Abstract:Objective: Advanced glycation end products (AGEs) and vascular adventitial fibroblasts (AFs) are involved in diabetes-related vascular complications. However, the effect of AGEs on AFs remains unclear. The aim of this study was to observe the impact of AGEs on cell migration capacity and associated inflammatory responses of AFs.Methods and results: Isolated vascular AFs of Sprague-Dawley rats were cultured, harvested after 24 h synchronization and challenged with AGE-HSA. AGE-HSA upregulated the expression of receptor for advanced glycation end products (RAGE), significantly increased the migration capacity and inflammatory mediators MCP-1, IL-6, VCAM-1 expressions on AFs. These effects could be significantly attenuated by anti-RAGE neutralizing antibody, p38, ERK1/2 and JNK MAPK inhibitors as well as by candesartan. AGE-HAS also upregulated NF-kappa B transcriptional activity and I-kappa B-alpha phosphorylation, effect that was significantly inhibited by candesartan.Conclusions: AGE-HSA increased the migration capacity and inflammatory responses of rat AFs via RAGE-MAPK-NF-kappa B pathways. Candesartan effectively inhibited these effects which might be a novel vascular protection mechanism of candesartan. (C) 2009 Published by Elsevier Ireland Ltd.
IgE Actions on CD4+ T Cells, Mast Cells, and Macrophages Participate in the Pathogenesis of Experimental Abdominal Aortic Aneurysms.

Abstract:Immunoglobulin E (IgE) activates mast cells (MCs). It remains unknown whether IgE also activates other inflammatory cells, and contributes to the pathogenesis of abdominal aortic aneurysms (AAAs). This study demonstrates that CD4+ T cells express IgE receptor FcεR1, at much higher levels than do CD8+ T cells. IgE induces CD4+ T-cell production of IL6 and IFN-γ, but reduces their production of IL10. FcεR1 deficiency (Fcer1a-/-) protects apolipoprotein E-deficient (Apoe-/-) mice from angiotensin-II infusion-induced AAAs and reduces plasma IL6 levels. Adoptive transfer of CD4+ T cells (but not CD8+ T cells), MCs, and macrophages from Apoe-/- mice, but not those from Apoe-/- Fcer1a-/- mice, increases AAA size and plasma IL6 in Apoe-/- Fcer1a-/- recipient mice. Biweekly intravenous administration of an anti-IgE monoclonal antibody ablated plasma IgE and reduced AAAs in Apoe-/- mice. Patients with AAAs had significantly higher plasma IgE levels than those without AAAs. This study establishes an important role of IgE in AAA pathogenesis by activating CD4+ T cells, MCs, and macrophages and supports consideration of neutralizing plasma IgE in the therapeutics of human AAAs.

Abstract:IgE has a key role in the pathogenesis of allergic responses through its ability to activate mast cells via the receptor Fc epsilon R1. In addition to mast cells, many cell types implicated in atherogenesis express Fc epsilon R1, but whether IgE has a role in this disease has not been determined. Here, we demonstrate that serum IgE levels are elevated in patients with myocardial infarction or unstable angina pectoris. We found that IgE and the Fc epsilon R1 subunit Fc epsilon R1 alpha were present in human atherosclerotic lesions and that they localized particularly to macrophage-rich areas. In mice, absence of Fc epsilon R1 alpha reduced inflammation and apoptosis in atherosclerotic plaques and reduced the burden of disease. In cultured macrophages, the presence of TLR4 was required for Fc epsilon R1 activity. IgE stimulated the interaction between Fc epsilon R1 and TLR4, thereby inducing macrophage signal transduction, inflammatory molecule expression, and apoptosis. These IgE activities were reduced in the absence of Fc epsilon R1 or TLR4. Furthermore, IgE activated macrophages by enhancing Na+/H+ exchanger 1 (NHE1) activity. Inactivation of NHE1 blocked IgE-induced macrophage production of inflammatory molecules and apoptosis. Cultured human aortic SMCs (HuSMCs) and ECs also exhibited IgE-induced signal transduction, cytokine expression, and apoptosis. In human atherosclerotic lesions, SMCs and ECs colocalized with IgE and TUNEL staining. This study reveals what we believe to be several previously unrecognized IgE activities that affect arterial cell biology and likely other IgE-associated pathologies in human diseases.
Effects of Autoantibodies Against AT1 Receptor from Patients with Refractory Hypertension on Intracellular Ca2+ Mobilization of Rat Vascular Smooth Muscle Cells

Abstract:Objective:To explore the effect of autoantibodies against AT1 receptor of patients with refractory hypertension on the intracellular Ca2+ mobilization in vascular smooth muscle cells (VSMC).Method:Forty-six patients with hypertension were recruited and divided into two groups based on whether their blood pressure was above or below 140/90 mmHg after combination drug therapy: a refractory hypertension group (n=22) and a non-refractory hypertension group (n=24). The epitope of the second extracellular loop of AT1 receptor was synthesized and used as antigen to screen the antibodies by ELISA. IgG from patients with the refractory hypertension was obtained by affinity purification, and was used to prove the affinity with AT1-receptor in the cultured vascular smooth muscles (VSMC) by immunocytochemistry. Then, Fluo-3/AM, a Ca2+ fluorescent indicator was used to load VSMC. IgG from patients was used to stimulate VSMC and the change of fluorescence was monitored by confocal microscopy.Result:Frequency of the autoantibodies against AT1 receptor in refractory hypertension group was higher than that in non-refractory hypertension group (P0.05). And VSMC showed increased intracelluar Ca2+ mobilization induced by the patients’ antibodies against AT1 receptor. Furthermore, we found the autoantibody-induced Ca2+ increase could be blocked by losartan, an AT1 receptor antagonist.Conclusion: Antibodies against AT1 receptor with the ability to activate AT1 receptor may account for Ca2+ increase in VSMC. And the autoantibodies may involve in the pathogenesis of hypertension, especially in patients refractory to drug treatment.

Abstract:Purpose: Endothelial-mesenchymal transition (EndMT) leading to endothelial dysfunction, inflammation and disruption of endothelial barrier, promotes the development of atherosclerosis. We have previously shown that inactivation of adenosine 2A receptor (ADORA2A) inhibits the formation of atherosclerosis lesion in mice. However, the underlying mechanism has not been fully studied. Here we investigated the role of ADORA2A-drive EndMT in the development of atherosclerosis. Methods: The expression of ADORA2A in vascular endothelial cells (EC) at atheroprone regions of mouse aortas were examined by en face staining with ADORA2A and CD31 antibodies. EC-specific Adora2a knockout mice and inhibitor KW6002 were used to investigate the role of endothelial ADORA2A in EndMT and atherosclerosis in vivo . In vitro studies, the role of ADORA2A in TGFβ-1-induced EndMT was examined by Western blot, qPCR and immunostaining. Results: ADORA2A expression in ECs of lesion-prone arteries was upregulated. The formation of atherosclerotic plaques and EndMT were inhibited in endothelial Adora2a knockout mice or mice treated with antagonist KW6002. In the in vitro studies, EndMT induction of human aortic endothelial cells (HAECs) was inhibited by Adora2a knockdown and antagonist KW6002, but was promoted by adenovirus encoding Adora2a. Mechanistically, ADORA2A promoted the expression of TGFβR1/ALK5 and the activity of its downstream P-Smad2/3 signaling pathway through the cAMP/PKA/CREB signaling pathway Conclusions: ADORA2A promotes EndMT through the cAMP/PKA/CREB/ALK5 signaling pathway, resulting in increased formation of atherosclerotic lesions.
Macrophage polarization is involved in liver fibrosis induced by β1-adrenoceptor autoantibody

Abstract:Accumulating evidence suggests that liver injury can be induced by the over-expression of β 1-adrenergic receptors (β 1-ARs). High titers of autoantibodies specific to β 1-adrenergic receptors (β 1-AA) are detected in the sera of heart failure patients, potentially playing agonist-like roles. However, the role of β 1-AA in liver function has not been characterized. In this study, we collect the sera of primary biliary cholangitis (PBC) patients, a condition which easily develops into liver fibrosis, and analyze the relationship between PBC and β 1-AA. A passive immunization model is established to assess the effect of β 1-AA on the liver. Subsequently, the effect of β 1-AA on macrophages is investigated in vitro. Results show that PBC patients have a high titer and ratio of β 1-AA, compared to controls. Liver injury and fibrosis are induced by β 1-AA. In vitro experiments with ROS probe demonstrate that β 1-AA induces macrophages to produce ROS and secrete TNFα. These effects can be partially reversed by metoprolol, a blocker for β 1-AR. Results from the transwell and phagocytosis assays show that β 1-AA promotes macrophage migration and phagocytosis. FCM tests suggest that β 1-AA induces the alteration of M1 rather than M2 markers in macrophages. Finally, the Annexin V/PI assay indicates that macrophage culture supernatants stimulated by β 1-AA cause hepatocyte apoptosis. Overall, these results suggest that β 1-AA is involved in PBC. The β 1-AA-induced activation, phagocytosis and phenotypic modification of macrophages may play an important role in the development of hepatic fibrosis and injury.

Pro-arrhythmic action of autoantibodies against the second extracellular loop of β1-adrenoceptor and its underlying molecular mechanisms

Abstract:Objectives: The incidence of arrhythmia is associated with autoantibodies against the second extracellular loop of β1-adrenergic receptor (β1-AAbs). The current study was designed to determine the mechanisms by which arrhythmia experimentally might be induced by β1-AAbs. Methods: Blood samples were collected from patients with varied arrhythmias or coronary heart disease (CHD) and healthy subjects. The titer of β1-AAbs was assessed. Passive immunization rat models with β1-AAbs were established to determine whether β1-AAbs induced arrhythmia. Conventional intracellular microelectrode technique and whole cell patch clamp were employed to record action potential duration (APD), resting potential (RP), L-type calcium current (ICa-L), sodium-calcium exchange current (INCX), transient outward potassium current (Ito), inward rectifier potassium current (Ik1) and delayed rectifier potassium current (Ik). Results: High levels of β1-AAbs were found in the sera of heart disease patients, especially in ventricular arrhythmia (VA). Transfusion with β1-AAbs could induce arrhythmias in normal rats in vivo. β1-AAbs purified from the sera of active immunized rats induced triggered activity (TA), delayed after depolarization (DAD), and prolonged APD in the papillary muscles of rats. β1-AAbs prolonged QT interval, increased ICa-L and decreased IK1, Ito and INa-Ca in rat ventricular myocytes in vitro. All these effects can be inhibited by β1-AR blocker metoprolol. Conclusions: These results demonstrate for the first time that β1-AAbs could directly induce ventricular arrhythmia by prolonging QT interval.

Abstract:Objective To study the relationship between anti-endothelial cell antibodies (AECA),anticardiolipin antibodies (ACA) and vascular endothelial cell injury diseases and the significance of the autoantibodies.Methods Serum AECA and ACA were detected by ELISA in 168 patients with vascular endothelial cell injury diseases.Result Positive rates of AECA in 168 patients with vascular endothelial cell injury diseases,including coronary heart disease,idiopathic thrombocytopenia purpura,hemolytic uremic syndrome,pregnancy in hypertension,were 32.9%,22.7%,22.2% and 38.9% respectively,and ACA were 38.2%,31.8%,33.3% and 44.4% respectively.They were significantly higher than those of normal controls.Conclusion AECA and ACA are related to the development and progress of vascular endothelial cell injury diseases,AECA and ACA are independent each other.
The functional subclasses of AT1 receptor autoantibody in patients with coronary heart disease

Abstract:Recently, the identification of autoantibodies (AT1-AA) targeting the second extracellular loop of angiotensin II type 1 receptor (AT1R-ECII) in patients with coronary heart disease (CHD) offers a novel perspective on the interplay between immunity and cardiovascular disease. However, much remains unknown regarding the functional diversity of AT1-AA. In this study, we measured the levels of AT1-AA in the sera of 306 CHD patients and purified AT1-AA from patient's sera (n = 127). The subclasses of AT1-AA were categorized based on their impact on intracellular calcium ([Ca2+]i) levels in mouse arterial smooth muscle cells (MASMCs). Our findings revealed 4 distinct [Ca2+]i response patterns indicating the existence of 4 functional subclasses named H1-, H2-, H3-, and H4-AT1-AA. The correlation analysis demonstrated a positive association between H1-AT1-AA and endogenous coagulation, as well as between H2-AT1-AA and exogenous coagulation; no significant correlation was observed between H3-AT1-AA and the indicators we analyzed. Conversely, H4-AT1-AA exhibited a negative correlation with both leukocyte number and bile acid levels. Logistic regression analysis showed that H2-AT1-AA possessed predictive value for severe CHD. Furthermore, in vitro experiments indicated that both H1- and H2-AT1-AA exerted cytotoxic effects on MASMCs, while H4-AT1-AA increased cell viability. Additionally, an AT1-AA-positive rat model was established by subcutaneously injecting with AT1R-ECII peptide, which produced four similar functional subclasses of rat AT1-AA upon active immunization. This study suggested that classifying different functional subclasses of AT1-AAs can facilitate more accurate evaluation of the condition and prognosis in patients with CHD, thereby providing a novel basis for clinical diagnosis and treatment.

Abstract:Monocyte/macrophage recruitment is closely associated with the degree of hypertensive renal injury. Here we investigated the direct role of macrophage using Liposome encapsulated clodronate (LEC) to deplete monocyte/macrophage in Ang II-induced hypertensive renal injury. C57BL/6 mice were treated with a pressor dose of angiotensin II (Ang, 1.4 mg/kg/day by mini-pump) plus LEC or the PBS-liposome for two weeks. Ang II mice developed hypertension (186 ± 5 vs. 110 ± 4 mmHg in control, p<0.05) , albuminuria (33.9 ± 4.9 vs. 83.6 ± 2.3 μg/mg creatinine in control, p<0.05), glomerulosclerosis and renal fibrosis. LEC reduced Ang II-induced albuminuria (59.6 ± 1.9 μg/mg creatinine, p<0.05) and protected against renal structural injury with a mild reduction in systolic blood pressure (153 ± 3 mmHg, p<0.05). Ang II significantly increased renal macrophage infiltration (MOMA2 + cells) and the protein expression of renal tumor necrosis factor α and interleukin β1, which were significantly reduced in Ang II mice treated with LEC. Ang II increased renal oxidative stress (as demonstrated by increased oxidative fluorescence densities, NADPH oxidative activity and the protein expression of NADPH oxidase subunits gp91phox and p22phox) and the expression of profibrotic factors transform growth factor β1 and fibronectin. Ang II also inhibited the phosphorylation of endothelial nitric oxide synthesis (eNOS, ser1177). LEC revised most of the changes in the Ang II-induced molecules as mentioned above. Our results suggest that renal macrophage is a main mediator to contribute to Ang II-induced hypertensive renal injury and fibrosis, the underlying mechanisms may involve reduction in macrophage-drove renal inflammation and restoration of the balance between renal oxidative stress and eNOS. These findings may lead us to develop a novel therapy directed at targeting the infiltration of macrophages or the macrophages-derived cytokines for treatment of hypertensive renal diseases.
Blocking VCAM-1 Prevents Angiotensin II-Induced Hypertension and Vascular Remodeling in Mice

Abstract:Adhesion of monocytes to the vascular endothelium frequently leads to an inflammatory response, which contributes to hypertension and vascular remodeling. Vascular cellular adhesion molecule-1 (VCAM-1) plays an important role in leukocyte adhesion and migration during inflammatory diseases. However, its role in angiotensin (Ang) II -induced hypertension and vascular dysfunction remains largely unknown. Wild-type (WT) mice were administered a VCAM-1 neutralizing antibody (0.1 or 0.2 mg/mouse/day) or IgG control and then infused with Ang II (490 ng kg −1 min −1 ) or saline continuously for 14 days. Systolic blood pressure (SBP) was measured with a tail-cuff system, pathological changes in the aorta were assessed by histological staining, and vascular relaxation was analyzed an aortic ring assay. Our results indicated that compared with saline infusion, Ang II infusion significantly upregulated VCAM-1 expression in the mouse aorta and serum. Moreover, Ang II infusion markedly increased arterial hypertension, wall thickness, fibrosis, infiltration of Mac-2 + macrophages, reactive oxygen species (ROS) production and vascular relaxation dysfunction. Conversely, blockade of VCAM-1 with a neutralizing antibody substantially alleviated these effects. In vitro experiments further confirmed that the VCAM-1 neutralizing antibody inhibited Ang II-induced macrophage adhesion and migration and DNA damage and oxidative stress in endothelial cells (ECs). In conclusion, these results indicate that blockade of VCAM-1 exerts a protective effect against Ang II-induced arterial hypertension and dysfunction by regulating monocytes adhesion and infiltration into the endothelium and represents a novel therapeutic approach for hypertension.

Abstract:Background: Angiotensin II type 1 receptor (AT1R) is a key mediator of the rennin-angiotensin-aldosterone system (RAAS). Autoantibodies against AT1R (AT1-AA), as an activator of AT1R, was found in abdominal aortic aneurysm (AAA) related diseases, such as hypertension and atherosclerosis. However, the role of AT1-AA in AAA is still unclear. Methods: We used angiotensin II (AngII)-infused male ApoE -/- mice model of AAA. Ultrasonography measurement, histological assessment, western blot, qRT-PCR and RNA sequencing were used to assess the severity of AAA. Nanoluciferase-based bioluminescence resonance energy transfer (NanoBRET) assay, radio assay of receptors and homogeneous time resolved fluorescence (HTRF) were used to measure the activation feature of AT1R and the affinity of AT1R with AT1-AA or AngII. Results: AT1-AA increased the incidence of AAA (vehicle vs. AT1-AA: 47% vs. 81%; P<0.05), which can be attenuated by AT1R blocker Candesartan. Furthermore, AT1-AA greatly increased severity of AAA, due to augmenting the diameter of abdominal aorta and the degradation of elastin by enhancing the expression of MMP2/9 (matrix metalloproteinase) and the accumulation of inflammatory cells. And AT1-AA could induce phenotypic switch of smooth muscle cells. But AT1-AA does not increase the plasma level of AngII. Mechanically, AT1-AA enhanced AngII-induced recruitment of Gq, but not β-arrestin1/2. AT1-AA potentiated the AngII-mediated Gq-dependent inositol phosphate (EC50 shift=3.829 nM) and the activation of PKC and ERK1/2. The responses were abolished by AT1R blocker and Gq inhibitor. The affinity of AT1-AA for the AT1R in cells was calculated to be 75.49 ± 4.7 nM. AT1-AA cannot compete for the binding of AngII to AT1R. However, AT1-AA significantly potentiated the binding of AngII with AT1R allosterically (mean K D ± SEM: 34.11 ± 0.9765 nM at 0 nM AT1-AA vs. 25.63 ± 1.826 nM at 100 nM AT1-AA; p<0.05), without altering the maximum binding. Conclusion: AT1-AA, as an allosteric activator of AT1R, promotes AAA through increasing the affinity of AngII with AT1R and activating Gq biasedly.

Abstract:Objectives Angiotensin (Ang) II and Ang-(1–7) are two of the bioactive peptides of the renin-angiotensin system. Ang II is involved in the development of cardiovascular disease, such as hypertension and atherosclerosis, while Ang-(1–7) shows cardiovascular protection in contrast to Ang II. We studied effects of Ang II and Ang-(1–7) on endothelial VCAM-1 expression, which are critical in the formation of atherosclerotic lesion. Methods VCAM-1 RNA expression was analysed by real time RT-PCR. The endothelial surface expression of VCAM-1 was measured by flow cytometry. VCAM-1 promoter with NF-κ B binding sites were cloned into pGL3 vector. Luciferase assay was used to analyse VCAM-1 promoter activity in endothelial cells. NF-κ B translocation was observed by immunocytochemistry. Results Treatment with Ang II resulted in an increase of VCAM-1 expression on endothelial cells, whereas Ang-(1–7) alone had no effects. However, preincubation with Ang-(1–7) inhibited Ang II induced VCAM-1 expression, which is demonstrated by flow cytometry and real time RT-PCR. In addition, Ang-(1–7) inhibited Ang II induced VCAM-1 promoter activity. Immunocytochemistry showed that Ang-(1–7) blocked Ang II induced translocation of NF-κ B from cytoplasm into nucleus, and the effects of Ang-(1–7) were abolished in the presence of MAS receptor antagonist A779. Conclusions These results suggest that Ang-(1–7) inhibited VCAM-1 expression, at least in part, through negative modulation of Ang II induced NF-κ B pathway.
Endothelial enriched microRNAs regulate angiotensin II-induced endothelial inflammation and migration

Abstract:The initial stage of atherosclerosis is characterised by recruitment of leukocytes to activate endothelial cells (ECs). MicroRNAs (miRNAs) are a class of 19 to 25 nucleotides, non-protein-coding RNAs that repress target gene expression by translational inhibition or mRNA degradation. The link between miRNA and endothelial functions is largely unknown. Northern blot showed that miR-155 and miR-221 were highly expressed in human umbilical vein endothelial cells (HUVECs) and vascular smooth muscle cells (VSMCs). Bioinformatics analysis proposed Ets-1, a key endothelial transcription factor for inflammation and tube formation, as a candidate target for miR-155 and miR-221/222 cluster. The effect was demonstrated by luciferase reporter assay and Western blot. By using Western blot, we also confirmed that angiotensin II type 1 receptor (AT1R) is a target of miR-155 in HUVECs. Quantitative PCR showed that Ets-1 and its downstream genes, including VCAM1, MCP1 and FLT1, were up-regulated in angiotensin II-stimulated HUVECs, and this effect was partially reversed by over-expression of miR-155 and miR-221/222. In addition, cell adhesion assay revealed over-expression of miR-155 and miR-221/222 effectively decreased the adhesion of Jurkat T cells to Ang II-stimulated HUVECs. Besides, by targeting AT1R, miR-155 can also decrease the HUVECs migration in response to Ang II. In summary, HUVECs highly expressed miR-155 may co-target AT1R and Ets-1 while miR-221/222 targets Ets-1, which indirectly regulate the expression of several inflammatory molecules of ECs, and therefore attenuate the adhesion of Jurkat T cells to activated HUVECs and reduce HUVECs migration. These findings present possible therapeutic targets in atherosclerosis.

Sustained Alleviation of Autoimmunity by Activating α2B-adrenergic Receptors

Abstract:Catecholamines binding to α- and β-adrenergic receptors on immune cells have recently been shown to play an important role in regulating immune responses. Although α2-adrenergic receptors are known to modulate the immune response in different ways, the therapeutic exploration of their utility has been limited by the lack of agonists selective for the three α2-adrenergic subtypes. We report in this study the identification of the agonist AGN-762, which activates α2B- and α2C-adrenergic subtypes, but not the α2A subtype. We show that AGN-762 reduced clinical disease in an experimental autoimmune encephalitis model of autoimmune disease via direct or indirect effects on T regulatory cells. The activity of AGN-762 was abrogated by depletion of T regulatory cells, which express the α2B-adrenergic receptor. Furthermore, a drug-induced shift to an anti-inflammatory phenotype was demonstrated in immune cells in the spleen of drug-treated experimental autoimmune encephalitis mice. AGN-762 does not display sedative and cardiovascular side effects associated with α2A subtype agonists. Immune modulation by selective α2-adrenergic agonists represents a novel, to our knowledge, approach for treating autoimmune disease.

Abstract:The autonomic nervous system has been studied for its involvement in the control of macrophages; however, the mechanisms underlying the interaction between the adrenergic receptors and alternatively activated macrophages (M2) remain obscure. Using FVB wild-type and beta 2 adrenergic receptors knockout, we found that β2-AR deficiency alleviates hepatobiliary damage in mice infected with C. sinensis . Moreover, β2-AR-deficient mice decrease the activation and infiltration of M2 macrophages and decrease the production of type 2 cytokines, which are associated with a significant decrease in liver fibrosis in infected mice. Our in vitro results on bone marrow–derived macrophages revealed that macrophages from Adrb2 −/− mice significantly decrease M2 markers and the phosphorylation of ERK/mTORC1 induced by IL-4 compared to that observed in M2 macrophages from Adrb2 +/+ . This study provides a better understanding of the mechanisms by which the β2-AR enhances type 2 immune response through the ERK/mTORC1 signaling pathway in macrophages and their role in liver fibrosis.

Endothelial Intracellular ANG (Angiogenin) Protects Against Atherosclerosis by Decreasing Endoplasmic Reticulum Stress

Abstract:ANG (angiogenin) is essential for cellular adaptation to endoplasmic reticulum (ER) stress, a process closely associated with cardiovascular diseases, including atherosclerosis. We aimed to investigate the role of ANG in the progression of atherosclerosis and elucidate its underlying molecular mechanisms. We constructed adenoassociated virus 9 ANG overexpression vectors and endothelial ANG- and ApoE (apolipoprotein E)-deficient mice to determine the effects of ANG on ER stress and atherosclerotic lesions. RNA sequencing of endothelial ANG- and ApoE-deficient mice identified ANG-dependent downregulation of ST3GAL5 (ST3 beta-galactoside alpha-2,3-sialyltransferase 5) expression, and the direct regulation of ST3GAL5 by ANG was verified by chromatin immunoprecipitation sequencing and luciferase reporter assay results. Reanalysis of expression profiling datasets indicated decreased ANG levels in patients' atherosclerotic lesions, and these data were validated in aortas from ApoE − /− mice. ER stress marker and adhesion molecule levels, aortic root lesions and macrophage deposition were substantially reduced in ApoE −/− mice injected with an adenoassociated virus 9 ANG without signal peptide (ANG-ΔSP) overexpression vector compared with empty and full-length ANG overexpression vectors. Endothelial ANG deficiency significantly elevated ER stress and increased adhesion molecule expression, which aggravated atherosclerotic lesions and enhanced THP-1 monocyte adhesion to endothelial cells in vivo and in vitro, respectively. Furthermore, ANG-ΔSP overexpression significantly attenuated oxidized low-density lipoprotein-induced ER stress and THP-1 monocyte adhesion to endothelial cells, which were reversed by ST3GAL5 inhibition. These results suggest that endothelial intracellular ANG is a novel therapeutic against atherosclerosis and exerts atheroprotective effects via ST3GAL5-mediated ER stress suppression.
Biased Activation of Β2-Ar/gi/grk2 Signal Pathway Attenuated Β1-Ar Sustained Activation Induced by Β1-Adrenergic Receptor Autoantibody

Abstract:Heart failure is the terminal stage of many cardiac diseases, in which β1-adrenoceptor (β1-AR) autoantibody (β1-AA) has a causative role. By continuously activating β1-AR, β1-AA can induce cytotoxicity, leading to cardiomyocyte apoptosis and heart dysfunction. However, the mechanism underlying the persistent activation of β1-AR by β1-AA is not fully understood. Receptor endocytosis has a critical role in terminating signals over time. β2-adrenoceptor (β2-AR) is involved in the regulation of β1-AR signaling. This research aimed to clarify the mechanism of the β1-AA-induced sustained activation of β1-AR and explore the role of the β2-AR/Gi-signaling pathway in this process. The beating frequency of neonatal rat cardiomyocytes, cyclic adenosine monophosphate content, and intracellular Ca2+ levels were examined to detect the activation of β1-AA. Total internal reflection fluorescence microscopy was used to detect the endocytosis of β1-AR. ICI118551 was used to assess β2-AR/Gi function in β1-AR sustained activation induced by β1-AA in vitro and in vivo. Monoclonal β1-AA derived from a mouse hybridoma could continuously activate β1-AR. β1-AA-restricted β1-AR endocytosis, which was reversed by overexpressing the endocytosis scaffold protein β-arrestin1/2, resulting in the cessation of β1-AR signaling. β2-AR could promote β1-AR endocytosis, as demonstrated by overexpressing/interfering with β2-AR in HL-1 cells, whereas β1-AA inhibited the binding of β2-AR to β1-AR, as determined by surface plasmon resonance. ICI118551 biasedly activated the β2-AR/Gi/G protein-coupled receptor kinase 2 (GRK2) pathway, leading to the arrest of limited endocytosis and continuous activation of β1-AR by β1-AA in vitro. In vivo, ICI118551 treatment attenuated myocardial fiber rupture and left ventricular dysfunction in β1-AA-positive mice. This study showed that β1-AA continuously activated β1-AR by inhibiting receptor endocytosis. Biased activation of the β2-AR/Gi/GRK2 signaling pathway could promote β1-AR endocytosis restricted by β1-AA, terminate signal transduction, and alleviate heart damage.

Abstract:Abdominal aortic inflammation (AAI) is a major arterial vasculitis characterized by chronic inflammation and fibrosis. Endothelial cells transform into mesenchymal cells (EMT) is one of the significant mechanisms of vasculitis fibrosis. Despite its importance, the molecular mechanism of EMT in AAI remains poorly understood. In this study, we induced AAI in mice through intraperitoneal injection of tumor necrosis factor-alpha (TNFα). To analyze protein expression, we performed Western blotting. Additionally, we extracted RNA using the nanomagnetic bead method to investigate the expression of functionally related genes. We conducted cell migration and invasion assays using scratch and Transwell techniques. Western blot analysis revealed the upregulation of microfibril-associated protein-3-like (MFAP3L) and tumor necrosis factor (TNF) receptor 2 (TNFR2), along with p38 signaling pathway activation. Notably, MFAP3L expression played a crucial role in the transforming growth factor-beta (TGFβ)-induced EMT process in endothelial cells. Furthermore, we identified that MFAP3L-mediated EMT relied on both TNFR2 expression and the activity of the TNFR2/p38 signaling pathway.

Abstract:Background: Artery stiffness causes cardiovascular pathology. Although mechanisms underlying endothelial dysfunction and arterial stiffness are well characterized, it is unknown if neural-immune interaction in aortic adventitia (AA) influences arterial stiffness. We hypothesized that sympathetic innervation and CD68 + macrophages are increased in AA of Angiotensin II (Ang II)-infused mice leading to increased arterial fibrosis and stiffness, and that voluntary wheel-running exercise (VWR) mitigates these effects. Methods and Results: In eight-week-old C57BL/6 male mice, Ang II was infused at 1.5 μg/g/day for 4 weeks with and without VWR. Mean arterial blood pressure [MAP] measured by tail-cuff averaged 135±11 mmHg in Ang II-infused mice (n=5) and 90±10 mmHg in vehicle-infused mice (n=4). Staining for markers of nerve fibers/bundles with Neurofilament A (NF200), sympathetic nerves with tyrosine hydroxylase (TH), nerve endings in AA with synapsin, and CD68 + macrophages were all increased in AA removed from Ang II-infused mice compared with vehicle-infused mice. The increases in CD68 + macrophages and extracellular matrix (ECM) were associated with elevated pulse wave velocity (PWV), an index for arterial stiffness (Ang II: p < 0.05 vs. vehicle). scRNA-seq analyses further corroborated that Ang II infusion promoted fibrosis and macrophage-associated genes in AA. VWR significantly attenuated the Ang II-induced increases in MAP, sympathetic innervation, CD68 + expression, ECM, and PWV (Ang II+VWR: all p<0.05 vs. Ang II). To chemically “denervate” the sympathetic nervous system, we injected 6-hydroxydopamine (6-OHDA, 100 mg/kg on day 0 and 250 mg/kg on day 2, then weekly for 4 weeks) or vehicle in Ang II-infused mice. 6-OHDA decreased MAP, sympathetic innervation, macrophage expression, ECM accumulation, and PWV (Ang II+6-OHDA: p<0.05 vs. Ang II+vehicle, n=3). Conclusion: Ang II-induced hypertensive mice develop increases in sympathetic innervation and macrophages in close proximity within AA that define an enhanced sympathetic neural-immune interface in association with vascular fibrosis, arterial stiffness, and hypertension. VWR significantly mitigates these pathophysiological responses.
