Mechanism of AT2R-induced Inhibition of Neointimal Hyperplasia in Rat Carotid Arteries after Balloon Angioplasty

Abstract:Objective:To study the mechanism of angiotensin Ⅱ type 2 receptor (AT2R) -induced inhibition of neointimal hyperplasia in rat carotid arteries after balloon angioplasty. Methods: After establishment of rat carotid balloon injury restenosis model, pAdCMV/AT2R with AT2R gene or control virus pAd-GFP was transduced into rat carotid arteries. The rats were divided into normal, non-transduced,pAd-GFP and pAdCMV/ AT2R groups. The arteries were harvested at 7,14 and 21 days after gene transfer. The expressions of AT2R, AT1R, extracellular regulated kinase(ERK1/2) and basic transcription element binding protein 2 were evaluated with RT-PCR and Western Blot. Results:The expressions of AT1R,AT2R,ERK1/2 and basic transcription element binding protein 2 in the non-transduced and pAd-GFP group were stronger than those in the normal group (P 0. 01).In the pAdCMV/AT2R group, the level of AT2R were much higher but the levels of ERK1/2 and basic transcription element binding protein 2 were obviously lower than in the non-transduced or pAd-GFP group (both P 0. 01). There was no difference in the AT1R expression between non-transduced, pAd-GFP and pAdCMV/AT2R groups. Conclusion: Mechanism underlying AT2R-induced inhibition of neointimal hyperplasia may be associated with the inactivation of ERK1/2,which make less mitogen-activated protein kinase into cell nuclei and down-regulate the expression of basic transcription element binding protein 2.
