Abstract:Background: Synaptic plasticity is the basis of memory and hypersensitive state of excitatory neurons in central nerve system (CNS), it also participates in central mechanism of visceral pain.We hypothesized that the synaptic plasticity and hypersensitive state may also exist in enteric nerve system (ENS) and contribute to peripheral mechanism of visceral hypersensitivity, which is an important biomarker of irritable bowel syndrome (IBS), since the ENS and the CNS have some similar structure and function.However, the molecular mechanisms remain elusive.Aims: We aimed to investigate the role of ephrinB2/ephB2 kinase signaling, which played an important role in axon guidance and angiogenesis, in modulating synaptic plasticity of ENS and visceral hypersensitivity in IBS.Methods: Post infectious-IBS models were induced by Trichinella spiralis, IgG-Fc and ephB2-Fc (inhibitor of ephrinB2/ephB2 signaling) were intraperitoneal administrated in tow groups respectively.The visceral sensitivity was assessed via abdominal withdrawal reflex (AWR) scores and mesenteric afferent nerve activity recording.The expression of ephrinB2 and ephB2 were detected by western blot and immunofluorescence.The markers of mature fiber (Tuj-1), newborn fiber (GAP43) and synapse (PSD95, synaptophysin and synaptohrevin) and synaptic plasticity-related immediate early factors (c-fos, arc, bdnf and ptgs2) were assessed.The morphologically synaptic plasticity was investigated via transmissionelectron microscopy.The functionally synaptic plasticity involving the expression and phosphorylation of NMDAR1 and NMDAR2B receptor were also detected.Results: EphrinB2/ephB2 were expressed in enteric fibers and increased in colon of PI-IBS rats, which had lower pain threshold and increased firing frequency of mesenteric afferent nerves.EphrinB2/ephB2 inhibition obviously improved the visceral hypersensitivity in PI-IBS rats.There were increased density of newborn fibers and sprout of synapses, and increased number of presynaptic vesicles and postsynaptic density in the colon of PI-IBS, while ephB2-Fc could inhibit the changes.The expressions of GAP43, PSD95, synaptophysin, NMDAR1, immediate early factors c-fos and arc were increased in the colon of PI-IBS.The synaptic plasticity of ENS in PI-IBS was blocked by inactivation of ephB2 receptors.Conclusions: The synaptic plasticity of ENS may participated in the formation of visceral hypersensitivity in PI-IBS, and ephrinB2/ephB2 activation played an important role in this process.
