Abstract:Programmed death-1 (PD-1, CD279), an inhibitory co-stimulatory molecule of the CD28 superfamily, plays a critical role in immune response. In this report, four novel mouse anti-human PD-1 monoclonal antibodies (MAbs) were prepared using hybridoma technology and immunological characteristics of the MAbs were determined. The results showed that all the MAbs (clones 9H1, 4B9, 8F5, and 1F8) were IgG1(kappa) and bound specifically to human PD-1. By mutual competition, we found that the antibodies recognized three different epitopes of PD-1 antigen and 9H1 MAb could block both PD-1/PD-L1 and PD-1/PD-L2 interaction. Cross-linking of PD-1 with MAb 9H1 markedly blocked PD-1 negative signal and promoted T cell proliferation. In addition, 4B9 and 9H1 MAbs were suitable for indirect ELISA detection. Thus, the MAbs against human PD-1 with high specificity and different activity would be useful for the further study of this molecule.

Abstract:Three hybridoma cell lines(1C1,2C11 and 2G7) secreting monoclonal antibodies(McAbs) against penicillin were produced by fusing mouse myeloma cells(SP2/0) with spleen cells from BALB/c immunized by ampicillin conjugate with cytochrome C.The three McAbs could specifically react with ampicillin.The titres of ascitic fluids of the three McAbs are up to 10 -6 in indirect ELISA.Isotypes and subclasses of 1C1 and 2C11 belong to IgG1 while those of 2G7 belong to IgM.The detection limits for ampicillin,calculated from the antibiotic concentration giving 50% binding inhibition,were 45.3(McAb 1C1),60.1(McAb 2C11) and 80.0 μg/L(McAb 2G71).The competitive inhibition enzyme-linked immunosorbent assay(CI-ELISA) analysis showed that the three McAbs can specifically react with ampicillin and its analogue, while no cross-reactivity was found with other antibiotics,indicating that the three McAbs can be used in detection of residues of the penicillin and its analogue in the immunoassay.

Abstract:Through immunization of BALB/c mice with synthesized CAP-ovalbumin(CAP-OVA),a hybridoma cell line,the 4C9 which could produce monoclonal antibody against chloramphenicol(CAP) and propagate stably,was established.With the myeloma cells,the ascites containing monclonal antibody(McAb) were prepared.The indirect competitive ELISA(ciELISA) test revealed the heavy chain and light chain of McAb were IgG1 and κ,respectively.The ELISA titer of ascites was 1∶1×106.The percentages of cross-activity to thiamphenicol,florfenicol and other antibiotics were all less than 0.01%.A horseradish peroxidase(HRP)-labeled antibody was synthesized by the sodium periodate reaction,then a direct competitive enzyme-linked immunosorbent assay(cdELISA) was developed.Quantization of the CAP was linear from 0.1 to 100 ng·mL-1,and the hemi-inhibitory concentration(IC50) was 5.81 ng·L-1.The recovery test showed that the detection limit for CAP was 0.1 ng·L-1 in cdELISA.Comparative test showed that detected sensibility of the monoclonal antibody was nearly equivalent to the commercial CAP ELISA kit.

Abstract:One hybridoma cell line (1G10) secreting monoclonal antibody (McAb) against chlorpyrifos was produced by fusing mouse myeloma cells (SP2/0) with spleen cells from BALB/C which were immunized by the artificial antigen conjugated with bovine serum albumin (BSA). Isotype and subclass of the monoclonal cell (1G10) were classified as IgGl. The light chain of the McAb was identified to be κ. Its titres of ascitic fluids were up to 1 × 10 -6 by indirect ELISA. The inhibition rate of 50% of the McAb with the insecticide chlorpyrifos detected by ELISA was 80.8 μg·L -1, the McAb could specifically react with chlorpyrifos but no cross-reactivity to chlorpyrifos analogues.

Abstract:To prepare a functional mouse anti-human PD-1 monoclonal antibody and to characterize its biological activities,a stable human PD-1 transfected cell line L929/PD-1 was used as an antigen to immunize BALB/c mice.By means of the cell fusion technique,multiple cell subcloning,repeated screening with L929/PD-1 as target cells and the L929/mock cells used as the negative control,the hybridoma specifically secreting mouse anti-human PD-1 monoclonal antibody was generated.Then its biological characterization was investigated by Western blotting,rapid murine Ig-subclass typing,indirect immunofluorescene assay,mutual competitive inhibition test and MTT incorporation assay.The results of characterization study showed that the monoclonal antibody(named as 6E2) could bind to human PD-1 specially,and it could recognize a new epitope of PD-1.Furthermore,the mAb 6E2 could suppress T cells proliferation and cytokine secretion.It is evident that the functional monoclonal antibody for human PD-1 has been generated,and it would provide the initial material for further study on the role of human PD-1.

Abstract:Objective To establish a hybridoma cell line secreting monoclonal antibody (McAb) against human PSCA. Methods BALB/c mouse was immunized by purified PSCA,A murine McAb designated D3 was generated and the specificity was tested by immunohistochemistry and Western bloting. Results A positive hybridome cell lines which contained McAb against PSCA were obtained.It belong to IgG1 according to the examination of immunoglobulin classification. Western-blot test showed that the McAb reacted specifically with PSCA without other crossing reactions.The antibody detected PSCA expression on the cell surface of normal and malignant prostate cells,PSCA protein expression was identified in 38/40(95%)primary prostate cancers.There was no statistical difference in PSCA expression level between normal prostate tissues and BPH,but the PSCA expression level of prostate cancer was of significant statistical difference. Conclusions It was showed the antibody could be used in diagnosis and immunotherapy of prostate cancer.

Abstract:OBJECTIVE:To obtain monoclonal antibodies against programmed cell death 10 (PDCD10) for further study of the structure and function of PDCD10 protein.METHODS:Balb/c mice were immunized with recombinant PDCD10, hybridoma cell lines secreting monoclonal antibodies against PDCD10 were screened by regular cell fusion and subcloning approach. The specificities of these monoclonal antibodies were determined by ELISA, Western blotting and Immunofluorescence assay.RESULTS:Three hybridoma cell lines (5G1, 4F7 and 3H5) stable in secreting specific monoclonal antibodies were successfully obtained. Subclass of IgG belonged to IgG1 (4F7 and 5G1) and IgG2b (3H5), respectively. The ascite titers of these monoclonal antibodies reached 1:10(7). They could specifically bind to recombinant PDCD10 and endogenous and overexpressed PDCD10 proteins proved by ELISA and Western blotting. They failed to react with E.coli lysates and glutathione S-transferase (GST). In addition, these three monoclonal antibodies could recognize different epitopes of PDCD10 proteins assessed by immune fluorescence competitive binding assay. Both endogenous and overexpressed PDCD10 protein mainly located in the nucleus.CONCLUSION:Monoclonal antibodies against PDCD10 with high titers and specificity have been successfully prepared, which has laid the foundation for further study of PDCD10 protein.

Abstract:To prepare specific antibodies against P1 protein of Mycoplasmas pneumoniae(Mp)for clinical diagnosis and correlative research. Based on the purification of protein, the antigens were emulsified with FCA or FIA and injected to BALB/c mice to obtain polyclonal antiserum against P1 protein. The McAbs obtained from the traditional hybridization technique were selected with recombinant P1 ELISA, and the specificity and sensitivity of the PcAb and McAbs were identified by ELISA and IFA. Result indicated the sensitivity of recombinant protein detected by ELISA was more than 1ng/ml. The titer of P1 antibody (in mouse serum) identified by Mp-ELISA reached 1∶1 600.And we obtained two groups of specific and stable McAbs against P1 protein of M. pneumoniae. ELISA analysis showed two McAbs had high affinity and specificity with recombinant P1 protein(5μg/ml),the titer of IgG reached 1∶800-1∶1 600. The PcAb and McAbs showed better affinity and specificity with Mp in IFA. In conclusion, the recombinant express protein show high immunogenicity and immunoreactivity. The anti-Mp P1 PcAb and McAbs has high specificity with Mp. The recombinant P1protein and the McAbs obtained from this study might be bases for the early clinical detection of Mp infection.

Abstract:The complete antigens were prepared by the mixed anhydride coupling method in which Neurotoxic Shellfish Poisoning Toxin PbTx-2 was conjugated with BSA and OVA respectively.A hybridoma cell line steadily secreting monoclonal antibodies against PbTx-2,coded 4B5,was obtained by fusing immunized SP2/0 cells with spleen cells from 8 weeks old BALB/c mice immunized with PbTx-BSA conjugate and subcloning for 3 to 4 cycles.The titer of the McAb in the ascites analyzed by indirect ELISA was 1:5.12×104,the antibody subclass was IgM,affinity constant was 2.32×107mol/L.

Abstract:Abstract Butyl benzyl phthalate (BBP) is widely used as a plasticizer in carrier of pesticide, food packing and many daily products. It could be hydrolysed to mono-butyl phthalate (MBP) inside the body of organisms, which is highly toxic to humans and other organisms, affecting the reproductive and developmental function. Immune spleen cells were collected from mice immunised intraperitoneally (i.p.) and multiple subcutaneously (s.c.). A hybridoma cell line named 2F7 was obtained via colony hybridisation of immune spleen cells and myeloma cells SP2/0. The mAb against MBP secreted by 2F7 was an isotype of IgG1, and showed 1.93×108 of an affinity and the CR values of the mAb with other analogues of MBP were not more than 5%, except for BBP (11.5%). The high affinity and IgG characteristics of the mAb against MBP could be used for MBP detection in the future.

Abstract:Objective To establish HSPC238 hybridoma cell lines and to characterize the activities of these monoclonal antibodies (McAbs). Method BALB/c mice were immunized with recombinant HSPC238 protein. Hybridoma cell lines were established by using hybridoma technique and monoclonal antibodies were prepared. Specificity of the antibodies was evaluated by Western-blot and enzyme linked immunosorbent assay (ELISA). Result Two stable hybridoma cell lines, E001 and E002, secreting IgG1 antibody against HSPC238 McAbs were obtained. Both E001 and E002 are HSPC238 specific. Using the ELISA method, the titer of the ascite fluid obtained from E001 and E002 was 1∶12800 and 1∶25600, respectively. Western blotting analysis confirmed that the two monoclonal antibodies detect human HSPC238. Conclusion High titer HSPC238 specific monoclonal antibodies were successfully prepared. The antibodies may be used in the future study of the HSPC238 protein.

Abstract:OBJECTIVE:Generation and Identification of Phage Engineering Antibodies Library against Hamster Prion Protein.METHODS:Fab antibodies were identified and confirmed. BALB/c mice were immuned with PrP proteins. After the third immunization, the total RNA was extracted from the mice spleens. The genes of heavy Fd Fragments and light chain of antibodies amplified by a series of specific primers of human IgG Fab fragment were cloned into phagemid vector pComb3.RESULTS:The combinatorial Fab library were constructed successfully and the cloning efficiencies both of light chain and Fd fragments were about near 10(6). The Fab library were panned by four cycles and screened with purified haPrP23-231 antigen on microtiter plates. 12 mAbs were isolated after four cycles of panning, five of which were sequenced and resulted sequence data were analyzed by alignment with GenBank immunoglobulin genes. Two strain of new heavy and light chain genes of Fab antibodies were identified and confirmed.CONCLUSION:The research in this article will provide foundation for study of diagnosis and therapy of prion.

Abstract:Purpose To prepare monoclonal antibodies specifically against extracellular peptide fragment of prostate specific membrane antigen (PSMA). Methods The extracellular fragment of PSMA protein expressed by a prokaryotic expressing system was used to immunize female Balb/c mice, splenocytes from immunized Balb/c mice were fused with SP2/0 cells. The hybridoma cells were primarily screened by enzyme-linked immunoabsorbent assay (ELISA) ,and the positive clones were further identified by immunocy-tochemistry, immnohistochemistry techniques. Results Fifty-five clones were primarily selected as positive by ELISA, and finally eight clones were identified as positive by immnohistochemistry and can be used in the slides of formalin-fixed,paraffin-embedded tissue of prostate carcinoma. Conclusions Eight hybridoma cell lines secreting monoclonal antibodies specifically against the extracellular peptide fragment of PSMA were prepared, which may be used in the diagnosis of prostate carcinoma,and as a tool for the immnotherapy of prostate carcinoma.

Abstract:To construct the gene sequence of a recombinant antibody,the light and heavy gene sequences of recombinant antibodies against coplanar polychlorinated biphenyls (Co-PCBs) were indexed and connected with a flexible linker peptide (Gly4Ser)3,then cloned and expressed in Escherichia coli M15. After purifying and renaturation,the purity of the recombinant antibody against Co-PCBs was more than 95%. Finally the antibody was assembled on a gold surface to prepare a protein chip for detecting PCB126 basing on SPR technology. The detection limit was about 5×10-4 pg·L-1,which meets the need of ultra trace detection of Co-PCBs in the environment. This detection method was fast and convenient,and less sample is needed because of its high-sensitivity. The work provides a good foundation for the real-time detection of PCBs in the environment.

Abstract:BACKGROUND AIM:For clinical diagnosis of malignant tumors and research the function of PAK4 protein,we prepared the McAbs of PAK4 protein.MATERIALS AND METHODS:The Balb/C mice were immunized with the recombinant plasmid of PAK4(the vector is P3XFLAG-CMV-10).By cell fusion and cell cloning,the McAbs of PAK4 were prepared.RESULTS:Two strains of hybridoma cells,producing anti-PAK4 antibody,named 2D1(PAK4)a subclass of IgG and 3H4(PAK4)a subclass of IgM,were obtained after fusion followed by three or four screening.The ascites of McAbs were prepared by injecting the hybridoma cells into the mice abdomen.Indirect ELISA results showed that the ascites of McAbs revealed high affinity with the PAK4 protein at the titer of 1∶ 1.0× 104.Immunohistochemistry showed that the McAbs of PAK4 reacted specifically with the cells of breast cancer.CONCLUSION:These results demonstrated that the McAbs of PAK4 could be a specific marker for diagnosis of cancer.
Specific Antibody for Pesticide Residue Determination Produced by Antibody-Pesticide Complex.

Abstract:A new method for specific antibody production was developed using antibody (Ab)-pesticide complex as a unique immunogen. Parathion (PA) was the targeted pesticide, and rabbit polyclonal antibody (Pab) and mouse monoclonal antibody (Mab) were used as carrier proteins. The Ab-PA complexes were generated by conjugating the two antibodies with an excessive dosage of PA. It was shown that the sensitivity of homologous enzyme-linked immunosorbent assay (ELISA) using the new antibodies was similar to that using original antibodies. However, the new mouse Pab had not only similar positive recognizing spectrum as the original Mab, but also a significantly improved sensitivity in heterologous ELISA when some recognizable competitors were applied. IC(50) value of ELISA based on a combination of the new mouse Pab and hapten 9 was 0.24 ng/mL, which was 445.54 times as that of the homologous ELISA. An Ab-pesticide complex may be a suitable alternative immunogen for producing highly specific antibody to improve the immunoassay sensitivity.

Abstract:Based on the structure of 4-methylphenol(4-MP) and the design principle of hapten,3-(4-Hydroxyphenyl)propionic acid(34-HPA) was selected as the hapten of 4-MP.34-HPA was coupled with bovine serum albumin(BSA) to prepare complete antigen 34-HPA-BSA and immunized 3~4 months old rabbits.The titer and identification of the polyclonal antibodies against 4-MP were characterized with indirect enzyme-linked immunosorbent assay(iELISA) which reached 1∶204 800.Indirect competitive ELISA(icELISA) was developed using 34-HPA coupled with ovalbmin(OVA) as the original coating.The results showed the best detection conditions for icELISA:the coating antigen 34-HPA-OVA concentration was 1.05 μg/mL,the antibodies 12 800 times diluted,0.1% gelatin solution blocking 1.5 h,competitive time was 1.0 h,horse radish peroxidase(HRP) labeled goat anti-rabbit IgG diluted 2 000 times and incubated 1.0 h.The linear range of icELISA to detect 4-MP was δ1.0×10-2~1.0×103 and the regression equation is y=-12.76x+66.66.The detection limit was δ0.02,the intra-assay and inter-assay coefficient of standard variation is 7.0% and 8.4%.

Abstract:Objectives:To generate specific monoclonal and polyclonal antibodies against human perforin. Methods: Balb/c mice and rabbits were immunized with recombinant human perforin C(PFP C), which is a truncated 125aa peptide in C terminal of human PFP. The spleen cells from immunized mice were fused with the HAT sensitive mouse myeloma cell line Sp2/0. Resulting hybridomas were screened for anti PFP C antibodies by an ELISA procedure. The monoclonal anti PFP C antibodies were identified by Western blot and immunohistochemistry. Results:Four hybridoma clones that secrete anti PFP C constantly were obtained, the acetic antibody titers in ELISA were 1∶3.2×10 3~1∶6.4×10 6. The titer of rabbit serum anti PFP C was 1∶1.2×10 4. Conclusions: Monoclonal and polyclonal antibodies with high titer and sensitivity were produced using recombinant human PFP C peptide.

Abstract:AIM:To prepare monoclonal antibodies (mAb) against recombinant human nucleoside diphosphate kinase-A(NDPK-A) and characterize their properties.METHODS:BALB/c mice were immunized with rhNDPK-A, and mAb was prepared by hybridoma technique. Ig subclass and specificity was analysed by double immunodiffusion and western blot respectively. The titres of mAbs in ascitic fluid, relative affinity and epitopes recognized by mAbs were determined by indirect ELISA.RESULTS:6 hybridoma cell lines secreting anti-rhNDPK-A mAbs were obtained. Their Ig subclass belonged to IgG1. The titers of 6 mAbs in ascitic fluid were 1x10(-4)-5x10(-6). Relative affinity of mAbs were 4.5x10(-9)-2.8x10(-10) mol/L. They recognized 3 different epitopes on rhNDPK-A molecule.CONCLUSION:6 mAbs against rhNDPK-A have been prepared successfully which provide useful reagent for clinical diagnosis and further research.

Abstract:To generate and characterize a monoclonal antibody (mAb) against humanin (HN), BALB/c mice were immunized with the purified pet-44a-HN in adjuvant and their splenic lymphocytes were fused with myeloma SP2/0 cells. The hybridoma cell lines were screened for HN-specific antibodies by indirect enzyme-linked immunosorbent assay (ELISA), and anti-HN mAb-producing hybridoma clones were obtained using a limiting dilution assay. The specificity and affinity of the antibodies were characterized by western blot assays and indirect ELISA. Following fusion, screening and cloning, four hybridoma clones were obtained, and the clone 5A8H3 was demonstrated to stably produce anti-HN IgG2a. Further characterization of 5A8H3 revealed that this mAb specifically recognized HN, the fusion proteins of pet-44a-HN protein and pGEMEX-1-HN, but not control (Escherichia coli proteins). This mAb interacted with HN at an affinity constant (Ka) of 2.0 x 10(8)M(-1). The HN-specific IgG2a mAb was successfully generated. It interacted with HN specifically and sensitively, providing a valuable tool for further study of the biological functions of HN.
