Detection of WT1 Gene by Real-Time Quantitative PCR in Childhood Leukemia

Abstract:Objectives To set up the plasmid with WT1 gene as reference standard and establish the method of real-time quantitative polymerase chain reaction(RQ-PCR)for detecting WT1 gene in childhood acute leukemia. To investigate preliminarily the characteristics of WT1 gene expression in childhood acute leukemia and its clinical significance. Methods The sequence of primer and probe were designed based on previous publication. Plasmid with WT1 gene was set up as positive reference standard using molecular cloning technique. With fluorescent probe,WT1 gene and internal control gene(GAPDH)were detected by RQ-PCR. The plasmids of WT1 and GAPDH gene were diluted step by step to establish the standard curve. Using the method of bio-standard curve,the absolute quantitaty of the WT1 gene was obtained. Result of the detected WT1 gene was shown as the ratio to the copies of GAPDH gene(WT1/104·GAPDH),and analyzed by SPSS 13.0 software. Results The plasmid with a part of WT1 gene was successfully established and was confirmed by electrophoresis after restriction and ribonucleotide determination. It was used as the positive control of the RQ-PCR. The detection of WT1 gene by RQ-PCR was also established. Ct value and initiation template from WT1 gene and GAPDH were opposite in linear correlation(coefficient of determination,R2 0.99). The repeatability and sensitivity of the experiment was satisfactory. The result of the detected WT1 gene in the childhood acute leukemia showed that the level of new acute myelogenous leukemia(AML)was obviously higher than the control group(9.2331 ± 37.6228 vs. 0.0482 ± 0.0348),a lower level of the chronic myelogenous leukemia with BCR-ABL(0.1080 ± 0.0616),and the new acute lymphoblastic leukemia(0.0605 ± 1.6124)had no difference from the control group. In addition,patients achieving complete responses after chemotherapy(0.3150 ± 0.7216)showed a lower level of WT1 gene expression than those at diagnosis(P = 0.000). Conclusions The WT1 plasmid made in this experiment could be used as the positive control to detect the expression of the WT1 gene by RQ-PCR. The result suggested that the expression of WT1 gene may be an indicator to monitor minimal residual disease in childhood AML.
