Abstract:Phosphorescence imaging technology has many advantages, including long lifetime, no real-time excitation, and elimination of biological self-fluorescence, which plays an important role in cell and organism monitoring. Phosphorescent probes provide a key material basis for the development of phosphorescent imaging. This review firstly introduces the luminescence mechanism of phosphorescence and its principle for biological imaging. Secondly, according to their composition and structures, phosphorescent probes are divided into groups of organic and inorganic. The research on their synthesis, performance, and application is summarized. Finally, the existing problems of organic and inorganic phosphorescent probes are put forward, and their development is prospected. This review can provide an idea for the design and application of advanced phosphorescent probes in the field of biological imaging.

Abstract:Bioanalysis based on optical imaging has gained significant progress in the last few decades. Luminescence probes are capable of detecting, monitoring, and tracing particular biomolecules in complex biological systems to figure out the roles of these molecules in organisms. Considering the rapid development of luminescence probes for bio-applications and their promising future, we have attempted to explore the working principles and recent advances in bio-applications of luminescence probes, in the hope of helping readers gain a detailed understanding of luminescence probes developed in recent years. In this review, we first focus on the current widely used luminescence probes, including fluorescence probes, bioluminescence probes, chemiluminescence probes, afterglow probes, photoacoustic probes, and Cerenkov luminescence probes. The working principles for each type of luminescence probe are concisely described and the bio-application of the luminescence probes is summarized by category, including metal ions detection, secretion detection, imaging, and therapy.

Abstract:Fluorescent imaging is a multidisciplinary integrated technology for bio-imaging. Application of fluorescent probes allows dynamic visualization of large bio-molecules inside cells and animals, which can benefit the diagnosis and treatment of various diseases.Satisfactory fluorophores and bio-orthogonal reactions play crucial roles in the construction of ideal fluorescent probes for in vivo imaging of cellular biopolymers and biological events. This review summarized the recent advance in these two areas with examples illustrating the applications in fluorescent bio-imaging.

Abstract:Peptide-based probes play prominent roles in biomedical research due to their promising properties such as high biocompatibility, fast excretion, favorable pharmacokinetics as well as easy and robust preparation. Considering the translation of imaging probes into clinical applications, peptide-based probes remain to be the most desirable and optimal candidates. This review summarized the development of peptide-based probes with promising imaging modalities and highlighted the successful applications for in vivo biomedical imaging.

Abstract:Some important biological species and microenvironments maintain a complex and delicate dynamic balance in life systems, participating in the regulation of various physiological processes and playing indispensable roles in maintaining the healthy development of living bodies. Disruption of their homeostasis in living organisms can cause various diseases and even death. Therefore, real time monitoring of these biological species and microenvironments during different physiological and pathological processes is of great significance. Fluorescent-probe-based techniques have been recognized as one of the most powerful tools for real time imaging in biological samples. In this Account, we introduce the representative works from our group in the field of fluorescent probes for biological imaging capable of detecting metal ions, small bioactive molecules, and the microenvironment. The design strategies of small molecule fluorescent probes and their applications in biological imaging will be discussed. By regulating the design strategy and mechanism (e.g., ICT, PeT, and FRET) of the electronic and spectral characteristics of the fluorescent platforms, these chemical probes show high selectivity and diverse functions, which can be used for imaging of various physiological and pathological processes. Through the exploration of the rational response mechanism and design strategy, combined with a variety of imaging techniques, such as super-resolution imaging, photoacoustic (PA) imaging, etc., we have realized multimode imaging of the important biological analytes from the subcellular level to the in vivo level, which provides powerful means to study the physiological and pathological functions of these species and microenvironments. This Account aims to offer insights and inspiration for the development of novel fluorescent probes for biological imaging, which could provide powerful tools for the study of chemical biology. Overall, we represent a series of turn-on/turn-off/ratiometric fluorescent/PA probes to visually and dynamically trace biological species and microenvironments in cells and even in vivo that seek higher resolution and depth molecular imaging to improve diagnostic methods and clarify new discoveries related to chemical biology. Our future efforts will be devoted to developing multiorganelle targeted fluorescent probes to study the mechanism of subcellular organelle interaction and employing various dual-mode probes of NIR II and PA imaging to investigate the development of related diseases and treat the related diseases at subcellular and in vivo levels.

Abstract:Carbon monoxide (CO) is an important gas transporter and is closely associated with a series of physiological and pathological processes. So far, it is clear that plenty of fluorescent probes have been prepared, but most of them still have some disadvantages and is unsuitable for imaging in vivo. Herein, a novel fluorecent probe named HPQ-BI-CO is synthesized and used to detect CO. In this probe, the benindole group is introduced to extend the wavelength. This probe possess a variety of advantages such as long emission wavelength, high sensitivity and selectivity, and rapid response capability. Moreover, the probe possesses a low cytotoxicity, which make it apply in the imaging of exogenous and endogenous CO in living 4T1 cells. Furthermore, this probe can be utilized as a promising tool for fluorescent bioimaging of CO in vivo like zebrafish.

Abstract:In recent years, with the rise of precision therapy and other frontier fields, the demand of the high quality fluorescent probes with extremely low toxicity, good biocompatibility, strong stability and high brightness is more urgent than ever. Proteins that with abundant active groups and special pockets can bind with different probes to alter the fluoresce properties, and some can even greatly enhance the fluorescence signals. Moreover, some proteins can simultaneously bind with two different molecules to fabricate fluorescence resonance energy transfer (FRET) probes. Recent progress witnessed that the protein-mediated strategies have provided a promising way for enhancing the performance of fluorescent probe. This article will review the up-to-date progress of protein-mediated fluorescent probes. We firstly provide a systemically on the principle of protein fluorescent probes fabrication and the interaction mechanisms between proteins and molecular dyes. Then the bioapplications of these fluorescent probes in bioimaging and biosensing were reviewed. And finally the current challenges and perspectives were discussed in-depth. We hope that this review can provide some new ideas and inspiration for relevant researchers to develop a new generation of fluorescent probes.

Abstract:Optogenetic nanoprobes are an important technique in optogenetics, utilized to deliver precise light stimulation to neurons in organisms, aiding neuroscientists in investigating the working mechanisms of the brain. Optogenetic nanoprobes have the potential to be used in the diagnosis and treatment of neurological disorders. To fulfill stimulation requirements such as stimulation intensity, stimulation range, stimulation patterns, and temporal and spatial resolution, researchers have developed probes capable of performing various optical functions. To fulfill functional requirements such as in-situ electrophysiological recording and delivery of chemical or biological molecules, scientists have developed multifunctional probes. In order to overcome the disadvantages of traditional optoelectronic devices, as they are rigid and easy to cause severe damage to organisms, flexible optical neural probes have been invented. This type of probe causes minimal harm to the organism during implantation and maintains a consistent level of light illumination, ensuring a prolonged life span. This paper provides an overview and the prospects for different types and functions of optogenetic probes, as well as their flexible technologies.

Abstract:Compared with short-lived emission probes featuring fluorescence imaging , the use of phosphorescent probes imparts the advantage of long-lived signal persistence that distinguishes against background fluorescence interference. However, the realization of ultralong organic phosphorescent (UOP) probes with an ultralong emission lifetime in an aqueous medium is still a challenge. Here, we present a rational strategy for obtaining UOP nanoparticles (NPs) in an air-saturated aqueous medium prepared using an organic phosphor (PDBCz) and a surfactant polymer (PVP), named PDBCz@PVP, showing an ultralong emission lifetime of 284.59 ms and a phosphorescence quantum efficiency of 7.6%. The excellent phosphorescence properties and water solubility of PDBCz@PVP make it a promising candidate for biological imaging. The as-prepared PDBCz@PVP NPs possess excellent luminescence intensity as well as illustrious biocompatibility both in vitro and in vivo. We demonstrate their use as an efficient phosphorescent nanoprobe both in living cells and zebrafish by capturing their afterglow emission signals under microscopy observation for the first time, realizing convenient and fast bioimaging with low cost, which allows for anti-fluorescence interference and shows promise for the future theragnostic applications in nanomedicine.

Abstract:Optical imaging essentially refers to in vivo fluorescence imaging and bioluminescence imaging. These types of imaging are widely used visualization methods in biomedical research and are important in molecular imaging. A new generation of imaging agents called multimodal probes have emerged in the past few years. These probes can be detected by two or more imaging modalities, which harnesses the strengths of the different modalities and enables researchers to obtain more information than can be achieved using only one modality. Owing to its low cost and the large number of probes available, the optical method plays an important role in multimodality imaging. In this mini-review, we describe the available multimodal imaging probes for in vivo imaging that combine optical imaging with other modalities. Copyright (C) 2011 John Wiley & Sons, Ltd.

Abstract:The development of super-resolution technology has made it possible to investigate the ultrastructure of intracellular organelles by fluorescence microscopy, which has greatly facilitated the development of life sciences and biomedicine. To realize super-resolution imaging of living cells, both advanced imaging systems and excellent fluorescent probes are required. Traditional fluorescent probes have good availability, but that is not the case for probes for live-cell super-resolution imaging. In this review, we first introduce the principles of various super-resolution technologies and their probe requirements, then summarize the existing designs and delivery strategies of super-resolution probes for live-cell imaging, and finally provide a brief conclusion and overview of the future.

Abstract:Organic long-lived luminescent probes featuring thermally activated delayed fluorescence (TADF) or room temperature phosphorescence (RTP) have emerged as highly promising tools for time-resolved luminescence imaging (TRLI) and sensing (TRLS). Their unique advantage lies in their ability to effectively eliminate interference from short-lived background signals. By introducing an appropriate delay time between the short-lived autofluorescence and measure of the long-lived luminescence emitted by the probes, a background-free approach emerges, enabling the accurate detection and imaging of diverse biologically relevant species. This review provides a comprehensive overview of metal-free organic compound-based responsive probes utilizing TADF and RTP for TRLI and TRLS. It offers detailed insights into the design strategies and sensing principles employed by these probes. Furthermore, the review discusses the current challenges faced by researchers in this rapidly expanding field and highlights the future prospects for advancements in this area of study.

Abstract:Since the advent of fluorescent probes, they have been widely used in the field of life sciences. Fluorescent probes can not only light up the cells, tissues and organs, but also be used to explore the bio‐distribution of specific types of disease markers. Therefore, the development of new fluorescent probes capable of recognition of bio‐markers are extremely important for the diagnosis of the related diseases. Herein, we focus on our recent works experienced the evolution from the beginning of simply fluorescent probes to the construction of theranostics platform. We have developed a series of new fluorescent probes for the detecting and imaging of reactive oxygen species (ROS) and specific enzymes in vitro and in living cells. With the shift of the fluorescence signal from visible to near‐infrared region, we expanded the application of the fluorescent probes from in vitro to in vivo. Abstract:Long-lived emissive probes have evolved as promising visualized tools for monitoring complex biological processes because of their distinctive features of large Stokes shifts and long luminescence lifetimes, which can effectively circumvent the interference of autofluorescence from biological tissue and organs. Particularly, purely organic room-temperature phosphorescence (RTP), especially in aqueous media, is of great interest due to its ultralong lifetimes, high quantum yields, low production cost, and satisfactory biocompatibility, making them serve as excellent candidates for in vivo deep-tissue time-resolved bioimaging. This review not only systematically summarizes recent advances of organic RTP probes, principally focusing on their in vivo bioimaging application of subcutaneous tissue imaging, wound healing monitoring, sentinel lymph node detection, thermometry, primary tumor visualization, assessment of tumor hypoxia and metastatic imaging. Various chemical structures, such as carbazole, phenothiazine, phenylpyrrole, polycyclic aromatic hydrocarbon, triphenylamine, difluoroboron β-diketonate, naphthalimide, and others, have been explored for these probes. The review also highlights the current strategies employed to enhance the properties of organic RTP molecules. These strategies include the polymer-assisted top-down method, bottom-up nanoprecipitation method, supramolecular nanoassemblies, and silica-skeleton nanocomposites. These approaches offer benefits such as ultralong lifetime, excellent water dispersibility, and biocompatibility under physiological conditions, thereby enhancing the performance of organic RTP probes for various applications. Furthermore, the review outlines the existing challenges and presents future prospects in this field. The aim is to inspire the development of more advanced and intelligent organic RTP probes with potential biomedical and clinical applications. By addressing these challenges and exploring new directions, researchers aim to contribute to the evolution of this field and the realization of its full potential in diagnostic and therapeutic settings.

Abstract:Many luminescent probes have been developed for intracellular imaging and sensing. During cellular luminescence sensing, it is difficult to distinguish species generated inside cells from those internalized from extracellular environments since they are chemically the same and lead to the same luminescence response of the probes. Considering that endogenous species usually give more information about the physiological and pathological parameters of the cells while internalized species often reflect the extracellular environmental conditions, we herein reported a series of cyclometalated iridium(iii) complexes as phosphorescent probes that are partially retained in the cell membrane during their cellular uptake. The utilization of the probes for sensing and distinguishing between exogenous and endogenous analytes has been demonstrated using hypoxia and hypochlorite as two examples of target analytes. The endogenous analytes lead to the luminescence response of the intracellular probes while the exogenous analytes are reported by the probes retained in the cell membrane during their internalization.

Abstract:In the second near-infrared spectral window (NIR-II; with wavelengths of 1,000–1,700 nm), in vivo fluorescence imaging can take advantage of reduced tissue autofluorescence and lower light absorption and scattering by tissue. Here, we report the development and in vivo application of a NIR-II phosphorescent probe that has lifetimes of hundreds of microseconds and a Stokes shift of 430 nm. The probe is made of glutathione-capped copper–indium–selenium nanotubes, and in acidic environments (pH 5.5–6.5) switches from displaying fluorescence to phosphorescence. In xenograft models of osteosarcoma and breast cancer, intravenous or intratumoral injections of the probe enabled phosphorescence imaging at signal-to-background ratios, spatial resolutions and sensitivities higher than NIR-II fluorescence imaging with polymer-stabilized copper–indium–sulfide nanorods. Phosphorescence imaging may offer superior imaging performance for a range of biomedical uses.

We review the properties of various nanomaterials that can be used for intracellular nanosensors in terms of the sensor design and the approaches to delivery of nanosensors based on engineering their surfaces. Abstract:Small-molecule based activatable fluorescence probes for detecting specific enzyme activity with high sensitivity can visualize the expression site of marker genes and cancers where the enzyme is highly expressed. However, the enzyme-catalyzed fluorescent hydrolysis product easily leaks out and diffuses from the reaction site, making it difficult to perform long-term tracking and immunohistochemical analysis which needs washing/fixation procedure. Our group have focused on quinone methide chemistry and developed series of activatable fluorescence probes with excellent intracellular retention that are converted to quinone-methide or aza-quinone-methide intermediates upon reaction with enzymes, which are then react with intracellular nucleophiles such as proteins and glutathione to be retained in cells and to exhibit significant increase in fluorescence. Based on this molecular design, we have developed fluorescence probes targeting β-galactosidase and γ-glutamyltranspeptidase with different colors. We also developed photo-functional probes such as activatable photosensitizers and caged fluorophores. These probes can visualize or kill target enzyme-expressing cells with high selectivity by suppressing the leakage of hydrolysis products from target cells, and fluorescence imaging in combination with immunostaining was possible due to the high tolerance of the obtained fluorescence signal even after washing and fixation.
