Abstract:Objective: Epsins are ubiquitin-binding endocytic adaptor proteins and regulate membrane receptor signaling. Methods: Tamoxifen-inducible Cre-driven global or constitutive Ve-cadherin Cre-driven endothelial-cell specific epsin 1 and 2 double knockout mice, iDKO or EC-DKO, respectively in ApoE-/- background were established and fed western diet (WD) for ten weeks. Aortic lesions were analyzed by histology and oil red O staining. Molecular mechanism was determined by molecular, cellular, biochemical and genetic approaches. Abstract:Epsins are endocytotic proteins for the internalization of plasma membrane receptors. Our previous studies have suggested that epsins play an important role in tumorigenesis and lymphatic development by modulating VEGFR2, VEGFR3 or Wnt signaling. Whether epsins affect atherosclerosis remains unknown. In an ApoE-deficient murine model, endothelial-specific double knockout mice (EC-iDKO/ApoE-/-) significantly attenuated atherosclerotic lesion and macrophage infiltration in aortic root and aortic arch compared to ApoE-/- mice fed Western diet for ten weeks. Endothelium-dependent relaxation is significantly improved in epsin-deficient mice. Mechanistically, the expression of epsins is upregulated in atheroma and mouse endothelial cells in a pro-inflammatory condition, mirrored in human atherosclerotic specimen. Proteomics study using Mass spectrometry revealed that epsins bind IP3R1 and facilitate its degradation in endothelium under inflammatory condition. Downregulation of IP3R1 by athero-prone substances augmented ER stress and NFkB signaling, reversible by ER stress inhibitor 4-PBA. In line with the findings from mouse studies, IP3R1 is dramatically downregulated in human atherosclerotic endothelium. Loss of epsins stabilized IP3R1, and attenuated ER stress-dependent NFkB activation and inflammatory signaling as evidenced by the reduced expression of P-selectin, adhesion molecules, and chemoattractant MCP-1 in primary mouse aortic endothelial cells (MAEC), and reflected by in vitro rolling analysis between isolated MAEC and macrophage from mouse. Molecular modeling and mapping analysis revealed that epsin UIM is critical to facilitate the degradation of IP3R1. In conclusion, we have identified a novel molecular mechanism regarding epsins in the regulation of atherosclerosis, and our data suggest that epsins are a potential target to treat atherosclerosis.

Abstract:Background: Epsins are a family of ubiquitin-binding, endocytic clathrin adaptors. Mice lacking both epsins 1 and 2 (Epn1/2) die at embryonic day 10 and exhibit an abnormal vascular phenotype. We have recently published that endothelial epsins function as critical regulators of tumor angiogenesis by controlling the endocytosis of VEGFR2 (J Clin Invest. 2012). The goal of this project is to define the novel role of epsins in endothelial cells (EC) and macrophages in regulating inflammation-mediated atherosclerosis. Methods and Results: To examine the role of endothelial and macrophage epsins in atherogenesis, we engineered mice with specific deletion of epsins in EC (EC-DKO) or myeloid cells (MΦ-DKO). Strikingly, these mice are viable but display markedly attenuated atherogenesis. Mice with either EC-DKO or MΦ-DKO on ApoE-/- background had a significant reduction in atherosclerotic lesion formation and macrophage infiltration compared to ApoE-/- mice fed a western diet. FACS analysis revealed that epsin deficiency greatly reduced TNFα and LPS-induced expression of adhesion molecules including ICAM-1, VCAM-1, P- and E-selectins, and CCR2 and MCP-1 in primary mouse aortic EC, and leukocyte recruitment in mouse aorta. Mechanistically, epsins promote TNFR/TLRs signaling and NF-κB and MAPK (JNK and p38) activation by enhancing recruitment of NEMO, an essential NF-kB activator. Conversely, in macrophages, epsin deficiency produces no change in LDL and scavenger receptor expression but impaired pinocytosis of Lucifer Yellow, indicative of a major defect in LDL lipid uptake. We also observed that a synthetic peptide comprising the epsin ubiquitin-interacting motif (UIM) and lesion homing sequence potently disrupts association of epsins with TNFR/TLR signaling complex in vitro, and inhibits atherosclerotic plaque in vivo. Our data show that epsins control atherosclerosis by promoting TNFR/TLR signaling in endothelial cells, facilitating LDL lipid uptake in macrophages, and provide a potential novel strategy to perturb atheroma progression. Conclusion: Our results demonstrate a novel proatherogenic role of epsins in atherosclerosis by potentiating endothelium inflammation, augmenting leukocyte recruitment, and promoting foam cell formation.

Abstract:Rationale: Low density cholesterol receptor (LDLR) in the liver is critical for the clearance of low-density lipoprotein cholesterol (LDL-C) in the blood. In atherogenic conditions, proprotein convertase subtilisin/kexin 9 (PCSK9) binds to LDLR on the surface of hepatocytes, preventing its recycling and enhancing its degradation in lysosomes, resulting in reduced LDL-C clearance. Our recent studies demonstrate that epsins, a family of ubiquitin-binding endocytic adaptors, are critical regulators of atherogenicity. Given the fundamental contribution of circulating LDL-C to atherosclerosis, we hypothesize that liver epsins promote atherosclerosis by controlling PCSK9-triggered LDLR endocytosis and degradation. Objective: We will determine the role of liver epsins in promoting PCSK9-mediated LDLR degradation and hindering LDL-C clearance to propel atherosclerosis. Methods and Results: We generated double knockout mice using albumin Cre in which both genes of epsin, namely, epsin 1 and epsin 2, are specifically deleted in the liver (Liver-DKO) on an ApoE -/- background. We discovered that western diet (WD)-induced atherogenesis was greatly inhibited, along with diminished blood cholesterol and triglyceride levels. Mechanistically, using scRNA-seq analysis on hepatocyte-derived data revealed elevated pathway involved in LDL particle clearance under WD treatment in ApoE -/- /Liver-DKO, which was coupled with diminished plasma LDL-C levels. Further analysis using the MEBOCOST algorithm revealed enhanced communication score between LDLR and cholesterol, suggesting elevated LDL-C clearance in the ApoE -/- Liver-DKO mice. In addition, we showed that loss of epsins in the liver upregulates of LDLR protein level. We further showed that epsins bind LDLR via the ubiquitin-interacting motif (UIM), and PCSK9-triggered LDLR degradation was abolished by depletion of epsins, preventing atheroma progression. Finally, our therapeutic strategy, which involved targeting liver epsins with nanoparticle-encapsulated siRNAs, was highly efficacious at inhibiting dyslipidemia and impeding atherosclerosis. Conclusions: Liver epsins promote atherogenesis by mediating PCSK9-triggered degradation of LDLR, thus raising the circulating LDL-C levels. Targeting epsins in the liver may serve as a novel therapeutic strategy to treat atherosclerosis by suppression of PCSK9-mediated LDLR degradation.

Abstract:The epsin family of endocytic adapter proteins are widely expressed, and interact with both proteins and lipids to regulate a variety of cell functions. However, the role of epsins in atherosclerosis is poorly understood. Here, we show that deletion of endothelial epsin proteins reduces inflammation and attenuates atherosclerosis using both cell culture and mouse models of this disease. In atherogenic cholesterol-treated murine aortic endothelial cells, epsins interact with the ubiquitinated endoplasmic reticulum protein inositol 1,4,5-trisphosphate receptor type 1 (IP3R1), which triggers proteasomal degradation of this calcium release channel. Epsins potentiate its degradation via this interaction. Genetic reduction of endothelial IP3R1 accelerates atherosclerosis, whereas deletion of endothelial epsins stabilizes IP3R1 and mitigates inflammation. Reduction of IP3R1 in epsin-deficient mice restores atherosclerotic progression. Taken together, epsin-mediated degradation of IP3R1 represents a previously undiscovered biological role for epsin proteins and may provide new therapeutic targets for the treatment of atherosclerosis and other diseases.
Single-Cell Analysis Reveals Critical Role of Macrophage Epsin in Regulating Origin of Foam Cell in Atherosclerosis

Abstract:Atherosclerosis is a chronic inflammatory condition characterized by the excessive accumulation of fat and lipid molecules, leading to the formation of foam cells and plaques in arterial walls. Dysfunction of vascular smooth muscle cells (VSMCs), fibroblast, endothelial cells, and macrophages is often associated with this pathology. We found that epsins accelerate atherosclerosis progression in individuals on a Western diet (WD). Using ApoE-deficient (ApoE ) and macrophage-specific epsin deletion in ApoE-/- backgrounds (LysM-DKO/ApoE ) mice fed a WD for 16 weeks, we observed significantly reduced foam cell formation in LysM-DKO/ApoE mice compared to ApoE mice. Single-cell RNA sequencing identified 20 major cell types, including seven VSMC and five macrophage subtypes. Among the VSMC subtypes, modulating VSMC1 was involved in inflammation and migration, while modulating VSMC2 was associated with VSMC phenotype switching. In atherosclerotic mice, populations of modulating VSMC1, VSMC2, foamy-Trem2, and inflammatory macrophages increased, but significantly decreased in epsin-deficient mice. Modulating VSMC2 transition into macrophages occurred with a probability of 0.57 in ApoE mice, compared to 0.01 in LysM-DKO/ApoE mice. Epsin deletion also reversed endothelial dysfunction and downregulated cholesterol and glucose-mediated signals, as well as inflammatory ligands Il1b and C1qa. Our findings suggest that epsin deletion reduces foam cell formation and rewires VSMC and endothelial functions, offering a novel therapeutic strategy for atherosclerosis.

Abstract:Epsin endocytic adaptor proteins are implicated in the progression of atherosclerosis; however, the underlying molecular mechanisms have not yet been fully defined. In this study, we determined how epsins enhance endothelial-to-mesenchymal transition (EndoMT) in atherosclerosis and assessed the efficacy of a therapeutic peptide in a preclinical model of this disease. Using single-cell RNA sequencing combined with molecular, cellular, and biochemical analyses, we investigated the role of epsins in stimulating EndoMT using knockout in Apoe −/− and lineage tracing/proprotein convertase subtilisin/kexin type 9 serine protease mutant viral-induced atherosclerotic mouse models. The therapeutic efficacy of a synthetic peptide targeting atherosclerotic plaques was then assessed in Apoe −/− mice. Single-cell RNA sequencing and lineage tracing revealed that epsins 1 and 2 promote EndoMT and that the loss of endothelial epsins inhibits EndoMT marker expression and transforming growth factor-β signaling in vitro and in atherosclerotic mice, which is associated with smaller lesions in the Apoe −/− mouse model. Mechanistically, the loss of endothelial cell epsins results in increased fibroblast growth factor receptor-1 expression, which inhibits transforming growth factor-β signaling and EndoMT. Epsins directly bind ubiquitinated fibroblast growth factor receptor-1 through their ubiquitin-interacting motif, which results in endocytosis and degradation of this receptor complex. Consequently, administration of a synthetic ubiquitin-interacting motif–containing peptide atheroma ubiquitin-interacting motif peptide inhibitor significantly attenuates EndoMT and progression of atherosclerosis. We conclude that epsins potentiate EndoMT during atherogenesis by increasing transforming growth factor-β signaling through fibroblast growth factor receptor-1 internalization and degradation. Inhibition of EndoMT by reducing epsin–fibroblast growth factor receptor-1 interaction with a therapeutic peptide may represent a novel treatment strategy for atherosclerosis.

Subsequently, gene ontology analysis of hepatocyte-derived data revealed elevated pathways involved in LDL particle clearance and very-low-density lipoprotein (VLDL) particle clearance under WD treatment in ApoE-/- /Liver-DKO, which was coupled with diminished plasma LDL-C levels. Abstract:Introduction: Endothelial dysfunction and arterial inflammation are critically important for cardiovascular diseases such as atherosclerosis. Of major importance, they are among the predominant culprits that promote the transition from stable to vulnerable atheromas. Our pilot studies revealed Disabled Homolog 2 (Dab2) deficiency in endothelial cells (EC) results in endothelial dysfunction. However, the molecular mechanisms that direct Dab2 to combat endothelial dysfunction during atherosclerosis are completely unknown. Hypothesis: Dab2 prevents the progress of atherosclerosis by maintaining EC function and reducing inflammatory factors. Methods and Results: We examined the expression of endothelial Dab2 in human atherosclerosis patients and ApoE -/- mice fed a western diet (WD) for 12 weeks by immunoflurecent co-staining of VE-cadherin and Dab2 and observed drastically reduced Dab2 expression in the endothelium and intima of human fatty streaks and mouse atherosclerotic aortae. Genetic deletion of Dab2 in the endothelium of ApoE -/- mice on a WD for 12 weeks significantly increased atherosclerosis as revealed by Oil Red O staining of aortic roots and arches, suggesting that Dab2 could be a atheroprotective flow-responsive gene. Similar results were found in WT and EC-iDab2KO mice injected with AAv8-PCSK9 virus. RNA-seq of primary mouse aortic ECs (MAECs) isolated from WT and EC-Dab2iKO mice revealed Dab2 deficiency upregulated many inflammatory genes. qRT-PCR confirmed TNF-α and IL-6 were significantly upregulated in Dab2KO MAECs. Additionally, we observed an increase in Dab2 binding to NEMO in the absence of endocytic adapter proteins epsins via co-IP with anti-Dab2 in WT and epsin-deficient DKO MAECs. We also found three Klf4 binding sites present in the Dab2 gene promoter/enhancer and that overexpression of Klf4 upregulated Dab2 expression in a dosage-dependent manner. Conclusions: Our findings indicated the essential role of Dab2 in protecting the atherogenic endothelium by reducing macrophage accumulation and increasing plaque stability in the aortic roots, which may provide new anti-atherosclerotic therapeutics.

Abstract:Epsins are a family of ubiquitin-binding, endocytic clathrin adaptors. Mice lacking both epsins 1 and 2 (Epn1/2) die at embryonic day 10 and exhibit an abnormal vascular phenotype. To examine the angiogenic role of endothelial epsins, we generated mice with constitutive or inducible deletion of Epn1/2 in vascular endothelium. These mice exhibited no abnormal phenotypes under normal conditions, suggesting that lack of endothelial epsins 1 and 2 did not affect normal blood vessels. In tumors, however, loss of epsins 1 and 2 resulted in disorganized vasculature, significantly increased vascular permeability, and markedly retarded tumor growth. Mechanistically, we show that VEGF promoted binding of epsin to ubiquitinated VEGFR2. Loss of epsins 1 and 2 specifically impaired endocytosis and degradation of VEGFR2, which resulted in excessive VEGF signaling that compromised tumor vascular function by exacerbating nonproductive leaky angiogenesis. This suggests that tumor vasculature requires a balance in VEGF signaling to provide sufficient productive angiogenesis for tumor development and that endothelial epsins 1 and 2 negatively regulate the output of VEGF signaling. Promotion of excessive VEGF signaling within tumors via a block of epsin 1 and 2 function may represent a strategy to prevent normal angiogenesis in cancer patients who are resistant to anti-VEGF therapies.

Abstract:Excess cholesterol accumulation in lesional macrophages elicits complex responses in atherosclerosis. Epsins, a family of endocytic adaptors, fuel the progression of atherosclerosis; however, the underlying mechanism and therapeutic potential of targeting Epsins remains unknown. In this study, we determined the role of Epsins in macrophage-mediated metabolic regulation. We then developed an innovative method to therapeutically target macrophage Epsins with specially designed S2P-conjugated lipid nanoparticles, which encapsulate small-interfering RNAs to suppress Epsins. We used single-cell RNA sequencing with our newly developed algorithm MEBOCOST to study cell-cell communications mediated by metabolites from sender cells and sensor proteins on receiver cells. Biomedical, cellular, and molecular approaches were utilized to investigate the role of macrophage Epsins in regulating lipid metabolism and transport. We performed this study using myeloid-specific Epsin double knockout (LysM-DKO) mice and mice with a genetic reduction of ABCG1 (ATP-binding cassette subfamily G member 1; LysM-DKO-ABCG1 fl/+ ). The nanoparticles targeting lesional macrophages were developed to encapsulate interfering RNAs to treat atherosclerosis. We revealed that Epsins regulate lipid metabolism and transport in atherosclerotic macrophages. Inhibiting Epsins by nanotherapy halts inflammation and accelerates atheroma resolution. Harnessing lesional macrophage-specific nanoparticle delivery of Epsin small-interfering RNAs, we showed that silencing of macrophage Epsins diminished atherosclerotic plaque size and promoted plaque regression. Mechanistically, we demonstrated that Epsins bound to CD36 to facilitate lipid uptake by enhancing CD36 endocytosis and recycling. Conversely, Epsins promoted ABCG1 degradation via lysosomes and hampered ABCG1-mediated cholesterol efflux and reverse cholesterol transport. In a LysM-DKO-ABCG1 fl/+ mouse model, enhanced cholesterol efflux and reverse transport due to Epsin deficiency was suppressed by the reduction of ABCG1. Our findings suggest that targeting Epsins in lesional macrophages may offer therapeutic benefits for advanced atherosclerosis by reducing CD36-mediated lipid uptake and increasing ABCG1-mediated cholesterol efflux. Abstract:Endocytosis has been traditionally considered as a mechanism to maintain plasma membrane homeostasis and internalize extracellular molecules. In the last few years, this view has been considerably expanded due to the accumulating evidence that endocytosis may also serve as a platform for intracellular signaling and actin dynamics. The multiplicity of endocytic actions relies on an extensive network of molecular interactions that are actively investigated both in vitro and in vivo. This network is supported by a variety of adaptor proteins that act as hubs for different cellular functions. Epsins are prototypes of such class of endocytic adaptors, with multiple interaction surfaces for factors implicated in endocytosis, actin dynamics, nuclear and ubiquitin function. By a genetic approach in mice, we systematically inactivated the three epsin genes in mammal. While single epsin knockouts (KO) do not show major phenotypic defects, double knockout (DKO) mice of the ubiquitously expressed epsins, i.e. the epn1/2 DKOs, are embryonic lethal at the beginning of organogenesis. Morphological, biochemical and expression analyses show defects that correlate with a disruption of the Notch signaling pathway. Strikingly, no alterations are observed in housekeeping internalization pathways as a consequence of epsin absence, strongly supporting the notion that epsins belong to a new class of endocytic adaptors with a specific function in intracellular signaling.

Abstract:Objective We previously showed that endothelial epsin deficiency caused elevated vascular endothelial growth factor receptor 2 (VEGFR2) and enhanced VEGF signaling, resulting in aberrant tumor angiogenesis and reduced tumor growth in adult mice. However, direct evidence demonstrating that endothelial epsins regulate angiogenesis specifically through VEGFR2 downregulation is still lacking. In addition, whether the lack of epsins causes abnormal angiogenesis during embryonic development remains unclear. Approach and Results A novel strain of endothelial epsin-deleted mice that are heterozygous for VEGFR2 (Epn1(fl/fl); Epn2(-/-); Flk(fl/+); iCDH5 Cre mice) was created. Analysis of embryos at different developmental stages showed that deletion of epsins caused defective embryonic angiogenesis and retarded embryo development. In vitro angiogenesis assays using isolated primary endothelial cells (ECs) from Epn1(fl/fl); Epn2(-/-); iCDH5 Cre (EC-iDKO) and Epn1(fl/fl); Epn2(-/-); Flk(fl/+); iCDH5 Cre (EC-iDKO-Flk(fl/+)) mice demonstrated that VEGFR2 reduction in epsin-depleted cells was sufficient to restore normal VEGF signaling, EC proliferation, EC migration, and EC network formation. These findings were complemented by in vivo wound healing, inflammatory angiogenesis, and tumor angiogenesis assays in which reduction of VEGFR2 was sufficient to rescue abnormal angiogenesis in endothelial epsin-deleted mice. Conclusions Our results provide the first genetic demonstration that epsins function specifically to downregulate VEGFR2 by mediating activated VEGFR2 internalization and degradation and that genetic reduction of VEGFR2 level protects against excessive angiogenesis caused by epsin loss. Our findings indicate that epsins may be a potential therapeutic target in conditions in which tightly regulated angiogenesis is crucial, such as in diabetic wound healing and tumors.
Epha2 Knockdown Attenuates Atherosclerotic Lesion Development in Apoe(-/-) Mice

Abstract:Background: The inflammatory response of vascular endothelial cells plays important roles in the initiation and progression of atherosclerotic lesions. EphA2 receptor activation promotes the endothelial cell inflammatory response, and its expression is increased in the endothelial cell layer of atherosclerotic plaques. However, the association between EphA2 and atherosclerosis has not been determined.Methods: Eight-week-old male ApoE(-/-) mice were systemically infected with adenoassociated virus serotype 9 carrying a small hairpin RNA specifically targeting the EphA2 gene to knock down EphA2 expression in aortic endothelial cells. These mice were then fed a high-cholesterol diet for 12 weeks. Blood was collected for the measurement of plasma lipids. The aortas were harvested to evaluate the atherosclerotic lesion size, macrophage components, and expression of proinflammatory genes using Oil Red O staining, immunofluorescence staining, and molecular biology analysis.Results: The lesions formed in the entire aorta and aortic sinus of the ApoE-/- mice with EphA2 knockdown were significantly smaller than those in the control mice (10.7% +/- 3.1% versus 25.1% +/- 4.2%; 0.51 +/- 0.02 mm(2) versus 0.85 +/- 0.03 mm2; n = 10; P <.05). Furthermore, the lesions in the ApoE-/- mice with EphA2 knockdown displayed reduced inflammation compared with the control mice, as reflected by the decreased macrophage infiltration (8.2% +/- 2.9% versus 22.7% +/- 4%; n = 10; P <.05); decreased nuclear factor-kappa beta activation; and diminished expression of vascular cell adhesion molecule-1, E-selectin, and monocyte chemotactic protein-1 (all P <.05).Conclusions: Our data demonstrate that the EphA2 receptor silencing attenuates the extent and inflammation of atherosclerotic lesions in ApoE-/- mice. Abstract:Introduction: Nonalcoholic steatohepatitis (NASH) is a chronic liver disease that is associated with high risks of cardiovascular diseases. Epsin is an evolutionarily conserved protein family that is involved in clathrin-mediated endocytosis. We found that both Epsin1/2 expressions in the liver are significantly up-regulated by the Western Diet (WD) induced NASH mice model, as well as in the liver biopsy of NASH patients. Hypothesis: We hypothesize that the expressions of Epsin1 and Epsin2 in the liver are required for NASH progression. Methods: We conditionally deleted Epsin1/2 using Albumin-Cre (Liver-DKO). We then induced a mouse model of NASH by feeding both WT and Liver-DKO mice a WD for 52 weeks. We used single cell multiome, combined with molecular approaches to study the mechanisms. We tested a strategy to deliver Epsin1/2 siRNAs encapsulated nanoparticles (NPs) targeting the liver via intravenous injection to treat diet induced NASH in mice. Results: We found WD-fed Liver-DKO mice have significant reductions in hepatic fibrosis formation and less lipid accumulation, and this was associated with better glucose and insulin tolerance, as well as lower serum aspartate transaminase (AST) and alanine aminotransferase (ALT) compared with WD-fed WT mice. Through analysis of the single cell multiome from the WT and Liver-DKO livers, we found more Kupffer cell in WT NASH mice compared with DKO. Interestingly, monocyte-derived inflammantory Kupffer cell (KCs) are enriched in WD-fed WT mice, however, embryonic-derived KCs are the enriched in liver DKO, and associate with dramatically reduced total KCs population. We also found that HNF4a was dramatically reduced in WD-fed WT mice, but its expression was preserved in WD-fed Liver-DKO mice, suggesting the loss of hepatocyte identity was inhibited by Epsins deletion. Therapeutically, we found significant resolution of hepatic fibrosis after knockdown of Epsin1/2 siRNA NP treatment. Conclusions: Liver specific deletion of Epsin1/2 significantly inhibits NASH progression by suppression of inflammation that protects hepatocytes from severe chronic injury. Our preclinical data suggests specially targeting Epsin1 and Epsin2 in the liver could be a promising therapeutic strategy for NASH treatment.

Abstract:E-selectin-1 (ESL-1), also known as golgi complex-localized glycoprotein-1 (GLG1), homocysteine-rich fibroblast growth factor receptor (CGR-1), and latent transforming growth factor-β complex protein 1 (LTCP-1), is a multifunctional protein with widespread tissue distribution. To determine the functional consequences of ESL-1 deficiency, mice were generated carrying an ESL-1 gene trap. After backcrossing to C57BL6/J for 6 generations, mice heterozygous for the gene trap (ESL-1(+/-)) were intercrossed to produce ESL-1(-/-) mice, however ESL-1(-/-) mice were not viable, even at embryonic day E10.5. To determine the effect of heterozygous ESL-1 deficiency on atherosclerosis, apolipoprotein E deficient (ApoE(-/-)), ESL-1(+/-) mice were generated and fed western diet. Compared to ApoE(-/-), ESL-1(+)(/)(+) mice, atherosclerotic lesions from ApoE(-/-), ESL-1(+/-) contained more collagen and fewer macrophages, suggesting increased plaque stability. In conclusion, heterozygous deficiency of ESL-1 is associated with features of increased atherosclerotic plaque stability while complete deficiency of ESL-1 leads to embryonic lethality.
Ablation of Myeloid ADK (Adenosine Kinase) Epigenetically Suppresses Atherosclerosis in ApoE-/- (Apolipoprotein E Deficient) Mice

Abstract:Objective- Monocyte-derived foam cells are one of the key players in the formation of atherosclerotic plaques. Adenosine receptors and extracellular adenosine have been demonstrated to modulate foam cell formation. ADK (adenosine kinase) is a major enzyme regulating intracellular adenosine levels, but its functional role in myeloid cells remains poorly understood. To enhance intracellular adenosine levels in myeloid cells, ADK was selectively deleted in novel transgenic mice using Cre-LoxP technology, and foam cell formation and the development of atherosclerotic lesions were determined. Approach and Results- ADK was upregulated in macrophages on ox-LDL (oxidized low-density lipoprotein) treatment in vitro and was highly expressed in foam cells in atherosclerotic plaques. Atherosclerotic mice deficient in ADK in myeloid cells were generated by breeding floxed ADK (ADKF/F) mice with LysM-Cre (myeloid-specific Cre recombinase expressing) mice and ApoE-/- (apolipoprotein E deficient) mice. Mice absent ADK in myeloid cells exhibited much smaller atherosclerotic plaques compared with controls. In vitro assays showed that ADK deletion or inhibition resulted in increased intracellular adenosine and reduced DNA methylation of the ABCG1 (ATP-binding cassette transporter G1) gene. Loss of methylation was associated with ABCG1 upregulation, enhanced cholesterol efflux, and eventually decreased foam cell formation. Conclusions- Augmentation of intracellular adenosine levels through ADK knockout in myeloid cells protects ApoE-/- mice against atherosclerosis by reducing foam cell formation via the epigenetic regulation of cholesterol trafficking. ADK inhibition is a promising approach for the treatment of atherosclerotic diseases.

Abstract:The vertebrate skin is a barrier-forming organ in which keratinocytes form a highly organized, stratified epithelium protecting the organism from the outside environment. One of the major regulators of this structure is Notch signaling. Notch is a transmembrane receptor interacting with ligands expressed on the surface of neighboring keratinocytes. Keratinocyte-specific deletion of Notch signaling pathway impairs epidermal differentiation, resulting in skin-barrier defects and skin carcinogenesis. Our laboratory, by a genetic approach in mice, demonstrates that combined inactivation of Epsin1 and Epsin2 genes leads to embryonic lethality around E9.5-10. The phenotype of Epn1;Epn2 double knockout is characterized by a subversion of the three main developmental programs active at this developmental stage, i.e., cardiovascular development, somitogenesis, and neural tube differentiation. Collectively, these morphological alterations resemble the developmental defects observed in mutants of Notch genes or in genes essential for the activation of the Notch signaling pathway, suggesting a crucial role of Epsin in enabling Notch signaling during embryogenesis. Intriguingly, the apparently healthy Epn1+/-;Epn2-/- shows an high incidence of squamous papillomas on their skin. Furthermore, expression of another epsin family member originally localized exclusively to surface epithelia, Epsin3, dramatically increases in the hyperplastic lesions of the three-allele mutants and in human basal carcinomas. In order to get further insight on Epn3 function we performed morphological expression analyses during mouse development. In contrast with initial reports, both in embryos and adults, we could detect various levels of Epn3 expression in several tissues, i.e., surface epithelia, neural tissue and heart. Moreover, in vitro studies performed on human keratinocytes in culture show a prominent role of this Epsin in the regulation of Notch signaling in this cell compartment.
Abstract 13426: Suppression of Epsin Expression Rescues Diabetes Triggered Impaired Lymphatic Function by Limiting VEGFR3 Degradation

Abstract:Rationale: Impaired lymphatic circulation causes lymphedema and contributes to pathological conditions, such as diabetes. VEGF-C/VEGFR3 signaling is a crucial regulator of lymphatic development; however, whether manipulation in signaling improves lymphatic circulation under pathological conditions is unclear. Objective: We recently uncovered that loss of epsins in lymphatic endothelial cells (LECs) caused heightened VEGFR3 signaling and delayed lymphatic formation during development. Herein, we investigated whether elevated VEGFR3 signaling caused by epsin loss is critical for pathological lymphatic circulation in adult mice. Methods and Results: Using innovative near-infrared lymphatic imaging, and tail lymphedema models, we demonstrated that epsin loss accelerated recovery from diabetic secondary lymphedema. High-glucose-induced oxidative stress results in reduced VEGFR3 signaling, impaired lymphangiogenesis and delayed recovery from diabetic secondary lymphedema. The dual luciferase reporter assay system and ChIP assay showed in STZ-induced diabetic mice drove Src- and AP-1- mediated upregulation of epsin expression. Epsin deletion in primary LECs caused heightened VEGFR3 signaling and promoted in vitro lymphangiogenesis by preventing VEGFR3 downregulation. Mechanistically, we showed that by protecting cell surface VEGFR3 from degradation, lack of epsin rescued high-glucose-induced VEGFR3 loss, defective lymphangiogenesis in vitro , and ameliorated STZ-induced diabetic secondary lymphedema in vivo . Conclusions: Epsins essentially regulate lymphatic circulation through the modulation of VEGFR3 signaling. In STZ- induced diabetic mice, Src induces epsin expression via transcription factor AP-1. Epsin depletion ameliorated diabetic secondary lymphedema under hyperglycemic condition by protecting VEGFR3 from oxidative stress-induced degradation. Our findings highlight the therapeutic potential for targeted inhibition of epsins within the lymphatic endothelium as a treatment for secondary lymphedema caused by uncontrolled diabetes.
