Relationship Between Protein Kinase C and Α1-Adrenoceptor Mediated Ca2+ Influx in Vascular Smooth Muscle Cells

Abstract:Phorbol dibutyrate(PDB),a proteinkinase C stimulator induced an increasc in [Ca2+]i in a con-centration-dependent manner with a maximum concentra-tion of 0.3 μmol·L-1. The effect of PDB was completelyblocked by 20 μmol·L-1 H7 [1-(5-isoquinolinesulfonyl)-2-methylpiperazine,a PKC inhibitor],and not inhibitedby prazosine or propranolol. PDB effect was signiflcantly re-duced by 1.25 μmol·L-1 verapamil and completely blockedby 2.5 μmol·L-1 verapamiL Phenylephrine(Phe)significantly increased [Ca2+]i and this cffcct was partiallyblocked by 10-20 μmol·L-1 H7.Phe increased the [Ca2+]i。due to intracellular Ca2+ release,and subsequent additionof CaCl2 into the medium (final concentration 1. 25mmol·L-1) furthcr increased the [Ca2+]idue toextrace11ular Ca2+ entry. H7 inhibited these effects of pheby 33 ± 3% and 58 ± 6% respectively KCl 20-100 mmol·L-1 increased [Ca2+]i concentration-dependently.H7 re-duced the effect of KCl.These results suggested that the ef-kct of PDE was due to PKC activation.but not due totransmitter release or activation of α-and β-adrenoceptors. PKC was involved in the [Ca2+]i increase induced by theα1-adrcnoceptor activation,and the effect of PKC on the[Ca2+]i increase was related to the intracellular Ca2+ re-lease and the extraceIlular Ca2+entry depending onvoltage-dependent Ca2+ channel.
