Abstract:Objective To prepare series of degradation products of sodium alginate,and to establish a determination method for their molecular weights by high performance liquid chromatography(HPLC). Methods Series of degradation products of sodium alginate were prepared by acid degradatoin under a high pressure.Six kinds of sodium alginate with different molecular weights were prepared by ultrafiltration, and their molecular weights were determined by HPLC.Results Each chromatogram showed one narrow peak with symmetrical shape,which indicated that those sodium alginates could be used as molecular weight standards.The linear regression equation between the log number of molecular weight (logMw) and the retention time(t) was established,which was logMw=—0.7119t+15.713,and R= 0.971.It indicated that the method had a good reproducibility and precise.Conclusion In this study, standards of sodium alginate were prepared and an HPLC determination method was established,which provided a theoretical and empirical basis for the molecular weight determination of sodium alginate.

Abstract:s: Sodium alginate and polyvinyl alcohol were separately used as carriers to entrap and immobilize the high efficiency degradating bacterium of succinonitrile. Results of degradation of succinonitrile with the bacterium entrapped and immobilized by different carriers were studied. The results showed that when the bacterium entrapped and immobilized with sodium alginate was used and the concentration of the bacterium was 1 g/L and 2 g/L at 30 ℃ with pH=6.5 and the rotating speed of shaker being 250 r/min, the degradation rate of succinonitrile was over 80%.

Abstract:Low-molecular-weight sodium alginate (LMWSA) has been reported to possess unique physicochemical properties and bioactivities. There is little information available about degradation of sodium alginate by ozonation. Effect of ozonation on molecular weight, molecular weight distribution, color change, M/G ratio, and chemical structure of sodium alginate was investigated. The molecular weight of sodium alginate decreased from 972.3 to 76.7 kDa in the 80-min period of ozonation at 25 °C. Two different degradation-rate constants were calculated. Molecular weight distribution of the LMWSA changed appreciably. Ozonation cannot lead to color change of LMWSA. The M/G ratio of LMWSA was not altered significantly, compared with that of the original alginate. The FT-IR and 13C NMR spectra indicated the chemical structure of LMWSA obtained by ozonation was not altered appreciably. New insight into the ozonation of alginate will be promisingly opened up. Ozonation of sodium alginate can be a alternative for production of LMWSA.

Abstract:Degradation is often a critical property of materials utilized in tissue engineering. Although alginate, a naturally derived polysaccharide, is an attractive material due to its biocompatibility and ability to form hydrogels, its slow and uncontrollable degradation can be an undesirable feature. In this study, the degradation behavior of hydrogel based on oxidized sodium alginate (OSA) crosslinked with Ca2+ was studied in phosphate buffer solution (PBS, pH = 7.4) and Tris-(hydroxymethyl) aminomethane-HCl (Tris-HCl, pH = 7.4) at 37 degrees C. The degradation behavior of OSA hydrogels with different degrees of oxidation was evaluated as a function of degradation time by monitoring the changes of molecular weight and weight loss. It was found that the degradation behavior relied heavily on the degree of oxidation and the surrounding medium. This result indicates that the degradation rates of OSA hydrogels can be controlled by changing the degree of oxidation. (C) 2009 Elsevier Ltd. All rights reserved.
Eco-friendly ozonation of alginate: Physicochemical characterization and degradation mechanism exploration through mass spectrometry

Abstract:The ozone degradation has been proven to be an effective degradation method for alginate, while the degradation mechanism remained to be unconfirmed. In this study, two high-molecular-weight alginates with different mannuronic/guluronic (G/M) ratios, HM and HG (G/M 0.49 vs 1.40), were depolymerized using established ozonation technology platform. Notably, HM can be degraded faster than HG especially within initial 30 min, indicating that the β-1, 4-mannuronic bonds are more susceptible to be ozonated than α-1, 4-guluronic bonds. However, HM/HG degraded to LMWA in 2 h and reached a plateau. Therefore, we employed mass spectrometry (MS) to profile the degraded products of LMWA polymannuronate (PM) and polyguluronate (PG) in more intense conditions. The results indicated that the oxidation process continued until all reducing ends were converted to carboxyl groups. The o-diol could directly oxidize to o-dialdehyde. This study provides a MS based elucidation of the mechanism by which alginate cleaves to oligosaccharides through ozonation.
Evaluation of Ultrasound As a Preventative Algae-Controlling Strategy: Degradation Behaviors and Character Variations of Algal Organic Matter Components During Sonication at Different Frequency Ranges

Abstract:To date, the removal of harmful algae and specific algae metabolites (e.g., microcystins, odorous compounds and phycocyanin) by ultrasound has been confirmed. However, the effects of ultrasound on algal organic matter (AOM) variation have not been systematically investigated, which is of great necessity for the management of water safety and the practical application of ultrasound for algae control. This study investigated the effects of ultrasound at three typical frequencies on the degradation behaviors and characters of AOM, including their fluorescence properties, molecular weight and especially sample toxicity. The results showed effective growth inhibition of algae over 7 days after ultrasonic treatment at frequencies of 29.4, 470 and 780 kHz; however, extracellular AOM increased by 1.4%-147.2%, 2.1%-82.5% and 0.9%-90.8%, respectively. High-frequency ultrasound (470 and 780 kHz) had significantly better AOM degradation. The fluorescence pseudo first-order rate constant refers to the decrease in fluorescence and implicates AOM degradation. The rate constants of AOM components were 0.00178-0.00295, 0.00880-0.01763 and 0.00670-0.01319 for the 29.4, 470 and 780 kHz treatments (0.059 W/mL), respectively. Low-frequency sonication was dominated by limited pyrolysis and supercritical water oxidation (SCWO) and mainly caused non-fracture damage. In addition to pyrolysis and SCWO, high-frequency sonication included conspicuous hydroxyl radical oxidation that accounted for more than 60% of the degradation. During high-frequency sonication, some AOM (similar to 25.1 kDa) was degraded into smaller products (0.8-2.0 kDa), and the degradation of the humic-like components was greater than that of the protein-like components. Moreover, high-frequency sonication caused an evident decrease in the toxicity of AOM. This study indicates that high-frequency ultrasound was a promising algae-controlling strategy with respect to effectiveness and safety.

Abstract:The effects of frequency of ultrasound on the precipitation process of prepared supersaturatedsodium aluminate solutions of practical concentration were studied experimentally under seeded,isothermal, batch crystallization conditions at various temperatures and initial αk(mole ratio ofNa2O/Al2O3). The decomposition and the particle number percentage for size below 2 μm at time of 15 hwere compared, particle size distribution and SEM photos of the product aluminum hydroxide were alsoanalyzed. The results indicate that the ultrasonic treatment at 16 kHz can enhance the decomposition rateof sodium aluminate solutions, and also has effects on particle morphology and particle size distributionof aluminum hydroxide precipitated.

Abstract:Oxidized sodium alginate (OSA) is selected as an appropriate material to be extensively applied in regenerative medicine, 3D-printed/composite scaffolds, and tissue engineering for its excellent physicochemical properties and biodegradability. However, few literatures have systematically investigated the structure and properties of the resultant OSA and the effect of the oxidation degree (OD) of alginate on its biodegradability and gelation ability. Herein, we used NaIO4 as the oxidant to oxidize adjacent hydroxyl groups at the C-2 and C-3 positions on alginate uronic acid monomer to obtain OSA with various ODs. The structure and physicochemical properties of OSA were evaluated by Fourier transform infrared spectroscopy (FT-IR), 1H nuclear magnetic resonance (1H NMR), X-ray Photoelectron Spectroscopy (XPS), X-ray Diffraction (XRD), and thermogravimetric analysis (TGA). At the same time, gel permeation chromatography (GPC) and a rheometer were used to determine the hydrogel-forming ability and biodegradation performance of OSA. The results showed that the two adjacent hydroxyl groups of alginate uronic acid units were successfully oxidized to form the aldehyde groups; as the amount of NaIO4 increased, the OD of OSA gradually increased, the molecular weight decreased, the gelation ability continued to weaken, and degradation performance obviously rose. It is shown that OSA with various ODs could be prepared by regulating the molar ratio of NaIO4 and sodium alginate (SA), which could greatly broaden the application of OSA-based hydrogel in tissue engineering, controlled drug release, 3D printing, and the biomedical field.

Abstract:Extraction method and the optimal bleaching condition of alginates from Scytosiphon Iomentsfius and their rheological characteristics were studied in this research. The results indicated that the optimal bleaching condition was conducted as follows-4% of sodium hypochlorite solution with pH of 10.5 was added to the alginate solution with a ratio of 3：600,and heated at 35℃ for 40min. The viscosity of sodium alginate solution from Scytosiphon Iomentsrius decreased with increasing of shearing rate,thus the sodium alginate solution behaved as pseudoplastic fluid and had thixotropism. The sodium alginate solution from Scytosiphon Iomentsrius possessed viscoelasticity,which primarily behaved viscocity in the low frequency area system and elasticity in the higher one. Furthermore,the viscosity of the sodium alginate solution also was influenced by temperature and the concentrations of alginate,salt and sucrose.

Abstract:The decomposition fraction and the particle size distribution during the precipitation process of sodium aluminate solution under different conditions with and without ultrasound were compared respectively. The experiments results indicate that while the conditions of ultrasound being kept constant, the ultrasonic effect on the precipitation process is enhanced as the concentration of sodium aluminate is reduced, and it is more significant at medium α_k. The effect of physical properties of sodium aluminate on ultrasonic enhancement on the precipitation process and ultrasonic cavitation was further investigated by the detection of surface tension coefficient and image of Luminol sonofluorescence.

Abstract:The aim of this study was to investigate the effects of ultrasound degradation on the physicochemical, rheological and antioxidant properties of Sargassum pallidum polysaccharides (SpPS). The results indicated that the ultrasound irradiation could significantly decrease the average molecule weight (MW), and particle size (Zavg) of native SpPS. The degradation pattern of SpPS was closely fitted to the first-order polymer degradation (random chain scission). The primary structure of SpPS before and after ultrasound degradation was not changed, and scanning electron microscopy (SEM) analysis showed that the morphology of SpPS was different from those of the degraded SpPS fractions. Rheological analysis indicated that the degraded SpPS solutions exhibited lower apparent viscosities than native SpPS solution at the same concentration, while the elasticity of the degraded fractions at a certain extent was enhanced. Furthermore, appropriately degraded SpPS fractions exhibited stronger DPPH and ABTS scavenging activity.

Abstract:In order to probe the mechanism of ultrasonic effects on the precipitation process from sodium aluminate solution, the viscosity, surface tension coefficient of sodium aluminate solutions with various α k(mole ratio of Na 2O/Al 2O 3) and Na 2O k concentration (caustic concentration represented by Na 2O) were detected, the sonofluorescence images and the precipitation process in corresponding solutions were also analyzed. The results of experiments indicate that the sonofluorescent phenomenon which prove the free radicals to be generated by ultrasonic treatment can occur in sodium aluminate solutions with composition in a given range. Physical properties of sodium aluminate solution can have effect on ultrasoniccavitation sonofluorescence and ultrasonic intensification on precipitation process. Cavitation and ultrasonic effects are significant when the Na 2O k concentration of sodium aluminate solution is relatively low, and when the α k at about 1.5.

Abstract:The hyaluronan at different concentrations were degraded in ultrasonic oscillator by changing ultrasonic frequency, water bath temperature and pH value. The hyaluronan at the concentration of 2.0 g/L degraded at the highest rates. High ultrasonic frequency and low temperature were propitious to hyaluronan degradation, while the pH value had little influence on it. Moreover, acid condition was a little more convenient for hyaluronan degradation than neutral and basic conditions.

Abstract:Oxidation reactions on –OH groups at C-2 and C-3 positions of the uronic units of sodium alginate were performed with sodium periodate, and the influence of degree of oxidation on physical properties of the oxidized derivatives was analyzed. The aim of this work was to find new active functional groups on alginates, which is a polymer of interest as support in drug-controlled delivery systems. The molar mass decreases rapidly until an oxidation of 10mol% and then remains nearly constant. Polymers with a degree of oxidation higher than 10mol% were no more able to form gels with calcium ions. Both elastic modulus (G′) and swelling degree (SD) of alginate gels decrease as the degree of oxidation increases. This behavior was attributed to the decreasing in the cooperative interactions between calcium ions and carboxylate groups due to a decrease in both the molar mass and the number of unreacted G units.

Abstract:To degrade the polysaccharide with high molecular weight the effectiveness of ultrasound for carboxymethyl cellulose (CMC) in aqueous solution has been studied at a time of 60 min and temperature of 25 ̊C and for polymer solution with concentration up 0.1 g/L. The Huggins equation was appropriate to apply to the intrinsic viscosity of CMC before sonication to understand the influence of salt on degradation, CMC solution was premixed with 0.1 M NaCl, before ultrasonication. Developed a kinetic model, successfully implement to predict and quantify rates of degradation and efficiency. The values of reaction rate constants and reaction orders were found related to the salt and the concentration used, suggesting that, the presence and absence of salts could increase or decrease the degradation by ultrasonic radiation through adjusting the molecular conformation of CMC

Abstract:The effects of ultrasound on the molecular weight distribution and emulsifying properties of both xanthan gum (XG) and propylene glycol alginate (PGA) were investigated. The results showed that ultrasonic treatment at different intensities decreased the apparent viscosity and narrowed the molecular weight distribution. Higher intensity increased the effectivity of the sonochemical effect. Ultrasound degradation did not change the primary structure of the PGA-XG complex, and SEM analysis showed that the morphology of the original polysaccharide differed from that of the degraded polysaccharide fractions. The ultrasonic intensities and treatment times had a substantial influence on the stability of the polysaccharide-stabilized oil-in-water (O/W) emulsions. The O/W emulsion stabilized by the polysaccharide treated with 270 W ultrasound waves for 7 min led to the smallest average particle size (detected via fluorescence microscopy) and showed stability against aggregation in O/W emulsions.

Abstract:In this study, molecular weight controllable degradation of algal Laminaria japonica polysaccharides (LPS) was investigated by ultrasound combined with hydrogen peroxide. Three main factors, i.e., ultrasonic power (A), ultrasonic time (B), and H2O2 concentration (C) were chosen for optimizing parameters by employing three-factors, three-levels BBD. The influence of degradation on structure change and antioxidant activities was also investigated. A second-order polynomial equation including molecular weight (Y) of Laminaria japonica polysaccharides and each variable parameter, i.e., ultrasonic power (A), ultrasonic time (B), and H2O2 concentration (C), was established: Y=20718.67−4273.13A−4000.38B−1438.75C+2333.25AB+1511.00AC+873.00BC+2838.29A2 + 2490.79B2+873.04C2. The equation regression coefficient value (R2 = 0.969) indicated that this equation was valid. The value of the adjusted determination coefficient (adjusted R2 = 0.914) also confirmed that the model was highly significant. The results of selected experimental degradation conditions matched with the predicted value. FT-IR spectra revealed that the structures of LPS before and after degradation were not significantly changed. Antioxidant activities of LPS revealed that low Mws possessed stronger inhibitory than the original polysaccharides. The scavenging effects on superoxide radicals was the highest when IC50 of crude LPS was 4.92 mg mL−1 and IC50 of Mw 18.576 KDa was 1.02 mg mL−1, which was fourfold higher than initial polysaccharide.

Abstract:In order to probe the mechanism of ultrasonic intensification on gibbsite precipitation process from sodium aluminate liquor,the effects of ultrasonic processing on the structure of sodium aluminate liquor were studied by Raman spectrum analysis,sodium nitrate were used as external standard to investigate the variation of in the liquor processed by ultrasound,The results show that ultrasonic processing can convert the structure of the sodium aluminate liquor to the direction advantage to decomposition,and the obvious change of the liquor structure can only occur when the power of the ultrasound exceeding a given value and the composition of the sodium aluminate liquor among a given range.

Abstract:This paper studied the rheological properties of low-concentration carrageenan with different concentration, shearing rate, temperature, pH or stirring time that would affect the viscosity of gum solution. The results showed the viscosity of low-concentration carrageenan was increased with concentration. It became pseudo-plasticity fluid and its viscosity was fluctuated with temperature. At low concentration, the viscosity of carrageenan solution was distinct increased when its pH value was under 5.0, the viscosity of low-concentration gum solution was constant when its pH value in range of 5.0~10.0 and its viscosity was gradually reduced when its pH value up 10.0. The viscosity was influenced with different stirring action.
