Abstract:Bioassay-guided fractionation, based on inhibition of -glucosidase, of Euonymus alatus led to the isolation of a new compound, catechin lactone A (7), and seven known compounds (1-6 and 8). Their structures were established on the basis of extensive 1D- and 2D-NMR experiments. The absolute configurations of compound 7 have been determined using vibrational circular dichroism spectroscopy. All these compounds were evaluated for their inhibition activities on -glucosidase and differentiation of 3T3-L1 cells. Compounds 1, 3-5, 7 and 8 showed potent yeast -glucosidase inhibition with IC50 values of 119.1, 39.6, 31.6, 23.0, 71.8 and 102.5 mu M, respectively; compounds 3-5, 7 and 8 can remarkably inhibit the differentiation of 3T3-L1 pre-adipocytes. This is the first report about -glucosidase and 3T3-L1 inhibitory activities of compounds 3, 7 and 8.

Abstract:Five new phloroglucinol derivatives, eucalyptins C-G (1-5), together with 13 known analogues (6-18) were isolated from the fruits of Eucalyptus globulus. The structures and absolute configurations of 1-5 were established by means of spectroscopic data analysis, computational calculation methods, and single-crystal X-ray diffraction. Compounds 1-18 were investigated for their immunosuppressive effects in vitro, and 1, 2, 6, and 7 displayed moderate inhibitory activities with IC50 values of 11.8, 10.2, 18.2, and 19.1 μM, respectively. The stimulation index (SI) of 1 was 64.2 and was compared to that of cyclosporine A (SI = 149.57). Further study demonstrated that 1 exhibited an immunosuppressive effect through inducing apoptosis and inhibiting cytokine secretion.

Abstract:Eucalyptus is one of the most important and highly exploited genus in family Myrtaceae. An UPLC/PDA/ESI-qTOF-MS method was adopted to identify Eucalyptus sideroxylon Cunn. ex Woolls leaves phytoconstituents. Cytotoxicity of E. sideroxylon leaves phloroglucinol-rich extract (PGRE) on VERO cells was determined. The antiviral effect of PGRE against hepatitis A (HAV), herpes simplex type 1 (HSV-I), herpes simplex type 2 (HSV-II), coxsackie (CoxB4), and adenoviruses was in vitro evaluated using MTT assay (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide). UPLC–MS analysis allowed the identification of 70 metabolites including: 26 triterpenes, 13 phloroglucinols, 8 fatty acids, 5 flavonoids, 5 oleuropeic acid glucosides, 3 gallic acid derivatives, and 10 miscellaneous. Twenty four metabolites identified in the leaves of E. sideroxylon and four in the genus Eucalyptus are reported herein for the first time. PGRE was found to be non-cytotoxic; the concentration that reduced the cell viability by 50% (CC50) was 0.808 mg/mL. Maximum non-toxic concentration (MNTC) of PGRE on Vero cells was 0.312 mg/mL. The best antiviral activity was observed against HSV-II. Its mechanism was through decreasing the viral replication (IC50 189.36 μg/mL, 87.65% inhibition) and attachment on Vero cells (IC50 199.34 μg/mL, 83.13% inhibition) rather than virucidal effect (IC50 293.1 μg/mL, 50.68% inhibition). This study provides a complete map for E. sideroxylon leaves composition. It also suggests the plant as a source of new antiviral agents.

Abstract:OBJECTIVE:To study the chemical constituents in the fruits of Eucalyptus globulus Labill.METHOD:The chemical constituents were isolated by various column chromatographic methods and structurally elucidated by IR, NMR and MS evidences.RESULT:Fifteen compounds were obtained and identified as beta-sitosterol (1), betulinic acid (2), stigmasterol (3), euscaphic acid (4), 2a-Hydroxybetulinic acid (5), macrocarpal B (6), macrocarpal A (7), oleanolic acid (8), 3,4,3'-O-trimethylellagic acid (9), 3-O-methylellagic acid 4'-O-(2"-O-acetyl )-alpha-L-rhamnopyranoside (10), camaldulenside (cypellocarpin C, 11), 3-O-methylellagic acid 4'-O-alpha-L-rhamnopyranoside (12), 3-O-methylellagic acid (13), ellagic acid (14), and gallic acid (15).CONCLUSION:Compounds 4 and 5 from genera Eucalyptus, 1, 3 and 11 from plant E. globulus, and 6, 7, 9 and 15 from the fruits of E. globulus were isolated for the first time.

Abstract:Eucaglobulin (1), a new complex of gallotannin and monoterpene, was isolated from the leaves of Eucalyptus globulus. Its structure was elucidated on the basis of spectral data. Four known hydrolyzable tannins [tellimagrandin I (2), eucalbanin C (3), 2-O-digalloyl-1,3,4-tri-O-galloyl-beta-D-glucose (4), 6-O-digalloyl-1,2,3-tri-O-galloyl-beta-D-glucose (5)], as well as gallic acid (6) and (+)-catechin (7), were also isolated. The antibacterial effects of some of these compounds were examined.

Abstract:Euphorboside A (1), an unusual meroterpenoid glycoside featuring the incorporation of an acylphloroglucinol moiety into a humulene skeleton to form a 6/6/11 ring system, was isolated from the roots of Euphorbia kansuensis. Its structure was elucidated by extensive spectroscopic analysis, chemical methods, and ECD calculations. Compound 1 was screened for the cytotoxicity against nine cancer cell lines, and 1 showed marked inhibitory activities against human colon cancer RKO and human breast cancer MDA-MB-231 cell lines with IC50 values of 3.70 and 4.15 μM, respectively.

Abstract:Acylphloroglucinols are well-known Eucalyptus secondary metabolites which exhibit a variety of structures and bioactivities. The investigation of a crude acetone extract of Eucalyptus camaldulensis leaves led to the isolation of two new acylphloroglucinols, eucalypcamals O and P ( 1 and 2 ) together with seven phloroglucinols ( 3 – 9 ), and a benzene derivative ( 10 ). Their chemical structures were elucidated by 1D and 2D nuclear magnetic resonance (NMR) spectroscopy and mass spectroscopy. The absolute configurations of compounds 1 and 2 were established by comparison of experimental and calculated electronic circular dichroism (ECD) data. Abstract:Three new formyl phloroglucinol meroterpenoids, eumaidials A-C ( 1 – 3 ), were isolated from the leaves of Eucalyptus globulus subsp. maidenii , along with ten known analogues ( 4 – 13 ). Their chemical structures were determined by various spectral data and electronic circular dichroism calculations. Eumaidial A ( 1) is the first β -caryophyllene-based formyl phloroglucinol meroterpenoids from the genus Eucalyptus . Abstract:Eleven new acylphloroglucinols, including six new formylated phloroglucinol-monoterpene meroterpenoids, eucalyprobusals A–F (1–6), one monomeric acylphloroglucinol, eucalyprobusone B (7), and four dimeric acylphloroglucinols, eucalyprobusones C–F (8–11) were purified from the fruits of Eucalyptus robusta. The establishment of the structures of 1–11 was achieved by a combination of NMR and HRESIMS data analyses, electron circular dichroism (ECD), and single-crystal X-ray diffraction. Compounds 6, 8, and an inseparable mixture of 10 and 11 were found to be potent AChE inhibitors with IC50 values of 3.22 ± 0.36, 3.82 ± 0.22, and 2.55 ± 0.28 μΜ, respectively. Possible interaction sites of 6, 8, 10, and 11 with AChE were investigated by means of molecular docking studies, and the results revealed that AChE residues Asn87, Ser125, Thr83, Tyr133, Tyr124, Tyr337, and Tyr341 played crucial roles in the observed activity of the aforementioned compounds.

Abstract:Five undescribed enantiomeric pairs of acylphloroglucinol-monoterpene meroterpenoids ((+)-/(−)-eucateretins A−E) resolved by chiral-phase HPLC were obtained from the leaves of Eucalyptus tereticornis Smith, along with nine known analogues. Their structures were elucidated by spectroscopic methods and ECD calculations. This is the first report of meroterpenoid enantiomers from this plant. Some of the isolates, (−)-eucateretin A, (+)-/(−)-eucateretins E, 7′ α -eucalrobusone X, eucalrobusone X, and robustadial B, exhibited inhibitory effects on ATP citrate lyase, and 7′ α -eucalrobusone X significantly suppressed the hepatocyte lipogenesis.

Abstract:Nine previously undescribed diterpenoids, euphorfiatnoids A−I, together with seven known diterpenoids, were isolated from the roots of wild Euphorbia fischeriana. Their structures were elucidated by the interpretation of HRESIMS, UV, and NMR data. Their configurations were determined by electronic circular dichroism (ECD) spectroscopy analysis and the structure of euphorfiatnoid A was confirmed by X-ray crystallography. To further understand the antitumor effects of E. fischeriana, we tested the cytotoxicity of these compounds against H460, HepG2, and MCF-7 cell lines in vitro using MTT assays. Euphorfiatnoid B exhibited the most promising inhibitory effect against H460 cells with an IC50 value of 9.97 μM. Euphorfiatnoid A and C also exhibited moderate cytotoxicity against HepG2 cells with IC50 values of 11.64 and 13.10 μM, respectively.

Abstract:(±)-Hypersines A–C (1–3), the three pairs of enantiomerically pure monoterpenoid polyprenylated acylphloroglucinols with an unprecedented 6/6/5/4 fused ring system, were isolated from Hypericum elodeoides. Their structures, including absolute configurations, were elucidated by comprehensive spectroscopic data, single-crystal X-ray diffraction, and quantum chemical calculations. The plausible, biosynthetic pathway of 1–3 was proposed. Moreover, the bioactivity evaluation indicated that 1a might be a novel DNA damage response inhibitor, and could enhance MCF-7 cell sensitivity to the anticancer agent, camptothecin.

Abstract:Phytochemical investigation on the nonpolar extracts of P. acidus leaves afforded three new cleistanthane diterpenoids, named phyllanacidins A–C (1–3), in addition to three known ones (4–6). Among them, compounds 2 and 3 represent the first examples of 17-nor cleistanthane diterpenoids. Their structures including absolute configurations were elucidated on the basis of extensive spectroscopic data and single crystal X-ray diffraction analysis. All the isolated cleistanthane diterpenoids were evaluated for their cytotoxic activities against five human cancer cell lines. Compounds 4–6 exhibited moderate cytotoxicity with IC50 values ranging from 16.11 to 30.82 μM.

Abstract:Two new tigliane diterpenes, named eupneonoids A (1) and B (2), together with four known analogues (3-6) were isolated from the whole plant of Euphorbia neorubella Bruyns. Their structures were deduced based on the detailed spectroscopic analysis. All the isolates displayed cytotoxic effects against A549 human tumor cell lines with IC50 values ranging from 1.318 to 7.042 mu M.

Abstract:The current study aimed to investigate the chemical composition, antioxidant, antibacterial, and cytotoxic properties of three extracts (hexane, dichloromethane, and methanol) from Cynoglossum tubiflorus. The composition of the methanolic extract was elucidated using HPLC-HESI-MS/MS analysis. The antioxidant effect was examined using NO, DPPH, FRAP, and TAC assays. Antimicrobial activity was evaluated by broth microdilution using various bacterial strains such as S. aureus, S. epidermidis, P. aeruginosa, E. coli, and K. pneumoniae. Structural disruptions in Gram-positive bacteria were visualized using scanning electron microscopy (SEM). Cytotoxic effects were evaluated on human MRC-5 in culture according to the MTT assay. The outcomes suggest that methanol extract contained a high amount of phenolic compounds (254.35 ± 0.360 mg GAE/g DE and 211.59 ± 0.939 mg QE/g DE). By applying the HPLC-HESI-MS/MS analysis, 32 compounds were identified, including phenolic acids, flavonoids, lignans, and fatty acids. This extract showed strong antioxidant (IC50 = 0.043 ± 0.001 mg/mL) and antimicrobial (MIC = 156 μg/mL) activities. The SEM suggests that cells exhibited membrane distortions characterized by surface depressions and alterations in bacterial shape, including dents, when compared to untreated cells. The in vitro cytotoxicity effect on human MRC-5 cells showed no toxicity effects at a concentration of 600 μg/mL. In silico analysis predicted low toxicity for all tested compounds across four different administration routes. This research indicates that this plant could be explored as a powerful source of natural drugs to target pathogens, with applications in the food, pharmaceutical, and medical industries.
