Abstract:Objective To investigate the effect of co application of nerve growth factor (NGF) and ciliary neurotrophic factor (CNTF) on the regeneration and functional recovery of transected sciatic nerve in rats. Methods A total of 120 Wistar rats were divided randomly into 4 groups. The sciatic nerve was transected and then bridge connected with silicone tube. The NGF+CNTF group received target muscle injection of NGF 100 ng/kg, and CNTF 100 ng/kg every day. CNTF group received 100 ng/kg, NGF group100 ng/kg. And NS group was given equal normal saline. The sciatic nerve functional index (SFI) measurement, electrophysiolgical assessment and morphometric analysis were performed for all the animals. Results The SFI, electrophysiologic parameters (nerve conduction velocity and amplitude of compound muscle action potentials), myelineted axons were significantly higher in NGF+CNTF group than in other groups. Conclusion Co application of NGF and CNTF can enhance peripheral nerve regeneration and functional recovery.
