthetic functional biomaterials have revealed instructive roles of individual extracellular biophysical and biochemical cues in regulating cellular behaviors. Development of integrated micro/nanoengineered functional biomaterials to study complex and emergent biological phenomena has also thrived rapidly in recent years, revealing adaptive and integrated cellular behaviors closely relevant to human physiological and pathological conditions. Working at the interface between materials science and engineering, biology, and medicine, we are now at the beginning of a great exploration using micro/nanoengineered functional biomaterials for both fundamental biology study and clinical and biomedical applications such as regenerative medicine and drug screening. In this review, an overview of state of the art micro/nanoengineered functional biomaterials that can control precisely individual aspects of cell-microenvironment interactions is presented and they are highlighted them as well-controlled platforms for mechanistic studies of mechano-sensitive and -responsive cellular behaviors and integrative biology research. The recent exciting trend where micro/nanoengineered biomaterials are integrated into miniaturized biological and biomimetic systems for dynamic multiparametric microenvironmental control of emergent and integrated cellular behaviors is also discussed. The impact of integrated micro/nanoengineered functional biomaterials for future in vitro studies of regenerative medicine, cell biology, as well as human development and disease models are discussed.
Biomaterial microarchitecture: a potent regulator of individual cell behavior and multicellular organization

Abstract:Insoluble cues from a cell's surrounding microenvironment have increasingly been shown to be important regulators of cell behavior. The microarchitecture of biomaterials used for 3D cell encapsulation, however, is often underappreciated as an important insoluble factor guiding cell activity. In this review, we illustrate that the subcellular physical features of a scaffold influence a range of cell behaviors, including morphology, cytoskeletal organization, migration, matrix remodeling, and long-range force transmission. We emphasize that the microarchitecture of stromal extracellular matrix (ECM)-specifically the fact that it consists of a network of long interconnecting fibers with micron and nanometer-sized diameters-is an important determinant of how cells naturally interact with their surrounding matrix and each other. Synthetic biomaterials with a microarchitecture similar to stromal ECM can support analogous cellular responses, suggesting that this fibrous microarchitecture is a key regulator of these cell behaviors. Drawing upon examples from in vitro, in silico, and in vivo studies, we compare these behaviors in fibrous matrices to those of cells cultured within nanoporous matrices (e.g., alginate and PEG gels) as well as macroporous scaffolds to highlight key differences in the cellular response to each type of microarchitecture. Understanding how microarchitecture affects cell behavior can lead to more efficient biomaterial selection when designing tissue engineered scaffolds for therapeutic applications. © 2016 Wiley Periodicals, Inc. Biomimetic Design for Bio-Matrix Interfaces and Regenerative Organs.

Abstract:The urgent demand for transplanted organs has motivated the development of regenerative medicine to biomimetically reconstruct the structure and function of natural tissues or organs. The prerequisites for constructing multicellular organs include specific cell sources, suitable scaffolding material, and interconnective biofunctional interfaces. As some of the most complex systems in nature, human organs, tissues, and cellular units have unique "bio-matrix" physicochemical interfaces. Human tissues support a large number of cells with distinct biofunctional interfaces for compartmentalization related to metabolism, material exchange, and physical barriers. These naturally shaped biofunctional interfaces support critical metabolic functions that drive adaptive human behavior. In contrast, mutations and disorders during organogenesis can disrupt these interfaces as a consequence of disease and trauma. To replicate the appropriate structure and physiological function of tissues and organs, the biomaterials used in these approaches should have properties that mimic those of natural biofunctional interfaces. In this review, the focus is on the biomimetic design of functional interfaces and hierarchical structures for four regenerative organs, liver, kidney, lung, heart, and the immune system. Research on these organs provides understanding of cell-matrix interactions for hierarchically bioinspired material engineering, and guidance for the design of bioartificial organs. Finally, we provide perspectives on future challenges in biofunctional interface designs and discuss the obstacles that remain toward the generation of functional bioartificial organs.

Abstract:Abstract The three‐dimensional (3D) multicellular platforms prepared by cells or biomaterials have been widely applied in biomedical fields for the regeneration of complex tissues, the exploration of cell crosstalk, and the establishment of tissue physiological and pathological models. Compared with the traditional 2D culture methods, the 3D multicellular platforms are easier to adjust the components and structures of extracellular matrix (ECM) because of the synthesis of ECM by cells and the use of biomaterials. Moreover, the 3D multicellular platforms also can customize the cell distribution and precisely design micro and macro structures of the systems. Based on these typical advantages of 3D multicellular platforms and their increasingly important position in the biomedical field, this review summarizes the present 3D multicellular platforms. Herein, current 3D multicellular platforms are divided into two major types: scaffold‐free and scaffold‐based 3D multicellular platforms. The specific characteristics and applications of different types of 3D multicellular platforms are thoroughly introduced to help readers understand how different models affect and regulate cell behaviors and inspire researchers on how to select and design suitable 3D multicellular platforms according to different application scenarios.
Enhanced osseointegration of three-dimensional supramolecular bioactive interface through osteoporotic microenvironment regulation

Abstract:Purpose: Osteoporosis is more likely to cause serious complications after joint replacement, mainly due to physiological defects of endogenous osteogenic cells and the pathological osteoclast activity. It is a feasible solution to design a prosthetic surface interface that specifically addresses this troublesome situation.Methods: A novel "three-dimensional (3D) inorganic-organic supramolecular bioactive interface" was constructed consisting of stiff 3D printing porous metal scaffold and soft multifunctional, self-healable, injectable, and biodegradable supramolecular polysaccharide hydrogel. Apart from mimicking the bone extracellular matrix, the bioactive interface could also encapsulate bioactive substances, namely bone marrow mesenchymal stem cells (BMSCs) and bone morphogenetic protein-2 (BMP-2). A series of in vitro characterizations, such as topography and mechanical characterization, in vitro release of BMP-2, biocompatibility analysis, and osteogenic induction of BMSCs were carried out. After that, the in vivo osseointegration effect of the bioactive interface was investigated in detail using an osteoporotic model.Results: The administration of injectable supramolecular hydrogel into the inner pores of 3D printing porous metal scaffold could obviously change the morphology of BMSCs and facilitate its cell proliferation. Meanwhile, BMP-2 was capable of being sustained released from supramolecular hydrogel, and subsequently induced osteogenic differentiation of BMSCs and promoted the integration of the metal microspores-bone interface in vitro and in vivo. Moreover, the osteoporosis condition of bone around the bioactive interface was significantly ameliorated.Conclusion: This study demonstrates that the 3D inorganic-organic supramolecular bioactive interface can serve as a novel artificial prosthesis interface for various osteogenesis-deficient patients, such as osteoporosis and rheumatoid arthritis.

Abstract:New generations of synthetic biomaterials are being developed at a rapid pace for use as three-dimensional extracellular microenvironments to mimic the regulatory characteristics of natural extracellular matrices (ECMs) and ECM-bound growth factors, both for therapeutic applications and basic biological studies. Recent advances include nanofibrillar networks formed by self-assembly of small building blocks, artificial ECM networks from protein polymers or peptide-conjugated synthetic polymers that present bioactive ligands and respond to cell-secreted signals to enable proteolytic remodeling. These materials have already found application in differentiating stem cells into neurons, repairing bone and inducing angiogenesis. Although modern synthetic biomaterials represent oversimplified mimics of natural ECMs lacking the essential natural temporal and spatial complexity, a growing symbiosis of materials engineering and cell biology may ultimately result in synthetic materials that contain the necessary signals to recapitulate developmental processes in tissue- and organ-specific differentiation and morphogenesis.

Abstract:Abstract Stem cell therapy is widely recognized as a promising strategy for exerting therapeutic effects after injury in degenerative diseases. However, limitations such as low cell retention and survival rates after transplantation exist in clinical applications. In recent years, emerging biomaterials that provide a supportable cellular microenvironment for transplanted cells have optimized the therapeutic efficacy of stem cells in injured tissues or organs. Advances in the engineered microenvironment are revolutionizing our understanding of stem cell-based therapies by co-transplanting with synthetic and tissue-derived biomaterials, which offer a scaffold for stem cells and propose an unprecedented opportunity to further employ significant influences in tissue repair and regeneration.

Abstract:Bone is a complex mineralized tissue composed of various organic (proteins, cells) and inorganic (hydroxyapatite, calcium carbonate) substances with micro/nanoscale structures. To improve interfacial bioactivity of bone-implanted biomaterials, extensive efforts are being made to fabricate favorable biointerface via surface modification. Inspired by microbially catalyzed mineralization, a novel concept to biologically synthesize the micro/nanostructures on bioceramics, microbial-assisted catalysis, is presented. It involves three processes: bacterial adhesion on biomaterials, production of CO32− assisted by bacteria, and nucleation and growth of CaCO3 nanocrystals on the surface of bioceramics. The microbially catalyzed biominerals exhibit relatively uniform micro/nanostructures on the surface of both 2D and 3D α-CaSiO3 bioceramics. The topographic and chemical cues of the grown micro/nanostructures present excellent in vitro and in vivo bone-forming bioactivity. The underlying mechanism is closely related to the activation of multiple biological processes associated with bone regeneration. The study offers a microbially catalytic concept and strategy of fabricating micro/nanostructured biomaterials for tissue regeneration.

Abstract:The extraordinary properties of biological materials often result from their sophisticated hierarchical structures. Through multilevel and cross-scale structural designs, biological materials offset the weakness of their individual building blocks and enhance performance at multiple length scales to match the multifunctional needs of organisms. One essential merit of hierarchical structure is that it can optimize the interfacial features of the building blocks at different length scales, from the molecular level to the macroscale. Understanding the roles of biological material interfaces (BMIs) on the determination of properties and functions of biological materials has become a growing interdisciplinary research area in recent years. A pivotal aim of these studies is to use BMIs as inspiration for developing bioinspired and biomimetic materials and devices with advanced structures and functions. Given these considerations, this review aims to comprehensively discuss the structure-property-function relationships of BMIs in nature. We particularly focus on the discussion of BMIs and their inspired materials from mechanical and optical perspectives because these two directions are the most well-investigated and closely related. The challenges and directions of design and fabrication of BMI-inspired mechanical and optical materials are also discussed. This review is expected to garner interest from advanced material communities as well as environmental, nanotechnology, food processing, and engineering fields.
Interfacial Bioelectrochemistry: Fabrication, Properties and Applications of Functional Nanostructured Biointerfaces

Abstract:Bioelectrochemistry is a new interdisciplinary field, which combines biotechnology with electrochemical science, while the emergence of nanotechnology is opening new horizons for the investigation of this field. Nanostructures (such as nanoparticles, nanotubes) have similar dimensions to those of biomolecules (such as proteins or DNA). The combination of nanostructures with biomolecules yields functional nanostructured biointerfaces with synergetic properties and functions. The recent surge of research interest in the bioelectrochemical field is focused on the advanced design and preparation of such potential nanostructured biointerfaces, as well as the fundamental understanding of the interfacial bioelectrochemical processes within these systems. This review describes recent advances in the development of nanostructured biointerfaces based on the nanostructure-biomolecule hybrid systems in the bioelectrochemical field. To begin with, a wide range of nanostructures, which derive from metal, semiconductor, carbon, and so on, are discussed, involving their synthesis, properties, and conjugation with biomolecules, respectively. Sequentially, the fabrication, properties and characterization of functional nanostructured biointerfaces on electrode surfaces are discussed. Particular emphasis is directed to the discussion and exploration of interfacial bioelectrochemical theories of these architectures, which are of fundamental importance to the unraveling of key biological processes and the comprehensive acquaintance of electron-transfer processes. In addition, the wide applications of nanostructured biointerfaces in the bioelectrochemical field are sketched, and prospects and further developments in this exciting interdisciplinary field are also suggested.

Abstract:The regulation of cellular behavior within a three-dimensional (3D) environment to execute a specific function remains a challenge in the field of tissue engineering. In native tissues, cells and matrices are arranged into 3D modular units, comprising biochemical and biophysical signals that orchestrate specific cellular activities. Modular tissue engineering aims to emulate this natural complexity through the utilization of functional building blocks with unique stimulation features. By adopting a modular approach and using well-designed biomaterials, cellular microenvironments can be effectively decoupled from their macro-scale surroundings, enabling the development of engineered tissues with enhanced multifunctionality and heterogeneity. We overview recent advancements in decoupling the cellular micro-scale niches from their macroenvironment and evaluate the implications of this strategy on cellular and tissue functionality.
