Abstract:Based on the growth observation of lily bulbs (Lilium Oriental hybrids, ‘Acapulco’, ‘Marco Polo’, ‘Siberia’, ‘Sobonne’ and ‘Tiber’), we found the mountainous condition was suitable for the bulb development. Total 288,020 bulbs (12-14 cm, 14-16 cm and 16-18 cm in circumference) of Oriental lily were harvested from 401,500 imported bulblets (6/9 cm), which were grown in the mountainous region (Jinyun, Zhejiang Province) having an elevation of 880 m. The average ratio for harvested flowering bulbs (>12 cm) and big bulbs (16-18 cm) was 71.7% and 15.4%, respectively. The concentrations of soluble sugar, sucrose and starch in the bulb scales, as well as leaf photosynthesis were investigated during the whole year cultivation. It was found that the fresh weight and circumference of harvested bulbs have the positive correlation between vegetative growth in lily. The bud removal improved the process of bulb development. Accumulation and transportation of carbohydrates in the bulbs also increased significantly from the bud-removal. The average concentrations of starch and total soluble sugar were 293 mg g FW -1 and 9.2 mg g FW -1 in the harvested bulbs, respectively. It was indicated that the quality of mountainous reproduction bulbs was similar to that of the commercial bulbs from Holland.

Abstract:Scaling is the most commonly used technique to artificially propagate lilies. Scales from different positions of the donor bulb vary in regeneration efficiency; however, the mechanism underlying bulblet formation remains unclear. To investigate the relationship between scale morphological characteristics, initial nutrient status and bulblet regeneration capacities during scale propagation of Lilium, we performed comprehensive morphological and correlation analyses using scales from three lily cultivars. Principal component analysis clearly distinguished middle scale (MS) from outer scale (OS) by morphological characteristics alone. Morphological results indicated that MS and OS differ significantly in terms of scale width, facial area, basal area, volume, length-to-width ratio and width-to-thickness ratio. Correlation analysis showed that scale width was significantly positively correlated with both the quantity and quality of regenerated bulblets. Among the cultivars, starch and soluble sugars accounted for 50–80% of scale DW. And a higher initial ratio of sucrose to starch in scales was more conducive to the bulblets formation. Although light had no effect on the incidence of bulblets, the formation of bulblets was positively enhanced, and better morphological consistency was obtained. This present study achieved a comprehensive morphological and nutritional analysis focused on bulblet formation capacities of scales from different positions of lily bulbs via scaling propagation, laying a foundation for future molecular studies on bulblet formation.

Abstract:The primary objective of this research was to study the female fertility of the odd-tetraploid cultivar Honesty of Lilium containing one set of Longiflorum chromosomes and three sets of Asiatic chromosomes (LAAA) to open a new approach to Lilium breeding. To assess its female fertility, 'Honesty' was hybridized with four autotetraploid Asiatic lily cultivars. The results showed that the fruit of all 'Honesty' x tetraploid (4x x 4x) combinations developed well, and viable seedlings could be obtained, suggesting that 'Honesty', despite being male-sterile, has considerable female fertility. Genomic in situ hybridization showed that the progenies of the 4x x 4x hybridizations were aneuploid. Considering that lily is vegetatively propagated and aneuploids often demonstrate considerable phenotypic variation, odd-tetraploid lilies such as 'Honesty' may be useful maternal parents for breeding new lily cultivars.

Abstract:Herbaceous peony (Paeonia lactiflora Pall.) is a renowned traditional Chinese flower, which has been cherished by people since ancient times. It holds significant value as both an agricultural cash crop and an ornamental and medicinal plant. However, the lack of in vitro propagation system has been long hindered the commercial production of this species and the development of new cultivars. In this study, the low chilling requirement cultivar Paeonia lactiflora 'Hang Baishao' that originated from low latitudes of China, was used to study the in vitro culture of embryos. The results of our experiment showed that the optimal medium for in vitro culture of embryos was a 1/2 Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 6-benzyladenine (6-BA), 1.0 mg/L gibberellic acid (GA3) and 2.0 g/L polyvinyl pyrrolidone (PVP). Additionally, histological observations revealed the crucial role of vascular bundle development in the survival of transplants. Plants with well-developed vascular tissue exhibited better transplanting success. This study presents, for the first time, a protocol for embryos propagation and transplanting of 'Hang Baishao', a cultivar with low chilling requirement (CR) during winter. This study provides an efficient method for in vitro culture of 'Hang Baishao', and makes it possible for large-scale production, utilization, and germplasm preservation of this species.

Abstract:Lilium spp. is a bulb flower with worldwide distribution and unique underground organs. The lack of an efficient genetic transformation system for Lilium has been an international obstacle. Because existing model plants lack bulbs, bulb-related gene function verification studies cannot be carried out in model plants. Here, two stable and efficient genetic transformation systems based on somatic embryogenesis and adventitious bud regeneration were established in two Lilium species. Transgenic plants and T-DNA insertion lines were confirmed by beta-glucuronidase (GUS) assay, polymerase chain reaction (PCR) and Southern blot. After condition optimization, transformation efficiencies were increased to 29.17% and 4% in Lilium pumilum DC. Fisch. and the Lilium longiflorum White Heaven', respectively. To further verify the validity of these transformation systems and apply the CRISPR/Cas9 (Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)-associated protein 9) technology in Lilium, the LpPDS gene in the two Lilium species was knocked out. Completely albino, pale yellow and albino-green chimeric mutants were observed. Sequence analysis in the transgenic lines revealed various mutation patterns, including base insertion, deletion and substitution. These results verified the feasibility and high efficiency of both transformation systems and the successful application of the CRISPR/Cas9 system to gene editing in Lilium for the first time. Overall, this study lays an important foundation for gene function research and germplasm improvement in Lilium spp.

Abstract:In this paper, the status of lily industry, such as cut-flower production, bulb production, edible lily production status, in mainland China is summarized. The lily flower yield in 2012 amounted to more than 600 million stems. This is the estimation from some companies of bulb trade in China, and we don’t have the accurate data from the government. It was estimated that the import quantity of lily bulb of main-land China in 2012, including from the Netherlands and the south hemisphere, was more than 200 million. In mainland China, at present time, lily flower production is mainly distributed at six provinces, Yunnan, Liaoning, Zhejiang, Guangdong, Jiangsu, and Fujian, and this situation has been sustained a couple of years. Among the six provinces, Yunnan is number one, and Liaoning is number two. Because of the distinctive environmental conditions, including the climate, soil and water, between north and south China, the equipments and some detailed tech-niques for cut lily production are quite different. Therefore, it has been formed two technology systems, the north China system and South China system. The total area of lily bulb production in China is still not so clear. It is estimated that lily bulb produced domestically is about 60 million in 2012 in mainland China. Many Chinese lily growers just re-use the bulbs for one or two years, some-times even three or four years in some region. There are mainly two methods of treat-ment for reusing lily bulbs after flowering. Edible lilies are a small branch of lily industry in China, but not very popular on the world. There are mainly three lily species, or varieties used for food with their bulbs in China. They are Lanzhou lily (Lilium davidii var. unicolor), which was mainly grown in the region around Lanzhou, Gansu province, Longya lily (L. brownie), which was mainly grown at Hunan and Jiangxi province, and Yixing lily (L. lancifolium), which was mainly grown in Jiangsu province.

Abstract:Male reproductive organs such as anther and pollen grain of lily (Lilium longiflorum L.) are generally much bigger than other plants'. For this reason, it becomes one of the most used materials by researches for revealing the mechanisms underlying sexual reproductive development. In recent years, several specifically expressed peptides, proteins, and cDNA fragments had been isolated by researchers through SDS-PAGE, 2 D-protein electrophoresis, SSH, and RT-PCR technologies in pollen, anther, and pollen tube and different developmental stages in lily. Furthermore, function of part of these peptides had also been predicted and studied. Besides, some proteins had also been isolated that related with pollen tube and stigma interaction during fertilization. Their function had been inferred as well. In this review was not only issued on the molecular biology mecha- nism of the sexual reproductive development in lily, but also discussed the problems occur during these study and their possible application in lily breeding.
Hybridization and Chromosome Doubling for Potted Asiatic Lilies(Lilium)

Abstract:Tetraploid plants usually grow more vigorously and produce larger flowers than their diploids.To breed tetraploid poted Asiatic lilies,the present study carried out the crosses among Asiatic lily(Lilium) cultivars Petit Brigitte,Orange Pixie,Black Bird,and Pollyanna with normal artificial pollination,then,the scales of the progenies obtained from three crossing combination were treated 4 h with 0.001%,0.002%,0.003% and 0.005% Oryzalin,and the regenerated bulblets were analyzed with chromosome preparation and fluorescence in situ hybridization(FISH) technique.The results showed that the chromosome doubling rates were 19% and 23% with 0.003% and 0.005% Oryzalin respectively,and FISH not only confirmed the chromosome doubled plants,but indicated that some particular chromosomes might not be doubled in some plants.The present result conformed that Oryzalin is effective to double lily chromosomes,and that cytological method,combined with FISH technique,enable individual chromosomes whether to be doubled or not more accurately.

Abstract:Chinese sweet lily (Lilium davidii var. unicolor), the only edible lily in the world, has high nutritional value. At present, the application of preservation technology is relatively backward, resulting in a large loss of lily. Therefore, the preservation has become one of the key factors limiting the development of the edible lily bulbs industry. This paper reviews the various application of pretreatment and storage technologies for the preservation of lily bulbs. It is considered that irradiation and proper preservative can be used for pretreatment. Vacuum packaging can be used for short-term storage and transportation. Phase temperature storage could be as a long-term storage mode of lily bulbs.

Abstract:Experiment was performed to investigate the bulblet development and its physiological mechanisms of Lilium Oriental hybrids(cv.'Sorbonne' and 'Siberia') in the mountainous region(Dayang Town,Jinyun County,Zhejiang Province) with an elevation of 800m.The morphological and physiological changes were entirely recorded in the growth process of bulblet development.By the analysis of the concentration changes of total sugar,starch,reducing sugar,sucrose in the scales,the accumulation and distribution of carbohydrates in bulblets were studied.It was found that the average fresh weight of the harvested bulbs was 45.2 g which was 3.4 times than that of bulblets.Their average circumference of harvested bulbs reached 15.5 cm,which was 1.5 times than that of bulblets.The starch content in harvested bulbs reached 293 mg·g~(-1)FW,which was about 3 times than that of bulblets.Concerned about the nutrient contents,the quality of harvested bulbs was equal to the imported commercial bulbs from Netherlands.Sucrose content was in the majority of total soluble carbohydrates,usually more than 60%.The change trend of sucrose was similar as the total sugar in the different growth stages.There is an obvious correlation between the leaf number and the size,fresh weight and plant height in harvested bulbs of lily.
Embryo Rescue Technology in the Cut Flower Lily

Abstract:Some lily cultivars in Oriental hybrids,Asiatic hybrids and Longiflorm hybrids as test material,with three culture methods of ovary slice culture(OSC),ovule with placenta culture(OPC) and embryo rescue method(ESR) were used to culture the immature embryo after cut-style pollination method in different interspecific hybrid groups,and the influencing factors of immature embryo culture in vitro were studied in different intraspecific hybrid groups.The results showed that the OSC and the OPC made some hybrid embryo germ in OA,LA crossings,and the OPC method was better than the OSC method.The ESR method made some hybrid embryo germ in OA,LA,LO crossings,and almost the entire hybrid seedlings died gradually except for the only one in LO continued to grow.Embryo age was about fifty days in OO crossings,in AA about forty days and in LL crossings it was necessary to add 0.01～0.1 mg/L NAA for culturing immature embryo,but wether to add 6-BA or not there existed differences among different crossing combinations.

Abstract:Different varieties of lily were used as materidls to breed new varieties according to the different color and cold resistance to breed new varieties. Conventional crossing menthod was used between hybrids and Asia lily. Three different methods were adopted between Asian and oriental lily. Under the analysis, the results showed that different cross combinations of lily interspecific hybridization were suit to different hybridization methods.

Abstract:Lily (Lilium spp.) has elegant flowers and beautiful colors, which makes it popular among people. However, the poor stress resistance and self-propagation ability of lily limit its application in landscaping to a great extent. In addition, transgenic technology is an important means to improve plant characteristics, but the lack of a stable and efficient genetic transformation system is still an important factor restricting the development of lily transgenic technology. Therefore, this study established a good lily regeneration system by screening different explants and plant growth regulators of different concentrations. Then, the genetic transformation system of lily was optimized by screening the critical concentration of antibiotics, the concentration of bacterial solution, and the infection time. Finally, the homologous lily cold resistance gene LlNAC2 and bulblet generation gene LaKNOX1 were successfully transferred to ‘Siberia’ and ‘Sorbonne’ to obtain lily transgenic lines. The results showed that when the stem axis was used as explant in ‘Siberia’, the induction rate was as high as 87%. The induction rate of ‘Sorbonne’ was as high as 91.7% when the filaments were used as explants. At the same time, in the optimized genetic transformation system, the transformation rate of ‘Siberia’ and ‘Sorbonne’ was up to 60%. In conclusion, this study provides the theoretical basis and technical support for improving the resistance and reproductive ability of Oriental lily and the molecular breeding of lily.

Abstract:Water shortage is a key environmental factor that negatively effects plant growth. Lanzhou lily (Lilium davidii var. unicolor) is a perennial herbaceous drought-tolerant crop widely used as a food and medicine in China. The effects of drought stress on plant growth, osmotic pressure, and secondary metabolite content differ. Here, we investigate alterations in basic physiological processes and in the accumulation of osmolytes by Lanzhou lily under drought stress. Plants were grown at three drought intensities, being irrigated at 5, 15, and 25 day intervals (either throughout the study or during specific growth stages). Water stress markedly decreased plant height and leaf length. With increasing drought stress, the chlorophyll content, bulb weight, and contents of soluble sugars, polysaccharides, and fructose decreased. In contrast, the proline, glucose, and trehalose contents increased under severe drought stress. In addition, glucose and trehalose contents differed significantly under drought stress at different growth stages, whereas other indicators differed significantly only under drought stress throughout the growth period. Our results demonstrate that Lanzhou lily may adapt to drought stress in different growth stages were different. During the shoot stage, the adaptation strategy was to reduce the growth of aboveground part to sustain the underground parts, but during the bulbs expansion stage Lanzhou lily appears to adapt to drought stress by consuming nutrients from underground bulbs to sustain the growth of the aboveground parts and complete the plant's life cycle and by changing osmotic regulation and the levels of secondary metabolites to improve resistance to drought stress.
