Abstract:MxIrt1 is a iron transport protein gene from Malus xiaojinensis.To study the functional characteristic of MxIrt1,the recombinant plasmid pGEX-MxIrt1 was constructed with 162 bp segments between 3rd and 4th TMR(Transmembrane region),following the expression in E.coli with high efficiency,purification using Glutathione Sepharose 4B,the recombinant GST-MxIRT1 protein was obtained.ELISA assay detected the reactivity of the polycolonal antibody and Western blot shows that specific band was corresponding to the expectation.The recombinant GST-MxIRT1 protein can also be used for immunohistochemistry.

Abstract:Objective To construct the eukaryotic expression vector of enhanced green fluorescent protein and HCMV IE1 fusion protein for exploring the expression and function of IE1 gene.Methods HCMV IE1 gene fragment obtained from pNEB193-IE1 by enzyme digestion was inserted into expression vector pEGFP-C1,then transformed into Escherichia coli JM109,a recombinant pEGFP-C1-IE1 was constructed and confirmed by SalⅠ/BamHⅠdigestion.Results The target gene IE1 fragment about 1476bp was obtained and IE1 was successfully subcloned into pEGFP-C1 vector.Conclusion The recombinant eukaryotic expression vector pEGFP-C1-IE1 was successfully constructed.

Abstract:A Full length cDNA of MxIrt1 gene from the iron-stressed root cDNA library of Malus xiaojinensis Chent et Jiang was cloned using the RACE method with primers designed based on the conserved domain sequences of plant IRT gene families. The recombinant prokaryotic expression vector pEIrt was constructed using vector pET30a. Restriction endonuclease analysis, PCR amplifying and sequencing confirmed that the construction was correct and had no base mutant. The prokaryotic expression analysis was carried out through transformation of E.coli (BL21). The SDS-PAGE electrophoresis analysis showed that the best expression was induced by 30℃ and 0.5 mmol·L-1 IPTG, under which a 40 kD recombinant protein was produced. These results will provide a foundation for further purifying and identifying objective protein.

Abstract:Objective To construct the eukaryotic expression vector of pEGFP-IRX1, and observe the IRX1 protein expression and its sub-cellular localization in SGC-7901 gastric cancer cell line. Methods The coding sequence of IRX1 1443 bp was obtained by PCR amplification,then the PCR product was in-serted into the pGEM-Teasy vector. After confirming the DNA sequence, IRX1 amplification product was sub-cloned into pEGFP-N1 vector. Finally, an IRX1 gene eukaryotic expression vector was obtained. The pEGFP-IRX1 expression vector was transfected into SGC-7901 gastric cancer cell line by Lipofectamine 2000,and the expression of pEGFP-IRX1 was detected by GFP fluorescence. Results The recombinant vector was highly expressed in SGC-7901 cells. The green fluorescence of EGFP-1RX1 fusion protein was clearly localized in nuclei of gastric cancer cells. Conclusion pEGFP-IRX1 expression vector was con-structed successfully, and the IRX1 protein was mainly observed in nuclei,which settled a basis for further functional study of gene IRX1 in gastric cancer.

Abstract:The full length of the maize γ-zein gene amplified by PCR from PROK.TG1LK plasmid was ligated into the intermediate vector pMD18-T, to obtain the recombinant plasmid named pMD18-γ-zein. After the recombinant plasmid and the improved pCAMBIAI1302,respectively were digested by Nco I and Bgl II restriction endonucleases, the plant expression vector pCB-GFP-γ-zein with the gene of green fluorescent protein (GFP) driven by 35S was built following the procedures of recovery, ligation, transformation and identification. The fusion of GFP-γ-zein gene was integrated into the onion epidermal cells by using the Biolistic approach; the fusion gene was expressed in onion epidermal cell wall, nucleus and cytoplasm confirmed by confocal microscopy.

Abstract:The total length sequence of γ-zein gene in pROK.TG1LK plasmid was amplified by PCR. The fragment was cloned into pMD18-T middle vector, and a new recombined vector named pMD18-T-zein was obtained. A new plant expression vector named pCB-GFP-γ-zein in which the reporter gene GFP is droved by 35s was constructed after cutting two vector pMD18-γ-zein and pCAMBIAI1302 with NcoⅠ and BglⅡ restriction enzymes, subsequently reclamation, ligation, transformation and identification. The recombined plant expression vector was incorporated into epidermic cell of onion by gene gun method and the fusion gene was transient expressed. The result of transient expression showed that the gene located in cell wall, cell, nucleus and cytoplasm.

Abstract:A pair of primers was designed according to the sequences published by Genbank in order to amplify Isopentenyl Transferase Gene(ipt gene) of the plasmid in Agrobacterium tumefaciens(strain C58).The gene was amplified by PCR and cloned into pMD18-T vector,then was digested and inserted into the pBI121 vector,and transformed into the E.coli component DH5α.The expressing vector was indentified by PCR and then transformed into Agrobacterium tumefaciens EHA105.After the identification by PCR from EHA105 and pBI121-ipt vector,the specific expression vector of ipt was constructed successfully.The construction of recombinant pBI121-ipt plant expression vector provides a basis for the research on functions of ipt gene.

Abstract:It is often necessary to construct more than one recombinant plasmids when investigating the characteristics, physchemical features and functional mechanisms of genes or proteins. Repeated sub-cloning procedures including design of primers, enzyme digestion, ligation and verification of recombinant plasmids, have to be involved with. For this reason, it has become a tendency to developing new genetic vectors which can be used in multitude of experiments. Therefore, by using pIRES vector as a backbone, here we reported the construction of a mammalian expression vector: pCMV-Myc-IRES-EGFP which contains the N-terminal c-Myc epitope tag and the enhanced green fluorescent protein (EGFP) translated in an IRES-dependent manner. This novel vector can be used to testify the efficiency of cell transfection, to collect successfully transfected cell population via cytometry, to conduct transcription and translation in vitro, to purify target proteins or to trap their interactional proteins. The availability of this vector can facilitate function study of genes.

Abstract:Vector is an indispensable tool in the process of transgenic research.Researchers usually connect foreign target genes with vectors and import vectors into target plants (animals) by technical means to observe the role played by target genes.Therefore,many factors,such as the success of vector selection,the efficiency of vector transformation and expression,the difficulty of vector detection and the level of metabolic burden,determine the success or failure of the test to a certain extent.Therefore,the use of a simple,easy to transform,high expression,easy to detect and small metabolic burden is very important.Based on this,the vector pCRI1210 was successfully constructed by inserting an interference frame and replacing reporter gene on the basis of the commonly used vector pBI121.pBI121 was commonly used in the study of genetically modified (gm),but its multiple cloning site was relatively small,and GUS gene detection was more complicated.In order to increase its multiple cloning loci,enhance its expression efficiency,and simplify the detection method,a plant binary expression vector,named pCRI1210,was constructed on the basis of pBI121 vector,which contained CaMV 35S promoter,cotton β-tubulin gene introns,CaMV 35S polyA terminator and interference expression box with GFP as reporter gene.Functional analysis in tobacco showed that the interference express cassette could normally expressed exogenous gene and GFP gene.The vector had multiple cleavage sites in a TUB intron 5'and 3',so that target gene could be positive and reverse inserted and hairpin structure of dsRNA was builded.In addition,the vector carries multiple cloning sites at both 5'and 3'of the CaMV 35S promoter,which could easily exchange 35S promoter to study other promoter functions.pCRI1210 could be used as expression vector or interference vector.Through validation in tobacco,skin cells of the onion and the healing of the cotton embryo,it would be a very practical tool for transgenic research.

Abstract:Both crtB and tp gene were amplified by PCR and cloned into pMD18-T vector respectively. The pMD18-tp and pMD18-crtB were digested and the crtB and tp gene were linked together by the pBlue-script KS vector. By digestion of pSK-tp-crtB and pBI121,the GFP gene in pBI121 was replaced with tp-crtB gene. The plant expression vector pBI121-tp-crtB was obtained,which was transformed into the E. coli component DH5α. The expressing vector was identified by sequencing and then transformed into Agrobacterium tum efaciens EHA105,which was constructed successfully by PCR identification. Therefore the construction of recombinant pBI121-tp-crtB plant expression vector provides a basis for the research on functions of crtB gene.

Abstract:OBJECTIVE:To construct the eukaryotic expression vector for Max interacting protein 1 (Mxi1).METHODS:The full length cDNA of Mxi1 gene obtained from fetal lymphocyte and KG1 cells were inserted into plasmid pDs-red2-N1 respectively to generate pDs-red2-N1/Mri1 (wild/mutation type). Then the recombinant vector was transfected into Cos-7 cells via liposome. 48 hours post transfection, mRNA of Mri1 gene was detected by RT-PCR and Mxi1 protein expression was detected by flow cytometry and fluorescence microscope in the Cos-7 cells.RESULTS:The eukaryotic expression vector of Mxi1 was constructed and transfected into eukaryotic cells successfully. The expression of red fluorescence protein in the transfected Cos-7 cells was observed under fluorescence microscope which implied the expression of Mxi1. The transfect efficiency of both wild and mutation type were in a high level in 3 days after transfected, which lasted to 6 d. RT-PCR amplified the total RNA extracted from the transfected Cos-7 cells could find increased mRNA level of Mxi1 gene.CONCLUSION:We successfully constructed the eukaryote expression vector for Mri1 gene; Cos-7 cells transfected with the vector via liposome could express Mxi1 protein. These could be useful for the further study of the Myc gene modulation.

Abstract:A plant expression vector, pLZ13, was constructed by replacing the GUS gene in pBI121 with the intron-GUS (iGUS) gene from pCambia1301. GUS staining of Agrobacterium cells containing pCambia1301 or pLZ13 indicated that GUS staining was prevented in both cases. A transient expression study was carried out with kumquat tissues as samples, and no difference was observed for the GUS expression patterns driven by the CaMV 35S promoters from the newly constructed vector pLZ13 and pCambia1301. The vector pLZ13 was a plant expression vector, both suitable for transient gene expression and transgenic research.

Abstract:Objective To construct a recombinant eukaryotic expression vector containing mOX40-hIgG1Fc fusion gene.Methods The gene fragment encoding the human IgG1Fc was amplified by RT-PCR from the total RNA of heallthy person's leukocytes and then inserted into an eukaryotic vector pcDNA3.1.The encoding region of mOX40 extracellular domain was cloned with PCR from pIRES2-EGFP-OX40 and then constructed eukaryotic expression vector pcDNA3.1-mOX40-hIgG1Fc with recombinant vector pcDNA3.1-hIgG1Fc.The recombinant vector was identified by PCR,enzyme-digestion and DNA sequencing.Results The recombinant eukaryotic expression vector was conformed by PCR,enzyme-digestion and DNA sequencing,which were repectively consistent with hIgG1Fc and mOX40 extracellular gene reported in GenBank.Conclusion A eukaryotic expression vector of mOX40-hIgG1Fc-pcDNA3.1 was constructed successfully,so to construct a solid basis for further studying.

Abstract:【Objective】To construct the green fluorescent protein fusion vector of LYRM1 and investigate the subcellular localization of LYRM1. 【Methods】The complete coding sequence of LYRM1 gene was amplified by RT-PCR from human omental adipose tissue and was subcloned into pEGFP-N2 vector.The recombinant plasmid was then transfected into 3T3-L1 preadipocytes by liposome.The subcellular localization of LYRM1 gene was observed by laser scanning confocal microscopy. 【Results】The recombinant plasmid was confirmed by PCR,restriction enzyme digestion and sequencing.The green fluorescence protein was distributed in the whole cell transfected with pEGFP-N2,but mainly distributed in nucleus transfected with pEGFP-LYRM1. 【Conclusions】The green fluorescent protein fusion vector of LYRM1 is constructed successfully and expressed primarily in nucleus.

Abstract:To construct the eukaryotic expression vector of wagyu Myogenin(myogenin,MyoG) gene,the experiments uses wagyu fibroblasts genomic DNA to amplify MyoG gene.MyoG gene was inserted into the eukaryotic expression vector pIRES2-EGFP with EGFP and the internal ribosome site(IRES),is was transfected into fetal fibroblast cells of Luxi huangniu identification with double restriction enzyme digestion,MyoG gene expression were detected by RT-PCR and Western blot.The result showed that the constructed eukaryotic expression vector pIRES-EGFP-MyoG wer confirmed by restriction enzymolysis and could expressed efficiently in fetal fibroblast cells of Luxi huangniu,MyoG was expressed in them successfully.It may lay a good foundation to further study MyoG for biologzcal function and mechanism.

Abstract:In order to construct generally efficient cell immortalization vector, pTP-hTERT, we modified the traditional piggyBac (PB) transposon using artificial synthesis, PCR and enzyme digestion. The modified vector contained the necessary transposon elements, a PB transposase expression cassette, a co-expression selectable element and a human telomerase reverse transcriptase (hTERT) expression cassette. The co-expression selectable element had two markers, enhanced green fluorescent protein (EGFP) gene and puromycin-resistance (Puro) gene, linked by porcine teschovirus-1 2A peptide (P2A). To validate the functionality of vector elements, we transfected pTP-hTERT into HEK293 cell, selected the positive cell clones and then conducted RT-PCR, Western blotting (WB) and Tail-PCR, methylene blue staining and statistic analysis on selected cells. The results of sequencing and cell culture show that the pTP-hTERT was constructed successfully and the positive cell could be selected by puromycin. The WB results, P2A cutting EGFP and Puro fusion protein with high efficiency, reflected the selectable element worked. The sequencing result of Tail-PCR confirmed the vector integrated into the genome through transposition. The results of methylene blue staining and statistic analysis indicated the clone of positive cells triggered by pTP-hTERT significantly increased (P < 0.01) compared with control group. The construction of pTP-hTERT provides an efficient tool for establishing immortalized cell lines and a demonstration for building other eukaryotic plasmids.

Abstract:A novel plasmid pGS/2SS-M4GFP was constructed in the present study by recombination of GS/2SS gene and enhanced green fluorescent protein (M4GFP) sequence. The GS/2SS fusion gene encoding two copies of somatostatin genes was firstly introduced into pVAX-asd vector in which the kanamycin resistance cassette was replaced by the asd cassette. The M4GFP gene was then fused into 3′ end of GS/2SS gene in the proper reading frame. After purified, plasmid pGS/2SS-M4GFP was transfected into different cell lines derived from pig kidney and human cancer cells. The transcription process of GS/2SS gene was confirmed by RT-PCR, and the localization as well as expression of GS/2SS-M4GFP fusion protein was observed by confocal microscopy and ELISA. Transfection results revealed that sole M4GFP was localized within the cytosol and the nucleus, while fusion protein GS/2SS-M4GFP was localized only in the cytoplasm. Furthermore, it should be noted that subcellular localization of GS/2SS-M4GFP was not specific to one cell line, but appeared to be common across a variety of cell lines. These results provide for the first time valuable evidence that M4GFP is a versatile tool to trace GS/2SS protein and pave the way for further study on its tissue distribution and immunological mechanism in vivo.

Abstract:PurposeTo construct Pichia pastoris expression vector of human intestinal trefoil factor.MethodsTotal RNA was extracted from human colon mucosa,hITF cDNA was isolated by RT-PCR,hITF gene was amplified with PCR and cloned into Pichia pastoris vector pPIC9.The expression plasmid pPIC9/hITF containing AOX1 promotor and the sequences of αsecreting signal peptides was constructed.ResultsThe nucleotide sequence of hITF was the same as expected.ConclusionThe successful construction of the recombinant plasmid will be further studied in this field.

Abstract:In this study,EGFP was cloned into pGEX-4T-1 vector to construct prokaryotic expression vector pGEX-4T-EGFP,the recombinant plasmids were transformed into E.coli BL21(DE3) and induced with IPTG for protein expression,then the EGFP protein was detected by UV irradiation and SDS-PAGE.It can be used to provide a new report gene and selective marker for prokaryotic expression system.

Abstract:The study of genomics depends on quick and high-flux screening of functional genes, and the rapid development of plant transient expression technology makes it possible to express exogenous genes in plants to widen the investigation of functional genes. The use of green fluorescent protein gene can indicate the expression of target gene, therefore we constructed a plant transient expression vector containing two separate expression frames, one with rare restriction enzyme cleavage sites for cloning and expression of target gene and the other frame with enhanced green fluorescent protein gene. Meanwhile, the vector transiently expressed green fluorescent protein gene in tobacco leaves and detached tomato fruits by agrobacterium-mediated method, indicating the potent feasibility of this vector for transient expression.
