Abstract:Phytic acid, a naturally occurring compound predominantly found in cereals and legumes, is the focus of this review. This review investigates its distribution across various food sources, elucidating its dual roles in foods. It also provides new insights into the change in phytic acid level during food storage and the evolving trends in phytic acid management. Although phytic acid can function as a potent color stabilizer, flavor enhancer, and preservative, its antinutritional effects in foods restrict its applications. In terms of management strategies, numerous treatments for degrading phytic acid have been reported, each with varying degradation efficacies and distinct mechanisms of action. These treatments encompass traditional methods, biological approaches, and emerging technologies. Traditional processing techniques such as soaking, milling, dehulling, heating, and germination appear to effectively reduce phytic acid levels in processed foods. Additionally, fermentation and phytase hydrolysis demonstrated significant potential for managing phytic acid in food processing. In the future, genetic modification, due to its high efficiency and minimal environmental impact, should be prioritized to downregulate the biosynthesis of phytic acid. The review also delves into the biosynthesis and metabolism of phytic acid and elaborates on the mitigation mechanism of phytic acid using biotechnology. The challenges in the application of phytic acid in the food industry were also discussed. This study contributes to a better understanding of the roles phytic acid plays in food and the sustainability and safety of the food industry.
Effects of Different Nitrogen and Water Treatments on Phytic Acid Contents and Protein Components in Rice Grain

Abstract:Two rice genotypes(Xiushui 110 and Chunjiang 15) were used to analyze the influence of nitrogen treatments,water management methods and their interaction on grain phytic acid content and protein components by a field experiments of two water treatments,including traditional irrigated cultivation(C1) and dried stress cultivation(C2) with 3 level nitrogen fertilizers(N1,0 kg/ha;N2,100 kg/ha;N3,200 kg/ha) for each water treatment.The results showed that C2 treatment(dried stress cultivation) had relatively higher phytic acid content in rice grain,while the effect of nitrogen treatments on grain phytic acid were water management-dependent,with slight high phytic acid content for 100 kg/ha nitrogen application(N2) and no nitrogen application(N1) relative to high nitrogen treatment(N3) under the condition of traditional irrigated cultivation(C1),but the opposite was true for the effect of N treatment to grain phytic acid when C2 treatment was exposed.The varying contents of grain phytic acid for different water and level nitrogen fertilizers were poorly correlated with grain total protein,4 protein components and rice yielding traits(available panicle per areas,grain number per panicle,1000-grain weight and seed setting rate),although the decreasing yield levels and low grain quality generally occurred at the extremely high application of N fertilizer;The varying extent of phytic acid,total protein contents and 4 protein components in different grains within a panicle was also related to water management and N fertilizer application,with relative high phytic acid for the bottom location grains compared to those located at upper and middle location within a panicle,implying that appropriate water management and N fertilizer application were generally benefit for the reduce of grain phytic acid content except for achieving a high yield level in rice production.

Abstract:This study was carried out to determine the effects of phytase on the growth performance, bone mineralization, digestion and metabolization of nutrientsubstance in Avian broilers. Each of 400, 1 day old male Avian broilers and 400, 1 day old female Avian broilers was randomly divided into 5 groups (Group I, available phosphorus-adequate group without phytase; Group II, available phosphorus-deficient group without phytase; Group III, available phosphorus-deficient group with 250 U kg(-1) phytase; Group IV, available phosphorus-deficient group with 500 U kg(-1) phytase; Group V, available phosphorus-deficient group with 750 U kg(-1) phytase). Each group included 4 replicates, each with 20 broilers. The results showed that the effect of phytase on the growth permance in male broilers was more obvious than that of female. Dry tibia weights of male and female broilers fed with Group II and III diets were decreased compared with Group I; Group IV and V were increased compared with Group I. Tibia phosphorus contents of male and female broilers fed with Group IT-V were decreased compared with Group I. Tibia calcium contents of male broilers fed with Group II and V diets were increased by compared with Group I; Group III and IV were decreased compared with Group I. Tibia calcium contents of female broilers fed with Group II, IV and V diets were increased compared with Group I; Group III was decreased compared with Group I. Apparent digestibility coefficients of total phosphorus of Group II-V in male and female broilers were increased compared with Group I. Phosphorus concentrations of faeces of Group II-V in male and female broilers were decreased compared with Group I. Apparent digestibility coefficients of calcium Group II-V in male and female broilers were increased compared with Group I. The result showed that phytase added to available phosphorus-deficient broiler diet could improve the growth performance, bone mineralization as well as calcium and phosphorus availability.
Effect of Agronomic Practices on Grain Yield and Phytic Acid Content of Early Rice Variety Jiayue 253 Under Direct Sown Cultivation

Abstract:Effects of the different sowing date,nitrogen level,sowing rate on grain yield,yield components and phytic acid content were studied in the field experiment with early rice variety Jiayu253.The yield,yield component factors,and the content of phytic acid in paddy rice,brown rice,and milled rice were measured.The main results were shown as follows: in term of single factor,the yield of rice reached the highest when the sowing date was on April 15,and the yield treated with 13kg / 667m2 N or 6kg / 667m2 sowing rate was significantly higher than those of other treatments,respectively;considering all factors,the yield of rice reached the highest under the treatment of sowing date on April 8,application of 13kg / 667m2 N,and 6kg / 667m2 seeds;phytic acid contents in brown rice were quite different among different sowing dates or nitrogen levels,showing a downward tendency with delaying sowing date or increasing nitrogen.However,there was no significant difference among sowing rates;on the contrary,the phytic acid content in milled rice was obviously insensitive to the sowing date,the nitrogen level,and the sowing rate.Thus,the results indicated that agronomic measures had little effects on the phytic acid content in milled rice,and wouldn’t affect the mineral availability.

Abstract:In cereals and legumes, phytic acid (PA) is the principal phosphorus reserve that serves the biosynthetic and nutritional demands of growing tissues during germination. Because of strong antioxidant activity and ability to chelate mineral ions, it provides several health and physiological benefits. Phytic acid is widely regarded as an anti-nutritional factor as it can bind minerals and proteins, thus decreasing their bioavailability. However, its chelating property is likely to provide several health benefits, including lowering the risk of diabetes and some malignancies, improving heart health and checking kidney stone formation. Furthermore, recent research has revealed that PA is effective against foodborne bacteria. These beneficial properties of PA open up the possibility of value-added applications in several new areas. Due to its various proven properties, PA has drawn more attention as an anti-nutrient than a beneficial compound. Nevertheless, the available food processing techniques for raw material preparation can be used to reduce the PA concentration in foods to mitigate its anti-nutritional effects. The resulting low PA products may find newer applications in the food industry. This paper aims to provide a general overview of phosphorus transport to different plant organs, biosynthesis of PA and its beneficial and anti-nutritional effects and a description of dephytinization methods.

Abstract:In the nutrition of monogastric animals phytate-P represents a poorly available source of phosphorus, especially in the case of diets low in phytase activity. Similarly the bioavailability of different minerals and trace elements is considerably reduced by phytate complexes. High concentrations of Ca increase the anti-nutritive effect of phytic acid on mineral and trace element bioavailability and thus impede the action of phytase. This effect can in part be compensated by an increased supply of vitamin D. There is also evidence for protective functions of phytic acid such as the prevention of the formation of free radicals, the delaying of post prandial glucose absorption, the decrease in plasma cholesterol and triglycerides as well as a change in the carry over of heavy metals. The basic mechanisms by which phytic acid may exert these effects are still not clear. In several studies reported in the literature, evidence for the nutritional significance and ecological importance of microbial phytase for pigs and poultry has been given. As the monogastric organism contains no or only negligible amounts of endogenous phytase in the stomach and small intestine, it is therefore dependent on plant or microbial phytase. Plant phytase, e.g. from rye, triticale, wheat or, in smaller amounts from barley, and supplemented Aspergillus-phytase display cumulative effects.

Abstract:Phytic acid is considered as one of the most abundant storage forms of phosphorous in the plant seed tissues. It accumulates throughout seed development until maturity and accounts for 60–70% of the total phosphorus in legume seed. Its amount ranges from 0.40 to 2.06% in legumes. It is a strong cationic chelator and negatively impacts the bioavailability of essential micronutrients like iron (Fe) and Zinc (Zn). It has very limited digestibility in humans and other non-ruminants which lack phytases. Besides, it also acts as an inhibitor of salivary and gastric enzymes during digestion. On the contrary, the beneficial effects of phytic acid include its role as an antioxidant, anti-carcinogen, and regulator of blood glucose and cholesterol levels. Several biotechnological strategies have been employed to reduce the phytic acid content in plants especially legumes for better absorption of essential micronutrients and in turn for bio-fortification to address the malnutrition challenges across the world. This review focuses primarily on the mechanisms governing phytic acid biosynthesis and its regulation/ reduction using various biotechnological, mutation breeding and computational approaches. This will help us in understanding the molecularity of the phytic acid development and will also contribute towards developing newer and more efficient strategies aimed at regulating phytic acid content in legumes.
Response of Phytic Acid to Nitrogen Application and Its Relation to Protein Content in Rice Grain

Abstract:Nitrogen (N) fertilization has been recognized as improving the grain beneficial micronutrients, including Zn and Fe, in rice. However, only a few studies have explicitly focused on N-induced variation in anti-nutritional components such as phytic acid (PA), PA synthesis-related gene expression, and variation in grain protein fractions. Therefore, in this study, two culture systems (hydroponic and detached panicle culture systems) were used to elucidate the influence of N application on PA concentration and its relation to the grain protein fractions, such as albumin, globulin, prolamin, and glutelin, and total protein in rice. Results showed that N application generally decreased the grain PA concentration in brown rice and down-regulated the PA synthesis-related genes in the lipid-independent pathway. In contrast, total grain protein and its fractions concentrations increased significantly. For grain positional distribution, PA and protein concentration were generally higher in the aleurone fraction than in the milled rice, regardless of N application. However, higher N application decreased the PA in both aleurone fraction and milled rice, while increased the grain protein fractions mainly in milled rice. These findings imply that N application could substantially improve the rice nutrition by reducing the PA while increasing the protein concentration. Hence, these findings may provide critical bases for rice nutritional improvement through optimal N management.

Abstract:Phytic acid (PA), extracted from oilseeds, legumes, cereals, nuts, and pollen by acid solutions under heating and/or stirring and then purified, has shown beneficial health and physiological effects due to its pronounced antioxidant activity and ability to chelate Fe3+ ions. Publications on PA have increased, especially the ones reporting its effect on disease prevention and treatment. Moreover, recent studies have suggested the PA efficacy as a foodborne pathogens inhibitor. Abstract:Background: Phytic acid is abundant in plant-based diets and acts as a micronutrient inhibitor for humans and non-ruminant animals. Phytases are enzymes that break down phytic acid, releasing micronutrients and enhancing their bioavailability, particularly iron and zinc. Deficiencies in iron and zinc are significant public health problems, especially among populations with disease-associated malnutrition or those in developing countries consuming phytic acid-rich diets. This narrative review aimed to summarize findings from human intervention studies on the interactions between phytic acid, phytase, and micronutrient bioavailability. Methods: An extensive PubMed search (1 January 1990 to 8 February 2024) was conducted using MeSH terms (phytic acid, phytase, IP6, "inositol hexaphosphate," micronutrient, magnesium, calcium, iron, zinc). Eligible studies included human intervention trials investigating the bioavailability of micronutrients following (a) phytase supplementation, (b) consumption of phytic acid-rich foods, or (c) consumption of dephytinized foods. In vitro, animal, cross-sectional, and non-English studies were excluded. Results: 3055 articles were identified. After the title and full-text review, 40 articles were eligible. Another 2 were identified after cross-checking reference lists from included papers, resulting in 42 included articles. Most studies exploring the efficacy of exogenous phytase (9 of 11, 82%) or the efficacy of food dephytinization (11 of 14, 79%) demonstrated augmented iron and zinc bioavailability. Most phytic acid-rich food-feeding studies (13 of 17, 77%) showed compromised iron and zinc bioavailability. Conclusions: Strong evidence supports decreased iron and zinc bioavailability in phytic acid-rich diets and significant improvements with phytase interventions. Studies of longer periods and within larger populations are needed.

Abstract:The field experiment with different water treatments and management methods,including traditional flooded cultivation(C1),film mulched cultivation(C2) and uncovered cultivation(C3),was conducted to investigate the effects of cultivation model and nitrogen application on grain nutritional quality and phytic acid content for japonica rice,with 3 levels of nitrogen fertilizer(N1,0 kg/hm2;N2,124 kg/hm2;N3,150 kg/hm2) being designed for each water management treatment.The results showed that C3 treatment(uncovered cultivation) had relatively higher total phosphorus content in rice grain,but much more decreasing phytic acid and inorganic phosphorus contents compared with traditional flooded cultivation(C1).For the same nitrogen level,total grain nitrogen and protein components of C3 treatment were lower than those of C2 and C3 treatments,implying that a reasonable fertilizer and water management in paddy production could improve grain nutritional quality,especially increase grain protein contents and decrease grain phytic acids.Additionally,the increasing application of nitrogen fertilizer should cause the remarkably enhancing accumulation of total phosphorus in rice grains.

Abstract:Thirty-one japonica cultivars with contrasting agronomic traits were used and 5 nitrogen (N) fertilizer treatments of level and timing were performed to determine the effects of IN application on grain phytic acid and proteins including albumin, globulin, prolamin, and glutelin. Variance analysis showed a large effect of N and a smaller effect of cultivar on grain phytic acid and proteins. With increased N level, grain phytic acid concentrations progressively reduced whereas concentrations of the 4 proteins and ratios of glutelin to total protein increased, indicating that N level has a beneficial effect on rice nutritional quality. In addition, substantial genotypic differences in response of grain phytic acid and proteins to N treatments were detected. Some cultivars such as Xiushui09, Zhendao10, and Yanjing5 exhibited more stability of grain proteins under contrasting N treatments, and the significance of this stability is discussed in regard to its use in rice quality improvement. Abstract:Development of rice cultivars with low phytic acid (lpa) is considered as a primary strategy for biofortification of zinc (Zn) and iron (Fe). Here, two rice genotypes (XS110 and its lpa mutant) were used to investigate the effect of P supplies on accumulations and distributions of PA, Zn, and Fe in rice grains by using hydroponics and detached panicle culture system. Results showed that higher P level increased grain PA concentration on dry matter basis (g/kg), but it markedly decreased PA accumulation on per grain basis (mg/grain). Meanwhile, more P supply reduced the amounts and bioavailabilities of Zn and Fe both in milled grains and in brown grains. Comparatively, lpa mutant was more susceptive to exogenous P supply than its wild type. Hence, the appropriate P fertilizer application should be highlighted in order to increase grain microelement (Zn and Fe) contents and improve nutritional quality in rice grains.
ANALYSIS OF NUTRITIONAL QUALITY OF LOW PHYTIC ACID MUTANTS IN MAIZE

Abstract:Major nutritional quality and components of eight low phytic acid(lpa) mutants and their corresponding wild types were studied.Compared to their corresponding wild types,the content of inorganic P(Pi) was all increased several times,while the content of total P(TP) in mutants was almost the same as their wild types.The contents of crude lipid and amylose were similar,but total starch was significantly different.Crude protein in some of mutants was increased significantly.Most of amino acids were increased,and essential amino acidLysine was increased except mutants derived from Q319 and X178.Mineral macronutrients and micronutrients were similar.All results showed that the lpa mutation in maize could enhance the nutritional quality and bioactivities.
The improved effect and its mechanism of phytic acid on DSS-induced UC mice

Abstract:Ulcerative colitis (UC) is an inflammatory bowel disease (IBD) with an unknown etiology that is currently difficult to treat effectively. Phytic acid, a natural compound found in high-fiber foods, such as grains, has been shown to have many pharmacological functions, such as anti-inflammatory and blood-milk barrier maintenance. Our study investigated the effects of phytic acid on UC as well as the underlying mechanisms. We found that phytic acid ameliorated weight loss, shortened colon length, increased clinical scores, and decreased the release of pro-inflammatory factors (Il-1β, Il-6, and TNF-α) in mice with DSS-induced UC. Phytic acid was also found to protect intestinal barrier integrity in mice with UC by maintaining the expression of tight junction proteins (occludin, claudin-3 and ZO-1) and mucin-2. In LPS-stimulated Caco-2 cells, phytic acid significantly inhibited the upregulation of pro-inflammatory factors and the downregulation of tight junction proteins. Further experimental results confirmed that phytic acid inhibits the activation of the AKT/NF-κB signaling pathway in both mice with UC and Caco-2 cells. To sum up, this study proved that phytic acid has a beneficial ameliorative effect on mice with DSS-induced UC, suggesting that it may be used to develop functional foods for the treatment or prevention of UC.
Effect Of Irrigation Regimes On Concentrations Of Phytic Acid And Minerals In Hulled And Naked Oats

Abstract:The effects of irrigation regimes, cultivar and irrigation x cultivar interaction on the concentrations of phytic acid, total phosphorus, crude protein, Ca, Mg, K, Zn, Fe, Cu and Mn in oat grains were determined. The relationships between the variables under study and irrigation quota were estimated with linear and quadratic regression models in hulled (Avena sativa L.) and naked (Avena nuda L.) oats. Significant differences were obtained among irrigation treatments, between the two oat varieties and irrigation X cultivar interaction for most of the variables under study. Changes in grain total phosphorus and copper contents with the increase of irrigation times and quota were linear while those in phytic acid, Ca, Mg, K, Zn, Mn were quadratic in naked oat. Changes in hulled oat grain (husked) Zn and Fe contents with the amount of irrigation were linear while those in phytic acid, total phosphorus, crude protein, Ca and Mg contents were quadratic. Phytic acid content of hulled oat was highly correlated with total phosphorus, crude protein, Ca, Mg and Zn (p < 0.01). In contrast, no significant correlations were observed in naked oat. Grain yield was positively and significantly correlated with phytic acid, total phosphorus and protein, but negatively correlated with Ca, Mg and Zn in hulled oat. The grain yield of naked oat was positively associated with phytic acid, Ca, Mg, K, Zn, Fe, and Mn contents.
The Response of Phytic Acid and Its Expression Profiles in Rice (oryza Sativa L.)Grain As Induced by Phosphorus Supply

Abstract:Phytic acid (PA)is the main storage form of phosphorus (P)in cereal grains.However,the relationship between external P supply and grain PA accumulation has not yet been fully understood.Two japonica rice cultivars were used to investigate the effects of phosphorus (P)treatments on grain phytic acid (PA)and its relation to inositol phosphate-related genes expression profiles using a culture of detached panicle of rice (Oryza sativa L.).Results showed that high P level caused a significant increase in grain PA,inorganic P (Pi)and total P contents.However,grain weight and PA content per grain were P concentration-dependent,and decreased under high P treatment (12P).High P supply decreased grain Zn and Fe concentrations,and their bio-availabilities.Moreover,P treatment also influenced all four PA related genes expression (RI NO1 ,ITP5/6 K-6 ,I PK2 and I PK1 ),especially for RI NO1 ,a rate-limiting gene in PA synthesis and highly expressed in rice grains.Specifically,RION1 gene expression was up-regulated at optimum P (3P ) concentration, but was significantly down-regulated under high P treatment (12P ), which corresponded with the variation of grain PA content in rice,suggesting RION1 was a key metabolic gene in PA synthesis with P supply.In contrast,only a slight contribution of I PK2 gene was exhibited in PA synthesis induced by P supply.
