Mouse Interleukin-2 Gene Therapy for Head Andneck Squamous Cell Carcinoma by Polycationicliposome-Mediated Transfection

Abstract:ve To investigate the characteristics of polycationic lipid and the opitimal lipid to DNA ratio of lipid-DNA complex (lipoplexes) by which mouse IL-2 gene was transferred to head and neck squamous cell carcinoma (HNSCC). Methods Murine models of HNSCC were established in the floor of mouth in C3H/HeJ mice with SCCⅦ cell line. Lipoplexes with various lipid to DNA ratios and the naked DNA were transfected both in vivo and in vitro. Tumors were subcultured. The supernatants and serums were collected for IL-2 secretion by enzyme-linked immunosobent assay (ELISA). Results IL-2 Secretion was consistently higher when lipid:DNA(L:D)complexes were formulated at higher L :D ratios(wt/wt) in vitro (L:D=5:1; L:D=8:1; L:D=12:1). IL-2 transgene expressed most when L:D ratio =3 and in a DNA dose-dependent manner. Conclusion Comparison between naked plasmid and lipid DNA complexes revealed that lipoplexes were more effective for intratumoral gene transfer to HNSCC. The results indicated that the formulation and dosage of polycationic L:D complexes night play a key role in determining the level of intratumoral transgene expression.
