Abstract:Flocculent is frequently used in oily wastewater treatment in domestic oilfields.In order to obtain high efficient and cheap flocculant,red mud is used to optimize the preparation of polyaluminum ferric chloride.Chemical analysis and X-ray diffraction analysis help to draw the aluminum and iron content in the red mud.Orthogonal experiment result shows that the factors affecting Fe dissolution from strong to weak are: the coefficient of amount of hydrochloric acid,the reaction temperature,the mesh number and the reaction time;and the optimum leaching conditions are: mesh number of 80-120,liquid-solid ratio of 5.5,reaction temperature of 95℃,reaction time of 3.5 h.

Abstract:Objective To study the best pre-treatment method and its technology before ultrafiltration of Reduning water extract.Methods To investigate such pre-treatment methods as chitosan flocculation on the impurity of prescription water extract,as well as polysulfone ultrafiltration membrane refination effect with molecular weight cut-off three to five million.Results The centrifugation and decompress filtration were not benefit to promote the geniposide reservation and reduce fouling degree of membrane.Absorption of active carbon could make the geniposide reservation deeply lower.The geniposide was reserved mostly and the total extracts reduced mostly for chitosan flocculation and microfiltration.But the flux was higher and the membrane fouling is smaller after chitosan flocculation.Conclusion Chitosan flocculation is the best pre-treatment method.The operating conditions are that temperature of Reduning water extract is 70 ℃,pH value 6,and the dosage of 1% chitosan is 70 mL/L.

Abstract:The purpose of this work was to develop a process to obtain Stevia extract of a better quality and quantity under influence of ultrasound technique. The chemical compositions, anti-nutritional factor and heavy metals of raw material were tested firstly in (ww−1) in our previous work. The extraction process was assisted by ultrasonication at power 360W for 12min using three different types of solvents (water, ethanol and isopropyl alcohol) in different concentrations, that is, 10, 20, 30, 40, 50, and 60% (vv−1) to optimize the extraction process. Stevia leaves extractions, analyzed by HPLC, indicated that isopropyl alcohol (60% vv−1) gave the highest rebaudioside A yield (35g100g−1). This optimum concentration was used in the next set of experiments to optimize ultrasonic power and time. Optimum applied power and sonication time was found 18min and 480W, respectively. The extraction yield obtained under optimum process conditions for water, ethanol and isopropyl alcohol were 32.79, 33.85 and 37.10 (g100g−1), respectively. Compared to classical methods like maceration and heat extraction, the utilization of ultrasound-assisted extraction proved to be a much simpler and efficient way to obtain rebaudioside A from Stevia rebaudiana leaves.

Abstract:Polysaccharide is the main component of the flocculation activated substances produced by bioflocculant-producing bacterium F6. It could be extracted from the supernatant of the fermented medium by cold acetone precipitation and purified by treating with Sevag reagent. The result showed that the optimum extracting conditions were that concentrating multiple is 1/3, acetone consistency is 100%, acetone quantity is 1:2, precipitating time is 2 h, and pH is 7; the optimum purifying conditions were that Vsample:Vsevag is 2:3, Vchloroform:Vbutanol is 4:1, purifying time is 30 min. After the preliminary extraction and purification, flocculating activity reached 98.89%.

Abstract:The strain F2 which produce excellent flocculating substance were isolated from soil samples and identified as Rhizobium radiobacter. The isolates F2 produced the flocculating substance during the late logarithmic growth phase and the early stationary growth phase. It was founded that the flocculation activated substances of BF distributed mainly in upper phase and were mainly composed of polysaccharide (83.0%) and protein (17.0%). The flocculation activated substances could be recovered from the supernatant of the fermented medium by cold acetone precipitation and purified by treating with Sevag reagent. It suggested that the optimum extracting conditions were that concentrating multiple is 1/4, acetone consistency is 100%, acetone quantity is 1:2, precipitating time is 2 h, and pH is 7, the rate of extraction for the polysaccharides was 30.2%; the optimum purifying conditions were that Vsample:Vsevag is 2:3, Vchloroform:Vbutanol is 5:2, purifying time is 30 min, the rate of eliminating for the protein was 74.2%. After the preliminary extraction and purification, BF remained high flocculating activity.

Abstract:The adsorption and desorption behaviour of macroporous adsorbent AB-8 for stevioside were investigated. The effects of pH, eluents and flow rate on the adsorption and desorption properties were studied. The results show that the adsorbent Ab-8 has a high adsorption capacity for stevioside and the elution is easy.
Study On Preliminary Extraction And Purification Of Compound Bioflocculant

Abstract:Two strains (designated F2, F6) which produce excellent flocculating substance was isolated from soil samples and identified as Rhizobium radiobacter and Bacillus sphaeicus. The compound bioflocculant (CBF) with higher flocculating activity was obtained by co culturing F2 and F6. The isolates F2 and F6 produced the flocculating substance during the late logarithmic growth phase and the early stationary growth phase. The flocculation activated substances of CBF distributed mainly in upper phase and was mainly composed of polysaccharide (90.6%) and protein (9.3%). The compound bioflocculant is thermo-stable and 75% of its activity remains after heating at 100 degrees C for 30 min. The flocculation efficiency of this bioflocculant increased with the pH within a range of pH 7-10 and was 86%-94%. The flocculation activated substances could be recovered from the supernatant of the fermented medium by cold acetone precipitation and purified by treating with Savage reagent. The experimental result showed that the optimum extracting conditions were that concentrating multiple is 1/2, acetone. consistency is 80%, acetone quantity is 1:1, precipitating time is 24h, and pH is 7, the rate of extraction for the polysaccharides was 55.5%; the optimum purifying conditions were that Vsample:Vsevag is 2:1, Vchloroform:Vbutanol is 5:2, purifying time is 90 min, the rate of eliminating for the protein was 92%. After the preliminary extraction and purification, CBF remained high flocculating activity.

Abstract:A new method for purification of steviol glycosides from Stevia Rebaudiana crude extracts by macroporous adsorption resin(MAR) was systematically investigated.The purification procedure consisted of two steps.In the first step,the main purpose was decolorization,and several types of macroporous resins including D293,D296,JN-1,JN-2,D890 and D201 were used in order to choose the best resin.JN-1 resin was selected according to their decolorization ratio,the retention of steviol glycosides ratio and desorption ratio.In order to increase the purity,a second step of refining was added by screening various types of macroporous resins(AB-8,JNF-1,X-5,NKA-II and D101).The results demonstrated that JNF-1 resin was the most suitable resin.The purity of steviol glycosides after the first step JN-1 was from 42.65% to 67.33% with a recovery of 84.87%.The purity of steviol glycosides increased to 92.92% with a recovery of 91.08% after the second step JNF-1.

Abstract:SummaryTo improve the shortness of the current purification technology of steviol glycosides (SG) extract, a two‐step purification procedure consisting of chitosan precipitation and reversed‐phase chromatography (RPC) was investigated. The impurity‐to‐SG ratio in the SG extract obtained from non‐destructive soaking was 5:1 ~ 6:1. It was found that 80.90% of the impurities could be removed by RPC using 20% (v/v) ethanol solution as the eluent with a high SG recovery of 96.98%. When 25% ethanol was applied, the SG recovery was reduced to 71.23%. An adsorption test using D280 anionic resin suggested that the impurities were negatively charged; thus, chitosan was used to co‐precipitate the impurities. An optimal decolourisation result of ~ 70% was obtained at 0.06 g·g−1 (w/w, to solids in extract) chitosan addition, shaking at 28 °C for 1 h. Afterwards, the extract was further purified by RPC. The total removal rate of impurities was increased to 91.20%, and the SG recovery was 85.15%.

Abstract:In order to realize the separation and purification of 2, 3-butaneidol from bioconversion, the flocculation of 2, 3-butanediol fermented broth with 10 flocculants was studied. With the analysis of flocculation rate and protein removing rate, the influence of different conditions on the flocculation effects, concentration of 2, 3-butanediol and subsequent extraction process was studied systematically. The suitable flocculation conditions were obtained as follows: FeCl 3 as flocculant, the dosage 23 g/L, pH=5.1, temperature 20-50°C, stirring for 15 min and settling for 20 min. Under those conditions, both flocculation rate and protein removing rate can be above 98%, which is helpful for the succeeding separation and purification process.
Screening of Bioflocculant-Producing Bacteria and Its Application in Clarification Process of Sugarcane Juice

Abstract:Two strains (N-25 and Z-24) of high bioflocculant-producing bacteria were isolated from activated sludge samples. The flocculation activities of bioflocculants from N-25 and Z-24 for kaolin suspension (5 g/L) were both more than 75%. Based on the biochemical properties and 16S rDNA sequence analysis, strains N-25 and Z-24 were identified as Proteus vulgaris and Klebsiella pneumoniae subsp. pneumoniae, respectively. The bioflocculants from N-25 and Z-24 were named BN-25 and BZ-24, and then applied in the clarification process of sugarcane juice. The optimal clarification conditions of bioflocculants BN-25 and BZ-24 were compared with polyacrylamide (PAM). Applications of bioflocculants BN-25 and BZ-24 were superior in the sedimentation time, the bulk of mud, the decoloration rate, while weaker in the turbidity removal rate. The bioflocculants BN-25 and BZ-24 were compounded with PAM and the expected clarification efficiency was achieved by significantly reducing the dosage of PAM. The results demonstrated that bioflocculants BN-25 and BZ-24 exhibited potential for application in the sugar industry.

Abstract:Polysaccharides have been widely used in many fields. In the field of food water treatment, polysaccharide-based flocculants have attracted extensive attention. Based on the enzymatic preparation of high molecular weight dextran, two safe, sustainable and eco-friendly dextran based high molecular flocculants were developed by introducing flocculating active groups into the molecular chain of dextran through cross-linking (DEPL) and esterification (Dex-PL). The prepared product was characterized by elemental analysis, Fourier transform infrared spectroscopy, thermogravimetric analysis and field emission scanning electron microscopy (FESEM). Because amino group is the main active group of flocculation, more amino groups are retained in esterification reaction. In the flocculation experiment of kaolin suspension, the efficiency of the two flocculants is very excellent, both of which are greater than 96%. The results of the study on the flocculation mechanism showed that charge neutralization, adsorption bridging and sweeping flocculation mechanisms occurred in the flocculation process. Dex-PL was applied to the clarification of sugarcane juice in sugar industry. At the dosage of 118 mg/L, the clarification efficiency reached 91.4%. The results of biodegradation experiment showed that the product can be biodegraded by enzymatic hydrolysis. This study provides experience for the development and application of new polysaccharide based flocculants, and provides safe and effective flocculants for sugar refining.

Abstract:Natural pectin, derived from fruit residue, presented a certain flocculation performance in previous studies. However, the process of extraction and treatment affected the flocculation performance considering the uncontrollable chemical composition and the molecule structure. In this study, degree of esterification (DE), degree of amidation (DA) were used as the internal factors affecting flocculation performance. The DE/DA values of pectin were obtained through FTIR, elemental analyses, H-NMR, and titration measurements. The kaolin suspension was employed for the coagulation jar tests, and the removal of NTU (Nephelometric Turbidity Unit) was used as the index of the flocculation performance. Results showed that the flocculation performance of pectin arising from different fruits was different, which was associated with the various preparation processes. By introducing polar groups into pectin, the flocculation was found to be related to bridging, adsorption, and charge neutralization. Based on the trends of three-dimensional response surfaces, the flocculation effect was improved with the decreased DE and the increased DA. The optimized amidated pectin was obtained by the amidation experiment, the turbidity reduction in wastewater was 99.63%. PRACTITIONER POINTS: The internal influencing factors of pectin as a flocculant were investigated. Different sources of pectin show different flocculation ability. Amidation modification can improve the flocculation performance of pectin. Response surface method to study the interaction of different influencing factors. Pectin may replace synthetic flocculants in water treatment.
Study on Purification Process of Yanyan Buccal Tablets

Abstract:Objective:To compare the fit and unfit of alcohol precipitation process and chitosan flocculation process used in purification of extract of yanyan buccal tablets.Methods:For alcohol precipitation process used in purification of extract of yanyan buccal tablets,to select optimal density of decoction and concentration after addition of alcohol by determination of extract yield and content of emodin;For chitosan flocculation process,to select optimal usage amount of chitosan with extract yield and content of emodin used for index,and we compare the two process.Results:Comparing alcohol precipitation process with the chitosan flocculation process,the retention rate of emodin is 97.16%,81.57%,and extract yield is 11.91%,19.21%.Conclusion:Alcohol precipitation process can remove foreign matter and reserve valid ingredient as far as possible,chitosan flocculation process can remove a trifle of foreign matter with loss of valid ingredient.Alcohol precipitation process is better than chitosan flocculation process used in purification of extract of yanyan buccal tablets.

Abstract:The extraction and purification of natural products with less content is a challenge.In order to solve the problem,such as pipeline jam by crystal products,30% andrographolide was selected to purify by supercritical extraction and crystallization.The result showed that there was synchronous gradient crystallization of andrographolide in supercritical CO 2 extraction,moreover,the higher the pressure,the smaller and shorter the crystal shape.The content of andrographolide was raised from 30% to 82.3% under 20 MPa,at 55 ℃,within 90 min,ata flux rate 15 L/min.The supercritical fluid extraction and crystallization is a new technique for separation of natural products in high purity.

Abstract:We isolated flocculants-producing bacteria F2 from soil. It shows high and stable flocculating activity for Kaolin clay suspension. In order to increase yield of flocculants, we need determine the optimal obtained time. We measured the changes of several parameters using shaking flask experiment, including flocculating- rate, the content of polysaccharide, protein and glucose. It's showed that the optimal obtained time of bio-flocculants is 21 h. And we built the model about the production of bio-flocculants. Through the comparison of experimental data and the corresponding calculated values from the models, we found that the data joint well and the model can provide theoretical basis for Industrialization of bio-flocculants.
