Abstract:The first catalytic enantioselective 1,2-reduction of enones with 0.45 mol equiv potassium borohydride solution catalyzed by a chiral N,N'-dioxide-Sc(III) complex catalyst was accomplished under mild reaction conditions. A number of optically active allylic alcohols were obtained in good to excellent enantioselectivities (up to 95% ee) with nearly quantitative yields.

Abstract:The relationship between the structure of the catalyst and the selectivity in the asymmetric borane reduction reaction of prochiral ketones is discussed. The variation of the catalyst itself at low temperature is observed by the in situ NMR method and the origin of the temperature effect of the reaction is proposed. It is concluded that the amount of the effective component of the catalyst present has an important effect on the enantioselectivity.

Abstract:The effect of temperature on the enantioselectivity of the oxazaborolidine-catalyzed asymmetric borane reduction of ketones was investigated in the presence of (5S)-3-oxa-1-aza-2-borabicyclo[3.3.0] octane (=(3aS)-tetrahydro-1H,3H-pyrolo[1,2-c][1,3,2]oxazaborole; 1a) and its 2-methoxy derivative (1b) as catalysts, which were synthesized from L-prolinol with borane and trimethyl borate, respectively. The results indicate that the two catalysts induce a different temperature-dependent enantioselectivity. The enantioselectivity of the B-unsubstituted (5S)-3-oxa-1-aza-borabicyclo[3.3.0]octane (1a) increases with increasing temperature, while its B-methoxy-substituted derivative 1b shows the highest enantioselectivity at ca. 50degrees. (5S)-3-Oxa-1-aza-2-borabicyclo[3.3.0]octane (1a) is more likely to dimerize than its 2-methoxy derivative 1b. The conversion rates of L-proline to L-prolinol in the presence of different amounts of borane were also determined in this study.

Abstract:The techniques for improving the stereoselectivity of asymmetric reduction of 3-oxo ester to chiral 3-hydroxy ester catalyzed by yeast cells were studied. Ethyl 4-chloro-3-oxobutanoate was chosen as the model substrate, and the effects of heating and enzyme inhibitor pretreatment of yeast cells on the stereoselectivity of the reduction reaction were investigated in detail. The results showed that the heating pretreatment and preincubation with allyl alcohol could remarkably improve the stereoselectivity of the S-(D)-product, respectively. The stereoselectivity of the S-form product increased with increasing temperature and treatment time in the heating pretreatment. Using allyl alcohol as enzyme inhibitor, the stereoselectivity of the S-form product also increased with increasing the concentration of allyl alcohol. Under appropriate pretreatment conditions, the ee of (S)-ethyl 4-chloro-3-hydroxybutanoate in the asymmetric reduction could reach as high as 97%.

Abstract:The secondary reduction in the direct and oxazaborolidine-catalyzed asymmetric borane reduction of ketones was investigated by the use of GC/MS tracing titration and control experiments. The results indicate that the secondary reduction affects the enantioselectivity only in noncoordinated solvents at low temperature and not under the usual catalytic reduction conditions because the intermediate alkoxyborane is unstable and quickly converts to borane and dialkoxyborane. The function of an alcohol additive in the asymmetric borane reduction of ketones is to consume excess borane in the reduction system thus inhibiting noncatalytic reduction, which leads to increased enantioselectivity in the catalytic reduction.

Abstract:The effect of borane source on enantioselectivity in the enantiopure oxazaborolidine-catalyzed asymmetric borane reduction of ketones has been investigated by using (S)-3,1,2-oxazaborobicyclo[3.3.0]octane and (S)-7,3,1,2-thiaxazaborobicyclo[3.3.0]octane as catalysts. The results indicate that the enantioselective order of different borane sources is borane-dimethyl sulfide < borane-N,N-diethylaniline < borane-THF for the asymmetric reduction of a ketone under the same conditions. Abstract:A series of alkyl 4-dialkylaminophenyl ketones were prepared and reduced asymmetrically by borane under the chiral oxazaborolidine catalysis. The results indicate that the ketones show a more obvious substituent effect on the enantioselectivity than the corresponding 4-alkyl/alkoxy/alkylthiophenyl ketones in, the asymmetric reduction because of the existence of a strong coordinate nitrogen atom with the boron atom in the catalyst and borane.

Abstract:During the synthesis of beta-chloro-alpha-phenylethanols by reduction of alpha-chloroacetophenones with sodium borohydride, many byproducts were produced unavoidably at relatively high temperature. Metal chlorides such as calcium, lanthanum, magnesium, manganese and zinc chloride could efficiently restrain side reactions, and so the product was obtained in high yield.

Abstract:Rationally tuning the electronic effect of catalysts is one of the most important strategies to improve stereoselectivity in asymmetric catalysis. (S)-2-aryl-3,1,2-oxazaborobicyclo[3.3.0]octanes, which can be considered as electronically tuned (S)-2-phenyl-3,1,2-oxazaborobicyclo[3.3.0]octane, were prepared and evaluated in the asymmetric borane reduction of ketones. An unexpected influence of the electronic effect of catalysts on the enantioselectivity was observed and attributed to the catalyst dimerization that was further confirmed experimentally. The unsuccessful tuning is accounted for by assuming that hydride transfer in the catalytic cycle is the rate-determining step in the reduction catalyzed by B-aryl catalysts.

Abstract:Enatioselective reduction of oxime ethers promoted by chiral spiroborate esters with an O3BN framework is reported for the first time. In the presence of (R,S)-1, 11 aralkyloxime ethers are reduced by borane-THF at 0-5 degrees C to give (S)-1-aralkylamine in high yield and excellent enatiomeric excess (up to 98% ee). Influence of reaction conditions on the enantioselectivity of the reduction is investigated, and a possible mechanism of the catalytic reduction is suggested.

Various aromatic and aliphatic conjugated nitroalkenes can be reduced to give the respective nitroalkanes with good yields under mild conditions. This protocol is not only practical, but may also provide insight into the mechanisms of redox transformations in biological systems.

Abstract:Density functional theory (DFT) has been applied to study the enantioselective reduction of 3-morpholin-4-yl-1-phenyl-1-propanone with borane catalyzed by (S)-4-benzyl-5,5- diphenyl-1,3,2-oxazaborolidine at the B3LYP/6-31G* level. All molecular species involved in the four reaction steps have been fully optimized and the structural parameters are provided, and the micro process of reaction was also investigated. The catalyst-alkoxyborane adduct formed in step III exhibits a B-O-B-N tetra-atomic ring. Reaction coordination calculations show that BH3 can react with 3-morpholin-4-yl-1-phenyl-1-propanone spontaneously, resulting in the need of 2 mol BH3 in the reaction.

Abstract:The chemoselective catalytic reduction of nitro scaffolds to value-added amine functions is desirable but challenging. We developed a combined KOtBu/BEt3 catalyst for deoxygenative reduction of nitroarenes, nitro heteroarenes and even notoriously challenging nitroalkanes using pinacolborane under mild reaction conditions. The novel transition-metal-free strategy is readily applicable to the preparation of commercially available pharmaceuticals.

Abstract:The enantioselective reduction of alpha-hydroxy ketones with catecholborane has been developed employing 5 mol% of an 1,1-bi-2-naphthol (BINOL)-derived ion-pair organocatalyst. This methodology provides a straightforward access to the corresponding aromatic 1,2-diols in high yields (up to 90%) with excellent enantioselectivities (up to 97%). Furthermore, the alpha-amino ketones also could be reduced with moderate ee values under mild reaction condition.

Abstract:Theoretical study on the enantioselective reductions of p-chlorophenyl ethyl ketone catalyzed by two chiral oxazaborolidines has been performed by AM1 MO method. The reaction includes four steps. The heat of reaction of each step and the activation energy of the 2nd step were obtained. It was found that the 2nd step and the 4th step are endothermic. Meanwhile, computations showed that the enantioselectivity of the reduction increased by using the PhO derivative of chiral oxazaborolidine as catalyst. The predicted chirality and optical activity of the final product are in agreement with the experimental result.
