Abstract:Several new 1,3,4-oxadiazole derivatives containing a pyrazole ring were designed and synthesized from ethyl acetoacetate and triethyl orthoformate as starting materials via multi-step reactions. The compound structures were confirmed by melting point, 1 H NMR and HRMS. They were evaluated for fungicidal and herbicidal activities. Four of the compounds exhibited moderate fungicidal activity against Colletotrichum species. Most of the compounds had moderate-to-good activity as a herbicide.

Abstract:Pyrroloquinoxalines have proved to be a very attractive scaffold for medicinal chemist in the recent past. These compounds were extensively studied as bioactive compounds and many of them are known to be biologically and medicinally useful molecules, such as anti-HIV agents, antimalarial agents, antagonist agents, anticancer agents, and PARP-1 inhibitors. Additionally, pyrroloquinoxalines are also important intermediates for the construction of 5-HT3 receptor agonists. In this review the developments in biological activities and synthetic methodologies of pyrroloquinoxalines are discussed.

Abstract:Pyrazole compounds are one kind of heterocyclic compounds with extensive bioactivity,and lots of pyrazole compounds have been commercialized both in the region of medicament and pesticide.In the process of finding out some novel pyrazole compounds,a~d four novel pyrazole intermediates were synthesized.Taking diethyl oxalate as starting substance,the novel compounds can be synthesized via condensation,cyclic condensation,alkylation,(bromination) and hydrolysis reactions.The yield of products was about 78%-90%.Their structures were confirmed by elementary analysis,1H-NMR,MS and IR.

Abstract:Within the past years, many researches on the synthesis, structure-activity relationships (SAR), antitumor, antiinflammatory and anti-bacterial activities of the pyrazole derivatives have been reported. Several pyrazole derivatives possess important pharmacological activities and they have been proved useful materials in drug research. Pyrazole derivatives play an important role in antitumor agents because of their good inhibitory activity against BRAFV600E, EGFR, telomerase, ROS Receptor Tyrosine Kinase and Aurora-A kinase. In addition, pyrazole derivatives also show good antiinflammatory and anti-bacterial activities. In this review, the bioactivities of the pyrazole derivatives mentioned above will be summarized in detail. We sincerely hope that increasing knowledge of the SAR and cellular processes underlying the bioactivity of pyrazole derivatives will be beneficial to the rational design of new generation of small molecule drugs. Abstract:The triazolopyrimidine ring is a ubiquitous structural feature of many active compounds with diversified pharmacology efficacy. These structures have aroused our / researchers interests in the development of novel compounds with anticancer, anti-inflammatory, antibacterial, antifungal, and other activities. A large number of published literatures were reviewed during the last few decades. This review contains various pharmacological and agrochemical activities of triazolopyrimidine and it may be regarded as the lead compound for the new research towards future medicinal and agrochemical development.

Abstract:Two series of N-pyridylpyrazole derivatives containing 1,2,3-triazole and quinazolin-4(3H)-one substructures were designed and synthesized. In total, 18 novel compounds were prepared, and all compounds were characterized by H-1 NMR, C-13 NMR and elemental analysis (EA). Preliminary bioassay results revealed that a few of new compounds with quinazolin-4(3H)-one moiety exhibited good insecticidal activity against the oriental armyworm (Mythimna separata). In addition, the compounds Ia-h with 1, 2, 3-triazole moiety showed broad-spectrum antifungal activities against Fusarium oxysporum f.sp.cucumerinum, Cercospora arachidicola Hori, Botryosphaeria dothidea, Alternaria solani, Gibberella zeae and Phytophthora capsici at 50 [mu g/mL concentration. The EC50 values of Ib and If against Botryosphaeria dothidea were 13.9 and 11.0 mu g/mL, respectively, which were comparable to Chlorothalonil. These results indicated the potential application of N-pyridylpyrazole derivatives as fungicide in further study.

Abstract:Over the past decades, pyrazole has displayed significant importance in various areas of chemistry and industry owing to its unique nature, reactivity, and properties. Pyrazole is versatile compound with a wide range of applications in pharmaceuticals, agriculture, materials science, and various branches of science. Specifically, pyrazole C-3/C-5 carbaldehydes can be used as a building block in organic synthesis to create more valuable and bioactive molecules. In this context, pyrazole C-3/C-5 carbaldehydes may be explored for achieving pyrazole fused/linked heterocyclic skeletons by employing different organic transformations. The investigation of pyrazole C-3/C-5 carbaldehydes has resulted in the formation of a range of biologically potent heterocyclic structures, which include pyrazole C-3/C-5-based silicon derivatives, pyrazole C-3/C-5-based Schiff base derivatives, pyrazole C-3/C-5-based metal complexes, pyrazole C-3/C-5-based imidazo[1,2-a]azines, pyrazole C-3/C-5-based sulphur containing derivatives, and pyrazole C-3/C-5-based thioamide and amides, as well as other pyrazole fused/tethered polycyclic systems. Previous reports have shown that derivatives synthesized from the pyrazole C-3/C-5 carbaldehyde have demonstrated favourable biological uses like 5-HT3A receptor antagonists, allosteric inhibitors, insecticidal activity, antioxidant, antifungal, antiproliferative and antimicrobial. Pyrazole C-3/C-5 carbaldehydes have the potential to be highly utilized in the medical field as well as material science if studied judiciously. This review focuses on the exploration of pyrazole C-3/C-5 carbaldehydes towards the synthesis of highly diversified pyrazole-based molecular hybrids till 2023 and describes their promising utilization in the field of pharmaceutical science.

Abstract:In order to find new pyrazole leading compounds, 14 1-[(un)substituted phenyl]-4-(pyrazol-5-ylcarbonyl)semicarbazide derivatives were synthesized from 4-chloro-3-ethyl-1-methyl-5-pyrazole carbonhydrazide and substituted phenylisocyanate. The structures of all the new compounds were confirmed by 1H NMR, IR, MS and elemental analysis. The preliminary bioassays indicated that, at 500 mu g/mL, 1-(o-tolyl)-4-(4-chloro-3-ethyl-1-methylpyrazol-5-ylcarbonyl)semicarbazide (4g) and 1-(2,4-dimethyl-phenyl)-4-(4-chloro-3-ethyl-1-methylpyrazol-5-ylcarbonyl)semicarbazide (4k) showed inhibitory activities 90% and 80% against Blumeria graminis, respectively; and at 25 mu g/mL, 4-phenyl-4-(4-chloro-3-ethyl-1-methylpyrazol-5-ylcarbonyl)semicarbazide (4c) showed inhibitory activity 70.1% against Botrytis cinerea and 1-(2-nitro-phenyl-4-(4-chloro-3-ethyl-1-methylpyrazol-5-ylcarbonyl)semicarbazide (4d) showed inhibitory activity 51.3% against Pyricularia oryzae.
Design,Synthesis and Antifungal Activity of Novel Pyrazole-Thiophene Carboxamide Derivatives

Abstract:Succinate dehydrogenase inhibitor is a low-toxic and high active fungicide. In order to develop novel and broad-spectrum succinate dehydrogenase inhibitor (SDHI) fungicides, thiophene was introduced into the structure of pyrazole carboxamide fungicides. Twenty four pyrazole-thiophene carboxamides were designed, synthesized and characterized by H NMR, C NMR and HRMS. The crystal structure of N-(4-methoxyphenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene2-carboxamide (7i) was determined by X-ray diffraction. The antifungal activity of all the synthesized compounds was determined against six plant pathogenic fungi, and preliminary bioassays suggested that some compounds exhibited good antifungal activity against Rhizoctonia solani, Fusarium graminearum and Botrytis cinerea. Among them, N-(4fluorophenethyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7c) exhibited the best antifungal activities against R. solani in vitro with EC50 value of 11.6 μmol/L, and N-(2-fluorophenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7j) against F. graminearum with EC50 value of 28.9 μmol/L. And N-(4-chlorophenyl)-4-(1-methyl-1H-pyrazol-4yl)thiophene-2-carboxamide (7h) showed similar inhibition abilities with thifluzamide against B. cinerea with EC50 value of 21.3 μmol/L. The molecular docking results showed that the high antifungal activitie compounds form stronger interactions with important amino acid residues of succinate dehydrogenase.

Abstract:Heterocyclic pyrazoles are an important class of biologically active scaffold with extensive biological functions such as antifungal, insecticidal, nematocidal, antiviral and antitumor activities. In this study, a series of pyrazole derivatives containing acylhydrazone or heterocyclic amide were designed and synthesized via a sequence transformation with good to excellent yields, and their structures were characterized by satisfied spectrum analyses. These newly obtained molecules were fully evaluated for their fungicidal activities against Phytophthora capsici , Sclerotinia sclerotiorum , Botrytis cinerea and Rhizoctonia solani , which indicated that the compounds 7 and 9 had appreciable selectivity against the basidiomycete species R . solani , and in vivo bioassay also demonstrated that the compound 9 could protect the horsebean against infection by R . solani .

Abstract:Pyrazolone is a well-known influential synthon for the synthesis of many molecules which hold remarkable potential in the advancement of functional materials, coordination chemistry, and pharmaceutical chemistry. Taking the advantage of diversity as well as multiple active sites of starting materials, a number of effective methods for synthesizing different pyrazolone compounds have been effectively established over the last few years. The chemistry of pyrazolone derivatives have attracted interests due to their diverse pharmacological functions such as analgesic, anti-inflammatory, anti-cancer, antibacterial, antifungal, anti-oxidant, anti-diabetic, anti-viral, anti-proliferative and many other biological activities. This review article provides an overview of this field, with the aim of highlighting the remarkable achievements that have been made with the multiple reaction sites of different pyrazolone synthons and the synthetic strategies for existing and novel pyrazolone derivatives. In addition, chemical synthetic schemes, important mechanisms, structure activity relationships (SAR) and promising biological investigations relating to the corresponding products are also discussed.

Abstract:In an attempt to search for potent fungicide, a series of novel N-aryl-1H-pyrazole-5-carboxylate derivatives was designed and synthesized. Their chemical structures were characterized by 1H NMR spectra and high resolution mass spectrometry(HRMS). The preliminary bioassay results indicated that some target compounds displayed better fungicidal activities against certain fungi at 50 µg/mL or favorable antitumor activities at 5 µg/mL compared with chlorothalonil and 5-fluorouracil, respectively. The structure-activity relationship demonstrated that the introduction of ester group and amide bond was favorable to the improvement of activities against Physalospora piricola and Phytophthora capsici.

Abstract:Abstract: Heterocyclic compounds containing nitrogen and their derivatives have been a rich source of medicines. Pyrazole, a five-membered ring structure, offers a variety of functionalities as well as stereo-chemical complexity. Studies conducted over the past 10 years revealed that an increasing amount of research has been performed on different pyrazole derivatives and their physiological and pharmacological activities. The objective of these studies is to uncover the full potential of pyrazole derivatives by elucidating the many drug-like properties and their link between the structure and mode of action. Here, we discuss different ways of synthesizing pyrazole derivatives. Due to recent advances in synthetic medicinal chemistry, this class of compounds can be readily developed and becomes a viable target for the discovery of novel drugs.

Abstract:To discover new compounds with favorable herbicidal activity, a range of phenylpyridine moiety-containing pyrazole derivatives were designed, synthesized, and identified via NMR and HRMS. Their herbicidal activities against six species of weeds were evaluated in a greenhouse via both pre- and post-emergence treatments at 150 g a.i./hm2. The bioassay revealed that a few compounds exhibited moderate herbicidal activities against Digitaria sanguinalis, Abutilon theophrasti, and Setaria viridis in post-emergence treatment. For instance, compounds 6a and 6c demonstrated 50% inhibition activity against Setaria viridis, which was slightly superior to pyroxasulfone. Thus, compounds 6a and 6c may serve as the new possible leading compounds for the discovery of post-emergence herbicides.

Abstract:In this research, with an aim to develop novel pyrazole oxime ether derivatives possessing potential biological activity, thirty-two pyrazole oxime ethers, including a substituted pyridine ring, have been synthesized and structurally identified through 1H NMR, 13C NMR, and HRMS. Bioassay data indicated that most of these compounds owned strong insecticidal properties against Mythimna separata, Tetranychus cinnabarinus, Plutella xylostella, and Aphis medicaginis at a dosage of 500 μg/mL, and some title compounds were active towards Nilaparvata lugens at 500 μg/mL. Furthermore, some of the designed compounds had potent insecticidal effects against M. separata, T. cinnabarinus, or A. medicaginis at 100 μg/mL, with the mortalities of compounds 8a, 8c, 8d, 8e, 8f, 8g, 8o, 8s, 8v, 8x, and 8z against A. medicaginis, in particular, all reaching 100%. Even when the dosage was lowered to 20 μg/mL, compound 8s also expressed 50% insecticidal activity against M. separata, and compounds 8a, 8e, 8f, 8o, 8v, and 8x displayed more than 60% inhibition rates against A. medicaginis. The current results provided a significant basis for the rational design of biologically active pyrazole oxime ethers in future.

Abstract:The pyrazolone structural motif is a critical element of drugs aimed at different biological end-points. Medicinal chemistry researches have synthesized drug-like pyrazolone candidates with several medicinal features including antimicrobial, antitumor, CNS (central nervous system) effect, anti-inflammatory activities and so on. Meanwhile, SAR (Structure-Activity Relationship) investigations have drawn attentions among medicinal chemists, along with a plenty of analogues have been derived for multiple targets. In this review, we comprehensively summarize the biological activity and SAR for pyrazolone analogues, wishing to give an overall retrospect and prospect on the pyrazolone derivatives.

Abstract:Pyrazoles are five-membered heterocyclic compounds that contain nitrogen. They are an important class of compounds for drug development; thus, they have attracted much attention. In the meantime, pyrazole derivatives have been synthesized as target structures and have demonstrated numerous biological activities such as antituberculosis, antimicrobial, antifungal, and anti-inflammatory. This review summarizes the results of published research on pyrazole derivatives synthesis and biological activities. The published research works on pyrazole derivatives synthesis and biological activities between January 2018 and December 2021 were retrieved from the Scopus database and reviewed accordingly.

Abstract:Heterocycles are common in the structure of drugs used clinically to deal with diseases. Such drugs usually contain nitrogen, oxygen and sulfur, which possess electron-accepting capacity and can form hydrogen bonds. These properties often bring enhanced target binding ability to these compounds when compared to alkanes. Pyrazine is a nitrogen-containing six-membered heterocyclic ring and many of its derivatives are identified as bioactive molecules. We review here the most active pyrazine compounds in terms of their structure, activity in vitro and in vivo (mainly antitumor activity) and the reported mechanisms of action. References have been downloaded through Web of Science, PubMed, Science Direct, Google Scholar and SciFinder Scholar. Publications reporting only the chemistry of pyrazine derivatives are beyond the scope of this review and have not been included. We found that compounds in which a pyrazine ring was fused into other heterocycles especially pyrrole or imidazole were the highly studied pyrazine derivatives, whose antineoplastic activity had been widely investigated. To the best of our knowledge, this is the first review of pyrazine derivatives and their bioactivity, especially their antitumor activity. This review should be useful for those engaged in development of medications based on heterocyclic compounds especially those based on pyrazine.
