Abstract:Momordica charantia L. (M. charantia), a member of the Cucurbitaceae family, is widely distributed in tropical and subtropical regions of the world. It has been used in folk medicine for the treatment of diabetes mellitus, and its fruit has been used as a vegetable for thousands of years. Phytochemicals including proteins, polysaccharides, flavonoids, triterpenes, saponins, ascorbic acid and steroids have been found in this plant. Various biological activities of M. charantia have been reported, such as antihyperglycemic, antibacterial, antiviral, antitumor, immunomodulation, antioxidant, antidiabetic, anthelmintic, antimutagenic, antiulcer, antilipolytic, antifertility, hepatoprotective, anticancer and anti-inflammatory activities. However, both in vitro and in vivo studies have also demonstrated that M. charantia may also exert toxic or adverse effects under different conditions. This review addresses the chemical constituents of M. charantia and discusses their pharmacological activities as well as their adverse effects, aimed at providing a comprehensive overview of the phytochemistry and biological activities of M. charantia.

Abstract:Momordica charantia ( M. charantia ), as a common edible vegetable and herb, is mainly distributed in tropical and subtropical regions of the world. M. charantia polysaccharides (MCPs), as the main pharmacologically active component in M. charantia , are water-soluble polysaccharides with an average molecular weight of 4–900 kDa. The extraction methods of MCPs mainly include hot water extraction, acid extraction, alkali extraction, ultrasonic extraction, enzyme extraction and three-phase partitioning extraction, and different extraction methods will affect the yield of MCPs. MCPs possess a variety of bioactivities, including antidiabetic, antiaging, antioxidant, antiviral, immunomodulatory and neuroprotective effects. The purpose of this review is to systematically summarize the latest research progress of MCPs in extraction, purification, structural characterization, and biological activity. In addition, the structure–activity relationship will be further discussed. We believe that this review will provide a useful reference for the investigation, production, and application of MCPs in functional foods and therapeutic agents.

Abstract:Momordica charantia L. is a kind of vegetable with medicinal value. As the main component of the vegetable, Momordica charantia polysaccharides (MCPs) mainly consist of galactose, galacturonic acid, xylose, rhamnose, mannose and the molecular weight range is 4.33 × 10 3 –1.16 × 10 6 Da. MCPs have been found to have various biological activities in recent years, such as anti-oxidation, anti-diabetes, anti-brain injury, anti-obesity, immunomodulatory and anti-inflammation. In this review, we systematically summarized the extraction methods, structural characteristics and physicochemical properties of MCPs. Especially MCPs modulate gut microbiota and cause the alterations of metabolic products, which can regulate different signaling pathways and target gene expressions to exert various functions. Meanwhile, the potential structure-activity relationships of MCPs were analyzed to provide a scientific basis for better development or modification of MCPs. Future researches on MCPs should focus on industrial extraction and molecular mechanisms. In East Asia, Momordica charantia L. is used as both food and medicine. It is not clear whether MCP has its unique biological effects. Further study on the difference between MCPs and other food-derived polysaccharides will be helpful to the development and potential application of Momordica charantia L .

Abstract:Momordica charantia (MC), a member of the Cucurbitaceae family, is well known for its pharmacological activities that exhibit hypoglycemic and hypolipidemic properties. These properties are largely because of its abundant bioactive compounds and phytochemicals. Over the years, numerous studies have confirmed the regulatory effects of MC extract on glycolipid metabolism. However, there is a lack of comprehensive reviews on newly discovered MC-related components, such as insulin receptor-binding protein-19, adMc1, and MC protein-30 and triterpenoids 3β,7β,25-trihydroxycucurbita-5,23(E)-dien-19-al, and the role of MC in gut microbiota and bitter taste receptors. This review offers an up-to-date overview of the recently reported chemical compositions of MC, including polysaccharides, saponins, polyphenolics, peptides, and their beneficial effects. It also provides the latest updates on the role of MC in the regulation of gut microbiota and bitter taste receptor signaling pathways. As a result, this review will serve as a theoretical basis for potential applications in the creation or modification of MC-based nutrient supplements.

Abstract:The Cucurbitaceae family, which includes Momordicacharantia Linn. (M. Charantia), has a widely geographical distribution throughout tropicaly and subtropicaly areas in the world. Since ancient times, its fruit used for vegetable diet, herbal therapy and is utilised in drugs to cure diabetes mellitus. Abstract:Background: Currently, balanced diet and healthy foods are attracting increasing attention due to their great health-beneficial effects. The tuber of Dioscorea (yam) is both edible and medicinal, as it contains various functional compounds with multiple pharmacological activities and disease targets. Moreover, yam tuber can serve as a vegetable or staple food due to its high starch content. Therefore, exploring the functional activities of yam tuber and its application in food and medicine industries is significant. Scope and approach: This review comprehensively summarizes the characteristics and application of yam tuber, as well as the findings about its primary nutrients, distinct functional components, extraction technologies, detection methods and biological activities, and provides some new prospects for further investigation. Besides, the development and comprehensive utilization of yam tuber products are elucidated. Finally, some future research directions for better utilization of yam tubers are suggested. Key findings and conclusions: The Dioscorea genus is distributed worldwide with a wide variety of species. Yam tuber is rich in nutrients and bioactive compounds, such as starch, protein, non-starch polysaccharides, steroid saponin, allantoin, and phenolic compounds. The most important health benefits include antioxidant activity, anti-inflammatory activity, gastrointestinal protection, gut microbiota regulation, cardiac diseases treatment, hypoglycemic activity, anti-tumor activity, and estrogen-like effect. We also summarize the applications of yam tuber in foods and medicine. Finally, the review indicates that better utilization of yam tuber is hindered by poor variety protection, limited research on its pharmacological activities, browning phenomenon, and some safety concerns.
Momordica Charantia Juice with Lactobacillus Plantarum Fermentation: Chemical Composition, Antioxidant Properties and Aroma Profile

Abstract:Momordica charantia L. has been consumed as a vegetable for thousands of years, and its fruit has various bioactive compounds. The aim of this study was to investigate the effect of fermentation with Lactobacillus plantarum (L. plantarum) NCU116 on the chemical composition, antioxidant activity, and volatile profile of M. charantia juice. Fermentation and sterilization increased the concentrations of short-chain fatty acids, while the total saponins in the juice was reduced. Fermentation increased lactic acid, total phenolics, and total flavonoids. Fermentation improved the phenolic composition of the juice, such as decreased caffeic and p-coumaric acids, and increased dihydrocaffeic and phloretic acids. Furthermore, the antioxidant properties of the juice and phenolic extracts increased after fermentation. Fermentation altered the aroma profile of the juice to confer a more desirable flavor by reducing aldehydes and ketones, while increasing alcohols and acids. Sterilization decreased lactic acid, total phenolic content, phenolic composition, antioxidant activities, and the aroma profile. These results suggested that L. plantarum fermentation had a beneficial effect on the physico-chemical properties, bioactive compounds, antioxidant property, as well as aroma composition of M. charantia.

Abstract:It[aut]Momordica charantia is a climbing, nearly or quite smooth, annual vine that is widely grown in Asia (especially Indian subcontinent), Africa, and in central Europe. Hindu physicians in India used the whole plant combined with cinnamon, long pepperPepper, rice and the oil of Hydnocarpus wightiana as an external application in scabies and other skin diseases. Fruits and leaves were used as anthelmintic, and externally applied in leprosy. Leaves juice was also rubbed in burning sole of the feet, and with black pepperBlack pepper was rubbed around the orbit as a cure for night blindness. Muslim physicians found it useful in rheumatism and gout, and in diseases of the liver and spleen, and as anthelmintic. Outer fruit coat and seeds, but not the fruit pulp, are cathartic, producing gastroenteritis with vomiting and diarrhea, the symptoms persisting for long periods. In Turkish folk medicine, mature fruits are used externally for wound healing and orally for the treatment of gastric ailments including peptic ulcers. It is part of the traditional diet of Okinawa Island (Japan) residents, is used to manage diabetes by Mauritian population, and a tea prepared from a wild variety, known as Cerasee is traditionally used for the treatment of diabetes mellitus in the West Indies and Central America. In Cuba and Puerto Rico, the plant is also used for the treatment of diabetes mellitus, wounds refractive to other treatments, skin diseases, chronic stomach ulcers, and for sterility in women. Whole plant is used in the management of depressive illness in traditional African medicine. Fruits are a good source of iron, calcium and phosphorus, contain substances with antidiabetic properties, such as charantin (a ribosome-inactivating peptide), vicine, and polypeptide-P, and other nonspecific bioactive components such as antioxidants. Lycopene is the major carotinoid of ripe seeds, and its concentration increases about 100-fold from the immature to the ripe stage. Drinking of homogenized water suspension of the pulp significantly reduced both fasting and postprandial serum glucose levels of NIDDM patients, and the fruit juice significantly improved glucose tolerance of Sri Lankan NIDDM patients. Addition of powdered fruit to standard therapy of newly diagnosed or poorly controlled type-2 diabetes patients with HbA1c levels between 7 and 9%, lowered HbA1c, but no significant improvement in mean FBS, TC, and body weight.
Emerging Potential of Momordica’s Bioactive Phytochemicals in Cancer Prevention and Therapy

Abstract:Worldwide, cancer incidence and mortality are rising quickly. Cancer remains the biggest cause of death despite advances in therapy. Plants produce bioactive phytochemicals, and as a result, the bioactive elements have long been the focus of cancer research, both for medication discovery and for the discovery of alternative chemo-preventive methods. The medicinal plant Momordica charantia or bitter melon contains a wide variety of phytochemicals, such as triterpenoids, triterpene glycosides, phenolic acids, flavonoids, lectins, sterols, and proteins. In many pre-clinical systems, the Momordica charantia extract exhibits an anti-cancer action against various malignancies. The bioactive components of the extract play a significant role in its anti-cancer properties. With an emphasis on underlying molecular pathways, we address the roles of Momordica's known bioactive components in several cancer models in this review. Through the inhibition of cancer cell proliferation and induction of cell death, several of active ingredients exhibit cancer prevention and therapeutic effects, at least in in-vitro models. Mechanistically examining the active components in pre-clinical systems may reveal a novel approach to cancer treatment.

Abstract:This article will review selected herbal products used in traditional Chinese medicine, including medicinal mushrooms ( bā xī mó gū; Agaricus blazei, yún zhī; Coriolus versicolor, líng zhī; Ganoderma lucidum, xiāng xùn; shiitake, Lentinus edodes, niú zhāng zhī; Taiwanofungus camphoratus), Cordyceps ( dōng chóng xià cǎo), pomegranate ( shí liú; Granati Fructus), green tea ( lǜ chá; Theae Folium Non Fermentatum), garlic ( dà suàn; Allii Sativi Bulbus), turmeric ( jiāng huáng; Curcumae Longae Rhizoma), and Artemisiae Annuae Herba ( qīng hāo; sweet wormwood). Many of the discussed herbal products have gained popularity in their uses as dietary supplements for health benefits. The review will focus on the active constituents of the herbs and their bioactivities, with emphasis on the most recent progress in research for the period of 2003 to 2011.

Abstract:Momordica charantia (bitter melon), a traditional medicinal plant, has been demonstrated to have potential in managing diabetes, gastrointestinal problems, and infections. Among its bioactive compounds, momordicine I, a cucurbitane-type triterpenoid, has attracted attention due to its substantial biological activities. Preclinical studies have indicated that momordicine I possesses antihypertensive, anti-inflammatory, antihypertrophic, antifibrotic, and antioxidative properties, indicating its potential as a therapeutic agent for cardiovascular diseases. Its mechanisms of action include modulating insulin signaling, inhibiting inflammatory pathways, and inducing apoptosis in cancer cells. The proposed mechanistic pathways through which momordicine I exerts its cardiovascular benefits are via the modulation of nitric oxide, angiotensin-converting enzymes, phosphoinositide 3-kinase (PI3K)/ protein kinase B (Akt), oxidative stress, apoptosis and inflammatory pathways. Furthermore, the anti-inflammatory effects of momordicine I are pivotal. Momordicine I might reduce inflammation through the following mechanisms: inhibiting pro-inflammatory cytokines, reducing adhesion molecules expression, suppressing NF-κB activation, modulating the Nrf2 pathway and suppressing c-Met/STAT3 pathway. However, its therapeutic use requires the careful consideration of potential side effects, contraindications, and drug interactions. Future research should focus on elucidating the precise mechanisms of momordicine I, validating its efficacy and safety through clinical trials, and exploring its pharmacokinetics. If proven effective, momordicine I could considerably affect clinical cardiology by acting as a novel adjunct or alternative therapy for cardiovascular diseases. To date, no review article has been published on the role of bitter-melon bioactive metabolites in cardiovascular prevention and therapy. The present work constitutes a comprehensive, up-to-date review of the literature, which highlights the promising therapeutic potential of momordicine I on the cardiovascular system and discusses future research recommendations.

Abstract:Plant-derived extracellular vesicles are functional nanovesicles that have significant applications in both disease prevention and treatment, as well as for use as drug carriers. Momordica charantia is a widely consumed food that has both medicinal and nutritional properties and has shown intervention in diabetes and inflammation caused by oxidative damage. In this study, Momordica charantia-derived extracellular vesicles (MCEVs) were extracted and demonstrated to have excellent antioxidant activity by characterization, lipid composition analysis, protein domain analysis, and in vitro antioxidant measurement. In addition, in vivo studies indicated that the MCEVs could restore ulcerative colitis by regulating oxidation and inflammatory factors. Therefore, the antioxidant properties of MCEVs may be important in protecting the colon from inflammation, which provides new insights into the application of MCEVs as drugs or vectors for intervention in ulcerative colitis.

Abstract:The separation and identification of bioactive substances that come from natural resources have aroused increasing attention. Momordica charantia ( M. charantia ) has a variety of bio-activities, containing anti-oxidation, immune regulation, anti-tumor, radiation protection, anti-diabetes, and liver protection. After the extraction new substances from Momordica charantia , their treatment effect on lung cancer combined with Hedyotis diffusa was determined and the related mechanism was explored as well in this study. Firstly, the CCK-8 assay was conducted and the inhibition of the new substances combined with Hedyotis diffusa . Next, the real time RT-PCR was conducted and the relative expression of the VEGF in the cancer cells was determined. In addition, the complete chloroplast genome sequence of M. charantia is followed by the phylogenetic analysis.

Abstract:Momordicacharantia L. belongs to the family Cucurbitaceae. This plant has widely been reported for use in traditional medical systems for the treatment of diabetes, rheumatism, intestinal gas and malaria. The ethnicity of M. charantiais Asia, Africa and some parts of Europe where it is considered edible. Sewage sludge, a by-product of waste water treatment plant, contains several nutrients. These nutrients are known as beneficial for the better growth and good yield of the plants. The objective of this study is to optimize soil with different ratios of sewage sludge for increased growth of bioactive components in M. charantia. The plant was grown at six places; in soil containing 20%, 40%, 60% and 80% sewage sludge ratio as well as in pure soil and in pure sewage sludge. Fruit harvesting was carried out after 75 days. Methanol extract of M. charantia (Fruit) was subjected to analysis using GC-MS. Comparative quantification of 7,11-hexadecadienal, oleic Acid and stigmasterol were carried out. It has been found that the soil containing 60% sewage sludge yielded maximum quantity of 7,11-hexadecadienal, oleic Acid and stigmasterol. Therefore, optimization of soil with different ratios of sewage sludge is carried out and 60% ratio of sewage sludge in soil is recommended for the promising growth of Momordicacharantia.

Abstract:Gac fruit (Momordica cochinchinensis Spreng) is a popular tropical fruit in southeast Asia. What is amazing is that its seeds (Momordicae Semen) and arils are traditional herbs with anti-tumor activity, and have protected human health for more than 1000 years. In recent years, its anti-tumor activity has received extensive attention and research. This manuscript summarized the chemical composition of saponins, fatty acids, volatile constituents, proteins, peptides, and other components from Momordicae Semen (MSE). The effect and mechanism of MSE and its extract on breast cancer, gastric cancer, lung cancer, esophagus cancer, melanomas, and human cervical epithelial carcinoma were discussed. In addition, its antioxidant, anti-inflammatory, and other pharmacological effects were also analyzed. We hope that this review will provide new ideas for the treatment of cancer and other diseases, and become a reference for the further research into complementary and alternative medicine.

Abstract:Diabetes mellitus (DM) is the most common of the endocrine disorders and represents a global health problem. DM is characterized by chronic hyperglycaemia due to relative or absolute lack of insulin or the actions of insulin. Insulin is the main treatment for patients with type 1 DM and it is also important in type 2 DM when blood glucose levels cannot be controlled by diet, weight loss, exercise and oral medications alone. Prior to the availability of insulin, dietary measures, including the traditional medicines derived from plants, were the major form of treatment. A multitude of plants have been used for the treatment of diabetes throughout the world. One such plant is Momordica charantia (Linn Family: Cucurbaceae), whose fruit is known as Karela or bittergourd. For a long time, several workers have studied the effects of this plant in DM. Treatment with M. charantia fruit juice reduced blood glucose levels, improved body weight and glucose tolerance. M. charantia fruit juice can also inhibit glucose uptake by the gut and stimulate glucose uptake by skeletal muscle cells. Moreover, the juice of this plant preserves islet β cells and β cell functions, normalises the systolic blood pressure, and modulates xenobiotic metabolism and oxidative stress. M. charantia also has anti-carcinogenic properties. In conclusion, M. charantia has tremendous beneficial values in the treatment of DM.
Cytotoxic and Anti-Inflammatory Triterpenoids in the Vines and Leaves of Momordica charantia

Abstract:The vines and leaves of Momordica charantia L. are used as herbal medicines to treat inflammation-related disorders. However, their safety profile remains uncharacterized, and the constituents in their extracts that exert anti-inflammatory and adverse effects remain unclear. This study isolated the characteristic cucurbitane-type triterpenoid species in the vines and leaves of M. charantia L. and analyzed their cytotoxicity, anti-inflammatory effects, and underlying mechanisms. Four structurally related triterpenoids—momordicines I, II, IV, and (23E) 3β,7β,25-trihydroxycucurbita-5,23-dien-19-al (TCD)—were isolated from the triterpenoid-rich fractions of extracts from the vines and leaves of M. charantia. Momordicine I was cytotoxic on normal cells, momordicine II exerted milder cytotoxicity, and momordicine IV and TCD had no obvious adverse effects on cell growth. TCD had anti-inflammatory activity both in vivo and in vitro. In lipopolysaccharide-stimulated RAW 264.7 cells, TCD inhibited the inhibitor kappa B kinase/nuclear factor-κB pathway and enhanced the expression of nuclear factor erythroid 2-related factor 2, heme oxygenase-1, and glutamate-cysteine ligase modifier subunit through the extracellular signal-regulated kinase1/2 and p38. Thus, the vines and leaves of M. charantia should be used with caution. An extraction protocol that can enrich TCD but remove momordicine I would likely enhance the safety of the extract.

Plant Foods for Human Nutrition

Abstract:Hypertension is one of the main factors of cardiovascular disease worldwide and is strongly related to the overall mortality. High salt intake is a major risk factors for hypertension. Identifying functional foods that can help prevent mechanistic abnormalities mediating salt-induced hypertension is an issue of considerable nutraceutical and scientific interest. Dietary Momordica charantia may be an alternative approach to avoid salt-induced hypertension. Dahl salt-sensitive (DSS) rats were used to determine whether Momordica charantia water extracts (ME) exerts anti-hypertensive effects in the present study. ME gavage could significantly prevented the increase of blood pressure, blood urea nitrogen, creatinine, and urine protein-to-creatinine ratio of DSS rats. Metabolomics analysis indicated that high-salt diet induced abnormal amino acid metabolism was related to nitric oxide (NO) deficiency, but ME gavage could upregulate the activities of nitric oxide synthase, aspartate aminotransferase, argininosuccinate lyase, argininosuccinate synthase and restore endogenous synthesis of arginine and NO. Meanwhile, renal function was improved after ME gavage. Citrulline, as one of the important component in ME, could attenuate salt-induced hypertension by increasing endogenous synthesis of arginine and NO. Antioxidants in ME, such as phenolic compound, may avoid high-salt induced oxidative stress in DSS rats, which may be another mechanism by which ME prevented blood pressure increase. Thus, the present study indicated that feeding Momordica charantia could avoid high-salt-induced hypertension in DSS rats.

Abstract:Momordica cymbalaria Hook F. (MC), belonging to the family Cucurbitaceae, is a plant with several biological activities. This detailed, comprehensive review gathers and presents all the information related to the geographical distribution, morphology, therapeutic uses, nutritional values, pharmacognostic characters, phytochemicals, and pharmacological activities of MC. The available literature showed that MC fruits are utilized as a stimulant, tonic, laxative, stomachic, and to combat inflammatory disorders. The fruits are used to treat spleen and liver diseases and are applied in folk medicine to induce abortion and treat diabetes mellitus. The phytochemical screening studies report that MC fruits contain tannins, alkaloids, phenols, proteins, amino acids, vitamin C, carbohydrates, β -carotenes, palmitic acid, oleic acid, stearic acid, α-eleostearic acid, and γ-linolenic acid. The fruits also contain calcium, sodium, iron, potassium, copper, manganese, zinc, and phosphorus. Notably, momordicosides are cucurbitacin triterpenoids reported in the fruits of MC. Diverse pharmacological activities of MC, such as analgesic, anti-inflammatory, antioxidant, hepatoprotective, nephroprotective, antidiabetic, cardioprotective, antidepressant, anticonvulsant, anticancer, antiangiogenic, antifertility, antiulcer, antimicrobial, antidiarrheal and anthelmintic, have been reported by many investigators. M. cymbalaria methanolic extract is safe up to 2,000 mg/kg. Furthermore, no symptoms of toxicity were found. These pharmacological activities are mechanistically interpreted and described in this review. Additionally, the microscopic, powder and physiochemical characteristics of MC tubers are also highlighted. In summary, possesses remarkable medicinal values, which warrant further detailed studies to exploit its potential benefits therapeutically.

Abstract:Diabetes mellitus is a highly prevalent metabolic disorder which is characterized by impaired glucose tolerance, with a relative or absolute insulin deficiency and profound changes in the metabolism of macronutrients. Traditional and complementary medicine is therapeutic strategies that have both been applied to improving glycemic control. Momordica charantia is one of the plant-based, folk medicines that used for improving glycemic control. We aimed to review, the effects of M. charantia on blood glucose with a clarification of the molecular pathways involved. Of the compounds derived from the plants, the insulin-like peptide, charantin, and the alkaloid vicine, have been reported to have hypoglycemic effects. Different mechanisms contribute to the antidiabetic activities of M. charantia, these include increasing pancreatic insulin secretion, decreasing insulin resistance and increasing peripheral and skeletal muscle cell glucose utilization, inhibition of intestinal glucose absorption and suppressing of key enzymes in the gluconeogenic pathways.
