Abstract:This paper deals with the rapid propagation ofAnoectochilus formosanus Hayata via tissue culture in vitro. The stem segments of A.frormosanus Hayata were used as explants to establish the sterile materials through the surface sterile and initial culture. Four basic media,including MS,H,B_5 and N_6were tested, and of which H was the optimal medium for the multiplication culture of A. formosanus Hayata. The order of the four media for the multiplication culture of A.formosanus Hayata was:H≥N_6B_5≥MS.16 hormone treatments, involving 3 kinds of hormone (KT, BA and NAA), and their different concentrations and different matchings,were tested. The experimental results showed that "2.0mg/L BA+1.0mg/L NAA" was the optimal hormone matching. Under this hormone condition, the axillary buds were germinated from the 3～4 nodes of the basic part of almost 100% of the plantlets of A. formosanus Hayata and the clustered shoots were formed. The recommended optional medium prescription was "H+100mg/L inositol+0.5mg/L nicotinic acid+1.0mg/L thiamine· HCI+0.5mg/L pryidoxine· HCI+2.0mg/L glycine+2.0mg/L BA+1.0mg/L NAA+2.5% sucrose+0.7%argar". 7 kinds of cultivated materials for the transplantation of A. formosanus Hayata were tested, of which the mixed cultivated material, "River sand:Garden soil:Coir=1∶1∶1", was the optimal cultivated material for the cultivation of A. formosanus Hayata.
