Abstract:Prompt and accurate detection of CA19-9 in human serum has great clinical significance for the early diagnosis and disease monitoring of cancer. Herein, we develop a convenient and antifouling electrochemical sensor for CA19-9 determination by immobilization of both an electrochemical redox probe [methylene blue (MB)] and immunorecognition element (CA19-9 antibody) on an electrostatic nanocage consisting of bipolar silica nanochannel array (bp-SNA). bp-SNA is composed of a negatively charged inner layer (n-SNA) and positively charged outer layer (p-SNA), which could be stably prepared on indium tin oxide (ITO) in several seconds using a two-step electrochemically assisted self-assembly approach and display asymmetric surface charges for confinement and enrichment of cationic MB into the inner n-SNA layer through electrostatic interaction. Modification of the CA19-9 antibody on the top surface of bp-SNA confers the sensing interface with specific recognition capacity. An antibody-antigen complex formed at the as-prepared immunosensor causes the decreased electrochemical signals of MB, achieving sensitive determination of CA19-9 with a wider linear dynamic range from 10 μU/mL to 50 U/mL and a low detection limit (3 μU/mL). Furthermore, accurate and feasible analysis of the CA19-9 amount in human serum samples by our proposed probe-integrated electrochemical immunosensor is realized.

Abstract:Convenient and sensitive detection of tumor biomarkers is crucial for the early diagnosis and treatment of cancer. Herein, we present a probe-integrated and label-free electrochemical immunosensor based on binary nanocarbon composites and surface-immobilized methylene blue (MB) redox probes for detection of carbohydrate antigen 199 (CA19-9), which is closely associated with gastric malignancies. Nanocarbon composites consisting of electrochemically reduced graphene oxides and carbon nanotubes (ErGO-CNT) are electrodeposited onto an indium tin oxide (ITO) electrode surface to form a 3D nanocomposite film, which could provide high surface area to immobilize abundant MB probes, facilitate the electron transfer of MB, and therefore, improve sensitivity. Polydopamine (PDA) served as a bifunctional linker is able to immobilize anti-CA19-9 antibodies and stabilize the inner probe, conferring the sensing interface with specific recognition capacity. Electrochemical detection of CA19-9 is achieved based on the decrease of the redox signal of MB after specific binding of CA19-9 with a wide linear range of 0.1 mU/mL to 100 U/mL and a limit of detection (LOD) of 0.54 nU/mL (S/N = 3). The constructed electrochemical immunosensor has good selectivity, repeatability, reproducibility, and stability. Furthermore, determination of CA19-9 in human serum samples is also realized.

Abstract:Developing non-invasive, sensitive and specific sensing strategies for cancerous cell detection with simple and low cost instrumentations provide great advantages in cancer research and early diagnosis of diseases. In the present work, gold nanoparticles (Au NPs) functionalized with recognition components (folic acid) and signal indicator (ferrocene) was designed to fabricate electrochemical cytosensor. The Au NPs can not only accelerate electron transfer between signal indicator and the underlying electrode but also accumulate more ferrocene on the cytosensor surface to magnify signal for improving detection sensitivity. The surface-tethered folic acid plays a key role in specific binding folate receptor-riched HeLa cells on the cytosensor surface, resulting in corresponding current signal change measured by differential pulse voltammetry method. A wide detection range from 10 to 10(6) cells/mL with a detection limit as low as 10 cells/mL for cancerous cells was reached in the presence of a large amount of normal ones with fast differential pulse voltammetry measurement. Detection of the captured cells can be finished within 1 min. The developed strategy provides a new way for operationally simple, rapid, sensitive and specific detection of cancerous cells.

Abstract:Carbohydrate antigen 19-9 (CA19-9) is an important marker for pancreatic cancer, ovarian cancer and other tumors, and its rapid and stable detection is the basis for early diagnosis and treatment. In this paper, a label-free electrochemical immunosensor for the sensitive detection of CA19-9 has been developed. First, the synthesis of two novel core-shell bimetallic nanomaterials, namely Ce-MOF-on-Fe-MOF and Fe-MOF-on-Ce-MOF, was accomplished using the MOF-on-MOF approach. The poor electrical conductivity of MOF materials was addressed by incorporating polyethylenimide (PEI) functionalized rGO with Ce-MOF-on-Fe-MOF and Fe-MOF-on-Ce-MOF nanomaterials. Simultaneously, toluidine blue (Tb) was employed as a redox probe and physically adsorbed onto the synthesized materials, resulting in the formation of two nanomaterials: rGO@Ce-MOF-on-Fe-MOF@Tb and rGO@Fe-MOF-on-Ce-MOF@Tb. The fundamental characterization reveals that the sensing performance of the rGO@Ce-MOF-on-Fe-MOF@TB-based immune sensor surpasses that of the rGO@Fe-MOF-on-Ce-MOF@TB-based immune sensor, which is attributed to the fact that, unlike the interlayer-constrained structure of Fe-MOF-on-Ce-MOF, in Ce-MOF-on-Fe-MOF, Ce-MOF penetrates into Fe-MOF to form a heterogeneous structure due to the relatively large pore size of Fe-MOF, which better combines the excellent biocompatibility and strong anchoring effect of Fe MOFs on antibodies, as well as the high electrochemical activity and conductivity of Ce-MOF, to enhance sensing performance. The proposed label-free immunosensor based on rGO@Ce-MOF-on-Fe-MOF@Tb has a wide linear range (1-100 000 mU mL-1), a low detection limit (0.34 mU mL-1), good stability, reproducibility, and repeatability, and satisfactory applicability, which provides a potential platform for clinical applications.

Abstract:In this study, a novel split-type electrochemical immunosensor based on controlled release strategy was proposed for sensitive analysis and detection of tumor marker carbohydrate antigen 199 (CA19-9). Specifically, glucose (Glu) was encapsulated in carrier mesoporous silica (MSN) with encapsulation technology, and surface functionalized Zinc sulfide (ZnS) caps were used as "gatekeepers". The complex is formed by encapsulating Glu within MSN with ZnS (ZnS@MSN-Glu) as a signal amplifier labeled on the signal antibody (Ab2). And the Ab2 can detect the presence of antibodies. To reduce the interference of biological analysis, the immune recognition process of ZnS@MSN-Glu-Ab2 bioconjugate and antigen was carried out in 96-well microplate, which did not interfere with the electrochemical analysis process. Therefore, the low sensitivity detection caused by biofouling of nanomaterials and immunoreaction on the testing platform is eliminated. Subsequently, the opening and timed release of mesopores were controlled by external stimuli, the disulfide bond cleavage by dithiothreitol (DTT), and glucose was effectively released. Then nickel cobalt layered double hydroxide (NiCo-LDH) were directly hydrothermally grown on carbon cloth (CC) electrodeposited with copper selenide (CuSe) nanosheets to construct three-dimensional (3D) cactus-like NiCo-LDH/CuSe/CC sensing platform. It can realize the catalytic oxidation of released glucose, triggering glucose-mediated signal amplification. The synergistic effect of the 3D cactus structure and active nanomaterials promotes electron conduction. Taking the detection of carbohydrate antigen CA19-9 as an example, the immunosensor shows a wide linear concentration range (0.001-100 U/mL) with the limit of detection of 0.0005 U/mL, realizing highly sensitive detection of CA19-9. This biosensing technique has considerable advantages and provides an innovative approach for trace detection of other biomarkers.

Abstract:Evaluating the levels of the biomarker carbohydrate antigen 19-9 (CA19-9) is crucial in early cancer diagnosis and prognosis assessment. In this study, an antifouling electrochemical immunosensor was developed for the label-free detection of CA19-9, in which bovine serum albumin (BSA) and graphene were cross-linked with the aid of glutaraldehyde to form a 3D conductive porous network on the surface of an electrode. The electrochemical immunosensor was characterized through the use of transmission electron microscopy (TEM), scanning electron microscopy (SEM), atomic force microscope (AFM), UV spectroscopy, and electrochemical methods. The level of CA19-9 was determined through the use of label-free electrochemical impedance spectroscopy (EIS) measurements. The electron transfer at the interface of the electrode was well preserved in human serum samples, demonstrating that this electrochemical immunosensor has excellent antifouling performance. CA19-9 could be detected in a wide range from 13.5 U/mL to 1000 U/mL, with a detection limit of 13.5 U/mL in human serum samples. This immunosensor also exhibited good selectivity and stability. The detection results of this immunosensor were further validated and compared using an enzyme-linked immunosorbent assay (ELISA). All the results confirmed that this immunosensor has a good sensing performance in terms of CA19-9, suggesting its promising application prospects in clinical applications.

Abstract:This work developed an electrochemical immunosensor to quantitatively detect CA19-9 in human serum, a pancreatic cancer biomarker, where the electrode materials were thionine (TH) and graphene sheets (GS), and horseradish peroxidase (HRP) and ferroferric oxide (Fe3O4) nanoparticles-loaded mesoporous silica nanoparticles (MSNs) served as labels to amplify signals. The electron transfer and immobilization of primary antibody of CA19-9 (Ab1) were enhanced by the as a substrate, the GS and TH hybrid (GS/TH). MSNs were used as a carrier for immobilization of secondary antibody of CA19-9 (Ab2), Fe3O4, and HRP. The sensitivity of the immunosensor could be enhanced by the synergistic effect between HRP and Fe3O4. The proposed technique is potential to be applied to clinical analysis or be used to detect other tumor markers.

Abstract:A label-free electrochemical immunosensor was successfully developed for sensitively detecting carbohydrate antigen 19-9 (CA19-9) as a cancer marker. To achieve this, a series of bimetallic cerium and ferric oxide nanoparticles embedded within the mesoporous carbon matrix (represented by CeO2/FeOx@mC) was obtained from the bimetallic CeFe-based metal organic framework (CeFe-MOF) by calcination at different high temperatures. The formed CeO2 or FeOx nanoparticles were uniformly distributed within the highly graphitized mesoporous carbon matrix at the calcination temperature of 500 °C (represented by CeO2/FeOx@mC500). However, the obtained nanoparticles were aggregated into large size when calcined at the temperatures of 700 and 900 °C. The CA 19-9 antibody can be anchored to the CeO2/FeOx@mC network through chemical absorption between carboxylic groups of antibody and CeO2 or FeOx by ester-like bridging. The CeO2/FeOx@mC500-based immunosensor displayed superior sensing performance to the pristine CeFe-MOF, CeO2/FeOx@mC700- and CeO2/FeOx@mC900-based ones. Electrochemical impedance spectroscopy results showed that the developed immunosensor exhibited an extremely low detection limit of 10 μU·mL-1 (S/N = 3) within a wide range from 0.1 mU·mL-1 to 10 U·mL-1 toward CA 19-9. It also illustrated excellent specificity, good reproducibility and stability, and acceptable application analysis in the human serum solution which was diluted 100-fold with 0.01 M PBS solution (pH 7.4) and spiked with different amounts of CA19-9. Consequently, the proposed electrochemical immunosensor is capable enough of determining CA 19-9 in clinical diagnostics.

Abstract:A novel label-free electrochemical immunosensor for the ultrasensitive determination of cancer antigen (CA724) was developed using a novel composite of CoAl layered double hydroxides (CoAl-LDH) and carboxyl-functionalized multiwall carbon nanotubes (MWCNTs-COOH) as platform and ferrocenecarboxylic acid (Fc-COOH) as signal label. The MWCNTs-COOH/Fc-COOH@CoAl-LDH composite was prepared by a convenient and simple one-step ultrasonic method, and various characterization techniques consisting of scanning electron microscopy (SEM), transimission electronic microscopy (TEM), TEM-Mapping, fourier transform infrared (FT-IR), X-ray diffraction (XRD) and X-ray photoelectronic energy spectrum (XPS) were applied to study the size and morphological features. Due to its large specific surface area and multilayer structure, the CoAl-LDH can be post-doped to embed a large amount of signal probe to realize the amplification of the internal reference signal Fc. In addition, the higher conductivity of MWCNTs-COOH compensates for the deficiency of CoAl-LDH, which effectively strengthened the electron transfer efficiency of electrochemical signaling substances. The optimal experimental conditions were detected to be 2.5 mmol of Fc-COOH, 4.0 mg/mL of concentration, pH 6.0, incubation time of 40 min, and incubation temperature of 37 °C. Under optimal conditions, the fabricated sensor exhibits linearity in a wide dynamic range covering the physiological concentration, from 0.001 to 100 U/mL and the limit of detection (LOD) was 0.03962 mU/mL, the calibration equation is stated as △I = 7.76363 log 10 C CA724 + 40.50351 (R 2 = 0.99674). The sensor is successfully detects trace levels of CA724 in human serum with excellent recovery rates ranging from 100.52 %-102.30 %, proving the synergy of MWCNTs-COOH/Fc-COOH@CoAl-LDH as a promising platform for electrochemical sensing for clinical detection of other disease markers.

Abstract:Here, a label-free immunosensor was conducted using rGO/Collard-like Bi2S3@Au nanocomposite system to detect carbohydrate antigen 19-9 (CA19-9) as a diagnostic biomarker for Pancreatic cancer. The unique morphology of Collard-like Bi2S3 increases its specific surface area, which allows it to immobilize more materials and biomolecules on its surface. To realize the covalent bonding of bismuth-based materials with biomolecules, bismuth-based materials were functionalized by Au NPs. Besides, the superior electrochemical activity and large specific surface area of Bi2S3@Au improved the stability of the sensor, which made it an ideal candidate for regulating the sensing interface in this study. Meanwhile, reduced graphene oxide (rGO) with remarkable electrical conductivity to improve the current response of the sensing interface, thus broadening the linear range of the immunosensor. Based on the above materials, a label free electrochemical immunosensor was constructed for detecting CA19-9. Under optimal conditions, it displays good linear relationship within the range of 0.01 U mL-1 to 100 U mL-1, the detection limit being 0.0012 U mL-1. Additionally, the immunosensor prepared in this work is characterized by superior interference immunity, reproducibility and storage stability, and has yielded satisfactory recovery rate in testing the serum samples of patients.
A Novel Electrochemical Immunosensor Based on COF-LZU1 as Precursor to Form Heteroatom-Doped Carbon Nanosphere for CA19-9 Detection

Abstract:Porous carbon sphere materials have a large variety of applications in several fields due to the large surface area, adaptable porosity, and good conductivity they possess. Obtaining a steady carbon sphere using the green synthesis method remains a significant challenge. In this experiment, covalent organic frameworks (COFs) were used as a precursor and Fe3O4NPs were integrated into the precursor in order to synthesize a porous carbon sphere material using the one-step pyrolysis method. COFs have an ordered porous structure, perpetual porosity, large surface area, and low density and display good environmental tolerance. These properties make them an excellent precursor for synthesizing porous carbon sphere, which maintains good morphology at high temperatures, and it is not involved in the removal of dangerous reagent and small size restrictions during the synthesis process. In addition to the formation of a porous carbon sphere, transition metal carbon material that contains N element can be an active catalyst. The composites exhibit better activity when Fe is doped into carbon materials containing N element than that of other doped transition metals including Mn and Co. In this situation, the integration of Fe3O4NPs and N element in the COF precursor exposed the active sites of the composites and the two substances synergistically improved the electrocatalytic properties, and the composites were named Fe3O4@NPCS. The constructed Fe3O4@NPCS/GCE immunosensor was applied as a means of detecting CA19-9 antigen and presented a wide linear range from 0.00001 to 10 U/mL with a low detection limit of 2.429 μU/mL (S/N = 3). In addition, the prepared immunosensor was utilized for detecting CA19-9 antigen in the real human serum, and the recovery rates were in the range from 95.24% to 106.38%. Therefore, a porous carbon sphere prepared by COFs as a precursor can be applied for the detection of CA19-9 antigen in real samples, which could be an excellent strategy for CA19-9 antigen detection and could potentially promote the development of COF materials in various electrochemical fields.

Abstract:Early cancer diagnosis is paramount for enhancing treatment efficacy, extending patient survival, and improving the quality of life. We developed a highly sensitive electrochemical biosensor for the detection of target DNA (tDNA) associated with gastric cancer. This advancement integrates dual signal amplification strategies: bio-barcode amplification (BCA) and surface-initiated enzyme polymerization (SIEP), with copper nanoclusters (CuNCs) serving as signal labels. Silica nanoparticles (SiO2) were covalently linked with polythymine (poly T) and complementary DNA to create bio-barcode probes. These probes, through hybridization, were immobilized on the reduced graphene oxide and Au nanoparticle (rGO-AuNPs) modified interface and marking the first amplification of the electrical signal. Subsequently, the extended poly T prompted by SIEP bound additional CuNCs through the combination of T-Cu2+, leading to a second round of signal amplification. The biosensor demonstrated a minimum detection limit of 0.13 fmol/L over a linear response range from 1 fmol/L to 1 nmol/L. It also showcased excellent specificity, repeatability, and stability, making it a promising tool for the sensitive detection of gastric cancer biomarkers.
Folic Acid-Functionalized Zirconium Metal-Organic Frameworks Based Electrochemical Impedance Biosensor for the Cancer Cell Detection

Abstract:The effective capture and sensitive detection of cancer cells are of great significance to the basic research as well as to the clinical diagnosis and therapy. Here we proposed a simple, low-cost, and high-efficient impedance cytosensor based on folic acid-functionalized zirconium metal-organic frameworks (MOF). Folic acid (FA)-functionalized zirconium MOFs (UiO-66) was utilized as the sensitive element due to the high surface area and porosity, in which FA can be effectively modified onto UiO-66 by the coordinative interaction. The successful preparation of FA@UiO-66 nanocomposite was carefully validated by power X-ray diffraction (XRD), Fourier transform infrared (FT-IR) spectra, UV spectra, zeta potential, transmission electron microscope (TEM), and scanning electron microscope (SEM). The obtained electrochemical impedance results indicated that this impedance sensor functionalized with FA@UiO-66 nanocomposite exhibited good analytical performances in a linear range from 1 x 10(2) to 1 x 10(6) cells/mL. The detection limit was calculated to be as low as 90 cells/mL. It also showed good selectivity in distinguishing FAR-rich cancer cells (e.g. HeLa and H1299 cells) and FAR-negative cells (e.g. HEK-293 cells). HeLa cells spiked in blood samples can also be detected with good recovery rates. All the results demonstrated that this simple and universal approach has great potential to develop disposable and portable biosensors and be applied in the early-stage cancer diagnosis.
A Novel Cytosensor for Capture, Detection and Release of Breast Cancer Cells Based on Metal Organic Framework PCN-224 and DNA Tetrahedron Linked Dual-Aptamer

Abstract:A convenient and efficient strategy for detecting cancer cells at preclinical stages is one of the dominant challenges in early cancer diagnostics. In this work, a sandwich-type cytosensor was structured to analyze cancer cells based on metal organic framework PCN-224 and DNA tetrahedron linked dual-aptamer. Firstly, the tetrahedral DNA nanostructures linked dual aptamers (AS1411 and MUC1) immobilizing on the gold electrode (GE) surface as bio-recognition elements can capture the MCF-7 cancer cells and improve the density and orientation of surface nanoprobes. Then, the novel metal organic framework PCN-224 homogeneously decorated by Pt nanoparticles was modified by the G-quadruplex/hemin DNAzyme, horseradish peroxidase (HRP) and dual-aptamer. The fabricated nanoprobes were applied to catalyze the oxidation of hydroquinone (HQ) with hydrogen peroxide (H2O2) for amplifying the electrochemical signal. The results indicated that the linear response of the aptasensor ranged from 20 to 1x10(7) cells/mL and the detection limit was 6 cells/mL. Finally, we performed an electrochemical reductive desorption to break gold-thiol bond and release all compounds on the electrode surface for analyzing the viability of collected cells and regenerating the cytosensor. This work can be extended to other cancer cells by using specific type aptamers and has widely applications prospect in point-of-care cancer diagnosis.

Abstract:Electrochemical methods have attracted considerable attention for developing cytosensing system since they can decrease the cost and time requirement for cell detection with simple instrumentation. Herein, a label-free electrochemical cytosensor with surface-confined ferrocene as signal indicator was developed for highly sensitive and selective detection of cancer cell. With layer-by-layer (LBL) self-assembly technique, positively charged poly(ethylene imine) functionalized with ferrocene (Fc-PEI) and negatively charged single-wall carbon nanotubes (SWNTs) were alternately assembled on 3-mercaptopropionic acid (MPA) modified gold substrate. Folic acid (FA) was covalently bonded onto SWNTs surface to specifically recognize cancer cells according to the high affinity of FA for folate receptor (FR) on cellular surface. The developed cytosensor presented high sensitivity and selectivity for the detection of human cervical carcinoma (HeLa) cell. By using fast-response differential pulse voltammetry (DPV) method, a wide detection range from 10 to 106cells/mL with a detection limit as low as 10cells/mL was reached even in the presence of a large amount of non-cancerous cells.

Abstract:The detection of tumor markers has significant importance in the early diagnosis of cancer. It is an important indicator for evaluating the extent of the disease and monitoring the efficacy of tumor treatment. Therefore, it is meaningful to find a convenient, fast, and sensitive method to detect tumor markers. Carbohydrate antigen 19–9 (CA19-9) is the first discovered carbohydrate antigen tumor marker with the highest sensitivity and specificity for pancreatic cancer. Carcinoembryonic antigen (CEA), as the most widely spectrum carbohydrate antigen tumor marker, can show an upward trend in many tumors. In this work, an electrochemical immunosensing platform was proposed for the detection of CEA and CA199. Nano-silicon sphere-cyclodextrin was synthesized for antibody capture. Graphene and gold nanoparticles were used to modify the electrode, improving the electronic conductivity of the electrode. The synthesized sensor can be used for highly sensitive detection of two tumor markers. This method also provides a new platform for clinical immune analysis of other biomolecules.
Electrochemical Immunoassay for Tumor Marker CA19-9 Detection Based on Self-Assembled Monolayer

Abstract:A CA19-9 electrochemical immunosensor was constructed using a hybrid self-assembled membrane modified with a gold electrode and applied to detect real samples. Hybrid self-assembled membranes were selected for electrode modification and used to detect antigens. First, the pretreated working electrodes were placed in a 3-mercaptopropionic acid (MPA)/β-mercaptoethanol (ME) mixture for 24 h for self-assembly. The electrodes were then placed in an EDC/NHS mixture for 1 h. Layer modification was performed by stepwise dropwise addition of CA19-9 antibody, BSA, and antigen. Differential pulse voltammetry was used to characterize this immunosensor preparation process. The assembled electrochemical immunosensor enables linear detection in the concentration range of 0.05–500 U/mL of CA19-9, and the detection limit was calculated as 0.01 U/mL. The results of the specificity measurement test showed that the signal change of the interfering substance was much lower than the response value of the detected antigen, indicating that the sensor has good specificity and strong anti-interference ability. The repeatability test results showed that the relative standard deviations were less than 5%, showing good accuracy and precision. The CA19-9 electrochemical immunosensor was used for the actual sample detection, and the experimental results of the standard serum addition method showed that the RSD values of the test concentrations were all less than 10%. The recoveries were 102.4–115.0%, indicating that the assay has high precision, good accuracy, and high potential application value.

Abstract:Developing ultrasensitive and highly selective biosensors is significant for the early diagnosis of cancers and monitoring of the treatment process of cancer patients. Metal–organic frameworks (MOFs), as emerging porous crystalline materials, have received increased attention on account of their potential applications in many fields, such as energy, environment, storage, separation, and sensing. In light of their unique merits, such as large surface areas, tunable pore scales, and good adsorption capabilities, MOFs have been applied as sensitive platforms for anchoring diverse probes (e.g., antibodies, DNA, or aptamers) for the construction of electrochemical (EC) biosensors. Herein, we present a comprehensive overview of MOFs-based EC biosensors for detecting diverse targets (e.g., cancer markers, microRNA, and living cancer cells) that are considered as the indicators for early diagnosis of cancers. Different strategies for construction of EC biosensors with monometallic MOFs, bimetallic MOFs, luminophore-combined MOFs, and MOFs-based nanocomposites are reviewed. Further, the fabrication of MOFs-based EC biosensors toward specific analytes is described to promote and guide future studies on MOFs for biosensing applications.

Abstract:In this work, a competitive and label-free electrochemical platform was performed for the ultrasensitive cytosensing of liver cancer cells based on DNA nanotetrahedron (NTH) structure and rolling circle amplification (RCA) directed DNAzyme strategy. The multifunctional nanoprobes were fabricated through a DNA primer probe, carboxyfluorescein (FAM) functionalized TLS11a aptamer and horseradish peroxidase (HRP) immobilized on the surfaces of the platinum nanoparticles (PtNPs). Then the NTH-based complementary DNA (cDNA) probe, complementary to the TLS11a aptamer, was attached on a disposable screen-printed gold electrode (SPGE) for increasing the reactivity and accessibility with the prepared nanoprobes. Due to the primer probe and the circular probe with G-quadruplex sequences for RCA, it can lead to the formation of numerous G-quadruplex/hemin DNAzyme, thus generating a remarkable electrochemical response. When the target cells were present, the nanoprobes were released from the SPGE due to the specific recognition of TLS11a aptamers for HepG2 cells, resulting in the electrochemical signal changes. The cytosensor was ultrasensitive for HepG2 tumor cell detection with a detection limit of 3 cell per mL. Furthermore, this strategy was also demonstrated to be applicable for cancer cell imaging. In summary, this electrochemical cytosensor holds great potential for circulating tumor cell detection in the early cancer diagnose.
