Abstract:Newly synthesized polymer-supported chiral amino alcohol catalysts 5a and 5b have been proved to be effective for the enantioselective addition of diethylzinc to aldehydes, affording the corresponding sec-alcohols in moderate enantiomeric excesses with good to excellent yields. The recycled catalyst could be reused in the same reaction without significant deterioration in performance.

Abstract:A new linear chiral polymer ligand was synthesized by the polymerization of (S)-5, 5'-dibromo-6, 6,-di(4-trifluoromethylphenyl)-2, 2'-binaphthol (S-M-l) with (S)-5, 5 '-diethynyl-6, 6'-dibutyl-2, 2'-dibutoxy-l, 1'-binaphthyl (S-M-2) via Pd-catalyzed Sonogashira reaction. The application of the chiral polymer ligand in the asymmetric addition of Et2Zn to various aromatic aldehydes had been studied. The results showed that this kind of soluble chiral polybinaphthols ligand in combination with Ti(OiPr)4 could exhibit good reactivity and enantioselectivity for the asymmetric addition of Et2Zn to aromatic aldehydes. Moreover, the chiral polymer ligand showed superior enantioselectivity over the model compound S-l and could keep similar reactivity as well as enantioselectivity after ten recycles.

Abstract:A series of chiral tridentate ligands derived from readily available enantiopure l-proline were designed and synthesized. The ligands together with Cu(OAc)2 were successfully used in asymmetric Henry reactions. Various structurally divergent aldehydes and nitromethane were converted into versatile β-nitro alcohols in MeOH at room temperature with very good yields (up to 85%) and enantioselectivities (up to 86%).

Abstract:Attachment of the proline-derived ligand 5 to two types of polymers, i.e. aminomethylated polystyrene resin and respectively MeO-PEG, was achieved via a succinyl linkage from the 4-hydroxy group. Both of the supported ligands (7 and 8) were proved to be highly active with good enantioselectivity (up to 90% ee) for the catalyzed asymmetric diethylzinc addition to aldehydes. The insoluble polymer-supported catalyst could be easily recovered and was reusable for several consecutive catalytic runs without significant loss in enantioselectivity.

Abstract:New Schiff base chiral polymers was synthesized. The asymmetric addition of diethyl zinc to aldehyde was achieved catalyzed by Ti(IV)/polymer which was formed in situ from chiral polymer and Ti(O-i-Pr)(4). Optical alcohols were obtained in a high yield with e.e. up to 84.5%, The effects of temperature, complex time, structure of substrate etc. on the catalytic activity and enantio selectivity were studied in detail. The reaction mechanism was proposed.

Abstract:As a structural analog of oxazoline, imidazoline (4,5-dihydroimidazole) has received much attention in the rational design of chiral ligands. The additional N-substituent provides broader space for fine-tuning of electronic and steric effects, and it offers a good handle for immobilizing onto solid supports. In the past decades, imidazoline ring has emerged as a powerful candidate for the design of chiral nitrogen-containing ligands, as well as a significant alternative for oxazoline ring. Various chiral imidazoline ligands have been designed and utilized in asymmetric organic reactions. These new catalysts can not only be applied in classical reactions, but also be employed to develop new organic reactions with high enantioselectivities. This review provides an overview of chiral imidazoline ligands. Their applications in asymmetric synthesis are also summarized.

Abstract:Heterogeneous asymmetric addition of diethylzinc to aldehydes catalyzed by chiral polymer catalysts has been achieved. The conjugated polymers based on a BINOL skeleton were synthesized in five steps and applied to this reaction with yield and e.e. up to 95 and 84.5%, respectively. These chiral polymers can be easily quantitatively recovered and reused without decreasing of e.e. or catalytic activity.

Abstract:Enantioselective conjugate addition reactions are powerful methods in synthesizing optically active molecules. Procedures that employ chiral homogeneous catalysts have been developed extensively, but ones that use chiral heterogeneous catalysts are less explored and remain a challenge. Here we report a polymer-supported chiral heterogeneous copper catalyst that demonstrated high reactivity and enantioselectivity in asymmetric conjugate addition reactions of both ketones and imines. It was found that reduced steric hindrance inside the micro catalytic environment and a 1:1 ligand to metal ratio was crucial for the high efficiency and reusability of the chiral heterogeneous catalyst. Various α,β-unsaturated carbonyl substrates, including pharmaceutical intermediates and natural products, could be transformed in high yields with high enantioselectivities. Successful application of this methodology in a continuous flow fashion further broadened its usage in dealing with sensitive organozinc reagents. In addition, production was successfully scaled up under the flow condition.

Abstract:Metal-catalyzed enantioselective allylation, which involves the substitution of allylic metal intermediates with a diverse range of different nucleophiles or S(N)2'-type allylic substitution, leads to the formation of C-H, -C, -O, -N, -S, and other bonds with very high levels of asymmetric induction. The reaction may tolerate a broad range of functional groups and has been applied successfully to the synthesis of many natural products and new chiral compounds.

Abstract:Novel C-2-symmetrical bissulfonamide ligands were easily prepared in three simple steps and applied in the enantioselective addition of alkynylzinc reagents to aldehydes. Compound 7a was found to be an outstanding ligand for this reaction. When the catalyst loading, 4 mol % of 7a, was used, high enantioselectivity with an ee value up to 97% was achieved under very mild conditions. When the amount of ligand 7a was lowered to 2 mol %, excellent levels of enantiocontrol up to 95% ee were still achieved. So far the most economic catalyst system was developed for the addition of terminal alkynes to aromatic aldehydes in terms of ligand loading and enantioselectivity. Moreover, ligand 7a in combination with Ti(O-i-Pr)(4) was found to be effective in promoting the addition reaction of an alkynylzinc reagent to unactivated simple ketones under very mild conditions. The corresponding chiral tertiary propargylic alcohols were obtained with enantiomeric excesses of up to 81%.

Abstract:A novel procedure for selective direct N,N-alkylation of the chiral Betti base was developed, and a new family of chiral ligands, (S)-1-(α-cycloaminobenzyl)-2-naphthols, were prepared. The ligands with five- and six-membered cyclic amines showed highly efficient asymmetric induction in the addition of diethylzinc to aromatic aldehydes in 93–96% yields and 91–99% ee.

Abstract:Pyrazolones are a vital class of heterocycles possessing various biological properties and much attention is paid to the diversified synthesis of enantiopure pyrazolone derivatives. We describe here the development of diphenylphosphinoalkanoic acid based chiral bisphosphine ligands, which are successfully applied to the palladium-catalyzed asymmetric allenylation of racemic pyrazol-5-ones. The reaction affords C-allenylation products, optically active pyrazol-5-ones bearing an allene unit, in high chemo- and enantioselectivity, with DACH-ZYC-Phos-C1 as the best ligand. The synthetic potential of the C-allenylation products is demonstrated. Furthermore, the enantioselectivity observed with DACH-ZYC-Phos-C1 is rationalized by density functional theory studies.

Abstract:A family of ferrocenyl amino alcohols (14 examples) has been prepared from the corresponding planar chiral ketones and evaluated as a new class of chiral ligands for asymmetric ethyl‐ and phenylzinc additions to aldehydes. The highly enantioselective synthesis of these ligands is facile, scalable, robust and relies on the use of cheaply available starting materials. Asymmetric ethyl additions proceeded smoothly at ambient temperature, with reaction times as short as 1 hour furnishing the desired products in up to 90% yield and 99% ee. The corresponding phenyl additions afforded products in up to 81% yield and 88% ee. The observed stereochemical outcome for the addition of diethylzinc was rationalized and the key proposed transition state was supported by an analysis of the X‐ray structures of a series of amino alcohol ligands.

Abstract:A chiral polymer ligand was synthesized by the polymerization of (S)-5,5′-dibromo-6,6′-dibutyl-2,2′-binaphthol (S-M-1) with (S)-2,2′-bishexyloxy-1,1′-binaphthyl-6,6′-boronic acid (S-M-2) via Pd-catalyzed Suzuki reaction. The application of the chiral polymer ligand to the asymmetric addition of phenylethynyl zinc to various aldehydes has been studied. The results show that the soluble chiral polybinaphthols ligand in combination with Et2Zn and Ti(OiPr)4 can exhibit excellent enantioselectivity for phenylacetylene addition to both aromatic and aliphatic aldehydes. The catalytically active center of the repeating unit S-1 used as a catalyst produced the opposite configuration of the propargylic alcohols to that of S-1, on the contrary, the chiral polymer gave the same configuration as the optically active binaphthol moiety of the polybinaphthols ligand. Moreover, the chiral polymer ligand can be easily recovered and reused without loss of catalytic activity as well as enantioselectivity.

Abstract:Catalytic enantioselective conjugate additions with easily accessible alkenylboronic acid pinacol esters as nucleophiles promoted by chiral copper complexes of N-heterocyclic carbenes are presented. These processes constitute an unprecedented instance of conjugate additions of a variety of functionalized alkenyl groups and afford desired products that are otherwise difficult to access in up to 98% yield and 99.5:0.5 enantiomeric ratio. The origins of ligand-controlled enantioselectivity are elucidated with density functional theory (DFT) calculations.

Abstract:A soluble chiral polymer ligand was synthesized by the polymerization of (R)-6,6′-dibutyl-3,3′-diformyl-2,2′-binaphthol (R-M-1) with 2,5-diaminopyridine (M-2) via a nucleophilic addition–elimination reaction. While arylboronic acids were used as the source of the transferable aryl group, the chiral polybinaphthols ligand in combination with Et2Zn without Ti(OiPr)4 exhibited higher enantioselectivity in asymmetric addition to aromatic aldehydes than alphatic aldehydes. When aromatic aldehydes with electron-withdrawing groups were chosen as substrates, the resulting diarylmethanols were produced in higher ee values than those with electron-donating groups as substrates. 2-Naphthaldehyde used as a substrate afforded product in 95% ee, which could be ascribed to the steric effect influence on this asymmetric arylation reaction. Moreover, the chiral polymer was easily recovered and reused, but exhibited a decrease of enantioselectivity in the third recycle.
