Abstract:Aminocoumarin and 3_hydroxycoumarin were synthesized by an improved method and Lossen rearrangement(yields 51%～80%). The novel approach for the synthesis of 3_aminocoumarin is successful and convenient. Meanwhile,5 derivatives of 3_aminocoumarin were prepared,their structures were confirmed.

Abstract:A facile and efficient domino cyclization between 3-aminocoumarins and arylglyoxal monohydrates in the presence of para-toluenesulfonic acid (p-TSA) as a promoter was employed for the synthesis of two libraries of chromeno[3,4-b]pyrrol-4(3H)-one derivatives. Simple workup procedures were used to obtain the products in moderate to excellent yields. The solvents toluene and ethanol served important roles in the selective formation of the pyrrolocoumarin derivatives characterized by their different substitution patterns.

Abstract:The condensation reaction of 4-hydroxycoumarin with each of a~h substituted benzalacetones in(absolute) ethanol gave coumarin analogues which were subsequently converted to their ethyl ethers(cyclocou-(marin)).The ethers can be quantitatively converted back to the coumarin analogues by acid hydrolysis.Four of them are new substances.The reaction mechanism is discussed in detail.All the products were characterized by HPLC,IR and ~(1)H NMR.Compared with the present process in the literature,the process for the synthesis of the coumarin analogues is obviously improved on the reaction time,temperature,solvent,(catalyst),and post-treatment.

Abstract:In this paper,3-amino-coumarin was prepared via the Schiff base hydrolysis method.Three derivatives of coumarin were synthesized by acylating 3-amino-coumarin with methacryloyl chloride,chloroacetyl chloride,and 2-bromoisobutyl bromide,respectively.The structure of these derivatives was characterized by FT-IR,MS and 1HNMR.The synthesized derivatives have potential application in the fields of fluorescent polymers.

Abstract:A series of new azocoumarins have been synthesized in 44.1% similar to 81.9% yields. Their structures were established on the basis of IR, H-1 NMR, MS and elemental analyses. The crystal structure of 2a was determined. The double function of 2a, trans-cis isomerization. of the azo group, and the torsion angle between the benzene and the coumarin ring suggested that azocoumarins might be uesful as efficient molecular devices.

Abstract:A mild and convenient Palladium-Catalyzed addition for the synthesis of biologically attractive 3-aroyl coumarins employing 3-cyanocoumarins as substrates has been developed. This new method to synthesize 3-aroyl coumarins has merits such as using green and non-toxic medium, displaying broad substrate scopes, and achieving moderate to excellent yields. In particular, this protocol shows the significant chemoselectivity to synthesize the halogen (e. g.,bromo and iodo) substituted 3-aroyl coumarins for further synthetic elaborations.

Abstract:The efficient synthesis of a 3-benzylsulfinylcoumarin and a 3-benzylsulfonylcoumarin starting from thiobenzyl alcohol using solvent-free Knoevenagel reaction as the key step is reported. Moreover, the molecular structure of 3-benzylsulfonyl-7-diethylaminocoumarin is confirmed by X-ray crystal analysis.

Abstract:A highly efficient and convenient procedure to access a series of coumarin-containing phthalides and derivatives is developed. The novel bis-heterocyclic products are obtained via reactions between 4-hydroxycoumarins (or 4-hydroxy-2-quinolones) with different 2-formylbenzoic acids in water as an environmentally friendly solvent. 4-Aryl substituted derivatives of 3-(4-hydroxy)coumarinylphthalide are obtained in excellent yields by initial conversion into the corresponding triflate and subsequent palladium-catalyzed Suzuki–Miyaura coupling with arylboronic acids.

Abstract:Coumarin-7,8-cyclophosphoramide derivatives were conveniently prepared by a facile method. 7-Hydroxycoumarin was aminomethylated regioselectively at the 8-C position using the Mannich reaction. The newly formed 8-(N-alkylaminomethyl) coumarins were then coupled with bis--chloroethyl dichlorophosphamide to form coumarin-7,8-cyclophosphoramide analogs. An efficient, highly regioselective method to synthesize coumarin-7,8-cyclophosphoramide derivatives is provided, and the approach has the merits of mild reaction conditions.

Abstract:8(or 6)-Chloro-7-hydroxy-4-methylcoumarin(1a or 1b) was prepared by Pechmann condensation between resorcinol chloride and ethyl acetoacetate. 1 reacted with dimethyl fulfate and produced 2a or 2b. Six new 7-alkylox-8(or 6)-chloro-4-methyl coumarins(3a~6a,3b and 4b) in yields of 66%~92% were synthesized via Williamson reaction of 1 and alkyl halides using tetrabutylammonium bromide as a phase transfer catalyst. The structures of 1～6 were characterized by 1H NMR,IR and GC-MS.

Abstract:4-Aryl-3,4-dihydrocoumarins(neoflavones) represent a minor class of natural compounds which based on the C6—C3—C6 skeleton with an characteristic aryl group in the 4 position.Structurally,4-aryl-3,4-dihydrocoumarins can also be regarded as the hybrids of isoflavones and coumarins,thus being expected to possess corresponding bioactivity.In this paper,ten 4-aryl-3,4-dihydrocoumarins were synthesized by two steps.The first condensation of substituted benzaldehydes and malonic acids in the presence of pyridine and piperidine at 70-90 ℃ gave the expected cinnamic acids in good yields.Then condensation of substituted cinnamic acids with appropriate phenols in the presence of BF3-Et2O and POCl3 gave the target molecule at room temperature.Their structures were confirmed by EI-MS,IR and 1H-NMR.The antioxidant and antitumor activity of these compounds were evaluated by DPPH and MTT assay,respectively.Some of 4-aryl-3,4-dihydrocoumarin derivatives showed strong scavenging activities against 1,1-diphenyl-2-picrylhydrazyl radical:(±)-7,8-dihydroxy-4-(3-hydroxy-4-methoxyphenyl)-3,4-dihydrocoumarin(3c) EC50=(2.69±0.09) μmol · L-1,(±)-7,8-dihydroxy-4-(4-methoxyphenyl)-3,4-dihydrocoumarin(3g) EC50=(2.32±0.10) μmol · L-1 and weak antitumor in vitro activity against BGC-823 cell line[(±)-7,8-dihydroxy-4-(3-hydroxy-4-methoxyphenyl)-3,4-dihydrocoumarin(3c) IC50=(136±21) μmol · L-1].The results showed that the 4-arylcoumarins which possessing the catechol moiety may be a beneficial scaffold for therapeutic purpose.

Abstract:A facile silver-catalyzed oxidative decarboxylation of arylthiodifluoroacetic acids or aryloxydifluoroacetic acids with coumarins/quinoxalin-2(1H)-ones was developed. This transformation provided a series of C-3 aryloxydifluoromethylated or arylthiodifluoromethylated coumarins/quinoxalin-2(1H)-ones containing various functional groups in moderate to good yields, featuring good functional group tolerance, easily accessible starting materials and operational simplicity.

Abstract:Nine new 7-alkoxy-4-ethoxycoumarins were synthesized from resorcinol and cyanoacetic by Pechmann reaction,hydrolyzation,ethyl etherization,and Williamson reaction.Optimum conditions for Williamson reaction were presented.In DMF(dimethylformamide),when n(4-ethoxy-7-hydroxycoumarin)∶n(alkylhalides)∶n(anhydrous potassium carbonate)∶n(TBAB,tetrabutyl ammonium bromide)=1∶1.5∶2.5∶0.1 and they were reacting at 60 ℃ to 80 ℃ for 11 h to 25 h,the yields were between 72%-95%.The structure of the intermediates and the target compounds were confirmed by IR,MS and 1H NMR.
