Abstract:Visible light promoted difunctionalization of alkynes is reviewed. The difunctionalization reaction is achieved by different reagents. Radicals such as carbon (sp3), carbon (sp2), and other heteroatom (P, S, N, Se, O, and halide) radicals initiated by visible light can undergo radical addition to a carbon-carbon triple bond. Upon further transformation, the desired difunctionalized products are obtained. Some organometallic complexes can be activated by visible light; the difunctionalization of alkynes is catalyzed by these species. Other reagents like 1,3-dipole precursors could also react with alkynes to give difunctionalization products; here, the 1,3-dipole derivatives are obtained by visible light photocatalysis. So far, the strategy has been succeeded in the formation of C–C bonds and C–X bonds. Several valuable chemical skeletons have been constructed under mild conditions. However, high regio- and stereoselectivities in some direct difunctionalization methodologies are yet to be achieved.

Abstract:We reported the visible-light-mediated photoredox-catalyzed oxidative radical-polar crossover and 1,5-hydrogen atom transfer combined site-selective remote C(sp3)-N cross-coupling alkylamination of alkenes. Various anilines and hydroxamides (1,5-hydrogen atom transfer reagents) could be tolerated. The mechanistic studies indicated the radical nature of the reaction and the indispensability of light and photocatalyst. Stern-Volmer fluorescence quenching and cyclic voltammetry experiments have been used to outline the proposed reaction pathway.

Abstract:Photochemistry has ushered in a new era in the development of chemistry, and photoredox catalysis has become a hot topic, especially over the last five years, with the combination of visible-light photoredox catalysis and radical reactions. A novel, simple, and efficient radical oxidative decarboxylative coupling with the assistant of the photocatalyst [Ru(phen)(3)]Cl-2 is described. Various functional groups are well-tolerated in this reaction and thus provides a new approach to developing advanced methods for aerobic oxidative decarboxylation. The preliminary mechanistic studies revealed that: 1)an SET process between [Ru(phen)(3)](2+)* and aniline play an important role; 2)O-2 activation might be the rate-determining step; and 3)the decarboxylation step is an irreversible and fast process.

Abstract:The synthesis and transformations of alkynes have held an important position in the field of organic synthesis. How to rapidly introduce alkynyl into organic molecules has been of great interest. Photoredox catalysis has been widely used in organic synthesis because it can generate high reactive free radical intermediates under mild conditions using green and clean energy. Visible light photocatalytic alkynylation has flourished in the last decade, which is believed to be a significant complement to the classic transition-metal-catalyzed Sonogashira reaction. This alkynylation could take place via in situ generated copper acetylide complex from terminal alkynes and copper, which also occurs through alkynyl radical species or radical addition elimination of non-terminal alkynes. Moreover, it was a good supplement to Sonogashira reaction that various radical precursors could be applied to alkynylation by visible-light photoredox catalysis. Based on the type of bonding of alkynylation, the research progress of alkynylation reactions by visible-light photoredox catalysis is reviewed, and the future development of this field is prospected.

Abstract:Visible-light-induced three-component 1,2-alkylarylation of alkenes and alkyl radical addition/cyclization of acrylamides have been realized via a photocatalytic halogen-atom transfer (XAT) process. This metal-free protocol utilizes readily available tertiary alkylamine as both an electron donor and XAT reagent for the activation of alkyl halides using naphthalimide (NI)-based organic photocatalysts. This process features broad substrate scope and good functional group tolerance under mild conditions, and could be effectively applied to a variety of medicinally relevant substrates.

Abstract:Selectivity control of the reactions of propargylic and/or allenylic radicals remains particularly elusive. Here, a visible light-mediated Cu-catalyzed protocol for the highly selective syntheses of tri- and tetra-substituted allenols from alkynol derivatives has been developed. Synthetic potentials of the products have been demonstrated. The control experiments, Stern-Volmer quenching, and cyclic voltammetry studies support a reaction pathway involving the generation of allenyl radicals via photo-mediated N-O bond scission, intramolecular 1,5-HAT process, the resonance to propargylic radicals, and coupling. Non-common intramolecular 1,6-HAT process has also been observed.

Abstract:Visible light-driven pyridoxal radical biocatalysis has emerged as a promising strategy for the stereoselective synthesis of valuable noncanonical amino acids (ncAAs). Previously, the use of well-tailored photoredox catalysts represented the key to enable efficient pyridoxal phosphate (PLP) enzyme-catalyzed radical reactions. Here, we report a PLP-dependent threonine aldolase-catalyzed asymmetric α-C-H alkylation of abundant amino acids using Katritzky pyridinium salts as alkylating agents. The use of engineered threonine aldolases allowed for this redox-neutral radical alkylation to proceed efficiently, giving rise to challenging α-trisubstituted and -tetrasubstituted ncAA products in a protecting-group-free fashion with excellent enantiocontrol. Mechanistically, this enantioselective α-alkylation capitalizes on the unique reactivity of the persistent enzymatic quinonoid intermediate derived from the PLP cofactor and the amino acid substrate to allow for novel radical C-C coupling. Surprisingly, this photobiocatalytic process does not require the use of well-established photoredox catalysts and operates through an unconventional photoinduced radical generation involving a PLP-derived aldimine. The ability to develop photobiocatalytic reactions without relying on classic photocatalysts or photoenzymes opens up new avenues for advancing stereoselective intermolecular radical reactions that are not known in either organic chemistry or enzymology.

Abstract:A mild and general strategy for the direct generation of alkoxy radicals from simple aliphatic alcohols enabled by visible-light-induced photoredox/persistent radical cation dual catalysis is developed. In this report, we present a mild and general strategy for the direct generation of alkoxy radicals from simple aliphatic alcohols enabled by visible-light-induced photoredox/persistent radical cation dual catalysis. The dual catalytic system can trigger alkoxy radical-mediated redox-neutral transformations, avoiding any metal catalysts and the requirement of proximal electron-rich arene moieties on the alcohol substrates. By employing this synergistic catalysis, the Giese reactions of diverse alcohols with electron-deficient alkenes via selective C–C bond scission of the corresponding alkoxy radical intermediates have been achieved. Different from the existing PCET and LMCT processes, mechanistic studies suggest the intermolecular reaction of thianthrene radical cations with alcohols and the subsequent single-electron reduction induced S–O bond homolysis of the formed 5-alkoxythianthrenium cations as the key steps to furnish alkoxy radicals.

Abstract:Allyl radicals served as a crucial intermediate in the radical functionalization of fundamental raw materials and received increasing attention. Despite the significance, allyl-radicals-participated radical cross-couplings were rare and mainly focused on couplings with aryl or benzyl radical analog. Herein, we realized radical 1,2-sulfonylacylation of allenes via unprecedented cross-coupling between NHCs-stabilized acyl radical and delocalized allyl radical. Cheap and stable sulfinates and readily accessible acyl sources could be installed into allene units with excellent regioselectivity by the cooperation of visible light catalysis and NHCs catalysis. This versatile protocol provides an attractive strategy for valuable sulfonyl-containing multi-substituted allyl ketones under extremely mild condi-tions with broad functional group tolerance, which might open a new avenue for the radical difunctionalization of allenes.

Abstract:Alkyl and acyl radicals play a critical role in the advancement of chemical synthesis. The generation of acyl and alkyl radicals by activation of C-O bonds using visible-light photoredox catalysis offers a mild and environmentally benign approach to useful chemical transformations. Alcohols, carboxylic acids, anhydrides, xanthates, oxalates, N-phthalimides, and thiocarbonates are some examples of alkyl and acyl precursors that can produce reactive radicals by homolysis of the C-O bond. These radicals can then go through a variety of transformations that are beneficial for the construction of synthetic materials that are otherwise difficult to access. This study summarizes current developments in the use of organic photocatalysts, transition-metal photoredox catalysts, and metallaphotocatalysts to produce acyl and alkyl radicals driven by visible light.

Abstract:We report herein an unprecedented protocol for radical cyclization of aldehydes with pendant alkenes via synergistic photoredox, cobaloxime, and amine catalysis. The transformation was achieved in the absence of external oxidants, providing a variety of 5-, 6-, and 7-membered ring products with alkene transposition in satisfactory yields. Abstract:A photoredox-catalyzed protocol for performing radical difunctionalization of alkenes using N-heterocyclic carbene (NHC) boranes and alkynyl bromines is described. The alkynylborylation difunctionalization reaction involves photoredox generation of boryl radical, with subsequent radical addition to the double bond followed by the capture of alkynyl bromide to form a C–C bond. This method features mild reaction conditions, remarkable chemoselectivity, broad substrate scope and good to excellent yields (up to 89%). The modification of coumarin derivatives indicates that this approach can provide a useful route for the synthesis of complex alkynylborylated products.
