Abstract:Sporopollenin exine capsules (SECs), extracted from plant pollen, are emerging as attractive and natural microcapsules for a wide range of bio-composite applications due to their abundant availability, chemical and mechanical robustness, species-specific architectural features, striking structural uniformity, and large inner cavity. However, SECs have a predominantly hydrophobic interface hindering their application in biomaterials. Thus, developing robust surface functionalization strategies to increase their hydrophilicity would open the door to interfacing microcapsules with cells. Herein, we create SECs with increased surface hydrophilicity coated by polypeptide multilayer films using layer-by-layer (LbL) self-assembly technology. Furthermore, human bone marrow-derived stem cells adhere more to these LbL-treated SECs acting as seed cells for tissue engineering. In summary, our findings demonstrate that LbL modification of SECs improves their functional properties and has broad implications across bio-composite applications.

Herein, we report an ultrafast, one-step, and effective modification strategy to render SECs bioactivation by coating them with ferric ions and tannic acids, which endow them with a better cell adhesion property. In summary, our results show that this ultrafast and one-step bioactivation strategy enhances the functional characteristics of SECs and holds wide-ranging implications for bio-composite applications.
Preparation of "micelles enhanced " polyelectrolyte capsules

Abstract:Hybrid CaCO3 particles were used as templates to fabricate two kinds of "micelles enhanced" polyelectrolyte capsules with three different polyel ectrolytes such as poly( allylamine hydrochloride), poly( sodium styrenesulfonate), and poly (di-allyldlmethylammonium chloride) via LbL technique. Both of (PAH/PSS)(6) and (PADA/PSS)(6) capsules had good spherical shape and good dispersion by CLSM. In contrast with conventional polyelectrolyte capsules, these novel micelles enhanced capsules had thick and compact capsule wall. Meanwhile, fine FESEM images showed that there was a significant difference in microstructure and topology between the two kinds of capsules made of different component polyelectrolytes.

Abstract:Sporopollenin exine capsules (SECs) are highly resistant to heat and various acids and bases. They are also cheap, highly porous, eco-friendly polymer biomaterials with stable microencapsulation capacity. Due to their strong and uniquely shaped exine layers, they can allow growth on metal oxide materials, as a biotemplate for use in different applications. In this study, first, a single SEC extraction method was applied to three different pollens from Pinus, Fraxinus excelsior, and Tilia. Scanning electron microscopy (SEM), Brunauer-Emmett-Teller (BET) analysis, and thermogravimetric/differential thermal analysis (TGA/DTA) measurements both before and after the extraction process were performed to observe changes in surface area, morphology, porous structure, and degradation properties. The protein content and removal were analyzed by elemental and spectrophotometric analyses. Then, SECs were loaded by passive and centrifuge loading for drug delivery, and the loading capacities were analyzed by Fourier transform infrared spectroscopy and spectrophotometry. The method was successful in opening the pores and maintaining the structural integrity of SECs. It was determined that the morphology and porosity affected the encapsulation efficiency. According to the loading capacities, Tilia SECs were the most efficient SECs for both loading methods. In addition, three different SECs were hydrothermally coated with cobalt and then heat-treated to obtain a metal oxide structure. A CO3O4 supercapacitor electrode constructed using CO3O4-F. excelsior SEC powder had the best surface area parameters. The electrode showed a maximum specific capacity of 473 F/g for over 3000 continuous cycles of galvanostatic charge-discharge (GCD).

Abstract:Cross-linked layer-by-layer (LbL) assemblies with a precisely tuned surface zeta-potential were fabricated to control the adsorption of proteins, mammalian cells, and bacteria for different biomedical applications. Two weak polyions including a synthesized polyanion and polyethylenimine were assembled under controlled conditions and cross-linked to prepare three robust LbL films as model surfaces with similar roughness and water affinity but displaying negative, zero, and positive net charges at the physiological pH (7.4). These surfaces were tested for their abilities to adsorb proteins, including bovine serum albumin (BSA) and lysozyme (LYZ). In the adsorption tests, the LbL films bind more proteins with opposite charges but less of those with like charges, indicating that electrostatic interactions play a major role in protein adsorption. However, LYZ showed higher nonspecific adsorption than BSA, because of the specific behavior of LYZ molecules, such as stacked multilayer formation during adsorption. To exclude such stacking effects from experiments, protein molecules were covalently immobilized on AFM colloidal probes to measure the adhesion forces against the model surfaces utilizing direct protein molecule surface contacts. The results confirmed the dominating role of electrostatic forces in protein adhesion. In fibroblast cell and bacteria adhesion tests, similar trends (high adhesion on positively charged surfaces, but much lower on neutral and negatively charged surfaces) were observed because the fibroblast cell and bacterial surfaces studied possess negative potentials. The cross-linked LbL films with improved stability and engineered surface charge described in this study provide an excellent platform to control the behavior of different charged objects and can be utilized in practical biomedical applications.

Abstract:Aim: This study aimed at constructing a novel disulfide-crosslinked collagen I/hyaluronic acid polyelectrolyte multilayer (PEM) coating incorporated with bFGF and arginine-glycine-aspartic acid on titanium via the layer-by-layer technique, and evaluating its biological effects. Materials & methods: The surface topography and components, thickness, degradation behaviors and bFGF release profiles of the PEM coating were investigated. The effects of the PEM coating on osteoprogenitor cell growth and bone implant interfacial binding strength in the rabbit femur model were evaluated separately. Results: The formation of disulfide bonds improved the stability of the PEM coating, resulting in a coating that can release bFGF in a slow and sustained manner. Biological evaluations revealed that the resultant PEM coating on titanium promoted various cell behaviors and enhanced the binding strength. Conclusion: The employed cotreatment regimen enabled bFGF and arginine-glycine-aspartic acid to have a synergistic effect on the cell responses, which, in turn, improved the osseointegration.

Abstract:A novel chitosan-based fluorescent bioconjugate (TPE-CS) with aggregation-induced emission (AIE) characteristic is synthesized and used as a fluorescent probe for monitoring layer-by-layer self-assembly process of natural polyelectrolytes. QCM results and contact angle measurement indicate that this AIE active TPE-CS bioconjugate can be assembled with alginate (ALG) through layer-by-layer deposition. Ellipsometry and fluorescence (FL) spectroscopy show an exponential growth of the TPE-CS/ALG multilayer films. Moreover, the exponential relationship between the FL intensity and the number of bilayers, which is in accordance with the thickness variation of multilayer films, provides solid evidence for its capacity to monitor the layer-by-layer self-assembly process.

Abstract:Cell adhesion and spreading are important events during cell-biomaterial interaction, which control survival, growth and differentiation of cells. Layer-by-layer technique was used to generate multilayer coatings for regulating adhesion of primary osteoblasts on biomaterials. Polyelectrolyte multilayers (PEM) were based on poly (ethylene imine) as primary polycation layer. PEM were then prepared from chitosan (CHI) as polycation and heparin (HEP), or sulfated HA (sHA) as polyanions. It was observed that attachment and spreading of primary osteoblasts (pOB) was highly dependent on the composition of multilayers, as well as pH values of polyelectrolyte solutions. Results presented in this paper may pave the way for application of PEM surface coatings for bone-contacting implant materials.

Abstract:Sequential layer-by-layer (LbL) deposition of polyelectrolytes followed by chemical cross-linking was investigated as a method to fabricate functional amphiphilic surfaces for marine biofouling prevention applications. A novel polyanion, grafted with amphiphilic perfluoroalkyl polyethylene glycol (fPEG) side chains, was synthesized and subsequently used to introduce amphiphilic character to the LbL film. The structure of the polyanion was confirmed by FTIR and NMR. Amphiphilicity of the film assembly was demonstrated by both water and hexadecane static contact angles. XPS studies of the cross-linked and annealed amphiphilic LbL films revealed the increased concentration of fPEG content at the film interface. In antifouling assays, the amphiphilic LbL films effectively prevented the adhesion of the marine bacterium Pseudomonas (NCIMB 2021).

Abstract:Novel "micelles enhanced" polyelectrolyte (PE) capsules based on functional templates of hybrid calcium carbonate were fabricated. Evidences suggested that the structure of capsule wall was different from that of conventional PE capsules, and the wall permeability of these PE capsules changed significantly. Lysozyme, a positively charged protein in neutral solution, was studied as a model protein to be encapsulated into the "micelles enhanced" PE capsules. Confocal laser scanning microscope was used to observe the entrapping process in real time, while UV-Vis spectroscope and scanning force microscope measurements suggested the high efficiency of encapsulation. In addition, the fluorescence recovery after photobleaching technique was employed to determine the existence form of deposited molecules. Further studies showed even negatively charged water-soluble peptides or proteins can be encapsulated into these hybrid capsules by modulating the pH value in bulk solution under its isoelectronic point as well.

Abstract:Controlling the adhesion of mammalian and bacterial cells at the interfaces between synthetic materials and biological environments is a real challenge in the biomedical fields such as tissue engineering, antibacterial coating, implantable biomaterials and biosensors. The surface properties of materials are known to profoundly influence the adhesion processes. To mediate the adhesion processes, polymeric coatings have been used to functionalize surfaces to introduce diverse physicochemical properties. The polyelectrolyte multilayer films built via the layer-by-layer (LbL) method, introduced by Moehwald, Decher, and Lvov 20years ago, has led to significant developments ranging from the fundamental understanding of cellular processes to controlling cell adhesion for biomedical applications. In this review, we focus our attention on the modification of surface physicochemical properties, using the LbL approach, to construct films which can either promote or inhibit mammalian/bacterial cell adhesion. We also discuss the emerging field of multifunctional surfaces capable of responding to specific cellular activity but being inert to the others.
Parallel Control over Surface Charge and Wettability Using Polyelectrolyte Architecture: Effect on Protein Adsorption and Cell Adhesion.

Abstract:Surface charge and wettability, the two prominent physical factors governing protein adsorption and cell adhesion, have been extensively investigated in the literature. However, a comparison between these driving forces in terms of their independent and cooperative effects in affecting adhesion is rarely explored on a systematic and quantitative level. Herein, we formulate a protocol that features two-dimensional control over both surface charge and wettability with limited cross-parameter influence. This strategy is implemented by controlling both the polyion charge density in the layer-by-layer (LbL) assembly process and the polyion side-chain chemical structures. The 2D property matrix spans surface isoelectric points ranging from 5 to 9 and water contact angles from 35 to 70°, with other interferential factors (e.g., roughness) eliminated. The interplay between these two surface variables influences protein (bovine serum albumin, lysozyme) adsorption and 3T3 fibroblast cell adhesion. For proteins, we observe the presence of thresholds for surface wettability and electrostatic driving forces necessary to affect adhesion. Beyond these thresholds, the individual effects of electrostatic forces and wettability are observed. For fibroblast, both surface charge and wettability have an effect on its adhesion. The combined effects of positive charge and hydrophilicity lead to the highest cell adhesion, whereas negative charge and hydrophobicity lead to the lowest cell adhesion. Our design strategy can potentially form the basis for studying the distinct behaviors of electrostatic force or wettability driven interfacial phenomena and serve as a reference in future studies assessing protein adsorption and cell adhesion to surfaces with known charge and wettability within the property range studied here.
Tailoring polyelectrolyte architecture to promote cell growth and inhibit bacteria adhesion.

Abstract:An important challenge facing the application of implanted biomaterials for tissue engineering is the need to facilitate desirable tissue interactions with the implant while simultaneously inhibiting bacterial colonization, which can lead to implant-associated infection. In this study, we explore the relevance of physical parameters of polyelectrolyte multilayers, surface charge, wettability and stiffness, in tissue cell/surface and bacteria/surface interactions, and investigate the tuning of the multilayer architecture to differentially control such interactions. Polyions with different side chain chemical structures were paired with polyethylenimine to assemble multilayers with parallel control over surface charge and wettability under controlled conditions. The multilayers can be successfully crosslinked to yield stiffer (the apparent Young's modulus was increased more than 3 times its original value) and more stable films while maintaining parallel control over surface charge and wettability. The initial adhesion and proliferation of 3T3 fibroblast cells were found to be strongly affected by surface charge and wettability on the non-crosslinked multilayers. On the other hand, these cells adhered and proliferated in a similar manner on the crosslinked multilayers (apparent Young's modulus ~2 MPa) regardless of surface charge and wettability. In contrast, Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) adhesion was primarily controlled by surface charge and wettability on both crosslinked and non-crosslinked multilayers. In both cases, negative charge and hydrophilicity inhibited its adhesion. Thus, a surface coating with a relatively high degree of stiffness from covalent crosslinking coupled with negative surface charge and high wettability can serve as an efficient strategy to enhance host cell growth while resisting bacterial colonization.
Structure Studies of Micelle-Enhanced Polyelectrolyte Microcapsules

Abstract:Hybrid calcium carbonate particles, synthesized III the presence of block copolymer polystyrene-b-poly(acrylic acid) (PS-b-PAA), were used to fabricate polyelectrolyte (PE) such as polylallyamine hydrochloride)/ poly(strene sulfonate sodium) (PAH/PSS) and poly (diallyldimethyl ammonium chloride)/poly (styrene sulforate sodium) (PADH/PSS) Capsules, by layer-by-layer (LbL) technique. Novel micelle-enhanced polyelectrolyte (PE capsules were thus obtained. The structure of the micelle-enhanced PE microcapsules was studied in this article. Due to the presence of a large amount of negatively charged micelles inside PE capsule interior, which were liberated from templates, the capsule wall was tightly re-constructed. Different from the wall properties of the conventional PE capsules, the wall of the micelle-enhanced PE capsules showed stability under an extensive pH range from I to 14, but the micelled enhanced capsules showed dented morphology in solutions with high NaCl concentration such as 0.5 mol/L and I mol/L. The state of the block copolymer in the capsules was investigated in dialysis bag by simulating the Solution condition of the capsule interior after the template being removed. It was found that the micelle size of the block copolymer In solution of templates dissolution was tens of nanometers after the solution was dialyzed for 24 h. With the dialysis time increasing up to at least 72 It, the block copolymer began to precipitate onto the dialysis bag from the solution The interaction between the 2-D polyelectrolyte multilayer film and micelles formed by block copolymer in templates solution was studied by UV-Vis measurements as well. It was found that PE multilayer could interact with micelles via electrostatic force. At the same time I the block copolymer remained in the capsules interior could be precipitated onto the PE multilayer via dialyzing in deionized water for a long time (pH similar to 6). A micelle-related layer was formed which was stabilized by the "bridge effect" of calcium ions. That is, the wall of the micelle-enhanced PE capsules had a bilayer structure and was formed by the polyelectrolyte multilayer and micelle-related layer. It seemed that the two layers with different properties clung to each other via electrostatic force. The formation mechanism of the bilayer structure was well illuminated oil the aforementioned studies. This is a key property of the micelle-enhanced PE capsules with which all the properties of the capsules were linked. Based on the bilayer structure of the capsules, the morphology changes of the capsules in solutions with different conditions such as high NaCl concentrations could be elucidated well. However, more direct proofs and further studies were needed to completely resolve the fine structure of the micelle-enhanced PE capsules.
Controlled Adhesion of HUVEC on Polyelectrolyte Multilayers by Regulation of Coating Conditions

Abstract:Adhesion of host cells on the surface of implants is necessary for a healthy ingrowth of the implanted material. One possibility of surface modification is the coating of the implant with a second material with advantageous physical-chemical surface properties for the biological system. The coverage with blood proteins takes place immediately after implantation. It is followed by host-cell interaction on the surface. In this work, the effect of polyelectrolyte multilayer coatings (PEMs) on adhesion and activity of human umbilical vein endothelial cells (HUVECs) was studied. The PEMs were formed from poly(styrenesulfonate) (PSS) and poly(allylamine hydrochloride) (PAH) from solutions with different concentrations of NaCl varying between 0 and 1.0 M. The adhesion of HUVEC and their viability on the PEM is related to the amount of adsorbed proteins from the applied cell growth medium. The amount of adsorbed proteins is controlled not only by the surface charge but also by the internal excess charge of the PEM. The internal excess charge of the PEM was controlled by changing the electrolyte concentration in the deposition solutions.
Layer by Layer Chitosan/Alginate Coatings on Poly(Lactide-Co-Glycolide) Nanoparticles for Antifouling Protection and Folic Acid Binding to Achieve Selective Cell Targeting

Abstract:Polyelectrolyte multilayers (PEMs) composed of two natural polysaccharides-chitosan (Chi) and alginate (Alg) were deposited by Layer by layer (LbL) assembly on top of biocompatible poly(lactide-co-glycolide) (PLGA) nanoparticles (NPs). Folic acid (FA) or FA grafted poly(ethylene glycol) (PEG-FA) were covalently bounded to the PEMs via carbodiimide chemistry. The assembly of biocompatible PEMs was monitored on planar surfaces by means of the quartz crystal microbalance with dissipation (QCM-D) technique and on top of PLGA NPs by means of c-potential measurements. BSA was used as model protein to characterize protein adsorption on PEMs. QCM-D showed protein deposition could not be observed on the Chi/Alg multilayer, for both Chitosan and Alginate as top layers. Finally, cellular uptake experiments were carried out by co-culture of HepG2 cells in presence of NPs. Flow Cytometry and confocal laser scanning microscopy (CLSM) were used to investigate the influence of the surface chemistry of the NPs on uptake. For the HepG2 cell line significantly less uptake of PLGA NPs coated with Chi/Alg than the bare NPs was observed but the uptake increased after attachment of FA molecules. (c) 2010 Elsevier Inc. Abstract:Chemically modified chitosan beads containing polyethyleneimine (PEI) were prepared to improve the metal ion adsorption capacity of the chitosan beads and their mechanical stability and to limit their biodegradability. The modified beads were encapsulated with the polymer material polysulfone by a novel surface coating method named the emulsion phase inversion method. The adsorption properties of the modified beads and the microstructures of the polysulfone coating layer were then analyzed. The experimental results showed that the PEI was successfully linked onto the chitosan beads. The density of the —NH2 groups in the modified beads was significantly increased, while the water content was reduced. The coating layer thickness was about 200 μm. The modified chitosan gel beads had excellent Cu(II) adsorption capacity, with a maximum Cu(II) adsorption capacity 1.34 times higher than that of the unmodified beads. The results show that even with the polysulfone coating the adsorption kinetics of the modified beads is still better than those of the unmodified beads. The modifications improve the mass transfer performance of the chitosan beads as well as the bead stability.

Abstract:The coating of capillary inner surface is considered to be an effective approach to suppress the adsorption of proteins on capillary inner surface in CE. However, most of coating materials reported are water-soluble, which may dissolve in BGE during the procedure of electrophoresis. In this study, a novel strategy for selection of physically coating materials has been illustrated to get coating layer with excellent stability using materials having poor solubility in commonly used solvents. Taking natural chitin as example (not hydrolyzed water soluble chitosan), a simple one step coating method using chitin solution in hexafluoroisopropanol was adopted within only 21 min with good coating reproducibility (RSDs of EOF for within-batch coated capillaries of 1.55% and between-batch coated capillaries of 2.31%), and a separation of four basic proteins on a chitin coated capillary was performed to evaluate the coating efficacy. Using chitin coating, the adsorption of proteins on capillary inner surface was successfully suppressed with reversed and stable EOF, and four basic proteins including lysozyme, cytochrome c, ribonuclease A and α-chymotrypsinogen A were baseline separated within 16 min with satisfied separation efficiency using 20 mM pH 2.0 H3PO4-Na2HPO4 as back ground electrolyte and 20 kV as separation voltage. What is more important, the chitin coating layer could be stable for more than two months during this study, which demonstrates that chitin is an ideal material for preparing semi-permanent coating on bare fused silica capillary inner wall and has hopeful potential in routine separation of proteins with CE.

Abstract:Soluble eggshell membrane protein (SEP) which posses se s good biocompatibility was immobilized on surface of polyethylene (PE) film via plasma pretreatment. Measurements with contact angle measurements, attenuated t otal reflectance infrared spectroscopy (ATR-FTIR), and X-ray photoelectron spe ctroscopy (XPS) confirmed the immobilization, which could be further improved by introducing nano-SiO x. Rinsing tests of immobilized PE film with 10 % ac etic acid (a good solvent of SEP) showed that SEP had been immobilized solidly on th e PE film surface and could endure the eroding of tissue fluid. The surface hydr ophilicity of SEP immobilized PE film was also suitable for the attachment and p roliferation of cells.
