Abstract:The aim of this study was to prepare and characterize nitrendipine nanosuspensions to enhance the dissolution rate and oral bioavailability of this drug. Nanosuspensions were prepared by the precipitation-ultrasonication method. The effects of five important process parameters, i.e. the concentration of PVA in the anti-solvent, the concentration of nitrendipine in the organic phase, the precipitation temperature, the power input and the time length of ultrasonication on the particle size of nanosuspensions were investigated systematically, and the optimal values were 0.15%, 30 mg/ml, below 3 degrees C, 400 W and 15 min, respectively. The particle size and zeta potential of nanocrystals were 209 nm (+/- 9 nm) and -13.9 mV (+/-1.9 mV), respectively. The morphology of nanocrystals was found to be flaky in shape by scanning electron microscopy (SEM) observation. The X-ray powder diffraction (XRPD) and differential scanning calorimetry (DSC) analysis indicated that there was no substantial crystalline change in the nanocrystals compared with raw crystals. The in vitro dissolution rate of nitrendipine was significantly increased by reducing the particle size. The in vivo test demonstrated that the C(max) and AUC(0-->12) values of nanosuspension in rats were approximately 6.1-fold and 5.0-fold greater than that of commercial tablets, respectively.

While the captopril concentration was detected at 0.5 h after the oral administration, and the peak blood level occurred in 1-3 h after the oral administration, the accumulate release percent in vitro was about 20% at 1 h, for the common delayed tablets. CONCLUSION: There are some correlation between release in vitro and absorption of the delayed-onset in vivo, sustained release tablets, and the remarkable lag time is observed, compared with the common delayed tablets.

Abstract:Hypertension continues to be a significant public health concern because of its associated morbidity, mortality, and economic impact on society. By the year 2025, 1.56 billion individuals will have hypertension. Nitrendipine, an antihypertensive drug, is used in this present study. Hydrophilic matrices are capable of controlling the release of the drug over an extended period. Microsphere based sustained drug delivery systems will provide a suitable alternative to reduce the side effects and increase the dissolution rate of the drug compared to oral drug delivery. We prepared drug encapsulated microspheres of acrylamide grafted chitosan (hydrophilic polymers) by the free radical mechanism. This mechanism involves the use of an initiator, potassium persulfate. Microspheres were synthesized from the grafted copolymers using water in oil emulsion technique with a cross-linking agent. Differential scanning calorimetry, scanning electron microscopy, and Fourier transform infrared spectroscopy were used to characterize the sample. The microspheres at stomach media showed low drug release characteristics, while at the intestinal organ, the drug release prolonged for 14 h. From the drug release kinetic models, Higuchi and zero-order were found to be the best model for the three ratios based on the correlation coefficient. The diffusion component is less than 0.5, which indicates quasi-fickian diffusion. From the kinetic study results, we concluded that the formed acrylamide grafted chitosan microspheres deliver the drug through the diffusion mechanism. Nitrendipine loaded microspheres prepared using water in oil emulsion technique showed favorable sustained drug release behavior.

Abstract:The orally disintegrating tablets of micronized levamlodipine besylate were prepared by direct powder compression with lactose and mannitol as fillers and polyplasdone as disintegrant. The formulation was optimized by orthogonal design. The uniformity of dosage units was good. The mean disintegrating time of the tablets in vitro and in oral cavity of six volunteers were both within 30 s. The in vitro dissolution of the tablets was above 90% in 5 min.

Abstract:OBJECTIVE To prepare captopril delayed-onset,sustained release tablets that start releasing after a lag time 5h. METHODS Captopril delayed-onset,sustained release tablets were prepared with dry-coated technology.Central composite design was used to optimize the formulation, artificial neural networks was used to predict the lag time of release,and multiple regression was performed using statistic analysis software (SAS).The observed values of the lag time of captopril delayed-onset,sustained release tablets prepared under the optimum condition were compared with the predicted.The out shell composition was confirmed. The composition of core tablets was optimized using captopril sustained release tablets as reference preparation and the similarity factor(f_ 2 ) as the evaluation standard. Then the formulation of captopril dry-coated tablets was finally confirmed.RESULTS The lag time in vitro of captopril delayed-onset,sustained release tablets prepared under the optimum condition was 5h. The tablets had similar release behavior to reference products(f_ 2 =64.06) and the drug release pattern was accorded with the first order kinetics.CONCLUTION The lag time of this dry-coated tablets is only controlled by the composition of out shell. The sustained release after release starting is controlled by both out shell and core tablet.
Preparation and characterization of nitrendipine solid lipid nanoparticles

Abstract:Nitrendipine, a dihydropyridine calcium channel blocker, has very poor oral bioavailability (10-20%) due to first pass effect. Solid lipid nanoparticle (SLN) delivery systems of nitrendipine have been developed using various triglycerides (trimyristin, tripalmitin and tristearin), soy phosphatidylcholine 95%, poloxamer 188 and charge modifiers stearylamine and dicetyl phosphate. SLNs were prepared by hot homogenization of melted lipids and aqueous phase followed by ultrasonication at temperatures above the melting point of lipids. Optimization studies of process and formulation variables were carried out. Particle size and zeta potential were measured by photon correlation spectroscopy (PCS) using Malvern zetasizer. Differential scanning calorimetry (DSC) and powder X-ray diffraction (PXRD) studies were performed to characterize state of drug and lipid modification. In vitro release studies were performed in phosphate buffer pH 6.8 using modified Franz diffusion cell. Stable nitrendipine SLNs of mean size range 79 to 213 nm and zeta potential -38.2 to +34.6 mV were developed. About 99% nitrendipine was entrapped in SLNs and were stable on storage at 4 and 25 degrees C. DSC and PXRD analyses revealed that nitrendipine is dispersed in SLNs in an amorphous state. The release pattern of drug is analyzed and found to follow Weibull distribution rather than first order and Higuchi equation.

Abstract:Objective To prepare sustained-release formulation of pramipexole dihydrochloride and study its in vitro release behaviors.Methods Pramipexole dihydrochloride sustained-release tablets were prepared with Hypromellose(HPMC) as matrix material,and Ethylcellulose(EC) as blockers.Technological parameters of tablets were screened with the accumulative releasing rate in vitro as index.Results In the optimal formulation,0.214% of pramipexole dihydrochloride,37.0% of HPMC K100M,38.0% of EC,23.0% of Pre-starch,1.00% of aerosil and 0.786% of magnesium stearate were selected as the main excipients.The release ratio of the optimal formulation was up to 90% in 24 hours and the pH value of release medium had no significant effects on the release of pramipexole dihydrochloride sustained-release tablets.The formulation demonstrated a typical extended release profile in the in vitro test.Conclusion The formulation had a good profile of extended releasing.The preparation process is simple,which is worth further development and application in the clinic.

Abstract:OBJECTIVE: To evaluate the pharmacokinetics and bioavailability of nimodipine nanostructured lipid carrier in rats. METHODS: A RP-HPLC method was established to determine the concentration of nimodipine in plasma. Nimodipine loaded nanostructured lipid carrier (NLC) were prepared by solvent dispersion method, and its pharmacokinetic parameters and bioavailability in SD rats were studied after administration. RESULTS: Compared with nimodipine tablet, the tmax of nimodipine loaded NLC was delayed by 2.3 h, and the bioavailability of nimodipine NLC was increased by 2. 26 folds. CONCLUSION: Nimodipine loaded NLC obviously enhanced the bioavailability of nimodipine in rats after oral administration.

Abstract:OBJECTIVE To study the drug release characteristics in vitro and the pharmacokinetic properties in vivo of self-made nifedipine controlled-release tablets using the imported preparation as the reference.METHODS To study the effect of pH value of media on the drug release characteristic of nifedipine controlled-release tablets,four types of release media with different pH values were used respectively.And for the release media of pH 6.8,three rotating speeds were taken to study its effect on the drug release.The blood drug concentrations of healthy male volunteers after administration with a single-dose of 30 mg nifedipine controlled-release tablet were determined by method of GC.RESULTS The drug release profiles in different media with various pH values were similar,and the drug release was of zero-order kinetics.The drug release was not affected by the rotating speed within the range studied.The main pharmacokinetic parameters of the self-made preparation and the imported preparation after single dose administration were as follows: t1/2β(13.0±4.4)h and(12.7±4.7) h;tmax(14.2±7.8) h and(16.7±7.6) h;Cmax(29.3±10.2) μg·L-1and(25.6±8.1) μg·L-1;AUC0-60(778.2±298.1) μg·L-1·h and(735.5±261.3) μg·L-1·h;AUC0-∞(834.3±334.1) μg·L-1·h and(798.8±283.6) μg·L-1·h.The relative bioavailability of the self-made preparation versus the imported preparation was as follows:F(AUC0-60)(106.4±17.0) % and F(AUC0-∞)(105.4±18.2) %.There was no significant difference in Cmax、AUC0-60、AUC0-∞ and tmax between the two preparations.CONCLUSION The pH values of release media and the rotating speed have no effect on the drug release.The test preparation is bioequivalent to the reference preparation.
Improvement of dissolution and bioavailability of nitrendipine by inclusion in hydroxypropyl-beta-cyclodextrin

Abstract:A significant increase in solubility and dissolution rate of nitrendipine, a slightly soluble calcium channel blocker, was achieved by inclusion complexation with hydroxypropyl-beta-cyclodextrin (HP-beta-CD). The inclusion complex was prepared by solvent evaporation method and characterized by phase solubility method, x-ray diffractometry, infrared spectroscopy, and differential scanning calorimetry. The solubility of nitrendipine increased linearly as a function of HP-beta-CD concentration, resulting in AL-type phase solubility diagram which revealed a formation of inclusion complex in a molar ratio of 1:1, with the apparent association constant of 108.3M(-1). The in vitro dissolution rate of nitrendipine in pH 7.4 phosphate buffer was in the order of inclusion complex, physical mixture, and nitrendipine powder. These three different forms of nitrendipine were administered orally to rats with a dose of 10 mg/kg equivalent to nitrendipine. The AUC of inclusion complex was significantly larger than that of nitrendipine powder. Tmax of inclusion complex was significantly shorter and Cmax was significantly higher than those of nitrendipine powder. Cmax of physical mixture was higher than that of nitrendipine powder. Tmax of physical mixture, however, remained the same. The results indicated that the bioavailability of nitrendipine could be improved markedly by inclusion complexation, possibly due to an increased dissolution rate.
Preparation of sustained-release nitrendipine microspheres with Eudragit RS and Aerosil using quasi-emulsion solvent diffusion method

Abstract:Sustained-release nitrendipine microspheres were prepared in liquid system by quasi-emulsion solvent diffusion method, in which the Aerosil was employed as an inert dispersing carrier to improve the dissolution rate of nitrendipine, and Eudragit RS as a retarding agent to control the release rate. The resultant microspheres were evaluated for the recovery, bulk density, average particle size, drug loading, and incorporation efficiency. And the factors affecting the formation of microspheres and the drug-release rate were investigated. It was observed by a scanning electron microscope (SEM) that the microspheres were finely spherical and uniform, and no entire nitrendipine crystals were observed visually. The results of X-ray diffraction indicated that nitrendipine in microspheres was disordered, suggesting that nitrendipine was highly dispersed in microspheres. The drug loading of microspheres was enhanced with increasing the ratio of drug to excipients, and the incorporation efficiency was always >90%. The formation of microspheres was mainly influenced by the amount of bridging liquid and sodium dodecyl sulfate (SDS) in poor solvent. The dissolution profiles could be modulated with adjusting the amount of retarding agent and dispersing carrier formulated.
Comparative Research on the Properties of Nitrendipine Solid Dispersions Prepared by Different Techniques

Abstract:OBJECTIVE To compare the properties of nitrendipine/PVP k25 solid dispemions prepared by three different technologies including spray drying,freeze-drying and co-precipitation technology.METHODS The characteristics of nitrendipine solid dispersions including micromorphology,crystalline profile and intermolecular interactions were analyzed by scanning electron microscopy (SEM),differential scanning calorimetry (DSC),powder X-ray diffraction (PXRD) and Fourier transform infrared spectroscopy (FF-IR).The dissolution and solubility characteristics were investigated by dissolution apparatus and constant temperature shaker.The crystallization inhibition effect of polymers against the drug in a supersaturated state was investigated by supersaturation crystallization experiment.RESULTS Nitrendipine existed in an amorphous form in the solid dispersion obtained by spray drying method.Compared with co-precipitation and freeze-drying method,the solid dispersion prepared by spray drying method significantly improved the dissolution and solubility of nitrendipine (P ＜ 0.05).The results of supersaturation crystallization inhibition test showed that PVP k25 could obviously inhibit the crystallization of nitrendipine in supersaturated state.CONCLUSION Based on the analysis of the above mentioned properties,nitrendipine solid dispersion prepared by spray drying technology displays the most optimal properties.Spray drying technology is more suitable for the practical production and industrial applications of nitrendipine solid dispersions.
Preparation and in vitro evaluation of a novel combined multiparticulate delayed-onset sustained-release formulation of diltiazem hydrochloride

Abstract:A combined delivery system, containing two kinds of diltiazem hydrochloride multi-layer coated pellets with different release characteristics, was developed to meet chronotherapeutic requirements. The dissolution studies in vitro indicated that the combined system could constantly release drug at a predetermined time in synchrony with the biologic rhythm of disease activity. These two kinds of pellets were mixed at the ratio 1:1. Pellet 2 could provide drug during the later phase of drug release from pellet 1, because the amount released was insufficient in the later phase (about 14-24 h after administration) for pellet 1. Addition of Tween 20 in the HPMC swelling layer was able to modify the hydration rate of HPMC layer which controlled the delayed time of the release system. It was found that the drug release kinetics followed Hixson-Crowell equation and this indicated that the main drug-transport mechanism inside the pellets was an erosion mechanism. The drug release rate was independent of the hydrodynamic conditions and pH of the external environment, and showed a decrease with increasing of osmotic pressure of the dissolution medium. These release characteristics indicated that the drug release from this system was driven by osmotic-pressure gradient.
Fast disintegrating tablets of nisoldipine for intra-oral administration

Abstract:Nisoldipine is a calcium channel blocker with low and variable oral bioavailability. This was attributed to slow dissolution and presystemic metabolism. Accordingly, the objective of this work was to enhance the dissolution rate of nisoldipine to formulate fast disintegrating tablets with rapid dissolution. Binary solid dispersions (SD) were prepared for the drug with hydroxypropyl methyl cellulose E5 (HPMC), polyvinylpyrrolidone (PVP), Pluronic F68 or polyethylene glycol 6000 (PEG 6000). SD formation increased the dissolution rate compared to pure drug with the corresponding physical mixtures failing to provide the same dissolution enhancement. This indicates that the SD enhanced dissolution is not due to the solubilizing effect of the polymer and can be due to physical change in the drug crystal which was confirmed by thermal analysis. SD with HPMC and PVP were selected for preparation of fast disintegrating tablets as they liberated most of the drug in the first 5 min. HPMC-based tablets disintegrated rapidly and released most of the drug in the first 2 min which correlated with the corresponding SD. In contrast, PVP-based tablets disintegrated slowly with gradual dissolution. This can be attributed to the binding effect of PVP. The study developed fast disintegrating tablet for intra-oral administration.

Abstract:Objective: To investigate the preparation of pulsatile release tablets, the release of the drug in vitro and the pharmacokinetics in vivo . Methods: Dil tiazem hydrochloride（DIL） was used as model drug. The pulsatile release tabl e ts were prepared by dry-coated method with carnauba wax, bee wax and hydrophil i c cellulose as coating materials. The effects of formulation and technology on t he release characteri stic of diltiazem hydrochloride was investigated. The mechanism of pulsatile rel ease of the drug was proved by erosion test. The pharmacokinetic study on four h uman subjects was done by means of HPLC measurement. Results: In vitro ， delayed-release ti me t 10 was 2.1　h, the maximum release time t rm 4.0　h and t he pulsed-releas e time t 10-90 1.7　h. In vivo ， delayed-release time t la g was 5.7　h, the p eak time 8.5　h and the pulsed-release time 2.6　h. Conclusion: The rele ase of drug from pulsatile-released tablets of diltiazem hydrochloride was in a pulsed way both in vitro and in vivo .

Abstract:OBJECTIVE To study the correlation between the dissolution in vitro and the absorption in vivo of nimodipine tablets.METHODS The dissolution of nimodipine in vitro was tested according to Chinese pharmacopoeia 2000, using the first method with RC dissolution tester. A single oral dose of nimodipine was given to 8 healthy male volunteers. Nimodipine concentration in plasma were determined by HPLC and the absorption percentage was calculated according to Loo-Reigelman formula. RESULTS The linear regressive equation established between the absorption percentage in vivo and the dissolution percentage in vitro of nimodipine tablets were Y= 2.230 4X -60.72,r = 0.9983(P0.01)and Y= 3.708 5X - 107.41, r = 0.895 1( P 0.05) ,respectively.CONCLUSION The results suggested that there was a significant correlation between the absorption in vivo and the dissolution in vitro of nimodipine tablets.

Abstract:The aims of this study were to enhance the solubility and dissolution rate of nimodipine (ND) by preparing the inclusion complexes of ND with sulfobutylether-b-cyclodextrin (SBE-β-CD) and 2-hydroxypropyl-b-cyclodextrin (HP-β-CD) and to study the effect of the preparation method on the in vitro dissolution profile in different media (0.1 N HCl pH 1.2, phosphate buffer pH 7.4, and distilled water). Thus, the inclusion complexes were prepared by kneading, coprecipitation, and freeze-drying methods. Phase solubility studies were conducted to characterize the complexes in the liquid state. The inclusion complexes in the solid state were investigated with differential scanning calorimetry (DSC), X-ray diffractometry (X-RD), and Fourier transform infrared spectroscopy (FT-IR). Stable complexes of ND/SBE-β-CD and ND/HP-β-CD were formed in distilled water in a 1:1 stoichiometric inclusion complex as indicated by an AL-type diagram. The apparent stability constants (Ks) were 1334.4 and 464.1 M−1 for ND/SBE-β-CD and ND/HP-β-CD, respectively. The water-solubility of ND was significantly increased in an average of 22- and 8-fold for SBE-β-CD and HP-β-CD, respectively. DSC results showed the formation of true inclusion complexes between the drug and both SBE-β-CD and HP-β-CD prepared by the kneading method. In contrast, crystalline drug was detectable in all other products. The dissolution studies showed that all the products exhibited higher dissolution rate than those of the physical mixtures and ND alone, in all mediums. However, the kneading complexes displayed the maximum dissolution rate in comparison with drug and other complexes, confirming the influence of the preparation method on the physicochemical properties of the products.

Pharmacokinetics and Bioequivalence Study of Nifedipine Controlled-release Tablets after Multiple Doses Administration in Healthy Volunteers

Abstract:OBJECTIVE Povidone/Copovidone were used as the main osmopolymers instead of Poly (ethylene oxide) (PEO) to prepare nifedipine controlled-release tablets of new osmotic pump type. The pharmacokinetic properties of nifedipine controlled-release tablets after multiple doses in healthy volunteers were studied, and the relative bioavailability of the test preparation was calculated and the bioequivalence was evaluated. METHODS Twenty four male healthy volunteers were enrolled in a randomized two-way crossover design. 30 mg nifedipine was administered per day during consecutive 7 d. The drug concentration in blood was determined with LC-MS/MS method. The pharmacokinetic parameters, relative bioavailability and the bioequivalence were calculated by DAS program. RESULTS The main pharmacokinetic parameters after multiple doses of test preparation and reference preparation were as follows:T1/2β (9.0±2.2)h and (9.5±3.8)h; Tmax (8.1±7.4)h and (6.7±4.1)h; Cssmax (52.7±28.2)μg·L-1 and (44.5±22.6)μg·L-1; Cssmin (29.4±22.2)μg·L-1 and (28.7±15.8)μg·L-1; AUC0-60 (1 087.4±671.6)μg·L-1·h and (1 040.2±518.4)μg·L-1·h; AUC0-∞ (1 113.1±677.2)μg·L-1·h and (1 074.5±519.8)μg·L-1·h , AUCSS (809.1±454.0)μg·L-1·h and (713.9±382.2)μg·L-1·h. The average relative bioavailability of the test preparation versus the reference preparation was as follows: FAUCss (115.2±29.2)%, FAUC0-60 (103.4±30.2)% and FAUC0-∞ (102.2±29.7)%. DF was (77.84±52.09)% and (55.48±30.54)%. The 90% confidence interval of Cmax, AUC0-60, AUC0-∞ and AUCss after longarithmic transform was (100.9-126.0)%, (87.5-111.8)%, (86.8-110.3)% and (98.8-124.4)% respectively. There was no significant difference in Tmax after non-parameter rank sum test. No adverse reaction was observed during the trial. CONCLUSION The test preparation was bioequivalent to the reference preparation. Povidone/Copovidone could replace PEO used as the main functional excipient of osmotic pump type controlled-release preparations.

Abstract:The development of a new HPLC-UV diode array procedure applied to follow the hydrolytic degradation of two well-known 4-nitrophenyl-1,4-dihydropyridine derivatives, nitrendipine and nisoldipine is reported. Hydrolysis of each drug were carried out in ethanol/Britton-Robinson buffer at different pHs, stored into amber vials at controlled temperatures of 40, 60 and 80 degrees C and periodically sampled and assayed by HPLC. Nitrendipine degradation in different parenteral solutions was also evaluated. The HPLC procedure exhibited an adequate selectivity, repeatability (<1%) and reproducibility (<2%). The recoveries were higher than 98% with CV of 1.13 and 1.54% for nitrendipine and nisoldipine, respectively. A significant degradation was observed at alkaline pH (>pH 8) with a first order kinetic for both drugs. At pH 12, 80 degrees C, k values of 3.56x10(-2) x h(-1) and 2.22x10(-2) for nitrendipine and nisoldipine, respectively were obtained. Also, activation energies of 16.8 and 14.7 kcal x mol(-1) for nitrendipine and nisoldipine, respectively, were calculated. Furthermore, from the results obtained from hydrolytic degradation in different solutions for parenteral use, we can affirm that solutions significantly increased the degradation of nitrendipine. In conclusion, the HPLC proposed procedure exhibited adequate analytical requirements to be applied to the hydrolytic degradation studies of nitrendipine and nisoldipine. Furthermore, all tested parenteral solutions significantly increased the hydrolytic degradation of nitrendipine, the composition of solution being a relevant factor.
