Abstract:Indole-based heterocycles containing sulfur or nitrogen atoms are important frameworks that occur widely in biological active natural products and drugs. Consequently, several novel chemical transformations and new catalytic strategies have been developed for the preparation of these skeletons in a highly efficient and selective manner. This review focuses on recent advancements for the construction of indole-fused heterocyclic compounds by using iminoindolines, 3-diazoindol-2-imines, 2-thioxoindolines, and their derivatives as starting materials.

Abstract:3-Diazoindolin-2-imines were constructed from indoles and sulfonylazides via an electronically matched 1,3-dipolar cycloaddition and a subsequent dehydroaromatization/ring-opening cascade. The reaction between 3-substituted indoles and sulfonyl azides provided 2-aminoindoles, while 2-substituted indoles furnished 3-aminoindoles. Moreover, 2,3-diaminoindoles could be prepared from the resulting 3-diazoindolin-2-imines and secondary amines via a rhodium-catalyzed amination. Further extension of 2,3-diaminoindoles led to the formation of imidazo[4,5-b]indoles.

Abstract:The development and utilization of new dienes and dienophiles for the controlled synthesis of isoquinuclidines is highly appealing. Herein, we describe a novel strategy for diastereoselective synthesis of indoline-fused isoquinuclidines via copper-catalyzed dearomative Diels-Alder reaction of cyclic amidines with indoles. This protocol avoids the use of unstable DHPs and activated alkenes, offering a more efficient and selective approach to synthesize isoquinuclidines.

Abstract:Synthetically valuable 3-haloindol-2-amines were prepared from readily available 3-diazoindolin-2-imines and a variety of alkyl halides through the palladium-catalyzed reaction. The broad substrate scope of both diazo compounds and alkyl halides, mild reaction conditions, and high efficiency are the main characters of this method. The synthesized 3-haloindol-2-amines can be conveniently transformed to more complex indole derivatives.

Abstract:A novel copper‐catalyzed dimerization of sulfoxonium ylides with 3‐diazoindolin‐2‐imines is described. Our reaction discriminates between the reactivity of sulfoxonium ylides and diazo compounds as both these classes are carbene precursors as well as nucleophiles. A wide range of spiro[cyclopropane‐1,3′‐indolin]‐2′‐imines were obtained in good to excellent yields with high diastereoselectivity under mild reaction conditions.

Abstract:The rhodium-catalyzed reactions of 3-diazoindolin-2-imines with β-enamino esters furnished 2,3-diaminoindoles in excellent yields with wide functional group tolerance. The synthesized 2,3-diaminoindoles could be converted into 5H-pyrazino [2,3-b]indoles via a sequential dehydrogenation/6π-ERC/aromatization process.

Abstract:This review paper provides a comprehensive overview of recent advancements in the synthesis, reactivity, and applications of N -isoindoline-1,3-dione heterocycles. These aromatic compounds, characterized by an isoindoline nucleus and carbonyl groups at positions 1 and 3, have gained significant attention for their potential use in diverse fields such as pharmaceutical synthesis, herbicides, colorants, dyes, polymer additives, organic synthesis, and photochromic materials. The review focuses on the period from 2015 to 2024, highlighting the diverse synthetic strategies employed to access N -isoindoline-1,3-diones derivatives. This review discusses the innovative methodologies and transformations that have enabled the efficient construction of these compounds with various substitution patterns. Furthermore, the reactivity of N -isoindoline-1,3-diones and their potential applications in different fields are explored. The paper emphasizes the importance of understanding the structure–activity relationships and biological properties of N -isoindoline-1,3-dione derivatives, with the aim of unlocking their potential as therapeutic agents. Additionally, the review underscores the need for sustainable and environmentally friendly synthetic approaches in this field.

Abstract:A novel and facile approach to 3-allyl-2-aminoindoles via the copper-catalyzed reaction of 3-diazoindolin-2-imines with allyltrimethylsilane is described. The domino reaction proceeds efficiently in one-pot and shows broad substrate scope with respect to the diazo substrates. Furthermore, the synthesized 3-allyl-2-aminoindoles can be conveniently transformed into 3-allyl-3-hydroxylindolin-2-imines and 3-allyl-3-fluoroindolin-2-imines.

Abstract:Symmetrical 3,3-diaryl-2-iminoindoles were prepared from 3-diazoindolin-2-imines and indoles via a copper-catalyzed cascade arylation/dehydrogenative cross-coupling process. By controlling the molar ratio of reactants and the operation procedure, 3-aryl-2-aminoindoles and asymmetrical 3,3-diaryl-2-iminoindoles could be approached.

Abstract:Rhodium-catalyzed reactions of 3-diazoindolin-2-imines with aromatic aldehydes and α,β-unsaturated aldehydes respectively furnished (E)-2-aminomethylene-3-oxoindoles and pyranoindoles in moderate to excellent yields with nice functional group tolerance. The synthesis of indoles and indole-involved synthetic methodologies in organic chemistry receive considerable attention all the time. Indole-N-carboxylic acids and derived indole-N-carboxamides are intriguing compounds, which have been widely used in organic synthesis, especially in multicomponent reactions and C-H functionalization of indoles. This Minireview summarizes the advances of novel reactions involving indole-N-carboxylic acids and indole-N-carboxamides in organic chemistry, and discusses the synthetic potential and perspective of this field.

Abstract:An efficient and facile synthesis of 2,3′-biindoles through Co(III)-catalyzed direct C–H coupling of N-pyrimidinylindoles with 3-diazoindolin-2-imines is reported. This method provided 2,3′-biindoles under mild reaction conditions and in moderate to excellent yields. The reaction tolerated a number of functional groups and could be scaled up with practical usage.

Abstract:A Lewis acid Sc(OTf)(3)-catalyzed annulation reaction of vinyl diazoacetates with in situ formed α,β-unsaturated imines for the preparation of indole-fused tricyclic rings has been reported. This strategy involves a sequential addition/rebound addition process of vinyl diazoacetates and an in situ dedinitrogenation. This annulation protocol features low-cost catalysts, mild reaction conditions, and facilely prepared substrates.

Abstract:A 1,3-dipolar cycloaddition reaction of arynes with 3-diazoindolin-2-ones under mild conditions in excellent yields has been developed, which allows facile access to a library of labile spiro[indazole-3,3′-indolin]-2′-ones. Spiro[indazole-3,3′-indolin]-2′-imines could be obtained as well following the same protocol. The isomerization reaction of spiro[indazole-3,3′-indolin]-2′-ones under thermal or acidic conditions has been efficiently achieved to afford a wide range of indazolo-[2,3-c]quinazolin-6(5H)-ones and the one-pot synthesis of indazolo-[2,3-c]quinazolin-6(5H)-ones from arynes and 3-diazoindolin-2-ones is also described. Whereas, spiro[indazole-3,3′-indolin]-2′-imines could not undergo the same rearrangement.
