Abstract:The comparison of the protective effects of four components of "green tea polyphenols' (GTP) - (-)-epigallocatechin gallate, EGCG; (-)-epicatechin gallate, ECG; (-)epigallocatechin, EGC; and (-)epicatechin, EC - against iron-induced lipid peroxidation in synaptosomes showed that: (1) the inhibitory effects of those compounds on TBA reactive materials from lipid peroxidation decreased in the order of EGCG > ECG > EGC > EC; (2) the scavenging effects of those compounds on lipid free radicals produced by lipid peroxidation could be classified as follows: ECG > EGCG > EC > EGC. Furthermore, we investigated the iron-chelating activity and the free radical scavenging activity of those compounds as their protective mechanisms against lipid peroxidation in synaptosomes. As for the iron-chelating activity, the ratio of EGC, EGCG, ECG or EC to iron(III) was 3:2, 2:1, 2:1 and 3:1, respectively. The hydroxyl radical (HO) scavenging activity of those compounds was investigated in a photolysis of the H2O2 system. It was found that their ability to scavenge hydroxyl radicals decreased in the order of ECG > EC > EGCG >> EGC. It was also found that they could scavenge lipid free radicals in the lecithin/lipoxidase system and their scavenging activity was classified as follows: ECG > EGCG >> EGC > EC. Moreover, we found that their antioxidant active positions were different from each other and the stability of the semiquinone free radicals produced by those compounds in NaOH solution decreased in the order of EGCG > ECG >> EC. The results indicated that the ability of those compounds to protect synaptosomes from the damage of lipid peroxidation initiated by Fe2+/Fe3+ was dependent not only on their iron-chelating activity and free-radical scavenging activity, but also on the stability of their semiquinone free radicals.

Abstract:Tea polyphenols is an ideal natural anti-oxidant,which has being widely applied in many fields.The methods of extraction of tea polyphenols from green tea have been become one of hot points of tea research.In the present paper,methods of extraction of tea polyphonols from green tea were reviewed,including solvent extraction,ionic precipitation,supercritical CO2 extraction,resin adsorption,ultrasonic assistant extraction,microwave assistant extraction and so on.We also discussed basic principle,producing routine and advantage or disadvantage of each method at the same time.

Abstract:The pu-erh tea was extracted with water and separated by the order of chloroform,ethyl acetate and n-butanol.The antioxidant activity of pu-erh tea extracts was investigated by the 2,2-diphenyl-1-picrylhydrazyl(DPPH) free radical-scavenging assay.The result pointed pu-erh tea ethyl acetate extract(PEAF)had the strongest DPPH scavenging activity.High performance liquid chromatography combined with diode array and electrospray-ion trap-mass spectra detection was used to separate and identify the compounds present in PEAF.There are three phenolic acids,i.e.,4-hydroxy-benzoic acid,dihydroxy-benzoic acid and gallic acid,seven catechins,i.e.,catechin,epicatechin(EC),gallocatechin,epigallocatechin(EGC),epicatechin gallate,epigallocatechin gallate and epiafzelechin-3-O-gallate,nine catechins derivatives,i.e.,procyanidins dimmer,puerin A,puerin B,EC-8,7-e-4α-(4-hydroxyphenyl)-3,4-dihydro-2(3H)-pyranone,EC-8,7-e-4β-(4-hydroxyphenyl)-3,4-dihydro-2(3H)-pyranone,Cinchonain Ib(α and β),EGC-8,7-e-4α-(3,4-dihydroxyphenyl)-3,4-dihydro-2(3H)-pyranone and EGC-8,7-e-4β-(3,4-dihydroxyphenyl)-3,4-dihydro-2(3H)-pyranone,and six flavonol glycosides,i.e.,quercetin,quercetin-glucoside,quercetin-rhamnoside,quercetin-rutinoside,quercetin-glucoside-rhamnoside-rhamnoside and kaempferol-rutinoside.
The Research of the Relationship between Antioxidation of Tea Extractive and Tea Polyphenols as Well as Catechins

Abstract:In this experiment, six kinds of tea extractive were tested as research objects, their total phenol content, catechins content, antioxidant activity and free radical scavenging activities were determined, and the relationships between them were analyzed according to the results. The analysis showed that the correlation coefficient between antioxidant activity and total phenol content is higher than that between antioxidant activity and total catechins content as well as EGCG by and large, indicating that tea polyphenols without catechins also has high antioxidant ability; the correlation coefficient between antioxidant activity and EGCG content is higher than that between antioxidant activity and total catechins, indicating that antioxidant activity of EGCG is higher than that of catechins' average; the results also showed that free radical scavenging ability of tea polyphenols, catechins and EGCG is different from each other.

Abstract:In the present study, we successively extracted the Pu-erh tea with acetone, water, chloroform, ethyl acetate, and n-butanol, and the extracts were then isolated by column chromatography. Our study demonstrates that the Pu-erh tea ethyl acetate extract, n-butanol extract, and their fractions had superoxide anion and hydroxyl radical scavenging activity: fractions 2 and 8 from the ethyl acetate extract and fractions 2, 4, and 5 from the n-butanol extract showed protective effects against hydrogen peroxide-induced damage in human fibroblast HPF-1 cells and increased the cells' viability under normal cell culture conditions. In addition, it is found that these fractions, except fraction 5 from the n-butanol extract, decreased the accumulation of intracellular reactive oxygen species in hydrogen peroxide-induced HPF-1 cells. Interestingly, the antioxidant effect of fraction 8 from the ethyl acetate extract on the above four systems was much stronger than that of the typical green tea catechin (-)-epigallocatechin-3-gallate, but there were almost no monomeric polyphenols, theaflavins, and gallic acid in fraction 8.

Abstract:The water-extracted pu-erh tea and black tea were separated by the order of chloroform,ethyl acetate,and n-butanol.The antioxidant activity of these teas and their fractions were investigated by the hydroxyl radical-induced luminol chemiluminescence assy and visible spectrophotometric method.The results revealed that black tea had slightly stronger antioxidant activity than pu-erh tea in water phase.The two types of tea had considerable antioxidant effect on some lipid.The ethyl acetate fraction was the most contributions to the antioxidant activity of the two types of tea in water phase which antioxidant activity was a little stronger from black tea than that from pu-erh tea,and had scarcely any antioxidant activities in oil phase.

Abstract:The pu-er tea extract treated with hot w at er was re-extracted with different organic regents step by step, got four kind of fractions, such as chloroform fraction, ethyl acetate fraction, n-butano l fraction and the other fraction. To compare the antioxidative activity of thes e fractions, the antioxidative effect was assayed in a Fenton-type reaction sys tem by chemiluminescence, and in the 2,2-diphenyl-1-picrylhydrazyl (DPPH) rad ical scavenging effect by spectrophotometer. The result showed that the strong s cavenging effect of the ethyl acetate fraction and the n-butanol fraction o n the DPPH radical and the hydroxyl radicals was observed.

Abstract:The antioxidative activity,aroma constituents and the major chemical components of three grades of Pu-erh tea,i.e.high,medium and low grade,were studied and compared.Results showed that the significant difference(P0.05)was shown in evaluation of the total antioxidative activity,scavenging hydroxyl free radical activity,scavenging DPPH free radical activity,total amount of alcohols compounds,total amount of heterocyclic oxygen compounds,total flavonoids and tea pigments between high and low grades.When compared with high or low grade,the medium grade of Pu-erh tea was found no significant difference(P0.05)in the others testing index except the total antioxidative activity,scavenging hydroxyl free radical activity,tea polyphenols and total flavonoids.Principal component analysis showed that the important indicator to distinguish the grade of Pu-erh tea seemed to be the scavenging hydroxyl free radical activity,the total antioxidative activity,tea polyphenols,total flavonoids and total amount of alcohols compounds.

Abstract:The antioxidant synergistic effects of Osmanthus fragrans flowers with green tea were evaluated, and their major antioxidant compounds contributed to the total amount of synergy were determined. The antioxidant compounds in O. fragrans flowers with green tea were identified by LC-MS and quantified by UPLC-PDA. The synergistic antioxidant interactions between O. fragrans flowers with green tea and their antioxidant compounds were tested using the Prieto's model after the simulated digestion. The main antioxidant compounds in O. fragrans flowers were acteoside and salideroside, whereas the main antioxidant compounds in green tea were caffeine, gallic acid, and L-epicatechin. The significant synergistic effect between O. fragrans flowers and green tea was observed and among nearly all of the combinations of their antioxidant compounds. Among the combinations, acteoside and gallic acid contributed most to the antioxidant synergy between O. fragrans flowers and green tea. However, the simulated digestion decreased this antioxidant synergy because it reduced the contents and the antioxidant capacities of their compounds, as well as the antioxidant synergy among the compounds.

Abstract:Extracts of raw and ripe Pu-erh tea were prepared from tea dust during processing.The antioxidtive activities of extracts were evaluated by ABTS and FRAP systems.The potentially protective effects of extracts on human umbilical vascular endothelial cells(HUVEC) were investigated in Na2S2O3-induction model.The results showed that the extract of raw Pu-erh tea had stronger antioxidtive activity than that of ripe Pu-erh tea in both test systems.But extract of ripe Pu-erh tea showed a less protective effect than that of extract of raw Pu-erh tea on the HUVEC cell damage induced by Na2S2O3,and relevent with tea polyphenols under certain concentrations.

Abstract:Oolong tea (OT) is unique for its flavor and healthy effects. We have reported that the polypheonols of OT possessed antioxidant anticancer, and hypolipidemic effects. In addition, tea polysaccharides (TPS) have long been recognized to have antioxidant, immunomodulatory and antidiabetic properties. Since there are three typical kinds of OTs, e.g. Tieguanyin, Fenghuangdancong and Dahongpao with the lightest, intermediate and the highest degree of fermentation respectively, it is important for us to know whether fermentation degree affected the production and their bioactivities of tea polysaccharide. In the present study, three polysaccharides named TTPS, FTPS and DTPS were isolated from these three typical kinds of oolong teas in the above order respectively. Physicochemical characteristics, in vitro antioxidant activity and alpha-glucosidase inhibitory effect of TTPS, FTPS and DTPS were studied. Analysis of the chemical compositions (protein, rot neutral sugars and uronic acid) revealed that they all belonged to acid heteropolysaccharides bound with protein. The contents of protein and uronic acid but rot neutral sugars proportionally increased with degree of fermentation, highest in DTPS, which significantly correlated with their potencies in antioxidant and alpha-glucosidase inhibitory activities. Analysis of neutral sugar compositions showed that they all contained seven monosaccharides with different molar ratio. Measurements of their molecular weight by high performance gel permeation chromatography indicated that 92.9% of TTPS and 94.4% of DTPS had a peak of molecular weight of 0.82×106 and 2.64×106 respectively, but that of FTPS had 34.2% with 0.93×106 and 68.8% with 0.01×106 respectively. Fourier Transform IR spectra exhibited that they all had similar characteristic absorption bands of uronic acid, protein, polysaccharide and pyran-glycosides respectively. Furthermore, measurements of antioxidant activities by assays of DPPH, ABTS and ferric-reducing antioxidant power (FRAP) showed that DTPS was the most potent, FTPS the next and TTPS the least respectively. Similarly, DTPS possessed the strongest inhibitory potential against alpha-glucosidase activity, followed by FTPS and then TTPS. The potencies of the bioactivities of these three OTPS were apparently proportional to their contents of protein and uronic acid, suggesting that the higher degree of fermentation of oolong teas produced higher contents of protein-bound acid heteropolysaccharides accompanied with higher antioxidant and alpha-glucosidase inhibitory activities. Perhaps, DTPS had a potential to play a role in prevention of type 2 diabetes mellitus. Further studies to confirm this contention is in progression.
Scavenging Effects of Tea Catechins on Superoxide and Hydroxy Radical

Abstract:Tea catechins, the most important compounds in tea polyphenols, can efficiently scavenge superoxide anion free-radical (O₂-·), hydroxyl radical. (·OH) The mechanism of scavenging active oxygen free radicals was investigated by ESR spin trapping technique and Chemiluminescence. Results showed that various tea catechins constitute an antioxidant cycle in accordance with the decreasing order of the first reductive potential, and produce the effect of cooperative strength each other. Esterificated catechins could scavenge active oxygen free radicals more effectively than the non-esterificated ones. When ·OH and O₂-·- were scavenged by (-)-epigallocatechin gallate [(-)- EGCG], the stoichiometric factors were 6, and the rate constants of scavenging reaction reached 7.71x10(6) and 3.52x10(11) L mmol-¹ S-¹, respectively. In the mean time, tea catechins could scavenge superoxide anion fiee radical (O₂-·) and hydroxyl radical (·OH) in a dose dependent manner. But at higher concentration or pH value, tea catechins can induce the prooxidant.

Abstract:Tea is among the most consumed drink worldwide, and its strong antioxidant activity is considered as the main contributor to several health benefits, such as cardiovascular protection and anticancer effect. In this study, the antioxidant activities of 30 tea infusions, which were obtained by the mimic of drinking tea of the public, from green, black, oolong, white, yellow and dark teas, were evaluated using ferric-reducing antioxidant power (FRAP) and Trolox equivalent antioxidant capacity (TEAC) assays, ranging from 504.80 ± 17.44 to 4647.47 ± 57.87 μmol Fe2+/g dry weight (DW) and 166.29 ± 24.48 to 2532.41 ± 50.18 μmol Trolox/g DW, respectively. Moreover, their total phenolic contents (TPC) were detected by Folin-Ciocalteu assay and were in the range of 24.77 ± 2.02 to 252.65 ± 4.74 mg gallic acid equivalent (GAE)/g DW. Generally, Dianqing Tea, Lushan Yunwu Tea, and Xihu Longjing Tea showed the strongest antioxidant activities among 30 teas. Furthermore, the phenolic compounds in tea infusions were identified and quantified, with catechins most commonly detected, especially in green tea infusions, which were main contributors to their antioxidant activities. Besides tea polyphenols, considerable content of caffeine also presented in 30 tea infusions.

Abstract:Tea (Camellia sinensis L.) is categorized into four types depending on the degree of fermentation, which in turn affects the physicochemical properties of tea. In this work, aqueous extracts of four typical teas, named green tea, oolong tea, black tea and puerh tea, were obtained, and their chemical compositions and bioactivities including antioxidant activity and inhibition on α-amylase and α-glycosidase were compared. It was found oolong and green tea not only possessed stronger antioxidant ability in assays of DPPH, ABTS and FRAP, but also had more power on inhibition on α-amylase and α-glycosidase. Pearson correlation analysis showed tea polyphenols and catechins had significant positive correlations on α-amylase inhibition. However, caffeine was observed to have a significant negative effect on α-glycosidase inhibition. These results indicated teas with no or slight fermentation (decaffeinated was better), were suitable for diabetes.

Abstract:Puerh tea is one of the most popular (or healthiest) beverages in Southeast Asia, because of its higher antioxidant, hypolipidemic and antidiabetic effects. One of the important active ingredients, tea polysaccharides (TPS) has long been recognized to possess immune modulatory and antidiabetic effects. In the present work, polysaccharides were isolated from puerh tea (PTPS) with aging (mild fermentation) for 1 year (PTPS-1), 3 years (PTPS-3) and 5 years (PTPS-5), respectively. The results obtained indicated that all of these three kinds of PTPS are acid heteropolysaccharides bound with proteins, with differences in molecular weights and in monosaccharide compositions. The longer aging process not only produced a higher yield of PTPS but also had higher contents of proteins and uronic acids which proportionally correlated with their antioxidant and α-glycosidase inhibitory activities (PTPS-5 > PTPS-3 > PTPS-1). The anti-α-glycosidase activities of PTPS-5 and PTPS-3 were found to be three times more and equally potent than that of the antidiabetic drug acarbose. All of these findings firstly indicate that the aging process not only induced changes in chemical compositions and proteins conjugation in PTPS, but also markedly enhanced antioxidant and anti-α-glycosidase activities. PTPS-5 exhibited potential antidiabetic effect and merits further studies.

Abstract:In this study, the chemical constituents of pu-erh tea, black tea, and green tea, as well as those of pu-erh tea products of different ages, were analyzed and compared using a chemical profiling approach. Differences in tea processing resulted in differences in the chemical constituents and the color of tea infusions. Human biological responses to pu-erh tea ingestion were also studied by using ultraperformance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOFMS) in conjunction with multivariate statistical techniques. Metabolic alterations during and after pu-erh tea ingestion were characterized by increased urinary excretion of 5-hydroxytryptophan, inositol, and 4-methoxyphenylacetic acid, along with reduced excretion of 3-chlorotyrosine and creatinine. This study highlights the potential for metabonomic technology to assess nutritional interventions and is an important step toward a full understanding of pu-erh tea and its influence on human metabolism.

Abstract:Pu-erh tea is a Chinese dark tea produced by sun-dried green tea-leaves. In this study, 17 phenolic compounds and 3 purine alkaloids in 36 Pu-erh teas were determined by high-performance liquid chromatography. Additionally, the in vitro antioxidant activities were evaluated by 5 various assays, respectively. PCA, HCA and OPLS-DA (R2X = 0.639, R2Y = 0.945 and Q2 = 0.940, respectively) could divide into fermented Pu-erh tea (FPT) and non-fermented Pu-erh tea (NFPT) groups absolutely. Compared with NFPT, solid-state fermentation caused the extremely significant (p < 0.001) decreases of 8 catechins, 3 other flavonoids (i.e. taxifolin, rutin and luteolin), gallic acid (GA), hydrolyzed tannin and theophylline, and enhanced ellagic acid, kaempferol and quercetin contents significantly (p < 0.05, <0.01 or <0.001) in FPT. Bivariate correlation analysis confirmed that 8 catechins, 3 flavonoids, theophylline and GA promoted the antioxidant activities of Pu-erh teas, while queretin showed extremely significantly (p < 0.001) negative correlation to the results of DPPH, ABTS and HSA assays. Furthermore, multivariate linear regression equations efficiently evaluated the results of FRAP, DPPH, ABTS and HSA assays in Pu-erh tea with a R2 value over 0.95. Metabolic fate of tea polyphenols in humans.

Abstract:Polyphenols, a ubiquitous group of secondary plant metabolites sharing at least one aromatic ring structure with one or more hydroxyl groups, represent a large group of natural antioxidants abundant in fruits, vegetables, and beverages, such as grape juice, wine, and tea, and are widely considered to contribute to health benefits in humans. However, little is yet known concerning their bioactive forms in vivo and the mechanisms by which they may alter our metabolome, which ultimately contribute toward disease prevention. Here we report a study to determine the metabolic fate of polyphenolic components in a Chinese tea (Pu-erh) in human subjects using a metabonomic profiling approach coupled with multivariate and univariate statistical analysis. Urine samples were collected at 0 h, 1 h, 3 h, 6 h, 9 h, 12 h, and 24 h within the first 24 h and once a day during a 6 week period including a 2 week baseline phase, a 2 week daily Pu-erh tea ingestion phase, and a 2 week "wash-out" phase, and they were analyzed by gas chromatography mass spectrometry and liquid chromatography mass spectrometry. The dynamic concentration profile of bioavailable plant molecules (due to in vivo absorption and the hepatic and gut bacterial metabolism) and the human metabolic response profile were measured and correlated with each other. This study demonstrates that the metabonomic strategy will enable us to integrate the overwhelming amount of metabolic end points as a systems' response to the absorption, metabolism, and disposition of a multicomponent botanical intervention system, leading to a direct elucidation of their mechanisms of action.
