Abstract:Prussian blue nanoparticles (PBNPs) with favorable biocompatibility and unique properties have captured the attention of extensive biomedical researchers. A great progress is made in the application of PBNPs as therapy and diagnostics agents in biomedicine. This review begins with the recent synthetic strategies of PBNPs and the regulatory approaches for their size, shape, and uniformity. Then, according to the different properties of PBNPs, their application in biomedicine is summarized in detail. With modifiable features, PBNPs can be used as drug carriers to improve the therapeutic efficacy. Moreover, the exchangeable protons and adsorbability enable PBNPs to decontaminate the radioactive ions from the body. For biomedical imaging, photoacoustic and magnetic resonance imaging based on PBNPs are summarized, as well as the strategies to improve the diagnostic effectiveness. The applications related to the photothermal effects and nanoenzyme activities of PBNPs are described. The challenges and critical factors for the clinical translation of PBNPs as multifunctional theranostic agents are also discussed. Finally, the future prospects for the application of PBNPs are considered. The aim of this review is to provide a better understanding and key consideration for rational design of this increasingly important new paradigm of PBNPs as theranostics.

Abstract:The utilization of metal organic framework (MOF) nanocomposites in various biomedical applications has been widely studied because of their unique properties. In recently years, Prussian blue (PB) is widely used in clinical studied due to its biosafety, which has already been approved by USA Food and Drug Administration (FDA). Therefore, it is great urgent to systematically summarize the resent progress to better serve the broad community of researchers. This review focuses on the properties of PB and PB analogues (PBA), and their applications in biological field. A systemic introduction of the properties is presented, including abilities of photothermal, scavenging of reactive oxygen species and adsorption. A variety of biomedical applications, such as tumor therapy, anti‐bacterial infection, biosensor, bioimaging, excretion, ischemic stroke treatment, etc. have been discussed as well. Furthermore, the challenges, key factors, and the prospects in the clinical transformation of PB or PBA are simultaneously discussed. This review will provide reference and guidance for the design and application of PB and PBA in the future.

Abstract:Prussian blue (PB) nanoparticles (NPs) and Prussian blue analogs (PBAs) can form metal-organic frameworks through the programmable coordination of ferrous ions with cyanide. PB and PBAs represent a burgeoning class of hybrid functional nano-systems with a wide-ranging application spectrum encompassing biomedicine, cancer diagnosis, and therapy. A comprehensive overview of recent advancements is crucial for gaining insights for future research. In this context, we reviewed the synthesis techniques and surface modification strategies employed to tailor the dimensions, morphology, and attributes of PB NPs. Subsequently, we explored advanced biomedical utilities of PB NPs, encompassing photoacoustic imaging, magnetic resonance imaging, ultrasound (US) imaging, and multimodal imaging. In particular, the application of PB NPs-mediated photothermal therapy, photodynamic therapy, and chemodynamic therapy to cancer treatment was reviewed. Based on the literature, we envision an evolving trajectory wherein the future of Prussian blue-driven biological applications converge into an integrated theranostic platform, seamlessly amalgamating bioimaging and cancer therapy. Statement of Significance Prussian blue, an FDA-approved coordinative pigment with a centuries-long legacy, has paved the way for Prussian blue nanoparticles (PB NPs), renowned for their remarkable biocompatibility and biosafety. These PB NPs have found their niche in biomedicine, playing crucial roles in both diagnostics and therapeutic applications. The comprehensive review goes beyond PB NP-based cancer therapy. Alongside in-depth coverage of PB NP synthesis and surface modifications, the review delves into their cutting-edge applications in the realm of biomedical imaging, encompassing techniques such as photoacoustic imaging, magnetic resonance imaging, ultrasound imaging, and multimodal imaging.

Abstract:Prussian blue (PB) is an USA Food and Drug Administration (U.S. FDA)-approved antidote for the treatment of radioactive element poisoning such as thallium, which has been proved to have outstanding biosafety. Controllable morphology, size and structure of PB-based nanomaterials (PB NMs), and their excellent physicochemical properties, make them play an important role in the biomedical fields, such as drug delivery nano-systems, molecular imaging, and cancer treatment. In recent years, PB NMs have made great progress in cancer theranostics. Therefore, it is a great urgent to systematically summarize the recent progress to better serve the broad community of researchers. This review begins with the synthesis methods, regulatory approaches, and modification methods of PB NMs to achieve more uniform size and morphology. Next, according to the different properties of PB NMs, its application in the cancer theranostics has been gradually expanded, mainly including PB NMs for cancer imaging (single-modal and multi-modal imaging) and cancer therapy (monotherapy and combination therapy). For cancer imaging, single-modal imaging (e.g., photoacoustic (PA) imaging, ultrasound (US) imaging, magnetic resonance (MR) imaging), and multi-modal imaging based on PB NMs are systematically summarized, as well as strategies to improve the diagnostic effectiveness. For monotherapy, PB NMs are extensively used in photothermal therapy (PTT), sonodynamic therapy (SDT), and photodynamic therapy (PDT), owing to their excellent photothermal performance and unique physicochemical properties. Combined cancer therapy based on PB NMs to further enhance the cancer-killing efficiency is also systematically summarized. Moreover, the in vivo long-term toxicity of PB NMs is emphasized. Finally, the challenges, key factors, and the future prospects in the clinical transformation of PB NMs as multifunctional theranostic agents are simultaneously discussed. This review will provide reference and guidance for the design and application of PB NMs in the future bioapplications.

Abstract:In recent years, Prussian blue nanoparticles (PBNPs), also named Prussian blue nano-enzymes, have been shown to demonstrate excellent multi-enzyme simulation activity and anti-inflammatory properties, and can be used as reactive oxygen scavengers. Their good biocompatibility and biodegradability mean that they are ideal candidates for in vivo use. PBNPs are highly efficient electron transporters with oxidation and reduction activities. PBNPs also show considerable promise as nano-drug carriers and biological detection sensors owing to their huge specific surface area, good chemical characteristics, and changeable qualities, which might considerably increase the therapeutic impact. More crucially, PBNPs, as therapeutic and diagnostic agents, have made significant advances in biological nanomedicine. This review begins with a brief description of the synthesis methods of PBNPs, then focuses on the applications of PBNPs in tissue regeneration and inflammation according to the different properties of PBNPs. This article will provide a timely reference for further study of PBNPs as therapeutic agents.

Abstract:Prussian blue is an iron-cyanide-based pigment steadily becoming a widely used electrochemical sensor in detecting hydrogen peroxide at low concentration levels. Prussian blue nanoparticles (PBNPs) have been extensively studied using traditional ensemble methods, which only provide averaged information. Investigating PBNPs at a single entity level is paramount for correlating the electrochemical activities to particle structures and will shed light on the major factors governing the catalyst activity of these nanoparticles. Here we report on using plasmonic electrochemical microscopy (PEM) to study the electrochemistry of PBNPs at the individual nanoparticle level. First, two types of PBNPs were synthesized; type I synthesized with double precursors method and type II synthesized with polyvinylpyrrolidone (PVP) assisted single precursor method. Second, both PBNPs types were compared on their electrochemical reduction to form Prussian white, and the effect from the different particle structures was investigated. Type I PBNPs provided better PEM sensitivity and were used to study the catalytic reduction of hydrogen peroxide. Progressively decreasing plasmonic signals with respect to increasing hydrogen peroxide concentration were observed, demonstrating the capability of sensing hydrogen peroxide at a single nanoparticle level utilizing this optical imaging technique.
Achieving Ultrasmall Prussian Blue Nanoparticles as High-Performance Biomedical Agents with Multifunctions

Abstract:Prussian blue nanoparticles (PBNPs), which belong to the iron-based metal–organic frameworks, are important biomedical agents. Reducing the size of PBNPs can bring improved functional properties, but unfortunately, has been a long-standing challenge. Herein, sub-5 nm ultrasmall PBNPs (USPBNPs) were successfully synthesized by using ethanol/water mixture as the solvent and polyvinyl pyrrolidone (PVP) as the surface capping agent. Adjusting the ethanol/water ratio is not only able to control the nucleation time and size of PBNPs but also tune the conformation of PVP molecules so as to prevent interparticle attachment and enlargement. At an ethanol/water ratio of 3:1, highly stable USPBNPs with a size of ∼3.4 nm were synthesized. Due to their large specific surface area, they demonstrated high peroxidase-like and catalase-like activities, which outperform PBNPs synthesized by a conventional method. Recent progress in Prussian blue films: Methods used to control regular nanostructures for electrochemical biosensing applications

Abstract:In the last decade, Prussian blue (PB) has attracted increased scientific interest in various research fields, such as fuel cells, gas separation and pollution treatment. Due to its advanced catalysis, biocompatibility, selectivity and stability, PB has been widely used in biosensor construction. However, the formation of regular PB nanostructures is challenging due to its fast crystallization rate. Recently, developments in this research area have increased due to emerging novel synthesis methods in nanoscale technology. Various regular nanostructures of PB films that show superior biosensing performance have been prepared. In this review, recent research progress in PB nanostructures is summarized, with special emphasis on the methodology of nanostructure control. The mechanism and key factors in regular PB crystallization are also discussed for each synthesis method. The performance of PB nanostructure-based biosensors is compared with others to show the advantages of nanostructure control. The methodology discussed in this review not only include the regular growth of PB films, but also provides information on the nanostructure control of more crystalline materials, including PB analogues, noble metals, metal oxides and coordination compounds. In addition to biosensing applications and the development of more advanced nanostructures, PB has also shown increased advanced properties in other scientific areas.

Abstract:In recent years, the researches on micro/nanoscaled Prussian blue (PB) and its analogues (PBAs) have received widespread attention due to their high specific surface area, multiple active sites, and diverse element types. Benefiting from their own structures, PB/PBAs materials are often used in fields such as electrode materials, catalysts, electrochromic materials, and cancer treatment. At present, researchers are still continuously exploring the potential of PB/PBAs materials. The synthesis strategy of PB/PBAs is a hot topic of discussion, which is of great significance to further explore the application value of PB/PBAs materials and large-scale commercial production. This work reviews the research progress of PB/PBAs materials in recent years. We focus on the synthesis strategies of PB/PBAs materials and discuss their structural properties and morphology control. Finally, we analyze existing vacancies and future development trends to provide a reference for future research.

Abstract:Owing to its extraordinary biocompatibility and promising applications, PB has been highly sought after. It is regarded as a valuable research target in the fields of sensing, energy and catalysis, particularly sensing. The introduction of PB improves the conductivity, stability, and electrocatalytic activity of biosensors, yet these properties are highly dependent on the morphology, size, chemical composition, etc. Therefore, to meet various requirements, engineering of the PB structure on the nanoscale to construct sensors with high stability, high sensitivity, and excellent electrochemical catalytic performance has received extensive attention. This review aims to offer an up-to-date comprehensive summary of PB materials with various nanostructures, with an emphasis on purpose-driven structure design and application to wearable devices/analytical chemistry. The contents of this review are based on a balanced combination of our studies and selected research done by worldwide academic groups. First, driven by the aim of breaking the limitations caused by the inherent defects of PB, various approaches, such as combining PB with functional materials or substances and constructing hollow or core-shell structures, to improve the electrochemical properties are explored. Then, we evaluate some key applications of PB-based nanomaterials in the area of electrochemical sensors for healthcare, environment monitoring and the food industry based on their different properties. Moreover, flexible wearable sensors based on PB are also discussed. The purpose of this review is to provide a better understanding and rational design of the increasingly important material PB as a key candidate for sensing materials.

Abstract:Prussian blue (PB) and its analogue (PBA) are a kind of representative cyanide-based coordination polymer. They have received enormous research interest and have shown promising applications in the electrochemical sensing field due to their excellent electrochemical activity and unique structural characteristics including open framework structure, high specific surface area, and adjustable metal active sites. In this review, we summarize the latest research progress of PB/PBA as an electrochemical sensor in detail from three aspects: fabrication strategy, synthesis method and electrochemical sensor application. For the fabrication strategy, we discussed different fabrication methods containing the combination of PBA and carbon materials, metal nanoparticles, polymers, etc., respectively, as well as their corresponding sensing mechanism for improving performance. We also presented the synthesis methods of PB/PBA materials in detail, such as: coprecipitation, hydrothermal and electrodeposition. In addition, the effects of different methods on the morphology, particle size and productivity of PB/PBA materials are also concluded. For the application of electrochemical sensors, the latest progress of such materials as electrochemical sensors for glucose, H2O2, toxic compounds, and biomolecules have been summarized. Finally, we conclude remaining challenges of PB/PBA-based materials as electrochemical sensors, and provide personal perspectives for future research in this field.

Abstract:With unique physical properties, chemical properties, and biological effects, magnetic nanomaterials are important functional materials in many fields. In the past decades, iron based magnetic nanomaterials have attracted much attention in the biomedicine field due to their superior magnetic properties and great potential in biomedical applications. In particular, magnetic iron oxide nanoparticles (MIONPs) have been playing a crucial role in the biomedicine field because of their diagnostic and therapeutic functions. Meanwhile, MIONPs are benign, low toxic, biocompatible, and biodegradable, so they are the only inorganic magnetic nanomaterials approved by the U.S. Food and Drug Administration (FDA) for clinical use at present. In this review, we mainly introduce the progress in the preparation of iron based magnetic nanomaterials for biomedical applications, including pure iron nanoparticles, iron-based alloy nanoparticles, and MIONPs, with a focus on MIONPs. Also, we summarize the preparation methods of MIONPs and point out the importance of their developments.

Abstract:Prussian blue nanoparticles (PBNPs) have attracted increasing research interest in immunosensors, bioimaging, drug delivery, and application as therapeutic agents due to their large internal pore volume, tunable size, easy synthesis and surface modification, good thermal stability, and favorable biocompatibility. This review first outlines the effect of tumor markers using PBNPs-based immunosensors which have a sandwich-type architecture and competitive-type structure. Metal ion doped PBNPs which were used as T1-weight magnetic resonance and photoacoustic imaging agents to improve image quality and surface modified PBNPs which were used as drug carriers to decrease side effects via passive or active targeting to tumor sites are also summarized. Moreover, the PBNPs with high photothermal efficiency and excellent catalase-like activity were promising for photothermal therapy and O2 self-supplied photodynamic therapy of tumors. Hence, PBNPs-based multimodal imaging-guided combinational tumor therapies (such as chemo, photothermal, and photodynamic therapies) were finally reviewed. This review aims to inspire broad interest in the rational design and application of PBNPs for detecting and treating tumors in clinical research.

Surface Modified Iron Oxide Nanoparticles As Fe Source Precursor to Induce the Formation of Prussian Blue Nanocubes.

Abstract:Nano-sized Prussian blue (PB) cubes were synthesized at room temperature by simply stirring the mixture of surface modified iron oxide nanoparticles (IONPs) and potassium ferrocyanide in an aqueous acid solution. The nanocubes were characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM) and X-ray diffraction (XRD). The influence of different molecules modified on the surface of IONPs on the cube formation was discussed. The surface modification with dimercaptosuccinic acid (DMSA), 3-aminopropyltriethoxysilane (APTS) and citric acid (CA) all displayed a key role in the formation precess of PB nanocubes, but which could not be formed as bare IONPs or Fe3+ were used as precursor. Combined with the reaction process tracing with UV-vis absorption spectroscopy and TEM, a possible kinetically controlled growth mechanism was proposed where slower formation rate of amorphous PB due to very low release rate of Fe ions from the surface modified IONPs and subsequent recrystallization are responsible for the obtained PB nanocubes. The peroxidase-like catalytic activity of the synthesized nanocubes was investigated and catalysis was found to follow Michaelis-Menten kinetics. The potential of using such PB nanocubes as an effective MRI contrast agent was also demonstrated.

Abstract:Prussian blue analogs (PBAs), the oldest artificial cyanide-based coordination polymers, possess open framework structures, large specific surface areas, uniform metal active sites, and tunable composition, showing significant perspective in electrochemical energy storage. These electrochemically active materials have also been converted to various functional metal containing nanomaterials, including carbon encapsulated metals/metal alloys, metal oxides, metal sulfides, metal phosphides, etc. originating from the multi-element compositions as well as elaborate structure design. In this paper, a comprehensive review will be presented on the recent progresses in the development of PBA frameworks and their derivatives based electrode materials and electrocatalysts for electrochemical energy storage and conversion. In particular, it will focus on the synthesis of representative nanostructures, the structure design, and figure out the correlation between nanomaterials structure and electrochemical performance. Lastly, critical scientific challenges in this research area are also discussed and perspective directions for the future research in this field are provided, in order to provide a brand new vision into the further development of novel active materials for the next-generation advanced electrochemical devices.

Abstract:Prussian Blue Nanozymes (PBNZs) have emerged as highly efficient agents for reactive oxygen species (ROS) elimination, owing to their multiple enzyme-like properties encompassing catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD) activities. As a functional nanomaterial mimicking enzyme, PBNZs not only surmount the limitations of natural enzymes, such as instability and high manufacturing costs, but also exhibit superior stability, tunable activity, low storage expenses, and remarkable reusability. Consequently, PBNZs have gained significant attention in diverse biomedical applications, including disease diagnosis and therapy. Over the past decade, propelled by advancements in catalysis science, biotechnology, computational science, and nanotechnology, PBNZs have witnessed remarkable progress in the exploration of their enzymatic activities, elucidation of catalytic mechanisms, and wide-ranging applications. This comprehensive review aims to provide a systematic overview of the discovery and catalytic mechanisms of PBNZ, along with the strategies employed to modulate their multiple enzyme-like activities. Furthermore, we extensively survey the recent advancements in utilizing PBNZs for scavenging ROS in various biomedical applications. Lastly, we analyze the existing challenges of translating PBNZs into therapeutic agents for clinical use and outline future research directions in this field. By presenting a comprehensive synopsis of the current state of knowledge, this review seeks to contribute to a deeper understanding of the immense potential of PBNZs as an innovative therapeutic agent in biomedicine.

Abstract:Coordination polymers (CPs) have gained a great deal of attention over the years because of their beneficial uses in catalysis, drug-delivery, and energy storage and separation. Recent research efforts have been devoted to architecting CPs that satisfy the needs of applications that have recently been developed. Herein, we announce a survey of the latest developments in the synthesis of Prussian blue (PB) and Prussian blue analogues (PBAs) cyano-bridged CPs. The recent synthetic concepts and the modest control over the shape and size of particles are demonstrated to the present. Various ways have been taken to construct nanostructured inorganic materials from PB and PBAs. We surveyed one such example: the thermal decomposition of these materials in different environments. The regulated thermal treatment of their nanostructures as precursors, in air or inert atmosphere, yields nanoporous metal oxides and carbides or alloys, respectively. The resulting metal frameworks participate as metal sources, and the organic components, which can be removed by a simple process of calcination, produce pores. The original morphologies are almost retained, even after the thermal treatments. This strategy has proved to be a promising solid-state chemistry for the fabrication of nanoporous metal oxides and carbides or alloys with the similar morphology.

Homogeneous and Nanogranular Prussian Blue to Enable Long-Term-Stable Electrochromic Devices

Abstract:The increasing demand for the state-of-the-art electrochromic devices has received great interest in synthesizing Prussian blue (PB) nanoparticles with a uniform diameter that exhibit excellent electrochromism, electrochemistry, and cyclability. Herein, we report the controllable synthesis of sub-100 nm PB nanoparticles via the coprecipitation method. The diameter of PB nanoparticles can be modulated by adjusting the reactant concentration, the selection of a chelator, and their purification. The self-assembled nanogranular thin films, homogeneously fabricated by using optimized PB nanoparticles with an average diameter of 50 nm as building blocks via the blade coating technique enable excellent performance with a large optical modulation of 80% and a high coloration efficiency of 417.79 cm2 C-1. It is also demonstrated by in situ and ex situ observations that the nanogranular PB thin films possess outstanding structural and electrochemical reversibility. Furthermore, such nanogranular PB thin films can enjoy the enhanced long-term cycling stability of the PB-WO3 complementary electrochromic devices having a 91.4% optical contrast retention after 16,000 consecutive cycles. This work provides a newly and industrially compatible approach to producing a complementary electrochromic device with extraordinary durability for various practical applications.

Abstract:As a presentative material of metal-organic frameworks (MOFs), Prussian blue (PB) draws great attention due to its low cost, facile preparation processes and hollow framework structure. Previous studies show that the PB-derived nanomaterials can inherit part of their features, exhibiting large surface area, inter-connected pores and hierarchical pore sizes, which may promote charge transfer when utilized in energy storage and conversion system. By tuning synthesis conditions (e.g. temperature and atmosphere), nano-materials with desirable structures and properties can be obtained, leading to various usage areas for energy storage and conversion applications (e.g. batteries, supercapacitors and catalysts). In this work, for the first time, various products obtained under different preparation conditions and their applications are summarized in detail in order to offer guiding significance for synthesizing nanoporous materials with special purposes.
