Abstract:Detailed chemical studies on the roots of Piper sarmentosum and Piper nigrum have resulted in several alkaloids. The roots of P. sarmentosum gave a new aromatic compound, 1-nitrosoimino-2,4,5-trimethoxybenzene (1). Piper nigrum roots gave pellitorine (2), (E)-1-[3',4'-(methylenedioxy)cinnamoyl]piperidine (3), 2,4-tetradecadienoic acid isobutyl amide (4), piperine (5), sylvamide (6), cepharadione A (7), piperolactam D (8) and paprazine (9). Structural elucidation of these compounds was achieved through NMR and MS techniques. Cytotoxic activity screening of the plant extracts indicated some activity.

Abstract:Two pairs of cyclohexene amide alkaloid enantiomers were obtained from the root of Piper nigrum . Their plane structures were established by NMR and HRESIMS spectra. The absolute configurations of 1a/1b and 2a/2b were determined by the comparison between the experimental and calculated electronic circular dichroism (ECD) spectra. All identified compounds were tested for inhibitory effects on acetylcholinesterase (AChE) in vitro . Notably, compounds 1b and 2b showed strong inhibitory effects on AChE and the interaction between proteins and compounds was discussed by molecular docking studies.

Abstract:Three new amides, (2E,14Z)-N-isobutyleicosa-2,14-dienamide (1), dipiperamides F and G (2 and 3), together with 30 known compounds were isolated from the fruits of Piper retrofractum. Their structures were elucidated by extensive spectroscopic analyses including 1D and 2D NMR as well as MS, and by comparison with the literature. A plausible biosynthetic pathway leading to the amides isolated in this study is discussed. All isolated compounds were screened for their antifungal and cytotoxic activities. Piperanine (9) showed growth inhibition against the fungus Cladosporium cladosporioides, while dipiperamides F and G (2 and 3), chabamide (30), nigramide R (31), dehydropipemonaline (24), pipernonaline (25), guineensine (22), brachystamide B (23), retrofractamide C (20), pellitorine (13), and pipericine (14) exhibited considerable cytotoxicity against L5178Y mouse lymphoma cells with IC50 values of 10.0, 13.9, 11.6, 9.3, 8.9, 17.0, 17.0, 16.4, 13.4, 28.3, and 24.2 mu M, respectively. Abstract:Three new C 19 –diterpenoid alkaloids, nagarumines A–C ( 1–3 ), together two known alkaloids, deoxyaconitine ( 4 ) and N-deethyldeoxyaconitine ( 5 ), were isolated from the roots of Aconitum nagarum . The structures of the new compounds were elucidated by spectral methods such as 1D and 2D ( 1 H- 1 H COSY, HMQC, and HMBC) NMR spectroscopy, as well as high resolution mass spectrometry. The in vivo pharmacological studies revealed that nagarumine C ( 3 ) possessed comparable antinociceptive activity (ED 50 = 76.0 μmol/kg) with the positive control drugs aspirin and acetaminophen. Abstract:OBJECTIVE:To study the chemical constituents in the roots and rhizomes of Notopterygium incisum.METHOD:The chemical constituents were isolated by various column chromatographic methods and structurally elucidated by NMR and MS evidences.RESULT:Twenty-four compounds were obtained and identified as isocalamenediol (1), pregnenolone (2), beta-sitosterol (3), bergapten (4), bergaptol (5), cnidilin (6), isoimperatorin (7), bergamottin (8), demethylfuropinnarin (9), nodakenetin (10), 7'-O-methylnotoptol (11), notopterol (12), notoptol (13), pabulenol (14), phenethylferulate (15), p-hydroxyphenethyl anisate (16), trans-ferulic acid (17), nodakenin (18), sucrose (19), 2-methoxy-4-(3-methoxy-1-propenyl)-phenol (20), 3, 4, 5 trimethoxy-trans-cinnamic acid (21), p-methoxycinnamic acid (22), 4-acetoxy-3-methoxy-trans-cinnamic acid (23), and p-hydroxy-trans-cinnamic acid (24).CONCLUSION:Compounds 1, 14, 20-24 were isolated from the roots and rhizomes of N. incisum for the first time. Compounds 1 and 14 are sesquiterpenoid and coumarin, respectively, and 20-24 were phenylpropanoid compounds.

Abstract:Two undescribed piperidine racemates, (±)-caulophines A and B (1 and 2), a new N-containing xanthone derivative (3), together with six known piperidines, were isolated from the roots of Caulophyllum robustum Maxim. Their structures were determined by extensive spectroscopic techniques. Compounds 3 and 7 exhibited weak cytotoxicities against human palace cancer hela cell line with inhibitory rates of 32.2% and 39.7%, respectively, at the concentration of 40 μM.

Abstract:A new alkaloid, (Z)-2-(1′-amino-2′-hydroxyvinyl)benzoic acid ( 1 ), along with 11 compounds, including five aporphines, (–)-nuciferine ( 2 ), (–)-nornuciferine ( 3 ), (–)-asimilobine ( 4 ), (–)-anonaine ( 5 ), and (–)-roemerine ( 6 ); two oxoaporphines, lysicamine ( 7 ) and liriodenine ( 8 ); two isoquinolones, northalifoline ( 9 ) and thalifoline ( 10 ) and two steroids, β -sitostenone ( 11 ) and stigmasta-4,22-dien-3-one ( 12 ) were isolated from the rhizomes of Nelumbo nucifera . The structure of the new alkaloid was elucidated by chemical and physical evidence.

Abstract:Two compounds (1 and 2) have been isolated from the stem bark of Dysoxylum binectariferum (Roxb.) Hook. f. ex Bedd. Compound 1 is rohitukine [5,7-dihydroxy-2-methyl-8-[4-(3-hydroxy-1- methyl)-piperidinyl]-4H-1-benzopyran-4-one], a known compound that has anti-inflammatory and immunomodulatory activities. Compound 2 is a new alkaloid, named rohitukine N-oxide, the structure of which was elucidated on the basis of spectral evidences and comparison with the 1H NMR data of rohitukine.

Abstract:Piperine, a natural alkaloid isolated from black pepper ( Piper nigrum ), was subjected to alkaline hydrolysis to yield piperic acid. A series of amides were synthesized by treatment of piperic acid with piperidine-4-carboxylic acid derivatives via different routes. The synthesized amides were characterized by various spectral techniques like FT-IR, 1 H NMR, and 13 C NMR spectroscopy and evaluated for their antifungal activity against pathogenic fungal species, viz. Fusarium , Penicillium , and Aspergillus , using poison food technique. All derivatives showed a significant activity against various fungal species in a dose-dependent manner. The results revealed that among various amide derivatives, compound 7a is more effective against Fusarium and Penicillium species, and compound 9b is more effective against Aspergillus species. It was observed that the mycelial growth of test fungi decreases with increasing the concentration of amide derivatives.

Abstract:Two new megastigmane glycosides, (6 R ,7 E ,9 R )-3-oxo- α -ionyl-9-O- α -L-rhamnopyranosyl-(1′′→4′)- β -D-glucopyranoside ( 1 ) and (6 R ,7 E ,9 R )-3-oxo- α -ionyl-9-O- β -D-glucopyranosyl-(1′′→6′)- β -D-glucopyranoside ( 2 ), together with six known analogues ( 3 – 8 ) were isolated from the leaves of Nicotiana tabacum . The structures of all metabolites were determined by comprehensive analysis of NMR and MS spectroscopic data as well as by comparison with those of previously reported. The in vitro anti-inflammatory activity of all isolates was evaluated using a lipopolysaccharide (LPS)-induced RAW264.7 cell inflammatory model, and the compounds 1 , 3 , 7 , and 8 exhibited inhibition of LPS-induced NO production in RAW264.7 macrophage cells with IC 50 values of 42.3-61.7 μM (positive control, dexamethasone, IC 50 = 21.3 ± 1.2 μM).

Abstract:Two triterpenoids were isolated from the roots of Peganum nigellastrum Bunge. Their structures were elucidated as 3alpha-acetoxy-27-trans-caffeoyloxyolean-12-en-28-oic acid methyl ester (1) and 3-oxotirucalla-7, 24-dien-21-oic acid (2) on the basis of spectroscopic evidence. 1 is a new triterpene ester and 2 is a known compound isolated for the first time from genus Peganum.

Abstract:Chemical investigation of the aerial part of Illigera aromatic S.Z. Huang & S.L. Mo has resulted in the isolation and characterization of one new aporphine alkaloid, illigerine B (1), along with three known analogues laurodionine B (2), N-formyl-laurolitsine (3) and illigerine A (4). Their structures were established by spectrometric means and physico-chemical properties. The in vitro cytotoxic activities of compound 1 against Hela, SMMC7721, and Bcap37 cell lines were evaluated. Compound 1 exhibited moderate cytotoxic activity against the three tumor cell types, with IC50 values of 12.40 +/- 0.78, 32.61 +/- 2.05, and 28.69 +/- 1.80 mu g/mL. This work shown that aporphine alkaloids might be useful as characteristic markers in chemotaxonomic research of the genus Illigera.
Nigramide J is a novel potent inverse agonist of the human constitutive androstane receptor

Abstract:The constitutive androstane receptor (CAR, NR1I3) is very important for drug development and for understanding pharmacokinetic drug-drug interactions. We screened by mammalian one hybrid assay among natural compounds to discover novel ligands of human constitutive androstane receptor (hCAR). hCAR transcriptional activity was measured by luciferase assay and mRNA levels of CYP2B6 and CYP3A4 in HepTR-hCAR cells and human primary hepatocytes were measured by real-time RT-PCR. Nigramide J (NJ) whose efficacy is comparable to those of hitherto known inverse agonists such as clotrimazole, PK11195, and ethinylestradiol. NJ is a naturally occurring cyclohexane-type amide alkaloid that was isolated from the roots of Piper nigrum. The suppressive effect of NJ on the CAR-dependent transcriptional activity was found to be species specific, in the descending order of hCAR, rat CAR, and mouse CAR. The unliganded hCAR-dependent transactivation of reporter and endogenous genes was suppressed by NJ at concentrations higher than 5 μmol/L. The ligand-binding cavity of hCAR was shared by NJ and CITCO, because they were competitive in the binding to hCAR. NJ interfered with the interaction of hCAR with coactivator SRC-1, but not with its interaction with the corepressor NCoR1. Furthermore, NJ is agonist of human pregnane X receptor (hPXR). NJ is a dual ligand of hCAR and hPXR, being an agonist of hPXR and an inverse agonist of hCAR.

Abstract:A new naphtho- γ -pyrone dimer, asperosperma A, and a new methyl nicotinate derivative, asperosperma B, with 12 known compounds were isolated from the endophytic fungus Aspergillus niger from the stem of Camellia flavida . Their structure was elucidated by NMR, ECD spectrum, and HR-ESI-MS data. Asperosperma A exhibited a highly cytotoxicity against H460 and 4T1 cancer cells with the IC 50 values were 0.37 ± 0.06 and 2.04 ± 0.79 μM, respectively. Moreover, it showed a highly sensitive against Staphylococcus aureus , Escherichia coli , Pseudomonas aeruginosa and methicillin-resistant S. aureus .

Abstract:A new alkaloid, named 2-methoxy-4-(2-(2-pyridine)-ethyl) phenol (1), together with two known compounds, was isolated from Zingiberis rhizoma. Their structures were elucidated on the basis of 1D and 2D NMR spectra and MS spectra. Compound 1 exhibited substantial bioactivity against Canidia albicans ATCC 10231 with a minimum inhibitory concentration of 1.0 mg/mL.

Abstract:Phytochemical investigation of the roots of Aconitum pseudostapfianum resulted in the isolation of two new C19-diterpenoid alkaloids, designated as pseudostapines A and B ( 1 and 2 ). Their structures were determined by spectral methods such as 1D and 2D ( 1 H– 1 H COSY, HMQC, NOESY, and HMBC) NMR spectroscopy, in addition to high-resolution mass spectrometry. The isolated alkaloids were tested in vivo for antinociceptive potential. The results show that pseudostapines A and B possess more potent antinociceptive effect than the positive control drugs aspirin and acetaminophen.

Abstract:Sixteen previously undescribed steroidal sapogenins along with two known ones were isolated from the stems of Solanum nigrum L. (Solanaceae). Their structures were identified using a combination of 1D and 2D NMR, HR-ESI-MS spectroscopy, the Mosher method, and X-ray diffraction analysis. Compounds 1 – 8 have an unusual F ring and 9 – 12 have a derived A ring, both of which are rare skeletons found in natural products. The biological evaluation showed that the isolated steroids exhibited inhibition of nitric oxide in the LPS-induced RAW 264.7 macrophages with IC 50 values from 7.4 to 41.3 μM. Further studies revealed that compounds 6 and 10 exhibited anti-inflammatory activity by blocking the nuclear translocation of NF- κ B, and down-regulating the expression of iNOS, COX-2, IL-1 β , and IL-6 in a concentration-dependent manner. These results suggest that the stems of S. nigrum may serve as a source of anti-inflammatory agents for use in healthy or medicinal products.

Abstract:A chemical investigation on an endophytic fungus Penicillium expansum isolated from the medicinal plant Plantago depressa Willd. (Plantaginaceae) afforded one new diketopiperazine-type alkaloid, namely penicimine A (1), as well as two known congeners (2 and 3). Their structures were elucidated by widespread spectroscopic data, and the absolute configurations of 1 and 2 were further confirmed by single-crystal X-ray diffraction analyses. Compound 1 represented the first example of benzyl-containing diketopiperazine-type alkaloid bearing a methyl group attached at C-15 position. Compound 1 showed anti-inflammatory activity against LPS-induced nitric oxide (NO) production in RAW264.7 mouse macrophages with an IC50 value of 25.65 μM.

Abstract:Piper longumL., known as long pepper, is an edible and medicinal plant used as spice and for the treatment of stomach disease and analgesia in traditional Chinese medicine.N-Alkylamides are the major secondary metabolites in this plant. Sixteen knownN-alkylamides were isolated fromP. longum. Their structures were established on the basis of spectroscopic data and comparison to reported literatures. Among them, five compounds were isolated from this plant for the first time. Ethanol extract, compounds1,2,3,7and11exhibited potent ability to increase the melanin content and weak stimulative effect on the tyrosinase activity in a concentration-dependent manner. Moreover, compound2also presented strong capacity to increase the tyrosinase activity in a concentration-dependent manner. These results indicated thatP. longummight be a good natural source of lead compound for skin disorder diseases.
