Abstract:The study of fast non-equilibrium solvent relaxation in organic chromophores is still challenging for molecular modeling and simulation approaches, and is often overlooked, even in the case of non-adiabatic dynamics simulations. Yet, especially in the case of photoswitches, the interaction with the environment can strongly modulate the photophysical outcomes. To unravel such a delicate interplay, in the present contribution we resorted to a mixed quantum-classical approach, based on quantum mechanically derived force fields. The main task is to rationalize the solvent reorganization pathways in chromophores derived from cyclocurcumin, suitable for light-activated chemotherapy to destabilize cellular lipid membranes. The accurate and reliable decryption delivered by the quantum-derived force fields points to important differences in the solvent reorganization, which further justify the different photo-isomerization quantum yields.

What problem does this paper attempt to address?
This paper aims to address the challenges in modeling and simulating the rapid non - equilibrium process of solvent reorganization of organic dye molecules after photo - excitation. Specifically, the research focuses on cyclocurcumin derivatives, a type of photoswitchable molecule. After being excited by visible light, how the interaction between them and the solvent affects their photophysical properties, especially the quantum yield of photoisomerization. These issues are crucial for the development of new drugs for light - activated chemotherapy (LAC) to destroy cell lipid membranes. ### Main research objectives: 1. **Understand the solvent reorganization path**: Through the method of quantum mechanically derived force fields (QMD - FFs), reveal the solvent reorganization path of cyclocurcumin derivatives after photo - excitation. This helps to explain the differences in the quantum yield of photoisomerization of different molecules in different solvents. 2. **Optimize the design of photoswitchable molecules**: Through detailed studies of natural cyclocurcumin (CYC) and its biomimetics (PYR), provide a theoretical basis for designing more effective photoswitchable molecules, especially those with high absorption efficiency, high quantum yield, and environmental independence in the visible and near - infrared regions. 3. **Simulate the photo - induced process**: Use a mixed quantum - classical approach, through non - equilibrium molecular dynamics (neqMD) to simulate the early solvent reorganization process after photo - excitation, so as to better understand the impact of photo - induced structural changes on biological systems. ### Research background: - **Applications of photoswitchable molecules**: Photoswitchable molecules have received extensive attention due to their potential applications in photodynamic therapy (PDT), smart materials, chemical or diagnostic probes, etc. - **Advantages of cyclocurcumin derivatives**: Cyclocurcumin derivatives have important photophysical properties, such as reversible cis - trans isomerization and a relatively high two - photon absorption cross - section, making them potential candidates for light - activated chemotherapy. - **Existing challenges**: Traditional continuum approaches and non - adiabatic dynamics simulations have limitations in describing non - equilibrium processes after photo - excitation, especially in capturing the anisotropic reorganization of the solvent. ### Methods and techniques: - **Quantum mechanically derived force fields (QMD - FFs)**: Generate force field parameters for specific electronic states through the JOYCE program. These parameters can accurately describe the geometric configurations and energy surfaces of molecules in different electronic states. - **Non - equilibrium molecular dynamics (neqMD)**: Simulate the solvent reorganization process after photo - excitation by switching the QMD - FF of the molecule from the ground state (S0) to the excited state (S1). - **Multiscale simulation**: Combine quantum mechanics and classical molecular dynamics methods to simulate the behavior of photoswitchable molecules in complex environments, including the explicit description of the solvent and the structural changes of the molecules. ### Expected results: - **Reveal the solvent reorganization mechanism**: Through detailed simulation results, reveal the solvent reorganization paths of cyclocurcumin derivatives in different solvents and their impact on the quantum yield of photoisomerization. - **Guide the design of new drugs**: Provide theoretical support for the development of efficient photoswitchable molecules, especially those that can maintain a high quantum yield in different environments. - **Promote the development of light - activated chemotherapy**: Through in - depth understanding of the interaction between photoswitchable molecules and biological systems, promote the development of light - activated chemotherapy technology and improve its application potential in cancer treatment and the treatment of other diseases. In conclusion, this paper uses advanced computational methods to deeply explore the solvent reorganization process of cyclocurcumin derivatives after photo - excitation, providing an important theoretical basis for designing more effective photoswitchable molecules.
