Eur J Cancer. 2013; 49: 946-954 Abstract Full Text Full Text PDF PubMed Scopus (64) Google Scholar assessed six selected single-nucleotide polymorphisms (SNPs) (rs1927914, rs4986790, rs4986791, rs11536889, rs1927911 and rs2149356) in TLR4 on risk of cancer. We searched the PubMed and CNKI (China National Knowledge Infrastructure) databases for all articles within a range of published years from 2000 to 2012 on the association between polymorphism in TLR4 and cancer risk (last search was update 29th February 2012). The following terms were used in this search: ‘TLR4 or Toll like receptor 4’ and ‘cancer’ and ‘polymorphism or polymorphisms’. Finally, 17 data in 16 articles with 6051 cases and 6385 controls on rs4986790 were available. Our meta-analyses found statistically significantly elevated overall cancer risk was associated with rs4986790. In terms of stratified analyses by cancer type, significantly elevated risk was observed in gastric cancer and ‘other cancers’. The update meta-analysis conducted by Zhao et al. was performed with comprehensive search up to December 2012, the search terms as follows: (‘Asp299Gly’ or ‘rs4986790’ or ‘896A/G’) and (‘cancer’ or ‘carcinoma’ or ‘tumour’ or ‘malignancy’ or ‘neoplasm’) were used. Ultimately, 29 case–control studies in 28 literatures including 10,102 cases and 11,667 controls were retrieved. Their results confirmed our findings.
