Abstract:Abstract In agro-ecosystem, plant pathogens hamper food quality, crop yield, and global food security. Manipulation of naturally occurring defense mechanisms in host plants is an effective and sustainable approach for plant disease management. Various natural compounds, ranging from cell wall components to metabolic enzymes have been reported to protect plants from infection by pathogens and hence provide specific resistance to hosts against pathogens, termed as induced resistance. It involves various biochemical components, that play an important role in molecular and cellular signaling events occurring either before (elicitation) or after pathogen infection. The induction of reactive oxygen species, activation of defensive machinery of plants comprising of enzymatic and non-enzymatic antioxidative components, secondary metabolites, pathogenesis-related protein expression (e.g. chitinases and glucanases), phytoalexin production, modification in cell wall composition, melatonin production, carotenoids accumulation, and altered activity of polyamines are major induced changes in host plants during pathogen infection. Hence, the altered concentration of biochemical components in host plants restricts disease development. Such biochemical or metabolic markers can be harnessed for the development of “pathogen-proof” plants. Effective utilization of the key metabolites-based metabolic markers can pave the path for candidate gene identification. This present review discusses the valuable information for understanding the biochemical response mechanism of plants to cope with pathogens and genomics-metabolomics-based sustainable development of pathogen proof cultivars along with knowledge gaps and future perspectives to enhance sustainable agricultural production.

Abstract:The pathogen Pseudomonas syringae , responsible for a variety of diseases, poses a considerable threat to global crop yields. Emerging biocontrol strategies employ antagonistic microorganisms, utilizing phyllosphere microecology and systemic resistance to combat this disease. However, the interactions between phyllosphere microbial dynamics and the activation of the plant defense system remain poorly understood. Here we show significant alterations in phyllosphere microbiota structure and plant gene expression following the application of biocontrol agents. We reveal enhanced collaboration and integration of Sphingomonas and Methylobacterium within the microbial co-occurrence network. Notably, Sphingomonas inhibits P. syringae by disrupting pathogen chemotaxis and virulence. Additionally, both Sphingomonas and Methylobacterium activate plant defenses by upregulating pathogenesis-related gene expression through abscisic acid, ethylene, jasmonate acid, and salicylic acid signaling pathways. Our results highlighted that biocontrol agents promote plant health, from reconstructing beneficial microbial consortia to enhancing plant immunity. The findings enrich our comprehension of the synergistic interplays between phyllosphere microbiota and plant immunity, offering potential enhancements in biocontrol efficacy for crop protection.

Abstract:Highlights • To overcome the major challenges of global food security and environmental and social problems caused by the extensive use of chemical pesticides, developments of modern green sustainable management strategies are urgently needed for the control of crop diseases. • A deeper understanding of the principles of plant immunity and induced host immunity offers potentials for reducing the use of chemical pesticides and paves the way for sustainable agriculture. • The sustainable environment-friendly disease prevention and control technologies based on plant immunity inducers are comprehensively summarized in this review. • The significance of the application of plant immunity inducers for plant disease control is systematically summarized. • By focusing on research advances of plant immunity inducers, this review offers future perspectives of plant immunity inducers and provides a theoretical reference for researchers. Background Crops are constantly attacked by various pathogens. These pathogenic microorganisms, such as fungi, oomycetes, bacteria, viruses, and nematodes, threaten global food security by causing detrimental crop diseases that generate tremendous quality and yield losses worldwide. Chemical pesticides have undoubtedly reduced crop damage; however, in addition to increasing the cost of agricultural production, the extensive use of chemical pesticides comes with environmental and social costs. Therefore, it is necessary to vigorously develop sustainable disease prevention and control strategies to promote the transition from traditional chemical control to modern green technologies. Plants possess sophisticated and efficient defense mechanisms against a wide range of pathogens naturally. Immune induction technology based on plant immunity inducers can prime plant defense mechanisms and greatly decrease the occurrence and severity of plant diseases. Reducing the use of agrochemicals is an effective way to minimize environmental pollution and promote agricultural safety. Aim of Review : The purpose of this work is to offer valuable insights into the current understanding and future research perspectives of plant immunity inducers and their uses in plant disease control, ecological and environmental protection, and sustainable development of agriculture. Key Scientific Concepts of Review : In this work, we have introduced the concepts of sustainable and environment-friendly concepts of green disease prevention and control technologies based on plant immunity inducers. This article comprehensively summarizes these recent advances, emphasizes the importance of sustainable disease prevention and control technologies for food security, and highlights the diverse functions of plant immunity inducers-mediated disease resistance. Abstract:Plants have developed a complex defense system against diverse pests and pathogens. Once pathogens overcome mechanical barriers to infection, plant receptors initiate signaling pathways driving the expression of defense response genes. Plant immune systems rely on their ability to recognize enemy molecules, carry out signal transduction, and respond defensively through pathways involving many genes and their products. Pathogens actively attempt to evade and interfere with response pathways, selecting for a decentralized, multicomponent immune system. Recent advances in molecular techniques have greatly expanded our understanding of plant immunity, largely driven by potential application to agricultural systems. Here, we review the major plant immune system components, state of the art knowledge, and future direction of research on plant⁻pathogen interactions. In our review, we will discuss how the decentralization of plant immune systems have provided both increased evolutionary opportunity for pathogen resistance, as well as additional mechanisms for pathogen inhibition of such defense responses. We conclude that the rapid advances in bioinformatics and molecular biology are driving an explosion of information that will advance agricultural production and illustrate how complex molecular interactions evolve.

Abstract:Microorganisms provide beneficial effects to plants through its mutualistic, commensalistic, or symbiotic association. However, the microbial diversity and the number of microbial populations can change over the different stages of plant growth. The multiple plant beneficial features including plant growth promotion, biocontrol activity, and the induction of plant disease resistance mediated by these microorganisms can make them to have the promises to prepare bioformulations for the agricultural applications. Since it is an environment-friendly approach, it also helps to reduce the heavy use of agrochemicals and thereby promotes sustainable agricultural practices. Among various plant beneficial microorganisms, endophytes are highly significant basically due to their distribution with in plants. Hence, deeper h understanding onthe plant protective mechanisms of endophytes is highly relevant for the development of active biocontrol formulations. Thus, the current chapter discuss in detail about the mechanisms employed by endophytic microorganisms for protecting the plants from wide range of pathogens. Detailed understanding on the mode of action and mechanism of endophytes against pathogens further helps to accelerate the application and commercialization of endophyte based formulation.

Abstract:Disease control is largely based on the use of fungicides, bactericides, and insecticides—chemical compounds toxic to plant invaders, causative agents, or vectors of plant diseases. However, the hazardous effect of these chemicals or their degradation products on the environment and human health strongly necessitates the search for new, harmless means of disease control. There must be some natural phenomenon of induced resistance to protect plants from disease. Elicitors are compounds, which activate chemical defense in plants. Various biosynthetic pathways are activated in treated plants depending on the compound used. Commonly tested chemical elicitors are salicylic acid, methyl salicylate, benzothiadiazole, benzoic acid, chitosan, and so forth which affect production of phenolic compounds and activation of various defense-related enzymes in plants. Their introduction into agricultural practice could minimize the scope of chemical control, thus contributing to the development of sustainable agriculture. This paper chiefly highlights the uses of elicitors aiming to draw sufficient attention of researchers to the frontier research needed in this context.

Abstract:Upon infection with insect-borne microbial pathogens, plants are exposed to two types of damage simultaneously. Over the past decade, numerous molecular studies have been conducted to understand how plants respond to pathogens or herbivores. However, investigations of host responses typically focus on a single stress and are performed under static laboratory conditions. In this review, we highlight research that sheds light on how plants deploy broad-spectrum mechanisms against both vector-borne pathogens and insect vectors. Among the host genes involved in multistress resistance, many are involved in innate immunity and phytohormone signaling (especially jasmonate and salicylic acid). The potential for genome editing or chemical modulators to fine-tune crop defensive signaling, to develop sustainable methods to control insect-borne diseases, is discussed.

Abstract:Plant growth and nutrition are adversely affected by various factors such as water stress, high temperature, and plant pathogens. Plant-associated microbes play a vital role in the growth and development of their hosts under biotic and abiotic stresses. The use of a rhizosphere microbiome for plant growth stimulation and the biological control of fungal disease can lead to improved crop productivity. Mechanisms used by plant-growth-promoting rhizobacteria (PGPR) to protect plants from soilborne pathogens include antibiosis, the production of lytic enzymes, indole-3 acetic acid production, decreasing ethylene levels by secreting 1-aminocyclopropane-1-carboxylate deaminase, competition for nutrients and niches, parasitism and induced systemic resistance. In this review, we emphasize the biological control of plant pathogens by root-associated microbes and discuss traits involved in pathogen reduction. Future research should focus on the effect of root exudation on plant–pathogen interactions under various abiotic factors. Moreover, the development of microbial fungicides with longer shelf lives will help farmers to opt for organic agriculture, reducing the use of chemical fertilizers. This trend is expected to drive the adoption of biological control methods in agriculture. The future prospects for the biological control of plant diseases are bright and are expected to play an increasingly important role in sustainable agriculture.

Abstract:Induced systemic resistance (ISR) of plants against pathogens is a widespread phenomenon that has been intensively investigated with respect to the underlying signalling pathways as well as to its potential use in plant protection. Elicited by a local infection, plants respond with a salicylic-dependent signalling cascade that leads to the systemic expression of a broad spectrum and long-lasting disease resistance that is efficient against fungi, bacteria and viruses. Changes in cell wall composition, de novo production of pathogenesis-related-proteins such as chitinases and glucanases, and synthesis of phytoalexins are associated with resistance, although further defensive compounds are likely to exist but remain to be identified. In this Botanical Briefing we focus on interactions between ISR and induced resistance against herbivores that is mediated by jasmonic acid as a central signalling molecule. While many studies report cross-resistance, others have found trade-offs, i.e. inhibition of one resistance pathway by the other. Here we propose a framework that explains many of the thus far contradictory results. We regard elicitation separately from signalling and from production, i.e. the synthesis of defensive compounds. Interactions on all three levels can act independently from each other.

Abstract:Plant beneficial microorganisms improve the health and growth of the associated plants. Application of beneficial microbes triggers an enhanced resistance state, also termed as induced systemic resistance (ISR), in the host, against a broad range of pathogens. Upon the activation of ISR, plants employ long-distance systemic signaling to provide protection for distal tissue, inducing rapid and strong immune responses against pathogens invasions. The transmission of ISR signaling was commonly regarded to be a jasmonic acid- and ethylene-dependent, but salicylic acid-independent, transmission. However, in the last decade, the involvement of both salicylic acid and jasmonic acid/ethylene signaling pathways and the regulatory roles of small RNA in ISR has been updated. In this review, the plant early recognition, responsive reactions, and the related signaling transduction during the process of the plant–beneficial microbe interaction was discussed, with reflection on the crucial regulatory role of small RNAs in the beneficial microbe-mediated ISR.

The effect of contact lens cleaners on lens parameters and surface quality of the Novalens.

Abstract:The purpose of this study is to evaluate the effect of one abrasive cleaner, one non-abrasive cleaner and one non-abrasive alcohol-based cleaner on the lens parameter stability of the NOVALENS. Twenty-four lenses, all NOVALENS material of identical design, were cleaned 100 times each. Eight lenses were cleaned with MiraFlow (CIBA Vision), eight lenses with Optic-Free Daily Cleaner and eight lenses with the Gas Permeable Daily Cleaner (PBH). The base curve radius, center thickness and power were verified immediately before, immediately after and (per manufacturer's recommendation) 4 hours after cleaning at baseline, 25, 50, 75 and 100 cleanings. Lens toricity of approximately 0.50D was found with all three cleaners after 100 cleanings. No change in lens power or center thickness was found. It was concluded that extreme care must be taken in handling the NOVALENS due to the potential of induced warpage.

Abstract:Biotic stressors pose significant threats to crop yield, jeopardizing food security and resulting in losses of over USD 220 billion per year by the agriculture industry. Plants activate innate defense mechanisms upon pathogen perception and invasion. The plant immune response comprises numerous concerted steps, including the recognition of invading pathogens, signal transduction, and activation of defensive pathways. However, pathogens have evolved various structures to evade plant immunity. Given these facts, genetic improvements to plants are required for sustainable disease management to ensure global food security. Advanced genetic technologies have offered new opportunities to revolutionize and boost plant disease resistance against devastating pathogens. Furthermore, targeting susceptibility (S) genes, such as OsERF922 and BnWRKY70, through CRISPR methodologies offers novel avenues for disrupting the molecular compatibility of pathogens and for introducing durable resistance against them in plants. Here, we provide a critical overview of advances in understanding disease resistance mechanisms. The review also critically examines management strategies under challenging environmental conditions and R-gene-based plant genome-engineering systems intending to enhance plant responses against emerging pathogens. This work underscores the transformative potential of modern genetic engineering practices in revolutionizing plant health and crop disease management while emphasizing the importance of responsible application to ensure sustainable and resilient agricultural systems.

Synthesis, phloem mobility and induced plant resistance of synthetic salicylic acid amino acid or glucose conjugates

Abstract:Background: The growing demand for food, combined with a strong social expectation for a diet produced with fewer conventional agrochemical inputs, has led to the development of new alternatives in plant protection worldwide. Among different possibilities, the stimulation of the plant innate immune system by chemicals represents a novel and promising way. The vectorization strategy of an active ingredient that we previously developed with fungicides can potentially extend to salicylic acid (SA) or its halogenated analogues. Results: Using the click chemistry method, six new conjugates combining SA or two mono- or di-halogenated analogues with L-glutamic acid or β-D-glucose via a 1,2,3-triazole nucleus have been synthesized. Conjugate 8a, which is derived from SA and glutamic acid, showed high phloem mobility in the Ricinus model, similar to that of SA alone despite a much higher steric hindrance. In vivo bioassays of the six conjugates against two maize pathogenic fungi Bipolaris maydis and Fusarium graminearum revealed that, unlike SA, the amino acid conjugate 8a with good phloem mobility exerted a protective effect not only locally at the application site, but also in distant stem tissues after foliar application. Moreover, compounds 8a and 8b induced up-regulation of both defense-related genes ZmNPR1 and ZmPR1 similar to their parent compounds upon challenge inoculation with B. maydis. Conclusion: The vectorization of salicylic acid or its halogenated derivatives by coupling them with an α-amino acid can be a promising strategy to stimulate SA-mediated plant defenses responses against pathogens outside the application site. © 2022 The Authors. Abstract:Different from the conventional biocidal agrochemicals, synthetic chemical inducers of plant immunity activate, bolster, or prime plant defense machineries rather than directly acting on the pathogens. Advances in combinatorial synthesis and high-throughput screening methods have led to the discovery of various synthetic plant immune activators as well as priming agents. The availability of their structures and recent progress in the mechanistic understanding of plant immune responses have opened up the possibility of identifying new or more potent chemical inducers through rational design. In this review, we first summarize the chemical inducers identified through large-scale screening and then discuss the emerging trends in the identification and development of novel plant immune inducers including natural elicitor based chemical derivation, bifunctional combination, and computer-aided design.

Abstract:Systemic acquired resistance is a powerful mechanism, based on the salicylic acid (SA) signaling pathway, which allows plants to resist to a wide range of pathogens. High SA, moreover, plays a key role in plant tolerance to abiotic stress. It seems, therefore, desirable to supply analogs of SA or stimulate the production of endogenous SA. Unfortunately, the chemical substances or physical means used for this effect often display a variable efficacy. After providing a review of them, we defend three major ideas: (i) plant resistance inducers (PRIs) must be combined for higher efficacy, notably for exploiting synergic effects between the SA and other signaling pathways, (ii) disease pressure can be reduced by exploiting the fungicidal properties displayed by some PRIs, (iii) biostimulants and crop management techniques should be used to ensure that plants have the resources they need to synthesize the compounds and structures required for efficient and lasting resistance. Some PRIs could also be used for their biostimulant effects in stress conditions. It could be concluded that holistic approaches which jointly address the issues of defense and tolerance stimulation, disease pressure and resource availability in plants are the ones that will allow for substantial reduction in fungicide use without sacrificing crop performance.

Abstract:This paper discusses the mechanisms by which fungi manipulate plant physiology and suppress plant defense responses by producing effectors that can target various host proteins. Effector-triggered immunity and effector-triggered susceptibility are pivotal elements in the complex molecular dialogue underlying plant-pathogen interactions. Pathogen-produced effector molecules possess the ability to mimic pathogen-associated molecular patterns or hinder the binding of pattern recognition receptors. Effectors can directly target nucleotide-binding domain, leucine-rich repeat receptors, or manipulate downstream signaling components to suppress plant defense. Interactions between these effectors and receptor-like kinases in host plants are critical in this process. Biotrophic fungi adeptly exploit the signaling networks of key plant hormones, including salicylic acid, jasmonic acid, abscisic acid, and ethylene, to establish a compatible interaction with their plant hosts. Overall, the paper highlights the importance of understanding the complex interplay between plant defense mechanisms and fungal effectors to develop effective strategies for plant disease management.

However, few natural pathogen-inducible promoters are available for this purpose. To address this problem, we have developed a strategy to synthesize dual SA- and JA-responsive promoters by combining SA- and JA-responsive cis elements based on the interaction between their cognate trans-acting factors. The resulting promoters respond rapidly and strongly to both SA and Methyl Jasmonate (MeJA), as well as different types of phytopathogens. When such a synthetic promoter was used to control expression of an antimicrobial peptide, transgenic plants displayed enhanced resistance to a diverse range of biotrophic, necrotrophic, and hemi-biotrophic pathogens. A dual-inducible promoter responsive to the antagonistic signals auxin and cytokinin was generated in a similar manner, confirming that our strategy can be used for the design of other biotically or abiotically inducible systems.
