Uniform Chitosan Microparticles Prepared by a Novel Micro-fluidic-jet Spray Dryer

Abstract:Chitosan is a natural polysaccharide with many applications in pharmaceutical and biomedical fields. Chitosan microparticles with narrow size distribution could realize their optimal applications as drug carriers for controlled/targeted release. It is imperative to be able to prepare these particles with defined size and morphology in order to systematically relate these properties to the release behaviour and other functionalities such as drug encapsulation and dissolution. In this study, we successfully prepared monodisperse, non-agglomerated chitosan microparticles in a single step by a novel micro-fluidic-jet spray dryer. The particle size can be tuned by changing the concentration of chitosan solution. The influence of chitosan concentration, storage time, and drying temperature on particle morphology were investigated. A series of measurements were used to observe the chemical and structural changes of chitosan in the precursors. Storage time of chitosan solution and dry temperature showed strong influence on particle morphology, while chitosan concentration had negligible effects. Changes to particle shapes from nearly spherical to mushroom-like (cap-shaped) were observed with increasing storage time and/or drying temperature.
